The Viburnum opulus (guelder rose) fruit is a Polish native fruit that grows across almost the entire territory of the country, except for the Tatra Mountains. The fruits are rich in nutrients and biologically active compounds, however, they are rarely used for food production or as a dietary ingredient. This study assessed this fruit for dry matter (17.3% ± 0.7), extract (13% ± 0.53), titratable acidity (1.78 g/100 g ± 0.48), pectin (1.10% ± 0.97), vitamin C (47.79 mg/100 g ± 1.02), the amount of sugar identified as fructose (6.85 g/100 g ± 0.67), antioxidant activity, ABTS (14.64 ± 0.61 μM Trolox/100 g), FRAP (211.14 ± 1.04 µM Trolox/100 g) and total polyphenolic content TPC (1695.88 ± 0.86 mg GAE/100 g). The analysis of phenolic identification was performed using the ACQUITY Ultra Performance LC (UPLC) system with the binary solvent manager and PDA detector.

Background. Barbary fig (Opuntia spp), called prickly pear, is a cultivated species belonging to family Cactaceae. It produces fruits one time per year during a short season. It is largely spreading in the Mediterranean countries. The aim of the study was to investigate the physicochemical and biochemical analysis of powder of prickly pear fruit peels of three Moroccan varieties (Aakria, Derbana, and Mles). Material and Methods. Both physicochemical analysis (humidity, water activity, Brix, ash content, pH, and total titratable acidity) and biochemical analysis (total carotenoid content, betalain content, total polyphenolic content, and ascorbic acid content) were effectuated according to protocols reported within the present research work. Results. Regarding the physiochemical analysis, the humidity of powder of prickly pear fruit peels ranged from 10.173 ± 0.002 to 15.27 ± 0.02%. The water activity (aw) ranged from 0.17 ± 0.002 to 0.336 ± 0.002. The values of Brix ranged from 68.67 ± 0.33 to 74.22 ± 0.51° Bx. pH values ranged from 5.41 ± 0.01 to 5.62 ± 0.01. The total titratable acidity values ranged from 0.644 ± 0.014 to 0.76 ± 0.007 g citric acid/100 g DM (dry matter). The ash content ranged from 1.02 ± 0.02 to 11.41 ± 0.03 g/100 g DM. Regarding the biochemical analysis, the total carotenoid content ranged from 5.14 ± 0.10 to 9.79 ± 0.06 mg/g of FM (Fresh matter). The total betalain content ranged from 2 ± 0.69 to 37.66 ± 2.65 mg/100 g of DM. The total polyphenolic content ranged from 1739.92 ± 13.69 to 2409.66 ± 43.65 mg gallic acid equivalent (GAE)/100 g of DM. The ascorbic content values ranged from 186.78 ± 13.23 to 294.04 ± 5.81 mg/100 g of DM. Conclusion. The results showed an interesting richness of the investigated powder in pigments (betacyanins, indicaxanthins, and carotenoids) and nutritional compounds including sugar, proteins, and vitamins that make this powder interesting for being used as a dye in food.

Background. Prickly pear (Opuntia spp.), called Barbary fig, is a cultivated species springing from family Cactaceae. It is native to Mexico and has been naturalized in other continents, especially the Mediterranean countries (North Africa). The aim of the study was to investigate the physical, physicochemical, and biochemical criteria of peels of three Moroccan prickly pear varieties (Aakria, Derbana, and Mles) growing in the Rhamna regions (dry area). Material and Methods. Both physicochemical characteristics (humidity, water activity, Brix, ash content, pH, and total titratable acidity) and biochemical characteristics (total carotenoid content, betalain content, total polyphenolic content, and ascorbic acid content) were were studied according to previously reported methods. Results. Regarding the physiochemical criteria, the moisture of the fresh peels of studied varieties ranged from 81.59 ± 0.02 to 83.47 ± 0.02%. The water activity (aw) ranged from 0.862 ± 0.001 to 0.872 ± 0.001. The values of Brix varied from 14.69 ± 0.05° Bx to 15.80 ± 0.03° Bx. pH values varied from 5.13 ± 0.01 to 5.32. The total titratable acidity values ranged from 0.130 ± 0.008 to 0.196 ± 0.014 g of citric acid/100 g of FM (fresh matter). The ash content values ranged from 8.92 ± 0.10 to 11.04 ± 0.06 g/100 g of FM. Regarding the biochemical criteria, the total carotenoid content ranged from 2.29 ± 0.01 to 2.87 ± 0.01 μg/g of FM. The total betalain content ranged from 6213.46 ± 58.86 to 8487.19 ± 51.71 μg/100 g of FM. The total polyphenolic content varied from 160 ± 3.55 to 243.79 ± 5.55 mg GA E/100 g of FM. The ascorbic content ranged from 58.21 ± 0.24 to 74.72 ± 0.17 mg/100 g of FM. Conclusion. The findings of physicochemical and biochemical criteria of the investigated varieties growing in Moroccan drylands showed promising results in terms of studied parameters.

