The aim of this study was to identify an anti-obesity peptide from Allomyrina dichotoma and investigate the lipid metabolic mechanism. Enzymatically hydrolyzed A. dichotoma larvae were further separated using tangential flow filtration and consecutive chromatographic processes. Finally, an anti-obesity peptide that showed the highest inhibitory effect on lipid accumulation was obtained, and the sequence was Glu-Ile-Ala-Gln-Asp-Phe-Lys-Thr-Asp-Leu (EIA10). EIA10 decreased lipid aggregation in vitro and significantly reduced the accumulation of body weight gain, liver weight, and adipose tissue weight in high-fat-fed mice. Compared with the control group, the levels of total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL), insulin, and homeostasis model assessment of insulin resistance (HOMA-IR) in the high-fat diet (HFD) group increased significantly, and the content of high-density lipoprotein cholesterol (HDL) in the serum decreased significantly. On the contrary, the levels of TC, TG, and insulin in the EIA10 group decreased significantly, and the HDL content increased significantly compared with the HFD group. Additionally, EIA10 dramatically decreased mRNA and protein levels of transcription factors involved in lipid adipogenesis. Taken together, our results suggest that EIA10 could be a promising agent for the treatment and prevention of obesity.

Diphlorethohydroxycarmalol (DPHC) is one of the most abundant bioactive compounds in Ishige okamurae. The previous study suggested that DPHC possesses strong in vitro anti-obesity activity in 3T3-L1 cells. However, the in vivo anti-obesity effect of DPHC has not been determined. The current study explored the effect of DPHC on high-fat diet (HFD)-induced obesity in C57BL/6J mice. The results indicated that oral administration of DPHC (25 and 50 mg/kg/day for six weeks) significantly and dose-dependently reduced HFD-induced adiposity and body weight gain. DPHC not only decreased the levels of triglyceride, low-density lipoprotein cholesterol, leptin, and aspartate transaminase but also increased the level of high-density lipoprotein cholesterol in the serum of HFD mice. In addition, DPHC significantly reduced hepatic lipid accumulation by reduction of expression levels of the critical enzymes for lipogenesis including SREBP-1c, FABP4, and FAS. Furthermore, DPHC remarkably reduced the adipocyte size, as well as decreased the expression levels of key adipogenic-specific proteins and lipogenic enzymes including PPARγ, C/EBPα, SREBP-1c, FABP4, and FAS, which regulate the lipid metabolism in the epididymal adipose tissue (EAT). Further studies demonstrated that DPHC significantly stimulated the phosphorylation of adenosine monophosphate-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in both liver and EAT. These results demonstrated that DPHC effectively prevented HFD-induced obesity and suggested that DPHC could be used as a potential therapeutic agent for attenuating obesity and obesity-related diseases.

The present study was intended to evaluate the effects of Paeonia emodi rhizome extracts on serum triglycerides (TGs), total cholesterol (TC), low density lipoprotein cholesterol (LDL-c), high density lipoprotein cholesterol (HDL-c), atherogenic index (AI), superoxide dismutase (SOD), and glutathione peroxidase (GPx). The plant was extensively examined for its in vitro antioxidant activity, and the preliminary phytochemical screening was carried out using standard protocols. Male Wistar rats were induced with hyperlipidemia using high-fat diet and were treated orally with hydroalcoholic and aqueous extracts at the dose of 200 mg/kg bw for 30 days. TGs, TC, LDL-c, and AI were significantly reduced while HDL-c, SOD, and GPx levels rose to a considerable extent. After subjecting to acute toxicity testing, the extracts were found to be safe. The observations suggest antihyperlipidemic and antioxidant potential of P. emodi in high-fat diet induced hyperlipidemic/oxidative stressed rats.

Objective: To investigate the effect of antibiotics and/or probiotics on broiler performance, some serum metabolites, cecum microflora composition, and ileum histomorphology under the Egyptian conditions. Design: Randomized controlled experimental study. Animals: Two hundred forty 1-day-old Ross (308) chicks were reared till 35 days of age. Procedures: The birds were randomly allocated into four main groups: a control diet without additives (CON); probiotic (Lactobacillus acidophilus) supplemented diet (PRO); antibiotic (Avilamycin) supplemented diet (ANT) and a mix group (AP) that received antibiotic in the diet form 1 to 4 days of age and treated during the rest of the experimental period with probiotics. Results: Chickens fed on probiotic or antibiotic diets had linear improvement in live body weight (LBW) and feed conversion ratio (FCR) compared with the control group, while the best LBW and FCR were in the AP group. An improvement in the nutrient digestibility was observed in the probiotic added groups (PRO and AP). Serum cholesterol and low-density lipoprotein cholesterol contents decreased when antimicrobial (probiotic or antibiotic) supplementations were used, while there was an increase in high-density lipoprotein cholesterol contents, serum total protein, and albumin levels. Among all groups, cecum Clostridium perfringens and Escherichia coli counts decreased; however, there was an increase in Lactobacillus count compared to the control group. In probiotic supplemented groups (PRO and AP), a significant (P<0.05) improvement in ilea architecture. Conclusion and clinical relevance: Using probiotic after initial treatment with an antibiotic in broiler diets had a positive effect on broiler growth performance, gut health (improved cecum microbial populations and ileum histomorphology), and nutrient digestibility.

