This study aimed to evaluate in vitro and in vivo dietary supplementation with different levels of natural or nano-zeolite forms on rumen fermentation patterns and nutrient digestibility. In the in vitro experiment, a basal diet (50% concentrate: 50% forage) was incubated without additives (control) and with natural zeolite (10, 20, 30 g/kg DM) or nano-zeolite (0.2, 0.3, 0.4, 0.5, 1.0 g/kg DM) for 24 h to assess their effect on ruminal fermentation, feed degradability, and gas and methane production using a semi-automatic system of in vitro gas production (GP). The most effective doses obtained from the in vitro experiment were evaluated in vivo using 30 Barki goats (26 ± 0.9 SE kg body weight). Goats were allocated into three dietary treatments (n = 10/treatment) as follows: control (basal diet without any supplementations), natural zeolite (20 g/kg DM diet), and nano-zeolite (0.40 g/kg DM diet). The in vitro results revealed that only the nano-zeolite supplementation form quadratically (p= 0.004) increased GP, and the level of 0.5 g/kg DM had the highest GP value compared to the control. Both zeolite forms affected the CH4 production, linear, and quadratic reductions (p < 0.05) in CH4 (mL/g DM), consistent with linear increases in truly degraded organic matter (TDOM) (p = 0.09), and propionate molar proportions (p = 0.007) were observed by nano zeolite treatment, while the natural form of zeolite resulted in a linear CH4 reduction consistent with a linear decrease (p = 0.004) in NH3-N, linear increases in TDOM (p = 0.09), and propionate molar proportions (p = 0.004). Results of the in vivo experiment demonstrated that the nutrient digestibility was similar among all treatments. Nano zeolite enhanced (p < 0.05) the total short-chain fatty acids and butyrate concentrations, while both zeolite forms decreased (p < 0.001) NH3-N compared to the control. These results suggested that both zeolite supplementation forms favorably modified the rumen fermentation in different patterns.

The rationale behind failure of cemented total hip replacement is still far from being well understood in a mechanical and molecular perspective. In the present study, the integrity of the stem–cement interface was investigated through an in vitro experiment monitoring fluid flow along this interface. The results indicated that a good mechanical bonding formed at the stem–cement interface before debonding of this interface was induced by physiological loadings during the in vivo service of the hip prosthesis.

This study investigated whether the mechanism underlying the neurotoxic effects of cadmium chloride (CdCl2) in rats involves p66Shc. This study comprised an initial in vivo experiment followed by an in vitro experiment. For the in vivo experiment, male rats were orally administered saline (vehicle) or CdCl2 (0.05 mg/kg) for 30 days. Thereafter, spatial and retention memory of rats were tested and their hippocampi were used for biochemical and molecular analyses. For the in vitro experiment, control or p66Shc-deficient hippocampal cells were treated with CdCl2 (25 µM) in the presence or absence of SP600125, a c-Jun N-terminal kinase (JNK) inhibitor. Cadmium chloride impaired the spatial learning and retention memory of rats; depleted levels of glutathione and manganese superoxide dismutase; increased reactive oxygen species (ROS), tumor necrosis factor α, and interleukin 6; and induced nuclear factor kappa B activation. Cadmium chloride also decreased the number of pyramidal cells in the CA1 region and induced severe damage to the mitochondria and endoplasmic reticulum of cells in the hippocampi of rats. Moreover, CdCl2 increased the total unphosphorylated p66Shc, phosphorylated (Ser36) p66Shc, phosphorylated JNK, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, cytochrome c, and cleaved caspase-3. A dose–response increase in cell death, ROS, DNA damage, p66Shc, and NADPH oxidase was also observed in cultured hippocampal cells treated with CdCl2. Of note, all of these biochemical changes were attenuated by silencing p66Shc or inhibiting JNK with SP600125. In conclusion, CdCl2 induces hippocampal ROS generation and apoptosis by promoting the JNK-mediated activation of p66Shc.

Fluoride-releasing composites lose fluoride very slowly over time. An interesting question is the possible change in mechanical properties related to the F release. If this happens, it might be expected that the mechanical properties of the outer surface of a fluoridating composite are affected first. The purpose of this study was to investigate in vivo and in vitro the changes in surface microhardness and surface structure of three fluoride-releasing composites and a non-F-containing control after 28 days. In the in vitro experiment, the composites were stored in tap water at 37°C. The results show that all composites stored in water were significantly softened after 28 days. In vivo, however, a very different picture emerged: The surface microhardness of the fluoride-releasing composites did not change significantly. In vitro, the data indicate that the amount of softening of the fluoridating composites is related to the amount of fluoride released. No relation was found between the amount of F released in one month in vitro and the microhardness changes in vivo. SEM micrographs of fluoridating composites do not reflect the microhardness changes mentioned.

