In aquatic environment, engineered nanoparticles (ENPs) are present as complex mixtures with other pollutants, such as trace metals, which could result in synergism, additivity or antagonism of their combined effects. Despite the fact that the toxicity and environmental risk of the ENPs have received extensive attention in the recent years, the interactions of ENPs with other pollutants and the consequent effects on aquatic organisms represent an important challenge in (nano)ecotoxicology. The present review provides an overview of the state-of-the-art and critically discusses the existing knowledge on combined effects of mixtures of ENPs and metallic pollutants on aquatic organisms. The specific emphasis is on the adsorption of metallic pollutants on metal-containing ENPs, transformation and bioavailability of ENPs and metallic pollutants in mixtures. Antagonistic, additive and synergistic effects observed in aquatic organisms co-exposed to ENPs and metallic pollutants are discussed in the case of “particle-proof” and “particle-ingestive” organisms. This knowledge is important in developing efficient strategies for sound environmental impact assessment of mixture exposure in complex environments.

The present review critically examines the state-of-the-art of the research concerning the likely environmental implications of engineered nanoparticles (ENPs) with specific emphasis on their interactions with phytoplankton in the aquatic environment. Phytoplankton plays a key role in the global carbon cycle and contributes to the half of the global primary production, thus representing some of the Earth ' s most critical organisms making the life on our planet possible. With examples from our own research and the literature, we illustrate what happens when aquatic organisms are unintentionally exposed to metal-containing ENPs, which are increasingly released into the environment from nano-enabled materials. We highlight the complexity of the ENPs behavior in the aquatic environment and focus on the three key steps of the bioavailability process: exposure availability, uptake availability and toxico-availability. The influence of the phytoplankton on the ENPs fate in the aquatic environment is discussed, too.

Background: Tocotrienols (TTs) are vitamin E derivatives naturally occurring in several plants and vegetable oils. Like Tocopherols (TPs), they comprise four isoforms, α, β, γ and δ, but unlike TPs, they present an unsaturated isoprenoid chain. Recent studies indicate that TTs provide important health benefits, including neuroprotective, anti-inflammatory, anti-oxidant, cholesterol lowering and immunomodulatory effects. Moreover, they have been found to possess unique anti-cancer properties.Objective:The purpose of this review is to present an overview of the state of the art of TTs role in cancer prevention and treatment, as well as to describe recent patents proposing new methods for TTs isolation, chemical modification and use in cancer prevention and/or therapy.Methods:Recent literature and patents focusing on TTs anti-cancer applications have been identified and reviewed, with special regard to their scientific impact and novelty.Results:TTs have demonstrated significant anti-cancer activity in multiple tumor types, both in vitro and in vivo. Furthermore, they have shown synergistic effects when given in combination with standard anti-cancer agents or other anti-tumor natural compounds. Finally, new purification processes and transgenic sources have been designed in order to improve TTs production, and novel TTs formulations and synthetic derivatives have been developed to enhance their solubility and bioavailability.Conclusion:The promising anti-cancer effects shown by TTs in several preclinical studies may open new opportunities for therapeutic interventions in different tumors. Thus, clinical trials aimed at confirming TTs chemopreventive and tumor-suppressing activity, particularly in combination with standard therapies, are urgently needed.

Although procedures for assessing the carcinogenic risks associated with exposure to individual chemicals are relatively well developed, risk assessment methods for mixtures of chemicals are still in the developmental stage. In this paper, we examine the difficulties in assessing the risks of exposure to complex mixtures, with special reference to the potential for synergistic effects among the compo nents of the mixture. Statistical models for describing the joint action of multiple exposures are reviewed, and their implications for low-dose risk assessment are examined. The potential use of pharmacokinetic models to describe the metabolism of mixtures is also considered. Application of these results in regulating mixtures of carcinogenic substances is illustrated using examples involving multiple contaminants in drinking water and polycyclic aromatic hydrocarbons produced from combustion sources.

Predicting the ecological responses to climate change is particularly challenging, because organisms might be affected simultaneously by the synergistic effects of multiple environmental stressors. Global warming is often accompanied by declining calcium concentration in many freshwater ecosystems. Although there is growing evidence that these changes in water chemistry and thermal conditions can influence ecosystem dynamics, little information is currently available about how these synergistic environmental stressors could influence the behaviour of aquatic organisms. Here, we tested whether the combined effects of calcium and temperature affect movement parameters (average speed, mean turning frequency and mean-squared displacement) of the planktonic Daphnia magna , using a full factorial design and exposing Daphnia individuals to a range of realistic levels of temperature and calcium concentration. We found that movement increased with both temperature and calcium concentration, but temperature effects became considerably weaker when individuals were exposed to calcium levels close to survival limits documented for several Daphnia species, signalling a strong interaction effect. These results support the notion that changes in water chemistry might have as strong an effect as projected changes in temperature on movement rates of Daphnia , suggesting that even sublethal levels of calcium decline could have a considerable impact on the dynamics of freshwater ecosystems.

FACTORS THAT AFFECTING THE TOXICITY OF POLLUTANTS TO AQUATIC ORGANISMS. There are a large number of pollutants in aquatic environment with various characteristics and factors that can modify and affect the toxicity of pollutants in this environment. The major factors affecting pollutant toxicity include physicochemical properties of pollutants, mode of exposure, time, environmental factors, and biological factors. Moreover, organisms in an aquatic ecosystem are seldom exposed to only single pollutant, and most cases the stress of pollution on aquatic ecosystems is related to the interaction and combined effects of many chemicals. The combined effects may be synergistic or antagonistic, depending on the pollutants and the physiological condition of the organism involved.

Understanding the combined effects of stressors on plants is important for understanding how they will tolerate herbivory and other damage under unfavorable conditions. When two stresses have synergistic effects, plants may experience particularly strong impacts. We examined individual and combined effects of drought stress and clipping (simulated herbivory) on two species of goldenrods (Solidago altissima L. and S. gigantea Ait.). Each stress reduced production of most plant structures, with drought stress having stronger impacts than clipping. The effects of the two stresses were additive for S. gigantea but synergistic for S. altissima, at least for aboveground biomass and inflorescence biomass. Both species, when under stress, reallocated resources toward asexual reproduction (rhizomes) and away from sexual reproduction (inflorescences). Our results suggest that even closely related plants may tolerate damage differently when under abiotic stresses, and that predicting the additive vs. synergistic action of combined stresses will be difficult.

The individual and combined effects of potassium sorbate and vanillin concentrations on the growth of Penicillium digitatum, P. glabrum, and P. italicum in potato dextrose agar adjusted to water activity 0.98 and pH 3.5 were evaluated. Inhibitory concentrations of potassium sorbate varied from 150 ppm for P. digitatum to 700 ppm for P. glabrum, and for vanillin from 1,100 ppm for P. digitatum and P. italicum and 1,300 ppm for P. glabrum. Fractional inhibitory concentration (FIC) isobolograms show curves deviated to the left of the additive line. Calculated FIC index varied from 0.60 to 0.84. FIC index as well as FIC isobolograms show synergistic effects on mold inhibition when vanillin and potassium sorbate are applied in combination.