Yerba maté extracts have in vitro antioxidant capacity attributed to the presence of polyphenolic compounds, mainly chlorogenic acids and dicaffeoylquinic acid derivatives. DPPH is one of the most used assays to measure the antioxidant capacity of pure compounds and plant extracts. It is difficult to compare the results between studies because this assay is applied in too many different conditions by the different research groups. Thus, in order to assess the antioxidant capacity of yerba maté extracts, the following procedure is proposed: 100 µL of an aqueous dilution of the extracts is mixed in duplicate with 3.0 mL of a DPPH 'work solution in absolute methanol (100 µM.L-1), with an incubation time of 120 minutes in darkness at 37 ± 1 °C, and then absorbance is read at 517 nm against absolute methanol. The results should be expressed as ascorbic acid equivalents or Trolox equivalents in mass percentage (g% dm, dry matter) in order to facilitate comparisons. The AOC of the ethanolic extracts ranged between 12.8 and 23.1 g TE % dm and from 9.1 to 16.4 g AAE % dm. The AOC determined by the DPPH assay proposed in the present study can be related to the total polyphenolic content determined by the Folin-Ciocalteu assay.

Resistance of bacteria, fungi and cancer cells to antibiotics and other drugs is recognized as one of the major problems in current medicine. Therefore, a search for new biologically active compounds able to either kill pathogenic cells or inhibit their growth is mandatory. Hard-to-reach habitats appear to be unexplored sources of microorganisms producing previously unknown antibiotics and other molecules revealing potentially therapeutic properties. Caves belong to such habitats, and Actinobacteria are a predominant group of microorganisms occurring there. This group of bacteria are known for production of many antibiotics and other bioactive compounds. Interestingly, it was demonstrated previously that infection with bacteriophages might enhance production of antibiotics by them. Here, we describe a series of newly isolated strains of Actinobacteria that were found in caves from the Tatra Mountains (Poland). Phage induction tests indicated that some of them may bear active prophages able to produce virions upon treatment with mitomycin C or UV irradiation. Among all the examined bacteria, two newly isolated Streptomyces sp. strains were further characterized to demonstrate their ability to inhibit the growth of pathogenic bacteria (strains of Staphylococcus aureus, Salmonella enterica, Enterococcus sp., Escherichia coli, and Pseudomonas aeruginosa) and fungi (different species and strains from the genus Candida). Moreover, extracts from these Streptomyces strains reduced viability of the breast-cancer cell line T47D. Chemical analyses of these extracts indicated the presence of isomers of dichloranthrabenzoxocinone and 4,10- or 10,12-dichloro-3-O-methylanthrabenzoxocinone, which are putative antimicrobial compounds. Moreover, various previously unknown (unclassified) molecules were also detected using liquid chromatography–mass spectrometry, suggesting that tested Streptomyces strains may synthesize a battery of bioactive compounds with antibacterial, antifungal, and anticancer activities. These results indicate that further studies on the newly isolated Actinobacteria might be a promising approach to develop novel antibacterial, antifungal, and/or anticancer drugs.

Tomato (Lycopersicon esculentum Mill.) is an economically important and widely cultivated vegetable crop that is consumed both fresh and processed. The nutritional value of tomato fruits is related to the content of carotenoids, polyphenols, sugars, organic acids, minerals and vitamins. Currently, there is a growing interest in the qualitative and quantitative increase in the content of health-promoting compounds in tomato fruits. VIR Lycopersicon (Tourn.) Mill. genetic resources collection includes 7678 accessions of one cultivated and nine wild species, which in turn provides ample opportunities for searching for information on the variability of the content of biologically active substances and searching for sources with a high content of them in the gene pool. Our work presents the results of the study of 70 accessions of cultivated and wild tomato on the main biochemical characteristics: the content of dry matter, ascorbic acid, sugars, carotenoids, chlorophylls and anthocyanins. As the basis for the selection of accessions for the study, accessions with various colors of fruits, including new accessions with varying content of anthocyanin, were taken. As a result of this study, the amplitude of variability in the content of dry matter (3.72–8.88 and 9.62–11.33 %), sugars (1.50–5.65 and 2.20–2.70 %), ascorbic acid (12.40–35.56 and 23.62– 28.14 mg/100 g), titratable acidity (0.14–0.46 and 0.33–0.48 %), chlorophylls (0.14–5.11 and 2.95–4.57 mg/100 g), carotenoids (0.97–99.86 and 1.03–10.06 mg/100 g) and anthocyanins (3.00–588.86 and 84.31–152.71 mg/100 g) in the fruits of cultivated and wild tomatoes, respectively, was determined. We have determined correlations between the content of dry matter and monosaccharides (r = 0.40, p ≤ 0.05), total sugars (r = 0.37, p ≤ 0.05) and ascorbic acid (r = 0.32, p ≤ 0.05); the content of ascorbic acid and carotenoids (r = 0.25, p ≤ 0.05). A high dependence of the content of chlorophyll a and b among themselves (r = 0.89, p ≤ 0.05), as well as between the content of chlorophyll b and anthocyanins (r = 0.47, p ≤ 0.05), the content of β-carotene (r = 0.26, p ≤ 0.05) and the content of monosaccharides (r = –0.29, p ≤ 0.05) has been noted. We have identif ied tomato accessions with a high content of individual chemical substances, as well as with a complex of traits that can be used as sources in breeding for a high content of dry matter, sugars, ascorbic acid, pigments and anthocyanins.

