Superalloys are used primarily for the aerospace, automotive, and petrochemical industries. These applications require materials with high creep resistance. In this work, evaluation of creep resistance and microstructural characterization were carried out at two new nickel intermediate content alloys for application in aerospace industry and in high performance valves for automotive applications (alloys VAT 32 and VAT 36). The alloys are based on a high nickel chromium austenitic matrix with dispersion of intermetallic L12 and phases containing different (Nb,Ti)C carbides. Creep tests were performed at constant load, in the temperature range of 675–750 °C and stress range of 500–600 MPa. Microstructural characterization and failure analysis of fractured surfaces of crept samples were carried out with optical and scanning electron microscopy with EDS. Phases were identified by Rietveld refinement. The results showed that the superalloy VAT 32 has higher creep resistance than the VAT 36. The superior creep resistance of the alloy VAT 32 is related to its higher fraction of carbides (Nb,Ti)C and intermetallic L12 provided by the amount of carbon, titanium, and niobium in its chemical composition and subsequent heat treatment. During creep deformation these precipitates produce anchoring effect of grain boundaries, hindering relative slide between grains and therefore inhibiting crack formation. These volume defects act also as obstacles to dislocation slip and climb, decreasing the creep rate. Failure analysis of surface fractures of crept samples showed intergranular failure mechanism at crack origin for both alloys VAT 36 and VAT 32. Intergranular fracture involves nucleation, growth, and subsequent binding of voids. The final fractured portion showed transgranular ductile failure, with dimples of different shapes, generated by the formation and coalescence of microcavities with dissimilar shape and sizes. The occurrence of a given creep mechanism depends on the test conditions. At creep tests of VAT 32 and VAT 36, for lower stresses and higher temperature, possible dislocation climb over carbides and precipitates would prevail. For higher stresses and intermediate temperatures shear mechanisms involving stacking faults presumably occur over a wide range of experimental conditions.

This paper examines the effect of equal-channel angular pressing (ECAP) on creep behaviour of pure aluminium, binary Al-0.2wt.%Sc alloy and ternary Al-3wt.%Mg-0.2wt.%Sc alloy. The ECAP was conducted at room temperature with a die that had a 90° angle between the channels and 8 repetitive ECAP passes followed route BC. Constant stress compression creep tests were performed at 473 K and stresses ranging between 16 to 80 MPa on ECAP materials and, for comparison purposes, on the initial coarse-grained materials. The results showed that the creep resistance of the ECAP processed Al-Sc and Al-Mg-Sc alloys was markedly deteriorated with respect to unpressed coarse-grained materials.

Positron emission tomography (PET) has a wide range of applications in the treatment and prevention of major diseases owing to its high sensitivity and excellent resolution. However, there is still much room for optimization in the readout circuit and fast pulse sampling to further improve the performance of the PET scanner. In this work, a LIGHTENING® PET detector using a 13 × 13 lutetium-yttrium oxyorthosilicate (LYSO) crystal array read out by a 6 × 6 silicon photomultiplier (SiPM) array was developed. A novel sampling method, referred to as the dual time interval (DTI) method, is therefore proposed to realize digital acquisition of fast scintillation pulse. A semi-cut light guide was designed, which greatly improves the resolution of the edge region of the crystal array. The obtained flood histogram shown that all the 13 × 13 crystal pixels can be clearly discriminated. The optimum operating conditions for the detector were obtained by comparing the flood histogram quality under different experimental conditions. An average energy resolution (FWHM) of 14.3% and coincidence timing resolution (FWHM) of 972 ps were measured. The experimental results demonstrated that the LIGHTENING® PET detector achieves extremely high resolution which is suitable for the development of a high performance time-of-flight PET scanner.

