Aster scaber is a wild vegetable cultivated in Korea and is known to contain phytochemicals with various biological activities. The potential antioxidant and anti-obesity effects of A. scaber and their mechanism are yet to be reported. We evaluated the total phenolic, flavonoid, and proanthocyanidin contents and oxygen radical absorbance capacity of A. scaber ethanolic extract (ASE), and analyzed the major phenolic compounds of ASE. Antioxidant activity was measured at the chemical level through 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power assay, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), and fluorescence recovery after photobleaching (FRAP) assay. In addition, it was measured in vitro through inhibition of Reactive oxygen species (ROS) production in 3T3-L1 adipocyte, and inhibition of lipid accumulation was also evaluated. ASE reduced the expression of enzymes involved in the production of ROS and increased the expression of antioxidant enzymes that reduce increased ROS levels. They also reduced the expression of adipogenesis transcription factors that regulate adipocyte differentiation in relation to ROS production, inhibited the expression of lipogenesis-related genes related to fat accumulation through AMP-activated protein kinase (AMPK) activity, and increased expression of lipolysis-related genes. Thus, ASE containing CGA (chlorogenic acid) inhibits ROS production in 3T3-L1 adipocytes, owing to its strong antioxidant activity, and inhibits lipid accumulation caused by oxidative stress. The extract can be used as a potential functional food material for reducing oxidative stress and obesity.

A method for the separation and quantification of acteoside by reverse-phase high-performance liquid chromatography (HPLC) was developed and validated. Hot water and ethanolic extracts of Osmanthus fragrans leaves and flowers were analyzed for acteoside content. Excellent linearity was obtained, with an r2 higher than 0.999. The precision, specificity, and accuracy of our method were excellent, suggesting that it can be conveniently used for the quantification of acteoside in the crude extract of O. fragrans. The hot water and ethanol extracts were analyzed, and their biological activities were tested. The extraction yields, marker (acteoside) contents, and antioxidant activities of the leaf and flower extracts were analyzed. The antioxidant activity was confirmed by measuring the 2,2-diphenyl-2-picrylhydrazyl radical scavenging activity, reducing power, and total phenolic content. The acteoside content tended to be higher in the 100% ethanol extract of O. fragrans compared to those with the other extraction conditions tested. Overall, almost all extracts prepared with ethanolic solvents tended to produce better antioxidant activity than those prepared with hot water. These results suggest that the ethanolic extract of O. fragrans could serve as a potential antioxidant and anti-inflammatory pharmaceutical source, and our validated method would be useful for the quality control of O. fragrans extracts.

Background: Biological activities of medicinal plants make them attractive alternative complementary medicines. Therefore, this study aimed to assess the antioxidant and antifungal activities of the methanol extract of Myrianthus arboreus, Allanblackia gabonensis, Gladiolus quartinianus, Peperomia fernandopoiana and Vepris soyauxii. Methods: Total phenolic contents (TPC) and flavonoid contents (FC) were evaluated using standard methods. The antioxidant activity of the extracts was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and ferric reducing power assays. Antifungal activity was evaluated by the broth micro-dilution method. Results: TPC of extracts ranging from 67.89 to 196.01 mgGAE/g TPC were very higher in P. fernandopoiana whole plant (PFW) extract (196.01±10.25 mgGAE/g) and MAL (165.71±10.68 mgGAE/g) whereas the A. gabonensis fruit extract (AGF) and M. arboreus bark extract (MAB) had the highest flavonoid content (66.54 ± 2.12 and 48.13 ± 1.65 mgEC/g of extract, respectively). Apart from M. arboreus leave extract (MAL), all extracts showed important DPPH radical scavenging activity (IC50 < 20 μg/mL). AGFl and PFW exhibited significant ferric reducing power compared to ascorbic acid and other extracts (P<0.05). Each of the tested extracts showed antifungal activity against atleast one of the five pathogenic fungi, with MICs ranging from 128 to 1024 µg/mL. MAL was active on all the tested fungi whereas MAB showed the highest antifungal activity (MIC=128 µg/mL). Conclusion: The findings of this study emphasize the evidence that the methanol extracts of the leave and bark of Myrianthus arboreus could be an alternative to fight against oxidative stresses and infections due to opportunistic yeast pathogens. Antioxidant-rich foods and beverages play an essential role in the prevention of diseases. This study assessed the influence of the addition of ethanolic extract of propolis (EEP) to beer at different concentrations (0.05, 0.15, and 0.25 g/L). Total phenolic content (TPC) and total flavonoid content (TFC) were determined. Antioxidant activity (AA) was evaluated by radical scavenging activity (DPPH and ABTS) and reducing power (FRAP). The addition of EEP in beer resulted in a linear increase in the TPC with values of 4.5%, 16.7%, and 26.7% above a control (no EEP added; 242 mg gallic acid equivalent/L). A similar increase was observed with TFC values 16.0%, 49.7%, and 59.2% above the control (16.9 mg quercetin equivalent/L). The FRAP assay indicated linear increases in AA relative to control with values of 1555, 1705, and 1892 μmol Trolox equivalent/L following EEP additions. The incorporation of EEP resulted in increases in the bioactive compounds and AA in beer without altering the physicochemical parameters of golden ale beer. The results indicate a promising use of propolis extract as a functional ingredient in beer.

