Apolipoprotein E (ApoE), a 34-kDa glycoprotein, as part of the high-density lipoprotein (HDL), has antioxidant, anti-inflammatory and antiatherogenic properties. The variability of ApoE expression in the course of some female fertility disorders (endometriosis, POCS), and other gynecological pathologies such as breast cancer, choriocarcinoma, endometrial adenocarcinoma/hyperplasia and ovarian cancer confirm the multidirectional biological function of ApoE, but the mechanisms of its action are not fully understood. It is also worth taking a closer look at the associations between ApoE expression, the type of its genotype and male fertility disorders. Another important issue is the variability of ApoE glycosylation. It is documented that the profile and degree of ApoE glycosylation varies depending on where it occurs, the type of body fluid and the place of its synthesis in the human body. Alterations in ApoE glycosylation have been observed in the course of diseases such as preeclampsia or breast cancer, but little is known about the characteristics of ApoE glycans analyzed in human seminal and blood serum/plasma in the context of male reproductive health. A deeper analysis of ApoE glycosylation in the context of female and male fertility will both enable us to broaden our knowledge of the biochemical and cellular mechanisms in which glycans participate, having a direct or indirect relationship with the fertilization process, and also give us a chance of contributing to the enrichment of the diagnostic panel in infertile women and men, which is particularly important in procedures involved in assisted reproductive techniques. Moreover, understanding the mechanisms of glycoprotein glycosylation related to the course of various diseases and conditions, including infertility, and the interactions between glycans and their specific ligands may provide us with an opportunity to interfere with their course and thus develop new therapeutic strategies. This brief overview details some of the recent advances, mainly from the last decade, in understanding the associations between ApoE expression and some female and male fertility problems, as well as selected female gynecological diseases and male reproductive tract disorders. We were also interested in how ApoE glycosylation changes influence biological processes in the human body, with special attention to human fertility.

The importance of zinc for male fertility only emerged recently, being propelled in part by consumer interest in nutritional supplements containing ionic trace minerals. Here, we review the properties, biological roles and cellular mechanisms that are relevant to zinc function in the male reproductive system, survey available peer-reviewed data on nutritional zinc supplementation for fertility improvement in livestock animals and infertility therapy in men, and discuss the recently discovered signaling pathways involving zinc in sperm maturation and fertilization. Emphasis is on the zinc-interacting sperm proteome and its involvement in the regulation of sperm structure and function, from spermatogenesis and epididymal sperm maturation to sperm interactions with the female reproductive tract, capacitation, fertilization, and embryo development. Merits of dietary zinc supplementation and zinc inclusion into semen processing media are considered with livestock artificial insemination (AI) and human assisted reproductive therapy (ART) in mind. Collectively, the currently available data underline the importance of zinc ions for male fertility, which could be harnessed to improve human reproductive health and reproductive efficiency in agriculturally important livestock species. Further research will advance the field of sperm and fertilization biology, provide new research tools, and ultimately optimize semen processing procedures for human infertility therapy and livestock AI.

Background: A human body keeps changing physiologically, biologically, and psychologically from birth to death. There is always a change in all human faculties. But adolescence is a phase earmarked where a human body experiences drastic changes among all these faculties. And any such bodily change carrying baggage of shame, insecurity, stigma, and concealment demands on-time awareness and intervention. Aim: The aim of the study is to understand the knowledge and level of awareness regarding sexual and reproductive health among the school-going adolescents of Kashmir Valley where majority of the population belongs to Muslim conservative culture. Methodology: Due to COVID-19 lockdown, the inability of access to schools and children gave the researcher the opportunity to utilize the alternate places and a total of 550 students from classes 8, 9, and 10 were selected for the purpose. The permission was sought from the respective teachers and then the parents. The sample was a combination of boys and girls, students from both private and government-run schools. The researcher collected the data and it was tabulated systematically and analyzed using Microsoft Excel. P value was evaluated by using an application “P Value: A Statistical Tool” from Play Store. Results and Conclusion: The results depicted that higher the level of class, more the knowledge they had about the matter. Girls had a little knowledge about sexual and reproductive health than boys. This study felt a dire need of educating both parents and adolescent boys and girls regarding sexual health. The shame does not lie in educating the children but in the mishaps that may result due to unawareness about the same.