We present an interactive Mathematica notebook that characterizes the electrical impulses along actin filaments in both muscle and non-muscle cells for a wide range of physiological and pathological conditions. The program is based on a multi-scale (atomic → monomer → filament) approach capable of accounting for the atomistic details of a protein molecular structure, its biological environment, and their impact on the travel distance, velocity, and attenuation of monovalent ionic wave packets propagating along microfilaments. The interactive component allows investigators to conduct original research by choosing the experimental conditions (intracellular Vs in vitro), nucleotide state (ATP Vs ADP), actin isoform (alpha, gamma, beta, and muscle or non-muscle cell), as well as, a conformation model that covers a variety of mutants and wild-type (the control) actin filament. The simplicity of the theoretical formulation and the high performance of the Mathematica software enable the analysis of multiple conditions without computational restrictions. These studies may provide an unprecedented molecular understanding of why and how age, inheritance, and disease conditions induce dysfunctions in the biophysical mechanisms underlying the propagation of electrical signals along actin filaments.

Failure analysis of laminated composite structures has attracted a great deal of interest in recent years due to the increased application of composite materials in a wide range of high-performance structures. Intensive experimental and theoretical studies of failure analysis and prediction are being reviewed. Delamination, the separation of two adjacent plies in composite laminates, represents one of the most critical failure modes in composite laminates. In fact, it is an essential issue in the evaluation of composite laminates for durability and damage tolerance. Thus, broken fibers, delaminated regions, cracks in the matrix material, as well as holes, foreign inclusions and small voids constitute material and structural imperfections that can exist in composite structures. Imperfections have always existed and their effect on the structural response of a system has been very significant in many cases. These imperfections can be classified into two broad categories: initial geometrical imperfections and material or constructional imperfections

Fibre-reinforced thermoplastic composite materials can be manufactured rapidly using a thermoforming process. The assortment of thermoplastic matrix systems is manifold and starts from bulk plastic like polypropylene (PP) up to high-performance systems like polyether ether ketone. High-performance thermoplastic polymers have durable properties but relatively high raw material costs. For structural application, engineering methods are needed to ensure the availability for use over the full range of the life cycle of parts. This equates to at least 15 years under exposure to varying climatic conditions for an automobile component. Bulk plastics have complex viscoelastic behaviour, which means that advanced methods are needed to ensure the long-term behaviour of both the pure plastic or fibre-reinforced materials with such a matrix system. In the following study, the creep behaviour of a glass fibre-reinforced PP material is investigated using different uniaxially loaded creep tests at different load and temperature levels. Starting from this empirical base, two characteristic creep functions are derived using a modified Burgers approach. To transfer the results of uniaxial creep situations to a three-dimensional multiaxial stress state, a method to interpolate the experimental creep curves is presented. This developed creep model is integrated into the implicit non-linear finite element program SAMCEF/Mecano and used to predict the creep behaviour of a complex laminate. The results are then validated against the performed experiments.

After developing a model neuron or network, it is important to systematically explore its behavior across a wide range of parameter values or experimental conditions, or both. However, compiling a very large set of simulation runs is challenging because it typically requires both access to and expertise with high-performance computing facilities. To lower the barrier for large-scale model analysis, we have developed NeuronPM, a client/server application that creates a “screen-saver” cluster for running simulations in NEURON (Hines & Carnevale, 1997). NeuronPM provides a user-friendly way to use existing computing resources to catalog the performance of a neural simulation across a wide range of parameter values and experimental conditions. The NeuronPM client is a Windows-based screen saver, and the NeuronPM server can be hosted on any Apache/PHP/MySQL server. During idle time, the client retrieves model files and work assignments from the server, invokes NEURON to run the simulation, and returns results to the server. Administrative panels make it simple to upload model files, define the parameters and conditions to vary, and then monitor client status and work progress. NeuronPM is open-source freeware and is available for download at http://neuronpm.homeip.net . It is a useful entry-level tool for systematically analyzing complex neuron and network simulations.

Pre-alloyed powders with a nominal composition of Ti-48Al-2Cr-2Nb+1W were consolidated by hot isostatic pressing (HIP). After the HIP process, a step cooled heat treatment (SCHT) with a carefully controlled cooling rate was applied to homogenize the HIP’ed microstructure and produce a fully lamellar microstructure. Following the SCHT, various isothermal aging at 950 °C and step aging processes form interfacial precipitates at the lamellar interfaces. The morphology, size, and distribution of the precipitates are dependent on the aging condition. Creep tests were carried out in air at 760 °C and 276 MPa to investigate the effect of interfacial precipitates. Primary creep resistance and creep life of the 8 and 144 hr aged conditions are improved substantially compared to the unaged condition due to the existence of the interfacial precipitates. However, the step aging process improves the creep resistance only slightly, probably because of the size and distribution differences of the interfacial precipitates compared to the 144 hr aged condition. Microstructure control is important since it has a substantial influence on creep behavior, especially primary creep resistance.

