The aim of this study was to determine how polyunsaturated fatty acids (PUFA) supplementation can affect the productive and reproductive performance in dairy cows subjected to a fixed-time artificial insemination (TAI) protocol under farm conditions. One hundred and ninety-eight Holstein non-pregnant cows were used. Treatments consisted of a control diet (CON), without added oil, and two diets supplemented with either 2.3% soybean oil (SOY) or 2.3% linseed oil (LIN) as dry matter. The diets were formulated to be isoenergetic and isoproteic. Dry matter intake and milk yield were similar among treatments (p > 0.05). Both the percentage of fat (p = 0.011) and protein (p = 0.022) were higher in milk from animals not fed with oil (CON). The greatest saturated fatty acid (SFA) concentration (p < 0.0001) was observed in milk from cows fed the control diet, without added oil. The monounsaturated fatty acids (MUFA), PUFA, and the n-3 PUFA content was higher (p < 0.0001) in the milk from animals fed with oil with respect to the control treatment. The C18:2 cis-9, trans-11 in the milk of animals fed with oil supplements was significantly higher (p < 0.0001) than in that of the control group. Animals supplemented with linseed oil tended to show higher plasma progesterone level (p = 0.09) and a higher number of pregnant cows on the first artificial insemination (p = 0.07). These animals tended to reduce the number of TAI (p = 0.08). In brief, results showed that vegetable oils rich in PUFA supplementation considerably improve the nutritional profile of milk. PUFA n-3 supplementation slightly improves some reproductive parameters in dairy cows subjected to the fixed-time artificial insemination (TAI) protocol.

Dairy cows fitted with ruminal, duodenal and ileal cannulas were utilized to investigate the effects of feeding with Ca soaps (CaS) of palm fatty acids (FA) and rapeseed FA. Diets compared were control diet based on maize silage and concentrate, and two diets with 40 g CaS of palm oil FA or rapeseed oil FA/kg diet, replacing part of the concentrates of the control diet. Total digestibilities of dry matter, fibre and fat, and ruminal fermentation were not significantly altered by giving CaS; the extent of ruminal biohydrogenation of total unsaturated C18 FA was significantly reduced by both CaS diets. Apparent intestinal digestibility of FA was not different among diets, although the amount of FA absorbed with the CaS diets was twice that with the control diet. No difference among diets was observed for milk production, or fat and protein contents. Giving CaS diets decreased the proportions of 4[ratio ]0 to 14[ratio ]0 FA in milk fat, and increased cis-18[ratio ]1n−9, compared with control diet. The rapeseed diet lowered the content of 16[ratio ]0, and increased the contents of 18[ratio ]0 and trans-18[ratio ]1n−7. CaS diets did not result in a marked increase of polyunsaturated FA content in milk fat. Butter from cows fed on the CaS diets contained more liquid fat at 6 and 14°C than butter from the cows fed on the control diet. Incorporating CaS, particularly those from rapeseed, in dairy cows' diets increased C18 FA in milk and improved butter spreadability.

As the co-enzyme of pyruvate formate-lyase under ruminal anaerobic condition, thiamine plays a critical role in carbohydrate metabolism in dairy cows. The objective of this study was to investigate the impacts of thiamine supplementation on ruminal carbohydrate-active enzymes. Twelve Holstein dairy cows were randomly assigned into three dietary treatments: control diet (CON; 20% starch, dry matter (DM) basis), high-concentrate diet (HC; 33.2% starch, DM basis) and a high-concentrate diet supplemented with 180 mg thiamine/kg DM (HCT; 33.2% starch, DM basis). Dry matter intake and milk production were recorded for 21 days. Rumen fluid samples were collected, and ruminal pH and volatile fatty acids (VFAs) were measured. The metagenome sequencing technique was used to detect the genes in ruminal microorganisms and identify putative carbohydrate-active enzymes. The total abundances of carbohydrate-active enzymes and fiber-degrading enzymes were both reduced by HC with no effect on starch-degrading enzymes compared with CON. However, the fiber-degrading enzymes and starch-degrading enzymes were both increased after thiamine supplementation. These results indicated that 180 mg thiamine /kg DM might effectively improve rumen carbohydrate metabolism through increasing the abundance of ruminal carbohydrate-active enzymes and consequently balanced the rumen volatile fatty acids and rumen pH, providing a practical strategy in preventing subacute ruminal acidosis in cows offered HC.

In beef herds, increasing animal welfare, improving reproductive performance and easing animal management are key goals in farm economics. We explored whether delaying the removal of the intravaginal progesterone device by 24 h in heifers synchronized with a 5d Co-synch 72-h protocol could improve reproductive efficiency of fixed-time artificial insemination (FTAI). In experiment 1, we examined the total synchronization rate (TSR) in cycling Holstein heifers. Heifers (13.4 ± 0.69 mo.) were randomly assigned to the standard 5d Co-synch 56-h protocol (5dCo56; n = 10), 5d Co-synch 72-h (5dCo72; n = 17), or the modified 5d Co-synch 72-h protocol, in which removal of the progesterone device was delayed by 24 h (6dCo48; n = 19). In experiment 2, 309 cycling beef heifers on 18 commercial farms were subjected to the 5d Co-synch 72-h or 6-d Co-synch 48-h protocol and conception rate (CR) studied. In experiment 1, the three protocols led no differences on TSRs of 80.0% (5dCo56), 88.2% (5dCo72), and 89.5% (6dCo48). In experiment 2, the CR from the beef heifers, observed during two consecutive reproductive seasons did not differ: 59.7% for 5dCo72 and 62.0% for 6dCo48 (p = 0.907). Therefore, delaying removal by 24 h provides satisfactory results without reducing reproductive efficiency of heifers.

