This study investigated the effects of dandelion on the ruminal metabolome and microbiome in lactating dairy cows. A total of 12 mid-lactation dairy cows were selected and randomly classified into two groups, supplementing dandelion with 0 (CON) and 200 g/d per cow (DAN) above basal diet, respectively. Rumen fluid samples were collected in the last week of the trial for microbiome and metabolome analysis. The results showed that supplementation of DAN increased the concentrations of ammonia nitrogen, acetate, and butyrate significantly. The rumen bacterial community was significantly changed in the DAN group, with Bacterioidetes, Firmicutes, and Proteobacteria being the main ruminal bacterial phyla. The abundance of Ruminococcaceae_NK4A214_group, UCG_005, and Christensenellaceae_R_7_group were relatively higher, whereas that of Erysipelotrichaceae_UCG_002 and Dialister were lower in the DAN than those in the CON. Metabolomics analysis showed that the content of d-glucose, serotonin, ribulose-5-phosphate, and d-glycerate were higher in the DAN group. These metabolites were enriched in the starch and sucrose metabolism, pentose phosphate pathway, tryptophan metabolism, and glycerolipid metabolism. The ribulose-5-phosphate and d-glycerate were correlated with Ruminococcaceae_NK4A214_group, UCG_005, and Christensenellaceae_R-7_group positively. This study demonstrated that the supplementation of dandelion impacts the ruminal microorganisms and metabolites in a way that rumen fermentation was enhanced in lactating dairy cows.

Friesian cows (16) in late lactation grazed pure white clover (Trifolium repens cv. Haifa) swards for 32 days in autumn and were supplemented with maize (Zea mays) silage. Four groups of 4 cows were offered either 19 or 39 kg dry matter (DM)/cow.day of white clover with either 0 or 4.4 kg DM/cow.day of maize silage. All cows were individually fed maize silage and grazed the pasture as individual groups according to treatment. When maize silage was fed, total intake increased, this occurring to a greater degree at the low pasture allowance. The level of substitution of silage for pasture was 0.14 and 0.40 kg DM reduction in pasture intake per kg DM of maize silage eaten at the low and high allowances, respectively. At the low pasture allowance, milk yields were 10.1 and 13.7 kg/cow. day when 0 and 4.4 kg DM/cow.day of maize silage were fed, respectively, and were 15.5 and 15.9 kg/cow.day at the high pasture allowance. Liveweight and body condition increased as plane of nutrition increased but there were no effects of feeding on milk fat or protein contents. Feeding maize silage had little effect on any rumen or faecal variable although there was generally less ammonia nitrogen in rumen fluid when cows were supplemented with maize silage.

Digestion and Balance trials were conducted with 18 lactating dairy cows in their 7-8 weeks of lactation using 7 day total collection to determine nitrogen and energy balance and digestibility of ration nutrients by lactating dairy cows fed three levels (0.8 and 16%) of corn condensed distillers solubles (CCDS). Cows were fed treatment rations for 4 weeks before collection. Following digestion and balance trials, rumen fluid samples were collected esophagially before feeding and 3 hours after feeding to determine the influence of CCDS on rumen pH, ammonia nitrogen and volatile fatty acids. Corn condensed distilers solubles significantly (P<0.05) improved the digestibility of the neutral detergent fiber and ether extract but depressed the digestibility of the ash. Apparent nitrogen retention was significantly (P <0.05) lower for animals on 8% CCDS ration. There were no significant differences in energy utilization among groups. Corn condensed distilers soluble did not significantly affect rumen pH, but depressed rumen ammonia nitrogen. On molar percentage basis, animals on the 8% CCDS ration had significantly (P< 0.05) higher acetate in the rumen 3 hours after feeding. Other VFA remained relatively unaffected by treatments.

