The presence of pharmaceutical products has raised emerging biorisks in aquatic environments. Fungi have been considered in sustainable approaches for the degradation of pharmaceutical compounds from aquatic environments. Soft rot fungi of the Ascomycota phylum are the most widely distributed among fungi, but their ability to biodegrade pharmaceuticals has not been studied as much as that of white rot fungi of the Basidiomycota phylum. Herein, we evaluated the capacity of the soft rot fungus Neopestalotiopsis sp. B2B to degrade pharmaceuticals under treatment of woody and nonwoody lignocellulosic biomasses. Nonwoody rice straw induced laccase activity fivefold compared with that in YSM medium containing polysaccharide. But B2B preferentially degraded polysaccharide over lignin regions in woody sources, leading to high concentrations of sugar. Hence, intermediate products from saccharification may inhibit laccase activity and thereby halt the biodegradation of pharmaceutical compounds. These results provide fundamental insights into the unique characteristics of pharmaceutical degradation by soft rot fungus Neopestalotiopsis sp. in the presence of preferred substrates during delignification.

White rot fungi of Marasmius sp. is a fungus which produce laccase in high activity. Laccase is one of the ligninolityc enzymes that capable to degrade lignin. This ability can be used for the pretreatment of lignocellulosic materials in the bioethanol production. Laccase was produced in flask by batch process using Solid State Fermentation (SSF). The optimisation was conducted by statistically of full factorial design. The particle size, moisture content, and Cu concentration were investigated in this study. Rice straw was used as solid substrate and the glycerol was used as the carbon sources in modified Kirk medium. The results showed that particle size of rice straw did not affect significantly to the enzyme activity. The highest laccase activity of 4.45 IU/g dry weight was obtained at the moisture content of 61% and Cu concentration of 0.1 mM.Keywords: laccase, Marasmius sp., optimisation, rice straw, solid state fermentation ABSTRAKJamur pelapuk putih, Marasmius sp. merupakan jamur yang menghasilkan enzim lakase dengan aktivitas tinggi. Lakase merupakan enzim ligninolitik yang dapat mendegradasi lignin. Kemampuan ini dapat digunakan untuk proses pengolahan awal bahan lignoselulosa pada pembuatan bioetanol. Produksi lakase dilakukan dalam labu dengan modus batch menggunakan fermentasi kultur padat. Optimisasi produksi enzim lakase dengan metode fermentasi padat dilakukan dengan  rancangan percobaan faktorial penuh. Pengaruh ukuran partikel, kelembapan, dan konsentrasi Cu diuji dengan medium penyangga jerami dengan menambahkan gliserol dalam medium Kirk termodifikasi sebagai sumber karbon. Penelitian ini menunjukkan bahwa ukuran jerami tidak berpengaruh signifikan terhadap aktivitas enzim. Aktivitas enzim lakase maksimum terjadi pada saat kelembapan 61% dan konsentrasi Cu 0,1 mM dengan aktivitas enzim lakase/berat kering tertinggi mencapai 4,45 IU/g.Kata kunci: lakase, Marasmius sp., optimisasi, jerami, fermentasi kultur padat

To obtain the basic information needed to estimate the degree of decay from compressive strength measured using a Fractometer (CS), relationships between CS and the contents of chemical components were analyzed for Magnolia wood decayed by three types fungi (brown rot, white rot, and soft rot fungi) at various decay levels. Weight loss ratio was significantly, negatively correlated with CS in woods decayed by brown rot and white rot fungi. In addition, a relatively high correlation coefficient was recognized between CS and holocellulose or α-cellulose content, except for wood decayed by soft rot fungus. The results obtained showed that Fractometer can detect the decrease of CS at relatively early stage of decay.