A comparative hormonal study of prostaglandin, gonadotropin releasing hormone and their combination was conducted on 30 randomly selected cross-bred dairy cows of SEMEX project for artificial insemination in Hillat Kuku, Khartoum North, Sudan. The study attempts to determine the response of the animals to hormonal induction and synchronization of estrous as well as fertility following fixed time artificial insemination. Three treatment groups of 10 cows each were undertaken. Groups 1, 2 and 3 involved administration of prostaglandin F2α (PGF2α), gonadotropin releasing hormone (GnRH) and a combination of PGF2α + GnRH, respectively. Descriptive statistical analysis was performed. The results revealed that all the three protocols induced and synchronized estrous were almost synchronous in all the treated groups. However, protocol 3 revealed a significantly (p<0.05) better result of 70% pregnancy rate compared to other two protocols that equally provided 50% pregnancy rates. The study resolves that protocol 3 be highly recommended to change the mindset of farmers about the spread of reproductive technology in Sudan. Protocols 1 and 2 provided acceptable pregnancy rates that can be enhanced with the improvement of management.

This study aimed to evaluate the effects of rectal temperature (RT) on conception rate (CR), as well as the effects of seasonality (spring-summer vs. autumn-winter) and timing of artificial insemination (AI) (morning vs. afternoon) on RT and CR in crossbred dairy cows (Holstein x Gyr). The experiment was conducted on a dairy farm in Centralina, MG, where 1,219 conventional and fixed-time inseminations were analyzed. The RT was measured immediately before AI using a digital thermometer. Pregnancy diagnosis was performed using ultrasonography between 28 and 60 days after AI. T The effects of seasonality and timing of AI on RT were analyzed with a Mann-Whitney U test and the effects of RT (above or below the average), seasonality and timing of AI on CR were analyzed with a Chi-squared test, both using the SAS program. The RT average was 39.4°C. Cows with RT ? 39.4°C had lower CR than cows with RT < 39.4°C (25.78% vs. 32.54%; P = 0.0096). During spring-summer, cows had higher RT (39.44°C ± 0.025 vs. 39.27°C ± 0.022; P < 0.0001) and lower CR (25.49% vs. 31.75%; P = 0.0146) compared with autumn-winter. Cows inseminated in the morning had lower RT (38.96°C ± 0.022 vs. 39.60°C ± 0.018; P < 0.0001) and higher CR (32.86% vs. 26.06%; P = 0.0102) than cows inseminated in the afternoon. In conclusion, crossbred dairy cows with rectal temperature equal to or greater than 39.4°C had lower conception rate. Moreover, rectal temperature and conception rate were affected by seasonality and insemination time.

The projected stagnation in the catch from global fisheries and the continuing expansion of aquaculture is considered against the background that fishmeal and fish oil are major feed stocks for farmed salmon and trout, and also for marine fish. The dietary requirement of these farmed fish for high-quality protein, rich in essential amino acids, can be met by sources other than fishmeal. However, the highly-polyunsaturated fatty acids eicosapentaenoic acid (20:5n-3) and docosahexaenoic acid (22:6n-3) present in high concentrations in fish oil are essential dietary constituents for marine fish and highly-desirable dietary constituents for salmonids. Currently, there is no feasible alternative source to fish oil for these nutrients in fish feeds. Vegetable oils rich in linoleic acid (18:2n-6) can partially substitute for 20:5n-3 and 22:6n-3 in salmonid and marinefish feeds. However, this is nutritionally undesirable for human nutrition because the healthpromoting effects of fish-derived 20:5n-3 and 22:6n-3 reflect a very high intake of 18:2n-6 relative to linolenic acid (18:3n-3) in Western diets. If partial replacement of fish oils in fish feeds with vegetable oils becomes necessary in future, it is argued that 18:3n-3-rich oils, such as linseed oil, are the oils of choice because they are much more acceptable lrom a human nutritional perspective, especially given the innate ability of freshwater fish, including salmonids, to convert dietary 18:3n-3 to 20:5n-3 and 22:6n-3. In the meantime, a more judicious use of increasinglyexpensive fish oil in aquaculture is recommended. High priorities in the future development of aquaculture are considered to be genetic improvement of farmed fish stocks with enhanced abilities to convert C18 to C20 and C22n-3 polyunsaturated fatty acids, enhanced development of primary production of 20:5n-3 and 22:6n-3 by single-cell marine organisms, and continuing development of new species.

Lactating crossbred Holstein-Friesian dairy cows (n = 331) were started on an Ovsynch regimen 68 ± 8.2 days after calving; 200 µg GnRH intramuscularly (i.m.) on Days 0 and 9, and 35 mg prostaglandin F2a i.m. on Day 7. Thirty-eight and 31 cows (11.5 and 9.4%, respectively) were in oestrus on Days 0 to 6 and 7 to 8, respectively, and inseminated, and the remainder were fixed-time inseminated (on Day 10). For these three groups, pregnancy rates (60-65 days after breeding) were 31.6, 38.7 and 34.0%, respectively (P = 0.82) and calving rates were 100, 100 and 89.9% (P = 0.23). In a preliminary trial, twelve lactating cows (45 to 60 days postpartum) with inactive ovaries were given 1500 IU eCG i.m.; 10 were in oestrus within 10 days after treatment (and inseminated) and eight of these were pregnant (30 days after breeding). The Ovsynch program resulted in acceptable reproductive performance in cyclic cows and eCG treatment has considerable promise for inducing oestrus in anoestrous cows.
