The antioxidative activity of Camelia sinensis tea and especially powdered tea extracts on the market, among others used as added value in functional foods, can considerably vary due to not only natural variance, but also adulteration and falsification. Thus, an effect-directed profiling was developed to prove the functional effects or health-promoting claims. It took 3–12 min per sample, depending on the assay incubation time, for 21 separations in parallel. Used as a fast product quality control, it can detect known and unknown bioactive compounds. Twenty tea extracts and a reference mixture of 11-bioactive compounds were investigated in parallel under the same chromatographic conditions by a newly developed reversed phase high-performance thin-layer chromatographic method. In eight planar on-surface assays, effect-directed tea profiles were revealed. Catechins and theaflavins turned out to be not only highly active, but also multi-potent compounds, able to act in a broad range of metabolic pathways. The flavan-3-ols acted as radical scavengers (DPPH∙ assay), antibacterials against Gram-positive Bacillus subtilis bacteria, and inhibitors of tyrosinase, α-glucosidase, β-glucosidase, and acetylcholinesterase. Further effects against Gram-negative Aliivibrio fischeri bacteria and β-glucuronidase were assigned to other components in the powdered tea extracts. According to their specifications, the activity responses of the powdered tea extracts were higher than in mere leaf extracts of green, white and black tea. The multi-imaging and effect-directed profiling was not only able to identify known functional food ingredients, but also to detect unknown bioactive compounds (including bioactive contaminants, residues or adulterations).

Polyphenols and triterpenoids in olive have relevant importance both in the physiology of the plant and the nutritional and biological value of its products. Olive leaf extracts are of special interest for their numerous health-promoting properties. The present research is investigating the occurrence of phytochemicals in supercritical fluid extracts from leaves with regard to collection time and drying temperature. The phytochemical profiles of the olive leaf extracts were determined by reversed-phase high-performance liquid chromatography (HPLC) coupled to electrospray ionization time-of-flight mass spectrometry (ESI-TOF-MS) detection. The main extracted phytochemicals were phenols and terpenoids. A significant variation in the amounts of the different components was observed as a function of the different drying temperature and collecting time (p < 0.05). Among samples, the maximal contents of polyphenols and secoiridoid derivatives were found in the extracts from olive leaves collected in November and dried at 120 °C, whereas triterpenoids showed the highest content in fresh leaves collected in August.

The pine (Pinus L.) is the largest and most heteromorphic plant genus of the pine family (Pinaceae Lindl.), which grows almost exclusively in the northern hemisphere. The demand for plant-based remedies, supplements and functional food is growing worldwide. Although pine-based products are widely available in many parts of the world, they are almost absent as food ingredients. The literature shows the beneficial effects of pine preparations on human health. Despite the wide geographical distribution of pine trees in the natural environment, there are very few data in the literature on the widespread use of pine in food technology. This study aims to present, characterise and evaluate the content of phytochemicals in pine trees, including shoots, bark and conifer needles, as well as to summarise the available data on their health-promoting and functional properties, and the potential of their use in food and the pharmaceutical industry to support health. Various species of pine tree contain different compositions of bioactive compounds. Regardless of the solvent, method, pine species and plant part used, all pine extracts contain a high number of polyphenols. Pine tree extracts exhibit several described biological activities that may be beneficial to human health. The available examples of the application of pine elements in food are promising. The reuse of residual pine elements is still limited compared to its potential. In this case, it is necessary to conduct more research to find and develop new products and applications of pine residues and by-products.