Wood-rotting fungi represent an important component of forest ecosystems. Among them, white-rot fungi are the most efficient lignin degraders. Biopulping using white-rot fungi in pretreatment of the materials, is one of the solutions to overcome disadvantages of traditional production methods. Today, the isolation and screening of lignin degrading fungi capable for application in biopulping are of keen interest in Vietnam. The use of non–wood, plant fibres in pulp and paper industry, special, agricultural residuces such as rice and wheat straw, sugarcane baggase, cornstalks etc is the new production toward, potential, serving sustainable development. The fungus CP9, which possessed high ligninolytic activity, was identified and studied in pretreatment of rice straw for biopulping. The fruiting bodies of strain CP9 were effuse on trunk. The hymenium was porous and brown white with short tubes, the white mycelia penetrated wood block. The colony was off-white, blossom, irregularly circular. The mycelia were thick and closely bound together. Beside lignin, this fungus could degrade other substrates such as casein, carboxymethyl cellulose and starch. Biological and morphological characteristics of the fungus CP9 suggested its placement in subdivision Basidiomycota. Combined with the results of phylogenetic analysis, which showed 99% similarity of the fungus with species Leiotrametes lactinea, our strain was named as Leiotrametes lactinea CP9. This fungus could grow well on rice straw under solid state fermentation. Pretreatment of rice straw using L. lactinea CP9 was based on the activity of fungal lignin peroxidase and laccase. After 20 days, the residual enzyme activity was of 21.6 and 18.4 nkat/g material for lignin peroxidase and laccase, respectively. Pretreatment significantly improved the quality of straw, as lignin loss of 38% while cellulosic fibers were comparatively well preserved.

Abstract In the last decades, many wood preservatives have been prohibited for their ecotoxicity. The present article is focusing on the conifer-derived condensed tannins as environment-friendly options for the substitution of artificial wood preservatives. Eight different tannin fractions were extracted from spruce cones, spruce barks, and pine cones. The parameters of tannin extraction, such as the methods of purification and concentration of active components in the extracts, have been investigated. The cone and bark extracts were tested for the growth inhibition of eight brown-rot fungi, three white-rot fungi, and four soft-rot fungi in liquid cultures. The cone tannins provided a more efficient fungal growth inhibition than bark tannins. Purification increased the antifungal properties of the extracts. The growth of brown-rot fungi was inhibited by the tannins already at low concentrations. However, the extracts were not effective against the white-rot or soft-rot fungi. More investigation is needed concerning the tannin source and the purification procedure of the extracts before tannins can be considered as an ecologically benign wood preservative.

SummaryOmphalina sp. a white-rot fungi (WRF)originated from oil palm plantation has abilityto degrade empty fruit bunches of oil palm(EFBOP) so that it is expected to producelaccase with high activity. The ability ofOmphalina sp. to produce laccase enzyme onliquid fermentation will be studied. The enzymewill also be partially purified andcharacterized. The research result showed thatthe highest enzyme activity (1.162 U/mL) wasobtained using glucose malt yeast (GMY)medium at room temperature for four days.The addition of 2,5-xylidine as an inducerproduced laccase earlier i.e two days, but theactivity of laccase was less active afterprolonged incubation compared to that ofcontrol. The laccase produced on mediumcontaining 2% EFBOP reached optimumactivity as much as 0.38 U/mL after 10 th daysof incubation. Partial purification of laccaseon Sephacryl S-200 HR column resulted58.23% of yield recovery with twice purity thanbefore. The optimum pH of laccase was 4.5.Laccase activity was stable even after heatedon 50 o C for 30 minutes, but then decreasedwhen heated until 60 o C. The efforts to break down the lignocellulosic complex found in the cell wall of straws, besides digestible cellulose and hemicellulose by rumen fermentation, improvement of straw digestibility by the degradation of indigestible lignin fraction of complex by using of biotechnological methods is one of the focus areas of animal nutritionists in recent years. Biological method sare prefer redover other methods due to the environmental friendliness. In the biological treatment methods of lignocellulosic complex, biodiversity of bacteria, enzymes and fungi gives opportunity to select lignin degrading species. Mycobacterium, Arthrobacter and Flavobacterium genre bacteria are used to degrade lignin by bacterial treatment. Lignocellulolytic enzymes isolated from different varieties of fungi are used in enzyme treatment. There are 3 genres of fungus that are white, Brown and soft rot in fungal treatments. Brown rot fungi prefer ably attack cellulose and hemicelluloses, but not lignin. White rot fungi attack the lignin and break up lignol bonds and aromatic ring. White rot fungi break down polysaccharides with hydrolytic enzymes such as cellulase, xylanase, and lignin with oxidative ligninolytic enzymes such as lignin peroxidase and laccase. Because of the fact that the microorganisms that can break down the lignocellulosic materials are the fungi and the cost is low, the application of white rot fungi is possible. In this paper, improvement the lignocellulosic comlex digestibility of straw by biological treatment with the advantage of biodiversity is discussed.
