The onion is one of the most cultivated and consumed vegetables, and is a very valuable source of antioxidant substances. Every onion variety is rich in flavonols—mainly quercetin derivatives which makes onions the main dietary source of these compounds. This study intends to develop an ultrasound-assisted extraction method (UAE), an economical, green, and efficient technique, that allows us to determine the quality of onion extracts in terms of flavonol composition and antioxidant activity. For this purpose, an ultrasound-assisted extraction method has been optimized to obtain extracts with a high amount of flavonols, as well as with a high antioxidant activity, not only separately using a Box–Behnken design, but also simultaneously, based on multi-response optimization. Multiple response optimization has not only demonstrated an efficacy level similar to that of the individual ones, but it also represents a considerable reduction in terms of cost, time and work. The optimal conditions for simultaneous extractions were determined as follows: 76.8% methanol as an extraction solvent at pH 2 and 58.5 °C temperature, 85% amplitude, 0.9 s cycle, and 0.2:13 g:mL sample mass/solvent volume ratio. Furthermore, the developed method exhibited a high precision level and great recoveries in a rather short extraction time (2 min). These results, together with the development of a fast and simple UHPLC analysis method, lead us to consider the developed UAE method as a suitable technique for the extraction of bioactive compounds from onion matrices.

Chokeberry (Aronia melanocarpa L.) contains a large number of bioactive compounds, which make of it a highly nutritional and antioxidant food with anti-cancer and anti-inflammatory properties. It has been recently seen its consumption increased because of its interesting composition and properties as a functional food. The ultrasound-assisted extraction method has been optimized, since it is a simple and fast technique to extract phenolic compounds, as well as anthocyanins, not only separately but also simultaneously. Multiple response has been optimized and demonstrated an effectiveness level similar to the individual ones. This represents a considerable reduction in costs, time and work. The optimal conditions for simultaneous extractions were: 54% methanol as extraction solvent at pH 2.72 and 69.4 °C temperature, 70% amplitude, 0.7 s cycle, and 0.5:18.2 g:mL sample mass/solvent volume ratio. The developed methods showed a high precision level with coefficients of variation lower than 5%. The methods were successfully applied to commercial samples. Additionally, the extraction of other compounds of biological interest, such as organic acids and sugars, was also studied.

Quercetin is a bioactive compound that has many beneficial effects on human health. Due to a lack of information about quercetin and quercetin derivatives in the Vietnam shallot, this work aimed to identify the quercetin family in shallot skin and flesh by liquid chromatography-mass spectrometry. Effect of extraction method (conventional solvent extraction and ultrasound-assisted extraction), including ethanol concentration (40–80%) and extraction time (15 to 60 mins) on the yield of quercetin were also investigated in this study. Quercetin, quercetin-glucoside and methylated-quercetin-hexose were commonly found in the extract of shallot flesh and skin. A very small amount of quercetin aglycone (0.7%) was found in flesh extract, while it was the most abundant in the skin sample (46.3%). The high content of quercetin-mono-glycoside was obtained in shallot extract (43.8% and 35.3% in the shallot flesh and skin, respectively). The flesh extract also contained high level of quercetin-di-glucoside (38.9%). About 14-16% of methylatedquercetin (-glucoside) were found in shallot bulb (both skin and flesh). It was found that ethanol concentration and extraction time directly affected on the quercetin extraction yield. The highest quercetin content was obtained in the sample which was extracted in 60% ethanol in combination with ultrasound-assisted extraction for 15 to 45 mins. The ultrasound-assisted extraction method improved quercetin yield by 13.38-15.64% and 49.46-56.88% for shallot skin and flesh compared to conventional solvent extraction. This study proved that ultrasound-assisted extraction could successfully be used for extraction of quercetin from shallot (both skin and flesh).