The article highlights comparative research results of milk quality indices obtained from the milking in specialized milking halls with such milking units as «Parallel», «Carousel» or in stalls with the milking unit «Molokoprovid». The fat and protein mass fraction, dry matter and fat-free dry matter, density, titratable and active acidity, heat resistance and freezing point have been determined according to the accepted techniques. The electrical conductivity of milk has been determined by using the analytical device MD-20 MAS-D-TEC. The total amount of milk bacteria has been determined by reductase reduction test and by seeding method in Petri dish. The milk quality has been investigated by the fermentation and rennet fermentation tests. The higher indices of the fat mass fraction, the protein mass fraction and the dry substance concentration of milk, obtained in specialized milking halls, have been established. This is not statistically significant. Milk, obtained from the milking unit «Molokoprovid», has higher index of titratable acidity, lower thermal stability in comparison with milk, obtained from specialized milking halls with milking units «Parallel» and «Carousel». It has been determined that there is the bacteria insemination increase in milk received from milking cows in stalls in comparison with milk, obtained from milking in specialized halls. Milk, obtained from the milking unit «Carousel», indicates the subclinical form of mastitis in cows or «Carousel» operation violationif there is in 1,8 mS/cm conductivity increase above average index 4,6 mS/cm. AbstractLakes located in the Polish and Slovak parts of the Tatra Mountains were included in the Tatra diatom database (POL_SLOV training set). The relationship between the diatoms and the water chemistry in the surface sediments of 33 lakes was the basis for the statistical and numerical techniques for quantitative pH reconstruction. The reconstruction of the past water pH was performed using the alpine (AL:PE) and POL_SLOV training sets to compare the reliability of the databases for the Tatra lakes. The results showed that the POL_SLOV training set had better statistical parameters (R2 higher by 0.16, RMSE and max. bias lower by 0.2 and 0.36, respectively) compared to the AL:PE training set. The better performance of the POL_SLOV training set is particularly visible in the case of Przedni Staw Polski where the curve of the inferred water pH shows an opposite trend for the period from the 1960s to 1990 compared to that based on the AL:PE dataset. The reliability of the inferred pH was confirmed by the comparison with current instrumental measurements.

AbstractStrong wind events frequently result in creating large areas of windthrow, which causes abrupt environmental changes. Bare soil surfaces within pits and root plates potentially expose soil to erosion. Absence of forest may alter the dynamics of water circulation. In this study we attempt to answer the question of whether extensive windthrows influence the magnitude of geomorphic processes in 6 small second- to third-order catchments with area ranging from 0.09 km2 to 0.8 km2. Three of the catchments were significantly affected by a windthrow which occurred in December 2013 in the Polish part of the Tatra Mountains, and the other three catchments were mostly forested and served as control catchments. We mapped the pits created by the windthrow and the linear scars created by salvage logging operations in search of any signs of erosion within them. We also mapped all post-windthrow landslides created in the windthrow-affected catchments. The impact of the windthrow on the fluvial system was investigated by measuring a set of channel characteristics and determining bedload transport intensity using painted tracers in all the windthrow-affected and control catchments. Both pits and linear scars created by harvesting tend to become overgrown by vegetation in the first several years after the windthrow. The only signs of erosion were observed in 10% of the pits located on convergent slopes. During the period from the windthrow event in 2013 until 2019, 5 very small (total area <100 m2) shallow landslides were created. The mean distance of bedload transport was similar (t-test, p=0.05) in most of the windthrow-affected and control catchments. The mapping of channels revealed many cases of root plates fallen into a channel and pits created near a channel. A significant amount of woody debris delivered into the channels influenced the activity of fluvial processes by creating alternating zones of erosion and accumulation.

The study assessed the changes in the quality and physical and chemical parameters of apples of four cultivars (‘Gala’, ‘Idared’, ‘Topaz’, ‘Red Prince’) subjected to mechanical vibration during transport under model conditions and after storage (shelf-life). Quality changes in apples were evaluated based on skin and flesh colour, total soluble solids, dry matter, firmness, titratable acidity, pH value, total polyphenol content, and antioxidant capacity. The vibration applied at a frequency of 28 Hz caused changes in the above parameters, which were visible also after storage and depended on the cultivar, but often did not show any clear trend or direction or were not statistically significant. The values of the total colour difference factor ΔE showed considerable variations in the skin colour but only small variations in the flesh colour of individual cultivars. Vibration resulted in a decrease in firmness. Variations in dry matter, total soluble solids, pH, and titratable acidity were small, often insignificant. Mechanical vibration and storage led to an increase in the polyphenol content and antioxidant capacity of all studied cultivars. The greatest stability of quality parameters, relatively high content of bioactive compounds, and antioxidant capacity were observed for ‘Red Prince’. The lowest quality parameters were noted for ‘Gala’. The analysed cultivars continued to show a high level of antioxidant capacity after treatment, which allows the conclusion that they remain a good source of bioactive compounds after transport and short-term storage.
