Due to their large spectrum of bioactive properties, much attention has recently been drawn to phlorotannins—i.e., phenolic compounds characteristic from brown macroalgae. This study aimed to evaluate the antioxidant and anti-inflammatory properties of F. vesiculosus phlorotannin extracts and purified fractions. Overall, the crude extract and its ethyl acetate fraction (EtOAc) showed good radical scavenging activity, particularly towards nitric oxide (NO•). Subsequent subfractions of EtOAc (F1 to F9) with different molecular weights were then shown to inhibit lipopolysaccharide-induced NO• production in macrophages, with stronger effects being observed for fractions of lower MWs. Of the three intracellular markers analyzed, inducible NO• synthase showed the highest sensitivity to almost all the phlorotannin-rich samples, followed by interleukin 1β and cyclooxygenase 2, which was only inhibited by F2. Furthermore, this subfraction inhibited the phosphorylation and degradation of inhibitory protein κBα, thus preventing the activation of NF-κB and blocking the inflammatory cascade at the transcriptional level. This sample was characterized by the presence of a major compound with a deprotonated molecular ion at m/z 507 with a fragmentation pattern coherent with that of a phlorotannin derivative. Overall, this work unveiled some of the mechanistic aspects behind the anti-inflammatory capacity of phlorotannins from F. vesiculosus, endorsing its use as a possible natural source of anti-inflammatory compounds.

Natural polyphenols, such as resveratrol (RES) or epicatechin (EPI), are attractive for treatments of various diseases, including vaginal infections and inflammation, because of their strong anti-oxidative and anti-inflammatory properties. However, their low solubility and consequent poor bioavailability limit their therapeutic uses. To overcome these limitations, a vaginal delivery system comprising either RES or EPI liposomes-in-hydrogel was developed. This system permits therapeutic action of both liposomal polyphenol (RES or EPI) and chitosan-based hydrogel. Liposomes of around 200 nm and entrapment efficiency of 81% and 77% for RES and EPI, respectively, were incorporated into chitosan hydrogel, respectively. Medium molecular weight chitosan (2.5%, w/w) was found to have optimal texture properties and mucoadhesiveness in ex vivo conditions. The in vitro release studies confirmed the sustained release of polyphenols from the system. Both liposomal polyphenols and polyphenols-in-liposomes-in-hydrogel exhibited only minor effects on cell toxicity. EPI showed superior radical scavenging activity at lower concentrations compared to antioxidants vitamin C and E. Anti-inflammatory activity expressed as the inhibitory activity of formulations on the NO production in the LPS-induced macrophages (RAW 264.7) confirmed the superiority of EPI liposomes-in-hydrogel. The plain liposomes-in-hydrogel also exhibited potent anti-inflammatory activity, suggesting that chitosan hydrogel acts in synergy regarding anti-inflammatory effect of formulation.

Amauroderma rugosumis a wild mushroom that is worn as a necklace by the indigenous communities in Malaysia to prevent fits and incessant crying by babies. The aim of this study was to investigate the nutritive composition and antioxidant potential and anti-inflammatory effects ofA. rugosumextracts on LPS-stimulated RAW264.7 cells. Nutritional analysis of freeze-dried mycelia ofA. rugosum(KUM 61131) from submerged culture indicated a predominant presence of carbohydrates, proteins, dietary fibre, phosphorus, potassium, and sodium. The ethanol crude extract (EE), its hexane (HF), ethyl acetate (EAF), and aqueous (AF) fractions of mycelia ofA. rugosumgrown in submerged culture were evaluated for antioxidant potential and anti-inflammatory effects. EAF exhibited the highest total phenolic content and the strongest antioxidant activity based on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) assays. HF showed dose-dependent inhibition of NO production in LPS-stimulated RAW264.7 cells and NO radical scavenging activity. Gas chromatographic analysis of HF revealed the presence of ethyl linoleate and ergosterol, compounds with known anti-inflammatory properties. In conclusion, the nutritive compositions and significant antioxidant potential and anti-inflammatory effects of mycelia extracts ofA. rugosumhave the potential to serve as a therapeutic agent or adjuvant in the management of inflammatory disorders.

