The effects of rutin on growth performance, hematological and biochemical profiles, antioxidant capacity, economics and the relative expression of selected antioxidants and lipid-related genes were studied in broiler chickens over 42 days. A total of 200 one-day-old female Ross-308 broiler chickens were distributed into four groups, with five replicates of 10 individuals per replicate. They were fed with 0 (control), 0.25, 0.5 or 1 g rutin/kg supplementation in their basal diet. Dietary rutin supplementation, especially the 1 g/kg diet, increased body weight gain, the protein efficiency ratio (p < 0.001) and both white blood cell and lymphocyte counts (p < 0.001). However, it had no effect on total protein, albumin, globulin, or alanine transaminase. A high concentration of rutin (0.5 and 1 g/kg) also significantly reduced serum total cholesterol, triacylglycerol and low-density lipoprotein cholesterol concentrations (p < 0.001), as well as malondialdehyde concentrations (p = 0.001). A high concentration diet also increased the activity of superoxide dismutase, catalase and glutathione peroxidase. Of the lipid-related genes examined, acetyl CoA carboxylase and fatty acid synthase were significantly down-regulated in the livers of rutin-fed individuals, whereas carnitine palmitoyl transferase 1 and peroxisome proliferator-activated receptor alpha were significantly up-regulated. Therefore, rutin supplementation at 1 g/kg has the potential to improve the productive performance and health status of broiler chickens.

Obesity is a common nutritional disorder associated with type 2 diabetes, cardiovascular diseases, dyslipidemia, and certain cancers. In this study, we investigated the effects ofCitrus ichangensispeel extract (CIE) in high-fat (HF) diet-induced obesity mice. Female C57BL/6 mice were fed a chow diet or an HF diet alone or supplemented with 1% w/w CIE for 8 weeks. We found that CIE treatment could lower blood glucose level and improve glucose tolerance. In the HF+CIE group, body weight gain, serum total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-c) levels, and liver triglyceride (TG) and TC concentrations were significantly (P<0.05) decreased relative to those in the HF group. To elucidate the mechanism of CIE on the metabolism of glucose and lipid, related genes expression in liver were examined. In liver tissue, CIE significantly decreased the mRNA expression levels of peroxisome proliferator-activated receptorγ(PPARγ) and its target genes, such as fatty acid synthase (FAS) and acyl-CoA oxidase (ACO). Moreover, CIE also decreased the expression of liver X receptor (LXR)αandβwhich are involved in lipid and glucose metabolism. These results suggest that CIE administration could alleviate obesity and related metabolic disorders in HF diet-induced obesity mice through the inhibition of PPARγand LXR signaling.

Carbamazepine is a drug that is widely used in the treatment of epilepsy and bipolar disorder. The prevalence of obesity in patients treated with carbamazepine has been frequently reported. However, whether carbamazepine affects adipogenesis, one of the critical steps in the development of obesity, remains unclear. Here, we show that carbamazepine increased the expression levels of peroxisome proliferator-activated receptor γ (PPARγ), CCAAT/enhancer-binding protein β (C/EBPβ), and fatty acid synthase (FASN) in 3T3-L1 cells. Notably, carbamazepine inhibited the expression levels of β-catenin, a negative regulator of adipogenesis, leading to enhanced adipogenesis. Conversely, β-catenin overexpression abolished the effect of carbamazepine on adipogenic gene expression. However, depletion of β-catenin further enhanced PPARγ expression. In addition, carbamazepine reduced β-catenin expression by lowering the levels of phospho-low density lipoprotein receptor-related protein 6 (p-LRP6) and phospho-glycogen synthase kinase 3β (p-GSK3β) in Wnt/β-catenin signaling. Moreover, carbamazepine reduced Wnt mRNA expression and decreased the promoter activities of TCF, the target of β-catenin during adipogenesis. These results suggest that carbamazepine enhances adipogenesis by suppressing Wnt/β-catenin expression, indicating its potential effects on obesity-related metabolism.

