Psoriasis is the most common inflammatory skin disease, characterized by the release of proinflammatory cytokines from lymphocytes, keratinocytes, and dendritic cells. Although psoriasis is considered an immune-mediated inflammatory disease, its effect on secretory activity of salivary glands and quantitative composition of saliva is still unknown. The aim of this study was to evaluate the secretion of saliva as well as several selected inflammation and nitrosative stress biomarkers in unstimulated and stimulated saliva as well as plasma of psoriasis patients. We demonstrated that, with progressing severity and duration of the disease, the secretory function of the parotid and submandibular salivary glands is lost, which is manifested as decreased unstimulated and stimulated saliva secretion and reduced salivary amylase activity and total protein concentration. The levels of tumor necrosis factor-alpha (TNF-α), interleukin-2 (IL-2), and interferon-gamma (INF-γ) were significantly higher, whereas interleukin-10 (IL-10) content was considerably lower in unstimulated and stimulated saliva of patients with psoriasis compared to the controls, and the changes increased with the disease duration. Similarly, we observed that the intensity of nitrosative stress in the salivary glands of psoriasis patients depended on the duration of the disease. By means of receiver operating characteristic (ROC) analysis, we showed that the evaluation of nitric oxide (NO), nitrotyrosine, and IL-2 concentration in non-stimulated saliva with high sensitivity and specificity differentiated psoriasis patients on the basis of the rate of saliva secretion (normal salivation vs. hyposalivation). In summary, the dysfunction of salivary glands in psoriasis patients is caused by inflammation and nitrosative stress.

Aim. To study the levels of pro- and anti-inflammatory cytokines in cervical secretion in females with cervical epithelial dysplasia and latent papillomavirus infection. Methods. The study included 120 females aged 20 to 40 years with cervical papillomavirus infection, who were assigned to two groups using «case-control» method. The first group included 60 females with latent disease, the second group - 60 females with mild and moderate cervical epithelial dysplasia (cervical intraepithelial neoplasia stages I and II). Cytokine levels in cervical secretion were measured by ELISA using «ProCon» test system. Results. ELISA test showed increased levels of interleukin-4 and interleukin-10 (43 [21; 74] and 48 [12; 88] pg/ml, respectively) and decreased levels of interleukin-2 (18.5 [5.5; 27.5] pg/ml), interleukin-6 (0.6 [0.06; 0.9] pg/ml), tumor necrosis factor alpha (88.5 [0; 123] pg/ml), interferon γ (2 [0; 4] pg/ml) in cervical secretion of females with cervical epithelial dysplasia compared to females with latent papillomavirus infection. Thus, females with cervical epithelial dysplasia showed increased levels of anti-inflammatory cytokines: interleukin-4 - by 2.7 times and interleukin-10 - by 2.4 times compared to females with latent disease, while levels of pro-inflammatory cytokines was decreased as following: interleukin-2 - by 1.4 times, interleukin-6 - by 4.5 times, tumor necrosis factor alpha - by 1.8 times, interferon γ - by 6.3 times (p 0.05). Conclusion. Imbalance of immune response cytokine regulation with anti-inflammatory cytokines prevailing might be an important factor facilitating persistence of papillomavirus in cervical epithelium and contributing to cervical epithelial dysplasia onset and progression.

Abstract Background: Graves’ orbitopathy (GO) is considered an autoimmune condition in close relationship with Graves’ disease (GD) affecting the thyroid. Several similarities exist between the two conditions, sharing the common antigen and the characteristics of the inflammation mediated by a number of cytokines. The result of the immune reactions will lead to the expansion of adipose tissue, production of glycosaminoglycans and soft tissue inflammation. Material and methods: In our study we examined the serum level of interleukin-1 (IL-1), interleukin 2 (IL-2), interleukin-4 (IL-4), interleukin-6 (IL-6), interleukin-8 (IL-8), interleukin-10 (IL-10), monocyte chemoattractant protein-1 (MCP-1), tumor necrosis factor-alpha (TNF-α), and interferon-gamma (IFN-γ) in correlation with the activity of disease and smoking habits in 25 patients with GD and GO. Results: We found that smokers had higher serum IL-6 and lower serum MCP-1, IL-8 and TNF-α level compared to non-smokers. Also, we found a weak positive correlation between serum IL-10, IFN-γ and disease activity (clinical activity score, CAS) and negative correlation between serum IL-1 and activity. Conclusion: Our findings support the fact that some cytokines (IL-10, IFN-γ, IL-1) play a role in active disease, while others are influenced by environmental factors, such as smoking (IL-6, IL-8, TNF-α). The discrepancy of cytokine profiles may reflect different patient characteristics, such as disease stage and disease activity and determination of serum cytokines would be useful in selecting patients who need more aggressive treatment protocols.