Experiments were conducted on an Al-0.2wt.%Sc alloy to evaluate the effect of equalchannel angular pressing (ECAP) on its creep behaviour. ECAP was conducted at room temperature with a die that had an internal angle of 90° between the two parts of the channel. The subsequent extrusion passes were performed by route BC up to 8 ECAP passes. Creep tests in tension were performed on the as-pressed samples at 473 K under an applied stress range between 10 to 50 MPa. For comparison purposes, some creep tests were performed also on the unpressed alloy. Following ECAP and creep testing, samples were prepared for examination by means of transmission electron microscopy (TEM) and scanning electron microscopy (SEM) equipped with an electron back scattering diffraction (EBSD) unit. The observation of the surface of the ECAPed samples after creep exposure showed the occurrence of mesoscopic shear bands. The EBSD data reveal that these bands are separated by high angle grain boundaries. The creep resistance of an alloy is a little decreased after one ECAP pass. However, successive ECAP pressing lead to a noticeable decrease of the creep properties. Thus, the Al-0.2wt.%Sc alloy processed by 8 ECAP passes exhibited faster creep rate by about two and/or three orders of magnitude than the unpressed alloy when creep testing at 473 K and at the same applied stress. The detrimental effect of ECAP on the creep resistance is probably a consequence of a synergetic effect of mesoscopic sliding of groups of grains along shear bands, more intensive grain boundary sliding and creep cavitation in creep of the ultrafine-grained material.

Hair is a unique biological matrix that adsorbs short-term exposures (e. g., environmental contaminants and personal care products) on its surface and also embeds endogenous metabolites and long-term exposures in its matrix. In this work, we developed an untargeted metabolomics workflow to profile both temporal exposure chemicals and endogenous metabolites in the same hair sample. This analytical workflow begins with the extraction of short-term exposures from hair surfaces through washing. Further development of mechanical homogenization extracts endogenous metabolites and long-term exposures from the cleaned hair. Both solutions of hair wash and hair extract were analyzed using ultra-high-performance liquid chromatography-high-resolution mass spectrometry (UHPLC-HRMS)-based metabolomics for global-scale metabolic profiling. After analysis, raw data were processed using bioinformatic programs recently developed specifically for exposome research. Using optimized experimental conditions, we detected a total of 10,005 and 9,584 metabolic features from hair wash and extraction samples, respectively. Among them, 274 and 276 features can be definitively confirmed by MS2 spectral matching against spectral library, and an additional 3,356 and 3,079 features were tentatively confirmed as biotransformation metabolites. To demonstrate the performance of our hair metabolomics, we collected hair samples from three female volunteers and tested their hair metabolic changes before and after a 2-day exposure exercise. Our results show that 645 features from wash and 89 features from extract were significantly changed from the 2-day exposure. Altogether, this work provides a novel analytical approach to study the hair metabolome and exposome at a global scale, which can be implemented in a wide range of biological applications for a deeper understanding of the impact of environmental and genetic factors on human health.

Drug biotransformation studies appear as an alternative to pharmacological studies of metabolites, development of new drug candidates with reduced investment as well as the most efficient production of chemical structures involves and drug quality control studies. A wide range of reactions in biotransformations process is catalyzed by microorganisms. Fungi can be considered as a promising source of new biotransformation reactions. The aim of this study was to evaluate the capacity of metronidazole biotransformation through the filamentous fungus Cunninghamella elegans ATCC 9245. The monitoring of metabolite formation was performed by high-performance liquid chromatography (HPLC) coupled to ultraviolet (UV) spectrophotometry. The results of the biotransformation of metronidazole showed drug consumption in culture and the formation of four new chromatographic peaks of chemical structures not elucidated. The method showed it became linear over 10-70 μg/mL (r = 0.999953). Accuracy, precision and stability studies agree with international guidelines.  Results are consistent in accordance with the principles of green chemistry as the experimental conditions had a low environmental impact, and few solvents use.