Background and Objective: Yoghurt, especially bio-yoghurt has long been recognized as a product with many health benefits for consumers. Also, honey and olive oil have considerable nutritional and health effects. So, the effect of administration of yoghurt made using ABT culture and fortified with honey (2 and 6%), olive oil (1 and 4%) or honey + olive oil (2+1 and 6+4% respectively) on some biological and hematological properties of rats was investigated.Methods:The body weight gain, serum lipid level, blood glucose level, serum creatinine level, Glutamic Oxaloacetic Transaminase (GOT) activity, Glutamic Pyruvic Transaminase (GPT) activity, leukocytes and lymphocytes counts of rats were evaluated.Results:Blending of bio-yoghurt with rats&#039; diet improved body weight gain. Concentrations of Total plasma Cholesterol (TC), High-Density Lipoprotein cholesterol (HDL), Low-Density Lipoprotein cholesterol (LDL), Very Low-Density Lipoprotein cholesterol (VLDL) and Triglycerides (TG) significantly lowered in plasma of rats fed bio-yoghurt. Levels of TC, LDL, VLDL, and TG also decreased in rat groups feed bio-yoghurt supplemented with honey and olive oil. LDL concentrations were reduced by 10.32, 18.51, 34.17, 22.48, 43.30% in plasma of rats fed classic starter yoghurt, ABT yoghurt, ABT yoghurt contained 6% honey, ABT yoghurt contained 4% olive oil and ABT yoghurt contained 6% honey + 4% olive oil respectively. The blood glucose, serum creatinine, GOT and GPT values of rats decreased while white blood cells and lymphocytes counts increased by feeding bioyoghurt contained honey and olive oil.Conclusion:The findings enhanced the multiple therapeutic effects of bio-yoghurt supplemented with honey and olive oil.

AbstractSulforaphane (SFN), a naturally-occurring isothiocyanate enriched in cabbage and broccoli, has been provided as food supplements to improve weight management and reduce lipid levels. However, its effects on serum lipid profiles are contradictory. In this review, a meta-analysis and systematic review of SFN on lipid reduction and weight control is assessed with mice and rats fed on high-fat diet. The effects of SFN supplementation were evaluated by weighted mean difference (WMD) in body weight (BW), liver weight (LW) and also by its effect on serum lipids. A random-effects model was applied to estimate the overall summary effect. SFN reduced BW (WMD: − 2.76 g, 95% CI: − 4.19, − 1.34) and LW (WMD: − 0.93 g, 95% CI: − 1.63, − 0.23) significantly in our ten trials. Its effects on serum total cholesterol (TC) (WMD: − 15.62 mg/dL, 95% CI: − 24.07, − 7.18), low-density lipoprotein cholesterol (LDL-C) (WMD: − 8.35 mg/dL, 95% CI: − 15.47, − 1.24) and triglyceride (TG) (WMD: − 40.85 mg/dL, 95% CI: − 67.46, − 14.24) were significant except for high-density lipoprotein cholesterol (HDL-C) component (WMD: 1.05 mg/dL, 95% CI: − 3.44, 5.54). However, species, disease model, duration, SFN dosage as well as route of administration did not explain the heterogeneity among studies. In summary, these findings provide new insights concerning preclinical strategies for treating diseases including obesity, diabetes, hypertension, non-alcoholic fatty liver disease as well as cardiovascular disease with SFN supplements.

The metabolic fitness (MF) is a component of athletes’ physical conditioning. This study aims to investigate the effects of quercetin supplementation on Turkish Junior athletes’ lipid and protein metabolism relating to MF after one month classic boxing training. Totally 20 voluntary junior male athletes were separated into two equal groups as the experimental group (EG) and control group (CG). The participants were supplemented with 500 mg quercetin fifteen minutes before each workout in one month boxing training program. Blood samples during pre and post training were taken from athletes in order to determine metabolic fitness related parameters. Lipid profile contains low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), trigliserid (TG), total cholesterol (TC) variables while protein metabolism includes the albumin, total protein, direct bilirubin and total bilirubin parameters. The Mann Whitney U analyzes were used for comparison of the means between experimental and control groups during pre and posttest and between pre and post test results in experimental and control groups. This study showed that EG had a similar physical characteristic with CG. There were significant decrease in TC and LDL-C and an increase in HDL-C in EG while there was only significant increase in HDL-C of in controls. A significant difference of HDL-C was observed between EG and CG during pretest. In other side, TC and LDL-C and HDL-C were significantly differentiated between EG and CG during posttest. Conclusion: it can be concluded that quercetin plays an important role on lipid metabolism not protein.

