Nucleotides are essential for the development of the gastrointestinal tract and immune function, but their intake with milk by piglets could be insufficient. The effect of nucleotides on growth and health was tested on 98 piglets divided into two groups: NU, orally administrated with 4 mL of a nucleotide-based product (SwineMOD®) at 10, 15, 18, 21, 27 days, or not (CO). Blood and feces were sampled at weaning (26 d, T1), and at 38 d (T2). Per each group and time-point, eight piglets were slaughtered and jejunal Peyer’s patches (JPPs) were collected. NU increased hemoglobin content and hematocrit, but not growth. At weaning, the NU fecal microbiota was characterized by the abundance of Campylobacteraceae, more typical of the growing phase, compared to CO, with a greater abundance of Streptococcaceae. For the blood transcriptome, an initial greater inflammatory activation was seen in CO, while at T2, NU enriched gene sets related to erythropoiesis. The activation of gene groups ranging from epigenetic response to transcriptional regulation evidenced an intense proliferative activity in NU JPPs. NU supplementation did not influence the growth performance of piglets but could have expressed a positive effect on pig microbiota anticipating its maturation at weaning. This immunostimulant activity in the JPPs could moderate the inflammation in the immediate pre-weaning.

Recent evidences suggest that gut microbiota plays an important role in regulating physiological and metabolic activities of pregnant sows, and β-carotene has a potentially positive effect on reproduction, but the impact of β-carotene on gut microbiota in pregnant sows remains unknown. This study aimed to explore the effect and mechanisms of β-carotene on the reproductive performance of sows from the aspect of gut microbiota. A total of 48 hybrid pregnant sows (Landrace × Yorkshire) with similar parity were randomly allocated into three groups (n = 16) and fed with a basal diet or a diet containing 30 or 90 mg/kg of β-carotene from day 90 of gestation until parturition. Dietary supplementation of 30 or 90 mg/kg β-carotene increased the number of live birth to 11.82 ± 1.54 and 12.29 ± 2.09, respectively, while the control group was 11.00 ± 1.41 (P = 0.201). Moreover, β-carotene increased significantly the serum nitric oxide (NO) level and glutathione peroxidase (GSH-Px) activity (P < 0.05). Characterization of fecal microbiota revealed that 90 mg/kg β-carotene increased the diversity of the gut flora (P < 0.05). In particular, β-carotene decreased the relative abundance of Firmicutes including Lachnospiraceae AC2044 group, Lachnospiraceae NK4B4 group and Ruminococcaceae UCG-008, but enriched Proteobacteria including Bilophila and Sutterella, and Actinobacteria including Corynebacterium and Corynebacterium 1 which are related to NO synthesis. These data demonstrated that dietary supplementation of β-carotene may increase antioxidant enzyme activity and NO, an important vasodilator to promote the neonatal blood circulation, through regulating gut microbiota in sows.

The intestinal microbiota plays several important roles in pig health and growth. The aim of the current study was to characterize the changes in the fecal microbiota diversity and composition of weaned piglets following an oral challenge with an ETEC: F4 strain and/or a treatment with colistin sulfate (CS). Twenty-eight piglets were used in this experiment and were divided into four groups: challenged untreated, challenged treated, unchallenged treated, and unchallenged untreated. Rectal swab samples were collected at five sampling times throughout the study. Total genomic DNA was used to assess the fecal microbiota diversity and composition using the V4 region of the 16S rRNA gene. The relative abundance, the composition, and the community structure of piglet fecal microbiota was highly affected by the ETEC: F4 challenge throughout the experiment, while the oral treatment with CS, a narrow spectrum antibiotic, resulted in a significant decrease of E. coli/Shigella populations during the treatment period only. This study was the first to identify some gut microbiota subgroups (e.g., Streptococcus, Lachnospiraceae) that are associated with healthy piglets as compared to ETEC: F4 challenged animals. These key findings might contribute to the development of alternative strategies to reduce the use of antimicrobials in the control of post-weaning diarrhea in pigs.

AbstractThe aim of the study was to assess the effect of fermented dried soybean (FSBM) and/or fermented rapeseed meal (FRSM) in diets for weaned piglets on production results, nutrient digestibility, gastrointestinal tract histology, and the composition of the gut microbiota. Piglets in the control group received standard diets with soybean meal. Animals in all experimental groups received diets in which a portion of the soybean meal was replaced: in group FR—8% FRSM; in group FR/FS—6% FRSM and 2% FSBM; in group FS/FR—2% FRSM and 6% FSBM and in group FS—8% FSBM. The use of 8% FRSM or 6% FRSM and 2% FSBM in the piglet diets had a positive effect on average daily gains. Piglets from the FR and FR/FS groups had the highest feed conversion rate. Group FS/FR and FS piglets had significantly lower mortality and lower incidence of diarrhoea. Piglets fed a diet with the fermented components, in particular with 8% FRSM or 6% FRSM and 2% FSBM, exhibited a positive effect on the microbiological composition and histology of intestines, which resulted in improved nutrient digestibility coefficients (ATTD and AID).