CRISPR/Cas9-mediated genome editing technology enables researchers to efficiently generate and analyze genetically modified animals. We have taken advantage of this game-changing technology to uncover essential factors for fertility. In this study, we generated knockouts (KOs) of multiple male reproductive organ-specific genes and performed phenotypic screening of these null mutant mice to attempt to identify proteins essential for male fertility. We focused on making large deletions (dels) within 2 gene clusters encoding cystatin (CST) and prostate and testis expressed (PATE) proteins and individual gene mutations in 2 other gene families encoding glycerophosphodiester phosphodiesterase domain (GDPD) containing and lymphocyte antigen 6 (Ly6)/Plaur domain (LYPD) containing proteins. These gene families were chosen because many of the genes demonstrate male reproductive tract-specific expression. AlthoughGdpd1andGdpd4mutant mice were fertile, disruptions ofCstandPategene clusters andLypd4resulted in male sterility or severe fertility defects secondary to impaired sperm migration through the oviduct. While absence of the epididymal protein families CST and PATE affect the localization of the sperm membrane protein A disintegrin and metallopeptidase domain 3 (ADAM3), the sperm acrosomal membrane protein LYPD4 regulates sperm fertilizing ability via an ADAM3-independent pathway. Thus, use of CRISPR/Cas9 technologies has allowed us to quickly rule in and rule out proteins required for male fertility and expand our list of male-specific proteins that function in sperm migration through the oviduct.

Background: In Bangladesh there is a scarcity of data on the degree of knowledge amongadolescents about reproductive health. Objective: To assess the awareness about reproductivehealth issues among adolescent girls in a rural area of Bangladesh. Materials and method: Thisdescriptive type of cross sectional study was conducted among 148 adolescent girls who wereselected purposively at PurbaChandara village of Kaliakairthana of Shafipurupazilla of Gazipurdistrict in Bangladesh. A pre-tested semi structured questionnaire was used to collect data byface to face interview. Results: Most of the respondents (76.35%) were within the age group of15-19 years. The average age of their menarche was 14 years. Majority of the respondents 136(91.89%) had history of menstruation and 97 (65.54%) mentioned the duration of menstrualcycle > 7 days. Regarding the hygienic practice during menstruation, majority of them (68.24%)were unhygienic. About knowledge on age at marriage, 131 (88.51%) reportedly mentionedcorrect answer that marriage age is 18 years or more for girls. Regarding the knowledge onfamily planning and family planning methods, most of them (41.89%) had no knowledge.About the infection of reproductive tract, 87 (58.78%) had no knowledge and about half of therespondents (92; 56.79%) had no knowledge about symptoms of STDs. Conclusion: it is ourresponsibility to improve the condition of adolescent girls by giving clear and correct knowledgeon reproductive health which will help them to maintain a good and sound reproductive healthin future. This research aims to get the results from the fertility age couples' motivation on cervical cancer early detection.The preparation of a scoping review adapted the Arksey O'Malley framework consisted of 5 stages: research questions with the PEOs framework (Population, Exposure, and Outcomes), searching literature using relevant databases. The 12 articles used to consist of 3 themes, namely sexual and reproductive health problems, sexual health problems, and sexual and reproductive health.

Endometrial adenocarcinoma is one of the most prevalent female reproductive tract cancers in the world, and the development of effective treatment is still the main goal of its current research. Epithelial-mesenchymal transition (EMT) plays a significant part in the occurrence and development of epithelial carcinoma, including endometrial adenocarcinoma. Recepteur d’origine nantais (RON) induces EMT and promotes proliferation, migration, and invasion in various epithelial-derived cancers, but its role in endometrial adenocarcinoma is still poorly studied. The purpose of this study is to verify the overexpression of RON in endometrial adenocarcinoma and to explore its specific roles. RON expression in tumor lesions was verified by immunohistochemical staining, HEC-1B cells were used to construct stable cell lines with RON overexpression or knockdown to investigate the effects of RON on the function of endometrial adenocarcinoma cells, and xenotransplantation experiment was carried out in nude mice to explore the effect of RON on the growth of endometrial adenocarcinoma in vivo. This study revealed that RON could promote the proliferation, migration, and invasion of HEC-1B cells and induce EMT, and these effects were regulated through the Smad pathway. RON overexpression could promote growth of endometrial adenocarcinoma cells in nude mice, while its inhibitor BMS777607 could restrict this role. RON played an important role in endometrial adenocarcinoma and had a potential to become a new therapeutic target for endometrial adenocarcinoma.