Abstract Backgroud: Saussurea involucrata Kar. et Kir. (Compositae) (CCSauI) cells are rich in caffeoylquinic acids (CQAs), which have plasma lipid reducing properties and anti-obesity effects, although the mechanisms remain unclear. Clarify CQA’s anti-obesity mechanism and provide new treatments for obesity. Methods: Sprague Dawley (SD) rat were fed a high-fat diet (HFD), then CQAs was intragastric administrated (0.1, 0.5 or 1 mg/mL). Rats were randomly divided into five groups (n=30): NC, MC, TPL (0.1 mg/mL), TPM (0.5 mg/mL) and TPH (1 mg/mL). Digestive enzyme inhibition was obtained by measuring the inhibition rate of α-amylase, α-glucosidase, and pancreatic lipase in vitro. Anti-obesity function was detected by determining the content of total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), malondialdehyde (MDA) superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). To analyze the related mechanisms qRT-PCR was employed. Results: From in vitro enzyme activity assays, the IC50 values of the CQAs extract for α-amylase, α-glucosidase, and lipase were 0.631, 0.31, and 0.438 mg/mL, respectively. CQAs administration for 8 weeks decreased retroperitoneal fat and serum and liver TC, TG, LDL-C, and MDA. Comparatively, levels of HDL-C, SOD, and GSH-Px were increased. Real-time fluorescent quantitative PCR showed inhibited expression of fatty acid synthase and 3-hydroxy-3-methyl glutaryl and coenzyme A reductase, peroxisome proliferator-activated receptor-α activation, and 7α-hydroxylase promotion. Conclusion: This study explored the role of CQAs in inhibiting digestive enzyme activities and up-regulating the expression of lipase, significant for prevention and treatment of diabetes and obesity.

The aim of this study was to determine the calcium requirements of broiler chickens in starter phase with the addition of phytase enzyme. This experiment used 1,200 broiler chickens (mixed sex) for 21 days rearing period in a closed house. The basal diets was corn and soybean meal which contained 0.097% Ca and 0.123% P-av. The treatments consisted of 6 diets treatment: P1 (basal diet with the addition of 0.42% non-phytate phosphorus (NPP), and then P2 to P6 were basal diet with the addition of 0.22% NPP and 1,000 FTU/kg phytase and the addition of calcium (Ca) at different levels: 0.82% (P2); 0.74% (P3); 0.66% (P4); 0.58% (P5) and 0.50% (P6). The growth performance, feed efficiency, and bone mineralization parameters were studied using Oneway ANOVA in a Completely Randomized Design. Duncan's new Multiple Ranges Test was used to separate means with significant differences. Results showed that 0.90% Ca supplementation without phytase and the reduction levels of Ca from 0.82% to 0.50% increased the amount of feed consumed by birds in all growth phases (P<0.05). The addition of phytase had beneficial effects on increasing body weight (P<0.05) and average of body weight gain (P<0.05), and decreased the value of FCR in 11 - 21 days and 1 - 21 days. The addition of phytase increased protein and energy consumption, followed by increased in the value of PER and  EER (P<0.05). When phytase was added in the diets, reduction levels of Ca in the diets up to 0.50% did not give any adverse effect on the tibia bone ash. It can be concluded that feeding with phytase can sustain growth performance, feed efficiency, and bone mineralization.

Conjugated linoleic acids (CLA) act as an important ligand for nuclear receptors in adipogenesis and fat deposition in mammals and avian species. This study aimed to determine whether similar effects are plausible on avian abdominal fat adipocyte size, as well as abdominal adipogenic transcriptional level. CLA was supplemented at different levels, namely, (i) basal diet without CLA (5% palm oil) (CON), (ii) basal diet with 2.5% CLA and 2.5% palm oil (LCLA), and (iii) basal diet with 5% CLA (HCLA).The content ofcis-9,trans-11 CLA was between 1.69- and 2.3-fold greater (P<0.05) than that oftrans-10,cis-12 CLA in the abdominal fat of the LCLA and HCLA group. The adipogenic capacity of the abdominal fat depot in LCLA and HCLA fed chicken is associated with a decreased proportion of adipose cells and monounsaturated fatty acids (MUFA). The transcriptional level of adipocyte protein (aP2) and peroxisome proliferator-activated receptor gamma (PPARγ) was downregulated by 1.08- to 2.5-fold in CLA supplemented diets, respectively. It was speculated that feeding CLA to broiler chickens reduced adipocyte size and downregulated PPARγand aP2 that control adipocyte cellularity. Elevation of CLA isomers into their adipose tissue provides a potential CLA-rich source for human consumption.

