Extraction solvent is a very important factor in the recovery of antioxidants from natural matrices. In this study, the effect of three solvents (ethanol, ethanol/water and water) on the phenolic composition, antioxidant and anti-cholinesterase activities and electrochemical behaviour of four winemaking byproducts (seeds, skins, stems, and pomace) was evaluated. Phenolic composition was determined by the Folin–Ciocalteu method and ultra-high-performance liquid chromatography (UHPLC), antioxidant activity by the capacity to scavenge 2,2-diphenyl-1-picrylhydrazyl and hydroxyl radicals, anti-cholinesterase activity by the Ellman’s method, and electrochemical behaviour by cyclic voltammetry. Eight phenolic compounds were quantified with higher content in water/ethanol extracts (e.g., epicatechin in pomace: 17 mg/100 g vs. 7 and 6 mg/100 g in ethanol and water extracts, respectively), although there were some exceptions (e.g., gallic acid in seeds was most abundant in water extracts). Moreover, the highest total phenolic content (TPC) and antioxidant activity were found in ethanol/water extracts (between 2 and 30-fold the values of the other extracts). Overall, the most active extracts in inhibiting both acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes were ethanol/water and ethanol extracts from seeds (between 31.11 and 53.90%). The electrochemical behaviour allowed for differentiating the extracts depending on the solvent and the byproduct. Our findings indicate that winemaking byproducts represent a source of phenolic compounds with antioxidant and anti-cholinesterase activities and suggest that cyclic voltammetry is a promising technique to evaluate the phenolic extraction process from these byproducts.

Morus alba L. (white mulberry) has been commonly used as a functional food and for medicinal purposes. Hence, the aim of the study was to compare the phenolic profile of white mulberry commercial samples in relation to their antioxidant potential and acetylcholinesterase (AChE) inhibitory activity. It is of interest to determine whether herbal products originating from different commercial sources differ in their phenolic profiles. For this purpose, a simple and rapid high-performance liquid chromatography (HPLC) method was used for the separation and determination of ten major phenolic compounds. Total phenolic (TPC), total flavonoid (TFC), and total phenolic acid contents (TPAC), as well as l(+)-ascorbic acid (ASA) contents, were determined. The antioxidant potential was assessed by DPPH (2,2-diphenyl-1-picrylhydrazyl radical) scavenging activity and ferric-reducing/antioxidant power (FRAP) assay, while the AChE inhibitory activity was determined by the Ellman assay for water extracts. The study revealed that excluding two herbal products containing fruits and a sample containing leaves of white mulberry, yerba mate and lemon, the remaining samples were generally consistent in terms of phenolic composition as well as antioxidant potential and AChE inhibitory activity. This reflects the health-promoting properties of the samples under study. Moreover, the results showed that the water extracts of white mulberry were richer in phenolic compounds and presented higher antioxidant activity than the hydromethanolic extracts. However, the water extracts showed low inhibitory activity against AChE. Moreover, the correlation analysis indicated a high positive relationship between phenolic composition and antioxidant activity in extracts of white mulberry. Overall, the obtained results may be useful in the evaluation of new dietary supplements and food products. The water extracts of white mulberry could be used for antioxidant purposes, while the hydromethanolic extracts could be incorporated in antioxidant formulations.

Agrimonia pilosa Ledeb (AL) has received considerable attention as a herbal medicine for its applications in ethnopharmacology with heath benefits. This study aimed to investigate antioxidant activities and A549 growth inhibitory capacity from its root extract (RE) and aerial parts extract (AE). The 50% ethanol extracts were used for the tests. The total polyphenol content and the antioxidant effects comprising ABTS+ and DPPH free radical scavenging activities were evaluated. Phenolic compounds in the extracts were isolated using high performance liquid chromatography (HPLC). Lactate dehydrogenase released in medium was also evaluated. Total phenolic and flavonoid content, and in vitro antioxidant potential of the RE were higher (p<0.01) than that of the AE. Two individual phenolic compounds consisting of 4-hydroxybenzoic acid and p-coumaric acid were firstly identified in both by HPLC. The RE exhibited higher A549 inhibitory capacity compared to the AE and activated the apoptotic proteins of bcl-2, bax, bad, caspase-3 and caspase-9 in A549. In conclusion, the AL extracts were more effective in antioxidant and A549 cells inhibitory capacity.