A number of experiments were undertaken to establish the productivity of lactating dairy cows when offered green, vegetative Persian clover (Trifolium resupinatum) herbage and to study its utilization by these cows. The research included indoor feeding and grazing experiments and intensive metabolism studies. Levels of daily intake varied from 1.3 to 4.5 kg DM/100 kg liveweight and in early-to-mid lactation, milk yields as high as 30 kg/day were sustained. In early-to-mid lactation, marginal returns to additional feeding were 1.24 kg milk, 0.046 kg fat and 0.034 kg protein for each additional kg DM of Persian clover eaten by cows weighing 500 kg. In late lactation, marginal returns were 1.10, 0.050 and 0.035 kg/kg DM for milk, fat and protein, respectively. In a grazing experiment, utilization of herbage on offer was very high, ranging from 58-88% as grazing intensity increased. Consideration of the products of digestion suggested that fermentation of Persian clover herbage in the rumen resulted in a balance of metabolites that is unlikely to be detrimental to animal production. However, there were two aspects of rumen fermentation that could influence the potential utilization of the herbage. First, for much of the year, levels of ammonia in rumen fluid (up to 500 mg/L) were much higher than microbial requirements. Second, rumen fluid pH often dropped to about 5.5, a level which may inhibit cellulolysis. The implications of manipulating these aspects of rumen fermentation are discussed. It was concluded that, while few problems exist with the use of vegetative Persian clover for lactating dairy cows, this herbage may be used more efficiently if fed in conjunction with supplements that provide a better balance of nutrients for fermentation in the reticulorumen.

This study evaluated the effects of intraruminal administration ofMegasphaera elsdeniion ruminal fermentation patterns, the profile of plasma metabolites, and milk yield and composition of mid-lactation dairy cows. Eight primiparous, ruminally cannulated Holstein cows were arranged in a paired 2×2 crossover design. Cows were randomly assigned to one of two treatments: 1) intraruminal inoculation of 35 ml suspension per day ofM. elsdeniiATCC 25940 (MEGA), containing 108cfu/ml of bacteria, dissolved in 35 ml of saline (0·15m), or 2) carrier alone (35 ml saline; CTR). Both postprandial and preprandial rumen volatile fatty acids (VFA) and plasma metabolite measurements were analysed. Postprandial VFA patterns were affected the most, with butyrate (P<0·01) and valerate (P<0·01) proportions increasing, and acetate (P<0·01), isobutyrate (P=0·05) and isovalerate (P<0·01) decreasing in MEGA cows. Preprandial data measured at various days showed that MEGA dosage tended to increase the molar proportion of propionate (P=0·09) and lower the acetate to propionate ratio (P=0·07) in the rumen fluid. There was no effect of treatment on rumen pH and on the concentration of lactate in the rumen as well as on selected preprandial plasma metabolites. Postprandial plasma concentrations of cholesterol tended to increase (P=0·07) in MEGA cows compared with CTR. Concentrations of non-esterified fatty acids (NEFA) in the plasma were lower in MEGA cows after the morning feeding (P<0·01). Sampling hour also affected plasma NEFA in this study. Plasma β-hydroxybutyrate (BHBA) were not affected by the treatment (P>0·05); however, after the morning feeding BHBA concentration was increased in both groups of cows. Dry matter intake and milk yield and composition were not affected by treatment. In conclusion, results indicate thatM. elsdeniihas the potential to modulate the rumen fermentation profile in mid-lactation Holstein cows, but these effects were only slightly reflected in changes in plasma metabolites and milk composition.

The objective of the present study was to investigate the effects of virginiamycin (VM ) supplementation on ruminal fermentation and microbial populations in steers. Four ruminally cannulated Chinese Luxi steers (BW 559.4 &plusmn; 30.1 kg) were used in a crossover design experiment with an experimental period of 28 days. The forage to concentrate ratio of the basal diet was 35:65 on dry matter basis. The experiment consisted of control treatment and treatment with control diet plus VM at a dose of 30 mg/kg concentrate (DM basis). Rumen fluid was collected at 07:30 prefeeding, at 11:30 and 17:30 postfeeding on day 27 and 28. A part of the pooled sample from rumen fluid was transferred to anaerobic culture by a roll-tube technique and analysed for species-specific real-time PCR quantification. The remaining pooled rumen fluid sample was analyzed for pH, VFA, ammonia N and l-lactic acid. The results showed that VM increased the ruminal pH (6.70 vs. 6.63; P &lt; 0.05), but it decreased ammonia nitrogen (4.94 vs. 6.19 mg/100 ml; P &lt; 0.01) and mean counts of amylolytic bacteria and proteolytic bacteria (P &lt; 0.01) as compared to the control. The additive VM did not affect the l-lactic acid concentration (1.39 vs. 1.26 mmol/l) in rumen fluid. Compared to the control, the steers receiving VM have altered a trend of quantification of Selenomonas ruminantium, Anaerovibrio lipolytica, Ruminococcus albus and Streptococcus bovis in rumen fluid (0.05&lt;p&lt;0.1) as compared to the control. However, VM had no significant effect on Lactobacillus spp. (P = 0.41), Butyrivibrio fibrisolvens (P = 0.35), on the genus Ruminococcus (P = 0.25), Ruminococcus flavefaciens (P = 0.52), Prevotella ruminicola (P = 0.54), on the genus Prevotella (P = 0.67) and Megasphaera elsdenii (P = 0.97). In this study, we found that VM had selective effects on ruminal bacteria and influenced ruminal fermentation by changing a part of the specific ruminal bacteria populations.