Abstract The aim of the study was to research the cytokine profile of cow blood in the treatment of acute postpartum endometritis with the use of recombinant α-, γ-interferons. Animals of the first group (n = 9) with a diagnosis of acute postpartum endometritis were intramuscularly injected with propranolol hydrochloride, denatured emulsified placenta, and nioxityl. Propranolol hydrochloride was administered for 4 days at a dose of 10 ml/animal at 24-hour intervals. The denatured emulsified placenta was injected subcutaneously on days 1-5-9 at a dose of 25 ml /animal. Nioxityl was injected intrauterine at a dose of 150 ml three times with a 48-hour interval. Cows of the second group (n = 11) with the same diagnosis were additionally injected intramuscularly with bovine recombinant α-, γ-interferons three times in 1–3 days at a dose of 5 ml/animal, 1 cm3 of which contains at least 1x104 IU/cm3 of the total antiviral activity of bovine recombinant α-, γ-interferons. Blood samples are taken from all groups before and at the end of the experiment. Blood samples are examined for the content of interleukin-2 (IL-2), interleukin-4 (IL-4), tumor necrosis factor alpha (TNFα), interleukin-10 (IL-10) using Bovine Elisa Kit Clood-Clone Corp (USA). The therapeutic effectiveness in the first group was 77.8%, in the second - 90.9%, which is 13.1% more. At the end of the course, the level of IL-2 decreased by 42.5% (43.5±4.2 pg/ml, P < 0.01), TNF-Α by 9.1% (457.9±34.6 pg/ml), the level of IL-4 increased by 14.8% (44.2±3.5 pg/ml, P < 0.05), IL-10 by 56.6% (35.7±2.8 pg/ml, P < 0.01). In the second group, the level of anti-inflammatory cytokines decreased: IL–2 by 48.7% (38.8±1.6 pg/ml, P < 0.01), TNFα by 26% (372.5±17.6 pg/ml, P < 0.05) and increased anti–inflammatory cytokines: IL-4 by 46.2% (56.3±4.1 pg/ml, P < 0.001) and IL-10 by 80.3% (41.1±3.5 pg/ml, P < 0.001), which indicates a decrease in the inflammatory response.

Introduction. “Thin” endometrium is known affect negatively to reproductive function. Treatment of “thin” endometrium remains a problem and subject of future major studies. Aim. To justify the possibility of applying of recombinant interleikina-2 person late reproductive age patients with “thin” endometrium before using assisted reproductive technologies. Matherials and Methods. We examined a total of 64 late reproductive age women. Three groups of patients were analyzed. The first investigated group included 25 women with histologically proven chronic endometritis; the comparison group included 20 patients with endometrium of the early proliferative phase; the second investigated group included 19 patients with “thin” endometrium, with indications for the conduct of in vitro fertilization. Interleukin 2, interleukin-4, tumor necrosis factor alpha, interleukin-1β, Interleukin-6, interleukin-8, interleukin-10, IFN-γ concentrations were determined by enzyme linked immunosorbent assay in serum and aspirates from the uterine cavity in first investigated group and comparison group. Vaginal ultrasonic examination was made for women of second investigated group in first and second phase of menstrual cycle before and after recombinant human interlejkinom-2 treatment. Statistical data processing was performed using application software package Statistica 6.0 (StatSoft, Ink. 1994–2001), adapted for biomedical research. P < 0.05 was considered significant. Results. Were found that concentrations of interleukin 2, interleukin-10 significantly reduced and concentrations of tumor necrosis factor alpha, interleukin-1β, Interleukin-6 and interleukin-8 significantly increased in aspirates from the uterine cavity in women of late reproductive age with chronic endometritis. All changes occur at the local level in chronic endometritis. Endometrial thickness a statistically significant increase after use of recombinant human interlejkinom-2 in patients with infertility and chronic endometritis. Conclusions. Application of recombinant interleukin-2 in patients with “thin” endometrium can become optimal schema therapy, which requires further study.

ABSTRACT In patients with chronic paracoccidioidomycosis (n= 10), levels of tumor necrosis factor alpha, interleukin-10, and interleukin-2 in serum, measured by enzyme-linked immunosorbent assay (in picograms per milliliter, as mean ± standard error of the mean), were higher than in normal controls (n = 8): 186 ± 40 versus 40 ± 7 (P < 0.05), 203 ± 95 versus 20 ± 8 (P = 0.001), and 96.3 ± 78.57 versus 1.19 ± 1.19 (P = 0.045), respectively. Gamma interferon and interleukin-4 levels were similar in patients and controls.