Broccoli is a source of bioactive compounds that provide an important nutritional value. The content of these compounds can vary depending on agronomic and environmental conditions, as well as on elicitation. In this study, three crop trials were carried out to evaluate the effects of the cultivation season, the application of different dosages of methyl-jasmonate (MeJA) on the overall quality and on the total content of bioactive compounds of ‘Parthenon’ broccoli cultivated under the field conditions of southeastern Spain. Color parameters, chlorophyll content, total phenolic compounds, total flavonoids and antioxidant activity were measured to evaluate the overall quality. Moreover, individual carotenoids, phenolic compounds and glucosinolates were evaluated by high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography equipped with diode array detector coupled to mass spectrometer using electro spray ionization (HPLC-DAD-ESI/MSn). The content of total carotenoids, phenolic compounds and glucosinolates were higher in autumn compared with spring, showing increases of 2.8-fold, 2-fold and 1.2-fold, respectively. Moreover, a double application of MeJA increased the contents of total carotenoids, phenolic compounds and glucosinolates by 22%, 32% and 39%, respectively, relative to the untreated samples. Considering our results, the controlled and timely application of 250 µM MeJA to the aerial parts of the plants four days before harvest, on two consecutive days, seems to be a valid agronomic strategy to improve the health-promoting capacity of Parthenon broccoli, without compromising its overall quality.

The present work had the objective of producing liqueurs from mango peels (varieties “Haden” and “Tommy Atkins”) by processes of alcoholic maceration and maceration with pectinase, as well as to evaluate bioactive compounds by reversed-phase high-performance liquid chromatography coupled to diode array detection and fluorescence-detection (RP-HPLC/DAD/FD) and in vitro antioxidant activity (AOX), for by-product potential reuse. Alcoholic maceration in wine ethanol (65% v/v) produced liqueurs with higher phytochemical and AOX content. Maceration with pectinase resulted in liqueurs with higher quercetin-3-O-glucopyranoside content. In relation to mango varieties, Haden liqueurs presented higher bioactive content than Tommy Atkins liqueurs. The liqueurs presented high antioxidant activity. The main bioactive compounds found were flavanols (epicatechin-gallate, epigallocatechin-gallate), flavonols (quercetin-3-O-glucopyranoside and rutin), and phenolic acids (gallic acid, o-coumaric acid, and syringic acid). The present study showed that the production of liqueur enabled the recovering of an important part of the bioactive content of mango peels, suggesting an alternative for the recovery of antioxidant substances from this by-product.

The beneficial effects of plant-rich diets and traditional medicines are increasingly recognized in the treatment of civilization diseases due to the abundance and diversity of bioactive substances therein. However, the important active portion of natural food or plant-based medicine is presently not under control. Hence, a paradigm shift from quality control based on marker compounds to effect-directed profiling is postulated. We investigated 68 powdered plant extracts (botanicals) which are added to food products in food industry. Among them are many plants that are used as traditional medicines, herbs and spices. A generic strategy was developed to evaluate the bioactivity profile of each botanical as completely as possible and to straightforwardly assign the most potent bioactive compounds. It is an 8-dimensional hyphenation of normal-phase high-performance thin-layer chromatography with multi-imaging by ultraviolet, visible and fluorescence light detection as well as effect-directed assay and heart-cut of the bioactive zone to orthogonal reversed-phase high-performance liquid chromato-graphy−photodiode array detection−heated electrospray ionization mass spectrometry. In the non-target, effect-directed screening via 16 different on-surface assays, we tentatively assigned more than 60 important bioactive compounds in the studied botanicals. These were antibacterials, estrogens, antiestrogens, androgens, and antiandrogens, as well as acetylcholinesterase, butyrylcholinesterase, α-amylase, α-glucosidase, β-glucosidase, β-glucuronidase, and tyrosinase inhibitors, which were on-surface heart-cut eluted from the bioautogram or enzyme inhibition autogram to the next dimension for further targeted characterization. This biological-physicochemical hyphenation is able to detect and control active mechanisms of traditional medicines or botanicals as well as the essentials of plant-based food. The array of 1,292 profiles (68 samples × 19 detections) showed the versatile bioactivity potential of natural food. It reveals how efficiently and powerful our natural food contributes to our homeostasis.