Abstract. In this study records of 58 925 litters of Austrian Large White and 17 846 litters of Austrian Landrace pigs were analysed. Regression models were used to determine the effects of litter, dam and sire inbreeding on total number of born, born alive and weaned piglets in Large White and Landrace. In both populations, litter and dam inbreeding showed a negative effect on all traits. Sire inbreeding had no effect in Large White, whereas a significant positive effect was observed in Landrace. On average, inbred sires with an inbreeding coefficient of 10 % had 0.45 more piglets born total and 0.43 more piglets born alive in comparison to non-inbred sires. In a further analysis the total inbreeding coefficients of the animals were divided into two parts: »new« and »old« inbreeding. »New« inbreeding was defined as the period of the first five generations. It was shown that the observed inbreeding effects were not only caused by recent inbreeding. Reproductive performance was also affected by »old« inbreeding. Finally partial inbreeding coefficients of four important ancestors in each population were calculated to investigate if inbreeding effects are similar among these ancestors. The results revealed a varation of inbreeding effects among the four ancestors. Alleles contibuting to inbreeding depression were descendent from specific ancestors.

Chronic constipation is a common gastrointestinal dysfunction, but its aetiology and pathogenesis are still unclear. Interestingly, the compositions of the gut microbiota in constipation patients and healthy controls are different. Various studies reported the different gut microbiota alterations in constipation patients, but most studies indicated that constipation patients showed the decreased beneficial bacteria and the reduced species richness of gut bacteria. Besides, the alterations in the gut microbiota may lead to constipation and constipation-related symptoms and the regulation of gut microbiota has a positive effect on gut functional diseases such as constipation. Microbial treatment methods, such as probiotics, prebiotics, synbiotics, and fecal microbiota transplantation, can be used to regulate gut microbiota. Increasing evidences have suggested that Chinese medicine (CM) has a good therapeutic effect on chronic constipation. Chinese medicine is well known for its multitarget and multimode effects on diseases as well as less side effects. In previous studies, after the treatment of constipation with CM, the gut microbiota was restored, indicating that the gut microbiota might be the target or important way for CM to exert its efficacy. In this review, we summarized the effects of microbial treatment and CM on the gut microbiota of constipation patients and discussed the relationship between CM and gut microbiota.

Abstract Valproic acid is an anti-epileptic drug with histone deacetylase inhibitor activity currently in clinical trial for the treatment of leukemia. In order to understand the effects of this agent in myeloid differentiation and leukemia therapy we determined the changes in gene expression that occurred when the myeloid cell line ML-1 was treated with valproic acid (VPA), all-trans retinoic acid (ATRA) or the combination for 24 or 96 hours. Each condition was compared to no drug treatment at each time point, as full biological triplicates. RNA samples were labeled and hybridized to Affymetrix HGU133A genechips. The data was processed using the Affymetrix MAS5.0 calculated intensity signals or was pre-processed using three different algorithms to normalize data: GC-RMA (GeneChip-Robust Microarray Analysis), RMA (Robust Microarray Analysis) or MBEI (Model-Based Expression Index), using the original Affymetrix CEL files. From the resulting normalized data sets, we then identified genes that were up or down regulated at least two-fold by the combination VPA + ATRA treatment when compared to the no drug condition or either drug alone. If any of these genes were affected by either ATRA or VPA, we selected only those with an additional change of 50% or more with the combination therapy. There were striking differences in the gene sets obtained from each normalization algorithm with respect to both list size and content. GC-RMA identified 100 genes significantly differentially expressed according to the above criteria at the 24 hour time point, MBEI found 19 and MAS5.0 found 39. Using the RMA algorithm, 33 genes were determined to be significantly affected by the combination treatment and all of these were also found with GC-RMA suggesting that GC-RMA has a greater sensitivity than RMA. This may be because GC-RMA, a refinement of RMA, takes into account mismatched probe set signals and probe set sequence content. Only 12 genes were common to all lists, including endothelin receptor B, myosin binding protein H, IL8, and G0S2. We are currently using real-time PCR to validate the extent of differential regulation of the commonly identified genes in reponse to VPA+ATRA. This will be contrasted with the levels of regulation of gene sets found by only one or two algorithms in order to determine the sensitivity and specificity of these methods. These data indicate the potential power of new bioinformatic tools for microarray analysis and the need for systematic validation to ensure biological relevance.