Diallyl disulfide (DADS) is the major organosulfur constituent in garlic, with a variety of pharmacological activities including antioxidant and anti-inflammatory. Here, we examined the potential antiedematous impact of DADS- versus carrageenan-mediated paw edema in mice. Carrageenan injection potentiated an inflammatory reaction as presented by the elevated serological C-reactive protein (CRP) levels and transcription of tumor necrosis factor-alpha (TNF-α, Tnfα), interleukin-1 beta (IL-1β, Il1b), interleukin-2 (IL-2, Il2), inducible nitric oxide synthase (iNOS), nitric oxide (NO), cyclooxygenase-2 (COX-2, Ptgs2), prostaglandin E2 (PGE2), monocyte chemoattractant protein-1 (MCP-1, Ccl1), nuclear factor kappa B (NF-κB), and myeloperoxidase (MPO) activity, while interleukin-10 (IL-10) was declined in the injured paw tissue. Additionally, carrageenan elevated lipid peroxidation in terms of malondialdehyde (MDA) and decreased glutathione content (GSH). Remarkably, DADS was found to inhibit the inflammatory signaling, suppressed the developed oxidative damage, and protected the histopathological alterations in the inflamed paw tissue in response to carrageenan injection. Our findings suggest that DADS could be used as an alternative therapy used to alleviate the pathophysiological changes associated with the genesis of paw edema through its potent anti-inflammatory and antioxidant impacts.

Aim: To compare the markers of inflammation and immune activation in virally suppressed HIV-infected children on antiretroviral therapy, who practiced regular structured exercise comprising running and yoga to those who did not over a 2-year period. Methods: This retrospective cohort study included 72 children aged 8 to 16 years divided into 2 groups, exercisers (n = 36) and the nonexercisers (n = 36) based on their intentional physical activity. The analyses were carried out at baseline and after 2 years (Y2) for the soluble biomarkers of inflammation and immune activation (tumor necrosis factor alpha, interleukin-6, interleukin-10, interferon gamma, sCD14, and sCD163). In addition, cell-associated biomarker (CD38), lipopolysaccharides, and the gene expression of interleukin-2 and brain-derived neurotrophic factor were also measured at Y2. Results: Reduction in levels of sCD14 (effect size [ES], −0.6; 95% confidence interval [CI], −1.08 to −0.14), tumor necrosis factor alpha (ES, −0.7; 95% CI, −1.18 to −0.23), interferon gamma (ES, −0.7; 95% CI, −1.17 to −0.22), and interleukin-10 (ES, −0.6; 95% CI, −1.08 to −0.14) was observed among exercisers as compared with nonexercisers at Y2. In addition, CD38+ expressing CD4+ T cells were found to be lower among exercisers (P = .01) at Y2. However, the differences in levels of interleukin-6, sCD163, lipopolysaccharides, interleukin-2, and brain-derived neurotrophic factor were not significantly different among the 2 groups. Conclusion: The study result suggests that regular structured physical activity improves the inflammatory profile of antiretroviral therapy-treated HIV-infected children.

Physical exercise reduces the biochemical markers of obesity, but the effects of multicomponent interventions on these markers should be explored. The present study aimed to elucidate how overweight/obese adolescents respond to a multicomponent program approach on body composition, physical fitness, and inflammatory markers, using a quasi-experimental study with 33 overweight/obesity adolescents (control group (CG) = 16; intervention group (IG) = 17). The intervention consisted of 24 weeks with physical exercises and nutritional and psychological guidance. Both groups were evaluated at the pre/post-intervention moments on body mass index (BMI); body fat (%Fat); waist circumference (WC); waist/hip ratio (WHR); waist-to-height ratio (WHtR), cardiorespiratory fitness (CRF); abdominal strength, flexibility; leptin; interleukin 6; interleukin 10; and tumor necrosis factor-alpha. Mixed-analysis of variance and generalized estimation equations were used for statistical analysis. There was an interaction effect between groups and time on %Fat (p = 0.002), WC (p = 0.023), WHR (p < 0.001), WHtR (p = 0.035), CRF (p = 0.050), and leptin (p = 0.026). Adolescents were classified as 82.4% responders for %Fat, 70.6% for WC, 88.2% for WHR, and 70.6% for CRF. Further, there was an association between changes in %Fat (p = 0.033), WC (p = 0.032), and WHR (p = 0.033) between responders and non-responders with CRF in the IG. There was a positive effect on body composition, physical fitness, and leptin. In addition, reductions in body composition parameters were explained by CRF improvements.