The effects of glycerol with fat sources as a feed alternative were investigated in the ration for Nellore steers (Bos indicus). Eight cannulated steers at 30 months of age with initial bodyweight of 554 ± 36.0 kg were used in a double 4 by 4 Latin square design with four consecutive 16-day periods. Treatments were three different diets with lipid sources (soybean, soybean oil and calcium salts of fatty acids) and one diet control without lipid sources. All diets formulated contained 10% crude glycerol and 5% ether extract with a forage:concentrate ratio of 35:65; corn silage was used as forage. The ruminal liquid was sampled for 24 h and ruminal fermentations were monitored by measuring pH, concentrations of ammonia nitrogenand volatile fatty acids in rumen fluid. Urine samples were obtained from the total collection for 24 h for estimation of rumen microbial protein supply using urinary purine derivatives. Our results showed that the use of lipid sources combined with glycerol did not induce significant changes in rumen pH, acetate molar proportion, ruminal microbial protein or dry matter intake. Although the acetate molar proportion was kept constant within normal parameters, the propionate molar proportion was increased by the diet containing lipid sources. Moreover, we found that there was a negative effect of lipid sources on crude protein and neutral detergent fibre corrected for ash and protein intake, and one positive effect on ether extract intake. We believe that association between fat and glycerol may affect rumen fermentation parameters through reducing fibre intake and increasing propionate production and ammonia nitrogen.

Context Several agro-industrial by-products such as oil palm meal could be useful as animal feeds in support of low cost of feed for livestock production. Aims This study investigated the effects of oil palm meal and urea pellet (PMUP) as a protein replacement for soybean meal (SBM) on feed intake, nutrient digestibility, rumen fluid characteristics, milk yield and milk composition in lactating dairy cows. Methods Five multiparous, early to mid-lactation, Holstein-Friesian crossbred dairy cows (75% Holstein-Friesian × 25% Thai Native Bos indicus) were randomly allocated in a 5 × 5 Latin square design to receive PMUP replacement for SBM at 0, 25, 50, 75 and 100% in concentrate. Key results The results show that roughage, total dry matter (DM), organic matter (OM) and acid detergent fibre (ADF) intakes in the cows fed with 25–75% PMUP were significantly higher than in the cows fed 100% PMUP (P &lt; 0.05), whereas the ether extract (EE) intake of the cows receiving the PMUP was higher than that of the control (P &lt; 0.05). Accordingly, the apparent digestibility, ruminal ammonia nitrogen (NH3-N) and total volatile fatty acid (VFA) of cows fed with PMUP replacement of SBM at 25–75% was higher than that in cows fed with 100% PMUP. In addition, milk yield and milk composition were not significantly different among treatments, whereas feed costs per kg milk yield was lowest in cow fed with 100% PMUP (P &lt; 0.05). Conclusions Replacing SBM with PMUP up to 50–100% in concentrate could reduce costs of feed per kg milk yield, but lower feed intake, nutrient utilisation and rumen fermentation were observed in cow fed with 100% PMUP. The use of PMUP from 25–75% replacement of SBM had no significant effects on the feed intake, nutrient utilisation, rumen fermentation and milk production. Therefore, the level of PMUP replacement of SBM in concentrate for lactating dairy cow should not exceed 75%. Implications PMUP could be used as a protein replacement for SBM in concentrate for lactating dairy cows. These findings should be applied further in practical farm condition in order to increase livestock production efficiency.