Oxidative stress and inflammation are related to several chronic diseases including cancer. Actinidia callosa var. ephippioides (ACE) is a special folk medicinal plant from Taiwan. The aim of this study is to evaluate the antioxidant, anti-inflammatory, and antiproliferative activities of the methanol extract and fractions from the stem of ACE. Trolox Equivalent Antioxidant Capacity (TEAC), 1,1-Diphenyl-2-picrylhydrazyl (DPPH) scavenging activity, total phenolic content, flavonoid content, inhibition on nitric oxide (NO) productions by LPS-induced RAW264.7 cell, and on lung cancer cell proliferation were employed. Among all fractions, ethyl-acetate fraction (EA-ACE) showed higher TEAC, DPPH radical scavenging activities, polyphenol and flavonoid contents, respectively. EA-ACE also decreased the LPS-induced NO production and expressions of inducible nitric-oxide synthase (iNOS) in RAW264.7 cells. EA-ACE had the highest antiproliferative activity with an IC50 (The concentrations required for inhibition of 50% of cell viability) of 469.17 ± 3.59 μg/mL. Catechin also had good effects in the antioxidant and anti-inflammatory activities. Catechin might be an important bioactive compound in the stem of ACE. The above experimental data indicated that the stem of ACE is a potent antioxidant medicinal plant, and such efficacy may be mainly attributed to its polyphenolic compounds.

Background: Spices and herbs are known to have anti-oxidant and anti-inflammatory properties. We studied their properties of Namya Kanom Jeen (NKJ), a spicy soup (Southern Thailand recipe) with water and ethanol (50% and 95%) extracts.Methods: This study aimed to assess functional properties of NKJ powder extract using in vitro model. These functional properties were anti-oxidant, α-amylase inhibition, and anti-inflammatory properties. Anti-oxidant activities were determined using free radical scavenging activity (DPPH) and Ferric Reducing Anti-oxidant Power (FRAP). Anti-inflammation effect was studied by measuring nitric oxide (NO) production inhibition on RAW264.7 macrophage cells after being exposed to lipopolysaccharide (LPS). Results: Water extract of NKJ powder demonstrated the highest activity in anti-inflammatory and anti-oxidant property by DPPH radical scavenging activity when compared with ethanol extract. Conclusion: NKJ powder extracts may ameliorate the oxidation and inflammation conditions which would be the combined effect of several bioactive compounds from many of the ingredients within these extracts.Keywords: Anti-oxidant; Anti-inflammation; Phenolic compound; Namya Kanom Jeen powder

Objective: The objective of this study was to evaluate the antioxidant and anti-inflammatory activities of the solid powder obtained from the ethyl acetate fraction from the flower Opuntia stricta. Methods: The flower extract was evaluated for antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay and reducing power assay was carried out by ferric-reducing capacity (FRC) assay method. The in vitro anti-inflammatory activity was evaluated using human peripheral blood mononuclear cells (PBMCs) stimulated by lipopolysaccharide (LPS) to evaluate nitric oxide (NO) production method. Results: The solid powder obtained from the ethyl acetate fraction from the flower O. stricta showed a good antioxidant activity in scavenging DPPH radical and FRC assay with compared standard sample. This solid powder also showed good anti-inflammatory activity in cell viability (LPS-induced PBMCs) assay and NO assay. Conclusion: These results suggest that the solid powder obtained from the ethyl acetate fraction from the flower O. stricta has significant antioxidant and anti-inflammatory activities.

Astragalus membranaceus (AM) has been used for anti-oxidative, anti-inflammatory, anti-cancer, and immunomodulatory activities. In this study, we confirmed that the anti-oxidative and anti-inflammatory effects of AM were enhanced after it was fermented by Lactiplantibacillus plantarum. The anti-oxidative effect was measured by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical levels, total phenolic contents (TPC), reducing power, and H2O2 levels. AM-LP MG5145 and MG5276 showed higher free radical scavenging activity than AM-NF (51.22%). In addition, AM-LP MG5145 and MG5276 showed higher TPC (49.11 mg GAE/mL), reducing power (OD700 = 0.37), and H2O2 (1.71 µM) than AM-NF. The calycosin contents determined to AM-NF (17.24 ng/mL), AM-LP MG5145 (139.94 ng/mL), and MG5276 (351.01 ng/mL) using UPLC-ESI-MS/MS. Anti-inflammatory effects were analyzed by investigating the inhibitory effects of fermented AM on cytotoxicity, NO production, and mRNA expression of COX-2, iNOS, NF-κB, and TNF-α in LPS-induced RAW 264.7 cells. AM-LP MG5145 and MG5276 showed no cytotoxicity. AM-LP MG5145 (50.86%) and MG5276 (51.66%) inhibited NO production in LPS-induced RAW 264.7 cells. Moreover, AM-LP MG5145 and MG5276 downregulated macrophage iNOS, COX2, TNF-ɑ, and NF-κB expression. In conclusion, A. membranaceus fermented by L. plantarum MG5145 and MG5276 can be used in cosmetics and health foods as natural antioxidant compounds.