The purpose of the present study was to assess the effect of kaolin feeding on health status, body weight gain (BWG), course of diarrhoeal infections caused by enterotoxigenic strains of <i>Escherichia coli</i> (ETEC) and the level of mycobacterial contamination in weaned piglets. The testing was performed in two experiments involving 40 weaned piglets at the age of 28 days. In the infection-free experiment, piglets were fed a diet without (C0) or with 1% content of kaolin (K0) for 20 days. Subsequently, all of them were fed the same diet without kaolin supplementation for 39 days. Identical diets were fed during the infection experiment, and moreover, both groups (CI and KI) were orally infected with ETEC (O141:F18ac, STa+) on Day 1 of experiment. The short-term feeding of kaolin to weaned piglets had a significant positive effect on their BWG. During the period of feeding the kaolin-containing diets, BWG in C0 and K0 were 0.20 and 0.29 kg, respectively (<i>P</i> < 0.05), and in CI and KI 0.13 and 0.19 kg, respectively (<i>P</i> < 0.05). There was no evidence of side effects to their health, neither was there any change in biochemical and haematological profiles. In the infection experiment, a protective effect of kaolin on the course of ETEC infection was evident. Colonization and shedding of ETEC by piglets fed the kaolin diet were milder and had a shorter duration in comparison with control piglets. The culture examination of pure kaolin and kaolin containing diets for mycobacteria were negative. Potentially pathogenic mycobacteria occurring in the environment were isolated from faeces and tissues of pigs. According to these results, supplementation of diets with 1% kaolin to prevent diarrhoea in piglets and to support their growth in the critical post-weaning period could be recommended.

Abstract Background: The inclusion of high-quality proteins and the protein restriction are commonly used in swine production. Our study was conducted to evaluate the effects of hydrolyzed wheat protein (HWP), fermented soybean meal (FSBM), and enzyme-treated soybean meal (ESBM) in low-protein diets on antioxidant capacity, immunity, fecal microbiota and metabolites of weaned piglets.Methods: A total of 144 weaned piglets were randomly assigned to 3 dietary treatments with 6 replications of 8 piglets per pen. The experiment was divided into phase 1 ( days 0-14) and phase 2 (days 15-28). The dietary treatments contained 15.90% HWP, 15.80% FSBM, and 15.10% ESBM in phase 1, and 7.90% HWP, 7.80% FSBM, and 7.50% ESBM in phase 2, respectively.Results: The ADG of piglets in the ESBM group was higher (P < 0.05) than HWP and FSBM during days 1-28. Compared with HWP and FSBM, piglets in ESBM had higher (P < 0.05) serum levels of FRAP and SOD on days 14, as well as higher (P < 0.05) serum FRAP level on days 28. Piglets in ESBM had lower (P < 0.05) serum levels of DAO and IL-1β than HWP on day 28. ESBM significantly increased the abundance of Bacteroidetes, Oscillospiraceae and Christensenellaceae and decreased the Clostridiaceae abundance in the feces compared with HWP and FSBM. The PICRUSt analysis revealed that the number of gene tags involved in valine, leucine and isoleucine degradation and lysine degradation in ESBM were lower (P < 0.05) than HWP and FSBM. Piglets in ESBM had higher (P < 0.05) fecal butyrate content compared with FSBM, and ESBM tended to decrease (P = 0.076) fecal cadaverine level.Conclusions: Overall, ESBM had advantages over HWP and FSBM in improving antioxidant capacity, immunity, and fecal microbiota and metabolites of weaned piglets.

The aim of the study was to evaluate the effect of additional selenium injection after weaning on the selenium (Se) status of piglets and to find whether the selected dose would be appropriate with respect to the level of oxidative stress. Another goal was to compare the efficacy and safety of sodium selenite and selenopyran as selenium sources for parenteral administration to piglets. Altogether 30 piglets were divided equally into three groups. Piglets in group 1 were injected i.m. with sodium selenite, piglets in group 2 were injected with selenopyran. The dose was 0.42 mg Se/kg body weight for both groups. Piglets in group 3 were given only saline. As expected, the study revealed low Se serum concentrations in weaned piglets. The injection of sodium selenite increased Se serum concentrations but did not have a positive effect on the peroxidase activities. Administration of selenopyran did not influence Se concentrations and gluthation peroxidase activities. The selected dose did not have a significant impact on the level of the oxidative stress. The piglets receiving Se only from the feed achieved comparable gluthation peroxidase activities during the trial. It seems that despite initially low Se concentrations, the physiological requirements for gluthation peroxidase synthesis were met with the feed consumption as the only Se source. The results of the study are important because until now it was unclear whether the selected dose would have negative effects on the organism with respect to the induction of oxidative stress in piglets.