     The aim of this study is to explain the effect of Ethanolic extract 70% of Metracaria chamomella on some physiological parameters in male rabbits. Twelve adult male rabbits were used in this study and were divided equally into two groups: First group was control (C) and received normal saline for four weeks, the second group (Treated group) was intubated orally with ethanolic extract of Metracaria chamomella in a dose 70 mg/kg B.W. for four weeks. Blood samples were collected by heart puncture from each animal at the end of experiment. Blood sample was divided into a part for hematological study and a part for biochemical analysis. The value of serum urea, and creatinine were reduced in animals that received Metracaria chamomella extract at dose of 70 mg/ kg B.W. as compared with the control group. Significant decrease in serum activity of aspartate aminotransferase alanine aminotransferase and alkaline phosphatase activity was observed in Metracaria treated animal as compared with the control group. This study explained that there was significant increase in serum total protein, serum albumin and serum globulin in treated animals as compared with the control group. The value of total cholesterol, serum triacylglycerol, serum low density lipoprotein cholesterol and very low density lipoprotein cholesterol concentration were reduced significantly in animal received Metracaria chamomella extract while the high density lipoprotein cholesterol was elevated significantly as compared with control group. While the effect of Metracaria chamomella extract on blood picture showed no changes in red blood cell count, hemoglobin concentration and hematocrit value but showed significant decrease in platelet count, and significant   increase in white blood cell count as compared with control group. Over all this study explained that Metracaria chamomella extract had Reno protective and hypolipidimic effect in male rabbit.

Two lactobacilli strains; Lactobacillus acidophilus ATCC 20079 and Lactobacillus plantarum ATCC 20179 and two bifidobacteria strains; Bifidobacterium bifidum GSGG 5286 and Bifidobacterium longum ATCC 15707 were studied their abilities to reduce the cholesterol content in vitro. It was investigated that the in vivo cholesterol-lowering effect of L. plantarum ATCC 20179, B. bifidum GSGG 5286 and mixture of both probiotics (L. plantarum ATCC20179 and B. bifidum GSGG5286) on hyperlipidaemic rats for 8 weeks. All lactobacilli and bifidobacteria strains assimilate the cholesterol content in laboratory media. It was observed the highest assimilation of cholesterol was in L. plantarum ATCC 20179 and B. bifidum GSGG 5286 strains. In vivo, L. plantarum ATCC 20179  group was more effective in improving serum lipid profile levels [total cholesterol (TC), triglycerides (TG), low density lipoprotein – cholesterol (LDL-C), high density lipoprotein – cholesterol                   (HDL-C), very low density lipoprotein – cholesterol (VLDL-C) and Atherogenic Index (AI)],                      liver enzyme activities (ALT, AST and ALP),  malonaldehyde (MDA), hydrogen peroxide (H2O2) and total antioxidants capacity (TAC) levels than mixed-organisms and B. bifidum groups, respectively of hyperlipidaemic rats. It was concluded that L. plantarum ATCC 20179 showed more                     favourable results than B. bifidum GSGG 5286 in relation to cardiovascular risk factors in hyperlipidaemic rats.

The current study investigated the role of guggulsterone (GS), a farnesoid X receptor antagonist, in the choline metabolism and its trimethylamine (TMA)/flavin monooxygenases/trimethylamine- N-oxide (TMAO) inhibiting potential in a series of in vitro and in vivo studies as determined by high-performance liquid chromatography (HPLC), mass spectroscopy (MS), and liquid chromatography (LC)-MS techniques. Atherosclerosis (AS) was successfully induced in a group of experimental animals fed with 2% choline diet for 6 weeks. Serum lipid profiles such as total cholesterol, triglycerides, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, and very low-density lipoprotein cholesterol were measured. Pro-inflammatory cytokines levels, markers for a hepatic injury, and oxidative stress markers were assessed. Interestingly, GS reduced the level of TMA/TMAO in both in vitro and in vivo studies as demonstrated by the peaks obtained from HPLC, MS, and LC–MS. Furthermore, GS exhibited cardioprotective and antihyperlipidemic effects as evidenced by the attenuation of levels of several serum lipid profiles and different atherogenic risk predictor indexes. GS also prevented hepatic injury by successfully restoring the levels of hepatic injury biomarkers to normal. Similarly, GS inhibited the production of pro-inflammatory cytokines levels, as well as GS, enhanced antioxidant capacity, and reduced lipid peroxidation. Histopathological study of aortic sections demonstrated that GS maintained the normal architecture in AS-induced rats. On the basis of results obtained from current investigation, we suggest that GS might have a great therapeutic potential for the treatment of AS.

Abstract: The objective of this work was to evaluate the effect of a dietary supplementation with soy lecithin (SL) on the productive performance and blood constituents of rabbit females and their offsprings. A total of 40 rabbits does were distributed into four treatments: control group, no dietary SL inclusion; and three groups with 0.5, 1.0, and 1.5% SL inclusion in the diets. The inclusion of 1.5% SL increased the count of blood cells and hemoglobin concentrations; 0.5-1.0% SL reduced the total cholesterol levels in the blood, as well as the low-density lipoprotein-cholesterol and the activities of the enzymes alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase, but increased the levels of total lipids, triglycerides, high-density lipoprotein-cholesterol, and the activities of the antioxidant enzymes. Supplementation with 1.0-1.5% SL resulted in higher milk production and heavier litters. Soy lecithin supplementation at 1% improves the physiological parameters and increases the milk production of rabbit does, also improving the performances of their offsprings.