Pewarna alami lebih disukai karena tidak memiliki efek yang negatif terhadap tubuh manusia sehingga mempunyai tingkat keamanan pangan yang tinggi, selain itu juga pewarna alami mudah diuraikan. Bahan pewarna alami bisa diperoleh dari berbagai jenis sumber, salah satunya yaitu spesies alga yang merupakan tumbuhan tingkat rendah di perairan. Alga terdapat 2 jenis yaitu makroalga yang berukuran besar dan mikroalga yang berukuran kecil (renik). Spesies mikroalga yang dapat menghasilkan pewarna alami salah satunya adalah Spirulina. Jenis pewarna alami yang terkandung pada mikroalga tersebut yaitu fikosianin. Pada penelitian ini, dilakukan ekstraksi fikosianin dari Spirulina platensis dengan menggunakan tiga metode berbeda yaitu Maserasi, Ultrasound-Assisted-Extraction (UAE) dan Freezing untuk mengetahui metode mana untuk menghasilkan yield dan antioksidan tertinggi dari ekstrak yang dihasilkan. Hasil dari penelitian ini didapatkan bahwa berdasarkan nilai yield tertinggi dan kandungan fikosianin, freezing merupakan metode ekstraksi terbaik dengan kandungan fikosianin yang paling tinggi yaitu sebesar 26,53%. Sedangkan nilai aktivitas antioksidan terbesar dihasilkan oleh metode ekstraksi maserasi dengan nilai IC50 sebesar 49,59 ppm. Fikosianin yang dihasilkan dari ketiga metode ekstraksi termasuk antioksidan kuat.KATA KUNCI: ekstraksi, Spirulina plantesis, fikosianin, biopigmen, antioksidanEXTRACTION OF PHYCOCYANIN FROM SPIRULINA PLANTESIS FOR BIOPIGMENT AND ANTIOXIDANTABSTRACTRecently, biopigment known as a pigment which has no impact and easy to degradated in human body and better than artificial pigment. Biopigment can be resulted from the extraction of algae that are living in the water. Spirulina plantesis is one of algae resulting a pigment and known as phycocyanin. In this research, extraction of phycocyanin was done by three method of extractions; maceration, Ultrasound-Assisted-Extraction (UAE) and Freeze-thaw. Those extraction methods were compared to decide which method that resulted highest yield of extraction and has highest antioxidant activities. From the resulted data, the highest yield and highest phycocyanin concentration was obtained by freeze-thaw extraction method. Phycocyanin concentration from freeze-thaw ectract was 26,53% (w/w). However, the highest antioxidant activities was obtained by maceraion extraction mehod. The IC50 of extract from maceration was 49,59 ppm. Hence, all extracts from different extraction methods resulted high antioxidant activities.

The extraction of chili oil and capsaicin from chili using some different method such as Soxhlet, maceration, ultrasound-assisted extraction, microwave-assisted extraction and supercritical carbon dioxide with ethanol as a modifier was studied in this work. The results showed that the main component of chili oil is polyunsaturated fatty acids (representing 77.4%); mainly oleic acid and linoleic acid. The highest yield of oil and capsaicin were 0,5358g/g and 2,9534mg/g material; achieved from microwave-assisted extraction method. However, the highest selectivity was 1.77% with supercritical fluid extraction method. With additional co-solvent, the concentration of capsaicin increased but the selectivity decreased.

Polyphenolic compounds, plant secondary metabolites essential for plant survival, are known for their high antioxidant and anti-inflammatory activity. In addition, several polyphenols, such as phloretin, also have potential antiviral effects, making these compounds potential ingredients of biofunctional foods. A promising source for the extraction of phloretin is a by-product of apple production—apple tree leaves. Focusing on green technologies, the first aim of the present study was to optimize the direct ultrasound-assisted extraction conditions to gain the maximum yield of phloretin from air-dried apple leaves. For the optimization of process parameters, we applied the response surface method with Box–Behnken design. The optimal extraction conditions were extraction time 14.4 min, sonication amplitude 10% and 10 g of sample per 100 mL solvent (70% ethanol, w/w). Using these conditions, we assessed the content of individual and total polyphenolic compounds along with antioxidant activity in the leaves of different autumn and winter apple cultivars grown in Estonia. The analyses were carried out with chromatographic (HPLC-DAD-MS/MS) and spectrophotometric methods. The phloretin concentration ranged from 292 to 726 µg/g and antioxidant activity from 6.06 to 11.42 mg GA eq./g, these being the highest in the local winter cultivars ‘Paide taliõun’ and ‘Tellissaare’, respectively.

Manililla is a mango variety whose residues contain bioactive compounds such as polyphenols and flavonoids, with high added value. The use of environmentally friendly extraction technology would be of great relevance; hence, this study aimed to evaluate the effect of solvent relation, sonication time and amplitude on the ultrasound-assisted extraction of total polyphenols in Manililla mango residues (peel, endocarp and kernel) and antioxidant activity. An experimental design 23 with a central point was used to evaluate the curvature behavior of the process variables. Conventional maceration was used as a control. The better conditions were obtained at the central point using 50% ethanol in water, 60% amplitude and 20 min of sonication time. We obtained values of up to 1814 mg GAE/100 g, 469 mg GAE/100 g and 672 mg GAE/100 g of total polyphenols and 1228 mg QE/100 g, 653 mg QE/100 g and 880 mg QE/100 g of total flavonoids for peel, endocarp and kernel, respectively. Mangiferin was quantified in ultrasound-assisted extraction at 150 mg/g in peel and 0.025 mg/g in the kernel, but it was not detectable in maceration. An antioxidant capacity of 87%, 14% and 83% inhibition for peel, endocarp and kernel, respectively, were obtained. Peel and kernel were the residues with higher potential as extraction material, while endocarp was not.