The aim of this study was to determine the best extraction conditions of phenolic compounds present in açaí, blueberry and goji berry fruits using the response surface methodology (RSM). The phenolic compounds profile by high-performance liquid chromatography, antioxidant activity was also determined. A factorial 23 design was used to analyze the effect of the solvent (ethanol and water), time (30 and 60 min) and temperature (30 °C and 60 °C) on the extraction of total phenolic compounds (TPC) and activity antioxidant (AA). The variables time and temperature had a positive effect on antioxidant activity (AA) in their highest levels, 60 min and 60 °C, respectively. The solvent ethanol 800 g/mL was more efficient in TPC extracting with AA in all matrices. Rutin was present in high amounts in blueberry and goji berry, and the myricetin in açaí. The açaí showed higher in vitro antioxidant activity when extracted at 60 °C for 60 min. The high correlation coefficient (0.98) of global response (GR) showed that we can find out single and global response in research with multiple dependent variables.  The GR analysis indicated the highest values of the TPC and AA when the fruits were extracted at 60°C for 60 min using ethanol as solvent and it was very useful for simplifying and improving the phenolic compounds extraction performance.

<abstract> <p>Vegetable oils are among the foodstuffs produced and consumed in abundance by the population in Burkina Faso. These edible oils are nutrient sources for health. However, the oxidation of edible oils is a phenomenon that leads to their degradation. Hence, the objectives of this study were to determine the antioxidants compounds, to evaluate the antioxidant activity of vegetable oils produced in Burkina Faso and to propose good manufacturing practices. Thus, 32 samples of crude peanut oils and refined cottonseed oils were analyzed. α-Tocopherol was determined by High performance liquid chromatography (HPLC), total phenolic compounds and DPPH by spectrophotometry. The α-tocopherol averages are 10.89 and 56.44 mg/100 g for peanut oils and cottonseed oils, respectively (<italic>p</italic> &lt; 0.05). For total phenolic compounds, the averages are 2.91 and 0.64 mg/100 g of gallic acid equivalent for peanut oils and cottonseed oils, respectively (<italic>p</italic> &lt; 0.05). The inhibition percentages are respectively 17.97% and 5.58% (<italic>p</italic> &lt; 0.05) for peanut oils and cottonseed oils. For antioxidant activity, the averages are 0.81 and 0.27 mg trolox/100 g for peanut oils and cottonseed oils, respectively (<italic>p</italic> &lt; 0.05). Cottonseed oils have the highest levels of α-tocopherol while peanut oils have high levels of total phenolic compounds and antioxidant activity. Vegetable oils contain acceptable levels of α-tocopherol and total phenolic compounds for their oxidative stability and health benefits for the consumer. However, the levels of biomolecules will be higher if production and preservation conditions are improved and controlled. The results could be used for future recommendations about human feeding programs in Burkina Faso.</p> </abstract>

Phenolic compounds are widely distributed in almost all higher plants and are present in different tissues. The quality and quantity of phenolics are used as criteria for variety identification. The present study has been planned to determine the phenolic composition of Pistacia vera. Total phenolic content and concentration of flavonoids of eight different extracts, from the floral buds and leaves of Pistacia vera. (Anacardiaceae) were determined using spectrophotometric methods. The total phenolic content ranged from 401.64 to 513.77 (mg GAE/g DW). The total flavonoid concentrations varied from 8.28 to 67.94 mg CE/g. Antioxidant activity was assessed by three methods: DPPH°+, β-carotene and ABTS and determined respectively as IC50 (mg/mL), CAA and PI (%), all extract have shown to be endowed by a high antioxidant activity similar to that of BHT. Ethanolic extract of male floral buds showed the highest phenolic and flavonoid concentration and the strongest antioxidant activity. The significant linear correlation was confirmed between the values for the total phenolic content and antioxidant activity of plant extracts. Phenolic compounds were investigated by LTQ Orbitrap LC-MS analysis and revealed 17compounds split into 5 groups: Abscisic acids, phenolic acids and their derivatives, flavonols, flavanones, flavones, flavanols. The high contents of phenolic compounds indicated that these last contribute to the antioxidant activity. The Pistacia vera can be regarded as promising candidates for natural plant sources of antioxidants with high value.