The aim of this study was to identify the optimal extraction conditions for leaves of Osmanthus fragrans var. aurantiacus. Inhibitory effects of various extracts on NO production were compared. Antioxidant evaluations for total phenol and flavonoid contents were carried out using various extracts of O. fragrans var. aurantiacus leaves obtained under optimal extraction conditions that showed the greatest effect on NO production. The optimal method for extracting O. fragrans var. aurantiacus leaves resulted in an extract named OP OFLE. OP OFLE showed DPPH and ABTS radical scavenging activities in a concentration-dependent manner. Phillyrin (PH) was isolated as a major compound from OP OFLE by HPLC/DAD analysis. OP OFLE and PH reduced inducible nitric oxide (iNOS) and cyclooxygenase (COX)-2 protein expression and downregulated proinflammatory cytokines such as interleukin (IL)-1β, IL-6, IL-8, and tumor necrosis factor (TNF)-α in LPS-stimulated RAW 264.7 and HT-29 cells. To determine the signal pathway involved in the inhibition of NO production, a Western blot analysis was performed. Results showed that OP OFLE decreased phosphorylation of extracellular regulated kinase (pERK) 1/2 and the expression of nuclear factor-kappa B (NF-κB). Our results suggest that extracts of O. fragrans var. aurantiacus leaves and its major components have biological activities such as antioxidative and anti-inflammatory properties.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

The present study was undertaken to explore the phytochemical screening, anti-bacterial and anti-oxidant activities of the hydro-methanolic leaves extract of Mangifera indica using standard screening methods such as disc diffusion and DPPH methods. In phytochemical screening, Mangifera indicaextract showed presence of secondary metabolites such as carbohydrate, phenols, tanins and proteins whereas Saponins were absent. It also showed antibacterial activities against almost all the test organisms. The extracts possessed potent hydroxyl radical scavenging activity against the positive control standard Ascorbic acid. Results denote the presence of hydroxyl radical scavenging principles in the extracts.

Inflammation has stimulated significant worldwide scientific interest because of its implication in many human diseases. Most inflammations are caused by reactive oxygen species or free radicals. Annona muricataleaf extracts were investigated for their in-vitroantioxidant and anti-inflammatory potentials. Annona muricataleavesweredried at room temperature, blended using a mill.and extracted with solvents of varying degree of polarities. The solventsused were hexane, ethyl acetate,and ethanol. After sequential extraction, the crude extracts were examined for their in-vitroanti-inflammatory activities on lipoxygenase inhibition, proteinase inhibition, albumin denaturation inhibition,and red blood cell membrane stabilization assays,while the antioxidant activities were examined using DPPH, ABTS and hydrogen peroxide assays. The results showed that the ethanol extract had significantlyhigher albumin denaturation inhibition activity at 500 μg/mL (p < 0.01). The activity of all the extracts on proteinase inhibition decreased with the increase in concentration of the extracts. Indomethacin (standard), ethanol extract,and ethyl acetate extract exhibited a dose dependent increase in lipoxygenase activity. The ethanol extract showed highred blood cell membrane stabilization activity at 500 μg/mL and the activity was comparable with that of the standard (diclofenac). Hydrogen peroxide scavenging activity of the extracts and standard (Vitamin C) were comparable at 20 –100 μg/mL. The ethanol extract showed significantly higher(p < 0.01) DPPH radical scavenging activity compared with other extracts. A similar trend was also observed for ABTS radical scavenging activity. Generally,the ethanol extract exhibited higher anti-inflammatory and antioxidant activities in most of the assays, this could be attributed to the polar compounds present in the extract.