Plants are major sources of bioactive organic molecules that are of interest to the pharmaceutical industry and are being screened for new drugs and chemicals. Glycyrrhiza glabra Linn. (Family: Fabaceae) also known as Liquorice, Mulaithi, or Yashtimadu is a well-known medicinal plant used in traditional medicine. Its roots and rhizomes are the medicinal parts used and are reported to possess antitumor, antimicrobial, antiviral, anti-inflammatory, immunoregulatory activities. This plant is also used as a flavoring agent due to its sweetness. In the present work, the ethanolic extract of Glycyrrhiza glabra was prepared, and its phytochemical analysis was done using HPLC. The in vitro antioxidant assays such as DPPH radical scavenging assay, Hydroxyl radical scavenging assay, total antioxidant activity assay, and total reducing power assay were done, and the results showed significant antioxidant activity of the extract. The extract was analyzed further to evaluate the ability to protect against oxidative stress in chicken liver tissue. The levels of glutathione and lipid peroxidation in H2O2 and/or G glabra extract-treated tissue indicated the potential to protect against oxidative stress under in vitro conditions. The in vitro comet assay results showed that the G glabra extract protected against H2O2 induced cellular DNA damage. These findings indicated promising antioxidant and antigenotoxic potential of G.glabra and need further exploration for translating these findings to its possible health benefits.

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

Active films based on carboxymethyl chitosan incorporated corn peptide were developed, and the effect of the concentration of corn peptide on films was evaluated. Physicochemical properties of the films, including thickness, opacity, moisture content, color, mechanical properties, water vapor permeability, and oil resistance, were measured. Biological activities of the films, including the antioxidant and antibacterial activities, were characterized in terms of 2, 2-diphenyl-1-picrylhydrazyl free radical scavenging activity, reducing power, the total antioxidant activity, and the filter disc inhibition zone method. The results indicated that the incorporation of corn peptide caused interactions between carboxymethyl chitosan and corn peptide in Maillard reaction and gave rise to the films light yellow appearance. Compared with the Control, the degree of glycosylation, browning intensity, thickness, opacity, tensile strength, antioxidant activity, and antibacterial activity of films were increased, but the elongation, vapor permeability, and oil resistance of films were decreased. The films based on corn peptide and carboxymethyl chitosan can potentially be applied to food packaging.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

The aim of this study was to investigate the biological and chemical activity on two species of fungi of the genus Penicillium isolated from wastewater. On the selected species of fungi the different antioxidant activity assays were carried out: DPPH free-radical scavenging activity, total antioxidant activity, Fe2+- chelating ability and Fe3+- reducing power. Total phenol content was also determinate for ethanolic extract of mycelia. Penicillium chrysogenum ethanolic extract contained higher total phenolic content and better total antioxidant capacity as well as ferrous ion chelating ability. Penicillium fumiculosum ethanolic extract showed higher DPPH free-radical scavenging activity, as well as reducing power. Based on the obtained results it can be concluded that two types of fungi are potential new sources of natural antioxidants.