AbstractDue to its complex biochemical properties, the bee pollen is considered one of the functional foods. Bee pollen collected from pollen grains from different botanical sources offers almost a full diet such as carbohydrates, proteins, amino acids, vitamins, minerals. In this study, methanol extracts of 13 honeybee pollen samples were evaluated for flavonoids, phenolic compounds, and antioxidant capacity. Caffeic acid, chlorogenic acid, vanillic acid, p-coumaric acid, gallic acid, quercetin, rutin, vanillin, and naringin were identified as main phenolic compounds in pollen extracts by High-Performance Liquid Chromatography (HPLC) analysis. The obtained results are: total phenolic content - 379.8 to 915.6 mg GAE/100 g, total flavonoid content - 207.1 to 550 mg QE/100 g, and antioxidant activity - 808.2 to 3311 mg GAE/100 g in bee pollen extracts.

The aim of the present study was to establish the polyphenolic profile and antioxidant activity of the leaves of Andrographis paniculata (kalmegh) that could be potentially used for the benefit of mankind. An ultra high performance liquid chromatographic method was used to identify and quantify the individual phenolic compounds of the kalmegh leaves. A total of 20 polyphenolic compounds were identified and quantified in kalmegh leaves, including hydroxybenzoic acids, hydroxycinnamic acids, flavonols and other group of phenolic compounds. The UPLC analysis of the phenolic compounds revealed that salicylic acid was the dominant phenolic compound present in the leaves extract followed by gallic acid, chlorogenic acid, veratric acid and protocatechuic acid. The kalmegh leaves were extracted with 4 different solvents (ethyl acetate, methanol, butanol and water) and screened for total phenolic content (TPC) and antioxidant activity. The antioxidant activity of kalmegh was assessed by evaluating the 1, 1-diphenyl-2-picrylhydrazyl (DPPH), reducing power and phosphomolybdenum assay. Side effects of drug-based treatment observed in patients during degenerative diseases treatments has directed towards to the identification of plants with antioxidant activity. Kaffir lime peel was selected in this study. This work was aimed to investigate the effect of solvent (hexane, ethyl acetate and n-butanol) polarity on the fractionation of ethanolic crude extract and assess its antioxidative property by DPPH radical scavenging assay. The results show semi polar solvent of ethyl acetate exhibited the best solvent to extract phenolic compounds from ethanolic kaffir lime peel with total phenolic content detected was 0.12 mg Gallic Acid Equivalent/mg. The employment of solvents possessing different polarity resulted several fractions, i.e. hexane, ethyl acetate, n-butanol and residue with different type of phenolic compounds in each fraction. Accordingly, each fraction exhibited different antioxidant activity against free radical compound of DPPH. The N-butanol fraction demonstrated the strongest activity which is shown by lowest IC50 value among the fractions tested. The fraction of n-butanol exhibited the IC50 value of 0.44 mg/mL which means that only 0.44 mg of the fraction/mL is required to inhibit the neutralization of DPPH by 50%. Phenolic identification by High Performance Liquid Chromatography indicates the different phenolic compounds in each fraction that contribute to antioxidative property to different extent. Further investigation to identify these phenolic compounds will lead to further development of kaffir lime as natural antioxidant to treat specific degenerative diseases.

Polyphenols from berries have proved healthy effects after“in vitro”and“in vivo”studies, such as preventing tumor growing and neurodegenerative and cardiovascular diseases. We compared four different kinds of berries—strawberry, raspberry, blackberry, and blueberry—with the aim to distinguish their phenolic composition, concerning their antioxidant capacity along with their“in vitro”availability. Folin-Ciocalteu method was used for the determination of phenolic compounds, and the antioxidant capacity was measured by ORAC method. Moreover, the determination of anthocyanins was accomplished with an HPLC-DAD. Finally, we carried out an“in vitro”digestion to simulate the gastrointestinal digestion. All berries showed good antioxidant capacity with significant differences, besides high total phenolic compounds. Content of anthocyanins measured by HPLC-DAD varied between the different berries, namely, blackberries and strawberries which showed higher anthocyanin concentration. After“in vitro”digestion, berries showed poor bioavailability of the analysis of anthocyanins (9.9%–31.7%). Availability of total phenolic compounds was higher than anthocyanins (33%–73%). Moreover, strawberries and blackberries presented the less availability grade. Decrease in antioxidant activity measured by ORAC method was about 90% in all berries studied. Therefore, bioavailability of phenolic compounds remains unclear and more correlation between“in vitro”and“in vivo”studies seems to be necessary.