Abstract Background: Rumen bacterial community is mainly affected by the type of diet consumed by the host animals. High concentrate diet increases the abundance of lactic acid producers and utilizers due to high level of non-structural carbohydrates thus reducing the number of fiber-degrading bacteria because of drastic decrease in pH. Dietary buffers are essential in regulating rumen pH through the compounds responsible in resisting drastic decrease in pH once cattle were fed with high-concentrate diet. However, no study has evaluated the effects of buffering capacity and efficiency in alleviating chronic acidosis in rumen. Ruminal metataxonomic and fermentation characteristics analyses were conducted to evaluate the effect of different buffering capacities on in vitro and in vivo experiments in high-concentrate fed Hanwoo steers. Results: Results revealed that BC0.9% and BC0.5% had similar and significant effect (P < 0.05) on in vitro ruminal fermentation at 3 to 24 h incubation. Both BC0.9% and BC0.5% had significantly highest (P < 0.05) buffering capacity, pH, and ammonia-nitrogen (NH3-N) than BC0.3% and CON at 24 h of incubation. Individual and total volatile fatty acids (VFA) were significantly lowest in CON. Increasing buffering capacity concentration showed linear effect on pH at 6 to 24 h while total gas and NH3-N at 3 and 12 h. Phylum Bacteroidetes dominated all treatments but a higher abundance of Firmicutes in BC0.5% than others. Ruminoccocus bromii and Succiniclasticum ruminis were dominant in BC0.5% and Bacteroides massiliensis in BC0.3%. The normalized data of relative abundance of observed OTUs’ representative families have grouped the CON with BC0.3% in the same cluster, whereas BC0.5% and BC0.9% were clustered separately which indicates the effect of varying buffering capacity of buffer agents. Principal coordinate analysis (PCoA) on unweighted UniFrac distances revealed close similarity of bacterial community structures within and between treatments and control, in which BC0.9% and BC0.3% groups showed dispersed community distribution. Conclusion: Our findings showed that increasing buffering capacity enhances rumen fermentation parameters and affects rumen microbiome by altering bacterial community through distinct structure between high and low buffering capacity, thus an important factor contributed to the prevention of ruminal acidosis during a high-concentrate diet.

ABSTRACT Controlling post-harvest papaya diseases without using agrochemicals is a challenge for producers. This study aimed at evaluating the effect of clove essential oil, biological fungicide (Trichodermil®), resistance inducer (Cob Sistem®) and chemical fungicide (Imazacure®) on the in vitro control of phytopathogenic fungi isolates from papaya as well as on the post-harvest quality of Tainung 1 papaya. The in vitro experiment was conducted in a complete randomized design, with five fungal species x five treatments and five replications. The in vivo experiment was conducted in a complete randomized design, with five treatments x five storage times, five replications and three fruits per replication. The fruits were stored under refrigeration at 10 ± 2 ºC and 90 ± 5 % of relative humidity and evaluated at 0, 7, 14, 21 and 28 days of storage, plus two shelf life days at 25 ± 2 ºC, to simulate marketing conditions. The inhibition of mycelial growth was evaluated in the in vitro experiment, while the diseases occurrence and post-harvest quality of the fruits were evaluated in the in vivo experiment. The clove essential oil and Trichodermil® were as efficient as Imazacure® in inhibiting the mycelial growth of Alternaria sp., Colletotrichum gloeosporioides and Rhizopus sp. The treatments with clove essential oil, Trichodermil® and Imazacure® were similar in controlling the pathogens up to 21 days of storage. The treatments had no effect on the fruits soluble solid contents.

We investigated the effect of diets containing organic zinc and a mixture of medicinal herbs on ruminal microbial fermentation and histopathology in lambs. Twenty-eight lambs were divided into four groups: unsupplemented animals (Control), animals supplemented with organic zinc (Zn, 70 mg Zn/kg diet), animals supplemented with a mixture of dry medicinal herbs (Herbs, 100 g dry matter (DM)/d) and animals supplemented with both zinc and herbs (Zn+Herbs). Each lamb was fed a basal diet composed of meadow hay (700 g DM/d) and barley (300 g DM/d). The herbs Fumaria officinalis L. (FO), Malva sylvestris L. (MS), Artemisia absinthium L. (AA) and Matricaria chamomilla L. (MC) were mixed in equal proportions. The lambs were slaughtered after 70 d. The ruminal contents were used to determine the parameters of fermentation in vitro and in vivo and to quantify the microbes by molecular and microscopic methods. Samples of fresh ruminal tissue were used for histopathological evaluation. Quantitative analyses of the bioactive compounds in FO, MS, AA, and MC identified 3.961, 0.654, 6.482, and 12.084 g/kg DM phenolic acids and 12.211, 6.479, 0.349, and 2.442 g/kg DM flavonoids, respectively. The alkaloid content in FO was 6.015 g/kg DM. The diets affected the levels of total gas, methane and n-butyrate in vitro (P &lt; 0.046, &lt; 0.001, and &lt; 0.001, respectively). Relative quantification by real-time PCR indicated a lower total ruminal bacterial population in the lambs in the Zn and Zn+Herbs groups than the Control group (P &lt; 0.05). The relative abundances of Ruminococcus albus, R. flavefaciens, Streptococcus bovis, and Butyrivibrio proteoclasticus shifted in the Zn group. Morphological observation found a focally mixed infiltration of inflammatory cells in the lamina propria of the rumen in the Zn+Herbs group. The effect of the organic zinc and the herbal mixture on the parameters of ruminal fermentation in vitro was not confirmed in vivo, perhaps because the ruminal microbiota of the lambs adapted to the zinc-supplemented diets. Long-term supplementation of a diet combining zinc and medicinal herbs, however, may negatively affect the health of the ruminal epithelium of lambs.

This study included the preparation of the mixture of alcoholic extracts of Peganum harmala seeds and Pericarp of Punica granutum at concentration (1+40), (1.5+45), (2+50) mgml. To study the influence of the mixture of alcoholic extracts of P. harmala and P. granutum on viability of the protoscolices of Echinococcus granulosus In vitro (In tubes), and In vivo (In albaino white mice injected intraperitoneal with protoscolices). In vitro experiment revealed complete inactivation of protoscolices (death) with concentration (2+ 50) mg/ ml after 30 minute. In vivo a significant reduction in weight of liver and spleen occurred in treated groups in comparison to control (untreated) infected group.