There is a growing demand to enhance pharmaceutical and food safety using synergistic compounds from Piper sarmentosum Roxb., such as polyphenols and water-soluble vitamins. However, information on standardized analytical methods to identify and quantify these compounds of interest is limited. A reversed-phase high-performance liquid chromatography with diode-array detection (HPLC-DAD)-based method was developed to simultaneously detect and quantify the amounts of tannin, flavonoid, cinnamic acid, essential oil, and vitamins extracted from P. sarmentosum leaves using methanol, chloroform, and hexane. Commercially and non-commercially-cultivated P. sarmentosum leaves were subjected to seven different drying treatments (shade; sun; air oven at 40 °C, 60 °C, 80 °C, and 100 °C; and freeze-drying) for three consecutive months. Most compounds were detected most efficiently at a detection wavelength of 272 nm. The developed method displayed good detection limits (LOD, 0.026–0.789 µg/mL; LOQ, 0.078–2.392 µg/mL), linearity (R2 > 0.999), precision (%RSD, <1.00), and excellent accuracy (96–102%). All P. sarmentosum leaf extracts were simultaneously tested and analytically compared without time-consuming fractionation. Methanolic plant extracts showed better peak area and retention time splits compared to chloroformic and hexanoic extracts. Differences in synergistic compound composition were dependent on the type of drying treatment but not on cultivation site and time of sampling. Flavonoid was identified as the dominant phytochemical component in P. sarmentosum leaves, followed by the essential oil, cinnamic acid, ascorbic acid, and tannin. Overall, we present a simple and reproducible chromatographic method that can be applied to identify different plant compounds.

Background: Polygonum maritimum is one of the spontaneous halophyte plants of the Algerian coast. Many studies were carried out to evaluate the contents and the quality of phenolic compounds of this plant around the Mediterranean region. Objective: This paper intends to identify, for the first time, the phenolic compounds from the flower part of P. maritimum. Methods: RP-HPLC-PDA (Reversed Phase-High Performance Liquid Chromatography-Photo Diode Array) material was used for this purpose. Many standards were used and their retention times were stored in a local database. Identification was made on the basis of retention times of retained compounds and those found in the literature, and UV spectra of each peak. Results: This study intends to identify five phenolic acids (gallic, ferulic, sinapic, caffeic and syringic acids), one flavonol (rutin) and one flavanone (naringenin). Conclusion: P. maritimum is an important source of natural bioactive compounds that can be exploited for the benefit of many fields.

Microalgae contain an abundance of valuable bioactive compounds such as chlorophylls, carotenoids, and phenolics and, consequently, present great commercial interest. The aim of this work is the study and optimization of recovering the aforementioned components from the microalgae species Chlorella vulgaris through conventional extraction in a laboratory-scale apparatus using a “green” mixture of ethanol/water 90/10 v/v. The effect of three operational conditions—namely, temperature (30–60 °C), duration (6–24 h) and solvent-to-biomass ratio (20–90 mLsolv/gbiom), was examined regarding the extracts’ yield (gravimetrically), antioxidant activity, phenolic, chlorophyll, and carotenoid contents (spectrophotometric assays), as well as concentration in key carotenoids, i.e., astaxanthin, lutein, and β-carotene (reversed-phase–high-performance liquid chromatography (RP–HPLC)). For this purpose, a face-centered central composite design (FC-CCD) was employed. Data analysis resulted in the optimal extraction conditions of 30 °C, for 24 h with 37 mLsolv/gbiom and validation of the predicted models led to 15.39% w/w yield, 52.58 mgextr/mgDPPH (IC50) antioxidant activity, total phenolic, chlorophyll, and carotenoid content of 18.23, 53.47 and 9.92 mg/gextr, respectively, and the total sum of key carotenoids equal to 4.12 mg/gextr. The experimental data and predicted results were considered comparable, and consequently, the corresponding regression models were sufficiently reliable for prediction.