This study was conducted to evaluate the effects of herb mixture (summer shield) supplementation on growth performance, nutrient utilisation, and plasma lipid profiles in broiler chickens. Thirty-six male chicks at 15&nbsp;days of age were divided into three groups with three replicates of four birds. The control group of chicks was fed a basal diet and the remaining two groups of chicks were fed the basal diet supplemented with summer shield at a concentration of 1 g/kg or 2 g/kg, respectively, until 37 days of age. Although feed intake was not different among the three groups, both body weight gain and breast muscle weight were increased in chicks fed a summer shield-supplemented diet. Summer shield supplementation decreased abdominal fat weight and consequently feed efficiency was improved. Both protein and fat utilisation was improved in chickens fed a summer shield-supplemented diet although dry matter content was not influenced. In addition, summer shield supplementation improved plasma lipid profiles, i.e., total cholesterol, triglyceride, and low-density lipoprotein cholesterol were decreased, whereas high-density lipoprotein cholesterol was increased. Furthermore, summer shield supplementation increased total protein, albumin, and globulin levels in plasma, while neither glutamate oxaloacetate transaminase nor glutamate pyruvate transaminase were affected. These results suggest that summer shield supplementation has positive effects on growth performance, nutrient utilisation, and plasma lipid profiles in broiler chickens.

Lead- (Pb-) induced oxidative stress is known to suppress growth performance and feed efficiency in broiler chickens. In an attempt to describe the specific underlying mechanisms of such phenomenon we carried out the current study. Ninety-six one-day-old broiler chicks were randomly assigned to 2 dietary treatment groups of 6 pen replicates, namely, (i) basal diet containing no lead supplement (control) and (ii) basal diet containing 200 mg lead acetate/kg of diet. Following 3 weeks of experimental period, jejunum samples were collected to examine the changes in gene expression of several nutrient transporters, antioxidant enzymes, and heat shock protein 70 (Hsp70) using quantitative real-time PCR. The results showed that addition of lead significantly decreased feed intake, body weight gain, and feed efficiency. Moreover, with the exception of GLUT5, the expression of all sugar, peptide, and amino acid transporters was significantly downregulated in the birds under Pb induced oxidative stress. Exposure to Pb also upregulated the antioxidant enzymes gene expression together with the downregulation of glutathione S-transferase and Hsp70. In conclusion, it appears that Pb-induced oxidative stress adversely suppresses feed efficiency and growth performance in chicken and the possible underlying mechanism for such phenomenon is downregulation of major nutrient transporter genes in small intestine.

Objective: To evaluate the effect of dietary supplementation of essential oils (EOs) of thyme, clove, and cinnamon to broiler chickens on growth performance, serum metabolites, and tissue lipid peroxidation and antioxidants enzyme activities. Design: Randomized controlled study Animals: One hundred-eighty, one-day-old Cobb broiler chicks were randomly allocated into 4 treatment groups (3 replicates, 15 chicks each). Broilers were reared in deep litter system - pens for 6 weeks of age. Procedures: The control group was fed on corn-soybean basal control diets (starter, grower and finisher) without EOs supplementation. In addition to the basal- control diets, the 3 experimental broiler groups were supplemented with thyme oil (2nd group), clove oil (3rd group) or cinnamon oil (4th group) at 100 mg/kg. Blood, liver and muscle (breast) samples were collected from 3 broilers of each replicate at 40 days of age for measurement of serum metabolites, malondialdehyde (MDA), and antioxidant enzyme activities (liver catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). Results: The broiler chickens fed on the diets supplemented with 100 mg/kg cinnamon oil had significantly higher body weight, body weight gain and feed intake, followed by broiler chickens fed on the diets supplemented with 100 mg/kg thyme and 100 mg/kg clove oils in comparison with control group, which was showed the lowest growth performance. Supplementation with EOs significantly improved feed conversion ratio (FCR), with increased liver CAT, SOD, GSH-Px activities, as well as serum level of high density lipoprotein (HDL) in comparison with control group, also, it induced a significant decrease in serum levels of cholesterol, triglycerides, low density lipoprotein (LDL), as well as both liver and muscle tissue lipid peroxidation (MDA). Conclusion and clinical relevance: The present results indicate that dietary supplementation of EOs (thyme, clove, and cinnamon) at 100 mg/kg resulted in significantly higher body weight, body weight gain, improved FCR, reduced serum cholesterol, triglycerides and LDL. Also, reduced lipid peroxidation in liver and muscle, and improved antioxidants status of broiler chickens.

Abstract This study aims to investigate the effects of commercial probiotic supplementation in water on the performance parameters, carcass traits, immune function, and antioxidant capacity of broiler chicks. In the experiment, 120 Arbor Acres (AA) broilers (60 male and 60 female) were randomly allocated into four groups (G) – G1: basal diet and G2, G3, and G4: basal diet with 1% Lactobacillus casei, 1% L. acidophilus, and 1% Bifidobacterium in the water, lasting 42 days. The experimental results revealed that probiotic additives produced positive impacts on body weight, average daily feed intake (ADFI), and average daily weight gain for female chicks, whereas these probiotics significantly reduced ADFI and the feed conversion ratio of male chicks (P < 0.05). Probiotics efficiently improved eviscerated yield and breast yield while reducing the abdominal fat (P < 0.05) for the male broiler chicks. A marked increase was observed in the weight of the spleen, bursa of Fabricius, and thymus in the treatment group (P < 0.05). Besides, probiotics produced a significant effect on the concentrations of immune-related proteins (P < 0.05) and markedly increased the concentrations of antioxidase and digestive enzymes when compared with the control (P < 0.05). The addition of probiotics dramatically reduced the total counts of Escherichia coli and Salmonella and increased the quantity of Lactobacilli (P < 0.05). The results of the present study demonstrated an increase in growth performance, carcass traits, immune function, gut microbial population, and antioxidant capacity by supplementing 1% probiotics (L. casei, L. acidophilus, and Bifidobacterium) in the water for broilers.

, , , , and Antibiotics as growth promoters in poultry diets are currently restricted, so other feed additives, such as prebiotics and probiotics, have been suggested as an antibiotics alternative to improve the performance and gut health of poultry. The current experiment was conducted to study the effects of adding Mannan oligosaccharides (MOS) as a potential replacement for an antibiotic on productive performance, nutrient digestibility, some blood parameters, and caecal microbiota of broiler chickens. For conducting the current research experiment, a total of 180 one–day old Ross broiler chicks were randomly divided into one control group fed a basal diet and four dietary treatments with six replicates for each treatment. The treatment groups were fed a basal diet supplemented with different levels of MOS 0.5, 1, and 2 g/Kg diet and Lincomycin 4.4 mg/Kg diet during 35 days of the feeding trial. With exception of the growing period, the group fed a basal diet supplemented with 2 g MOS/Kg feed had considerably higher body weight and weight gain, while having the lowest feed consumption and best feed conversion ratio compared to the other treatment groups, during all experimental periods. Moreover, dietary supplementation of MOS resulted in a significant decrease in the counts of caecal E. coli and Enterococcus, while Lactobacillus and Yeast bacteria counts were significantly higher, compared to non-supplemented groups. Broiler chicks having a 2 g MOS/kg diet recorded lower values of cholesterol, low-density lipoprotein (LDL), urea, and liver enzymes, including Aspartate transaminase (AST) and Alanine aminotransferase (ALT), while they recorded significantly higher high-density lipoprotein (HDL), compared to the other experimental groups. Group treated with MOS 2 g/Kg feed improved digestibility of crude protein, ether extract, crude fiber, nitrogen-free extract, and organic matter, compared to the control group, Additionally, MOS supplementation also increased the intestinal villi length, width, and crypt depth and decreased intestinal inflammation, compared to the control group. In conclusion, supplementation of MOS at 2 g/kg diet improved growth performance, digestibility, and blood parameters without having adverse effects on the intestine of broiler chickens, comparable to the Lincomycin.
