This study examined the effects of various types, quality, and levels of dietary oils on laying performance and the expression patterns of antioxidant-related genes in Hy-line brown laying hens. A total of 720 40-week-old Hy-line brown laying hens were fed the same corn-soybean basal meals but containing 0.5 or 1.5% normal or oxidized soybean oil or lard, a total of 8 treatments. The results showed that laying rate (LR) and fatty acids of raw yolk were significantly correlated dietary type of oil (p < 0.05). With the increasing concentration of normal oil, it significantly increased LR and decreased feed conversion ratio (FCR, feed/egg) and albumen height of laying hens. The oxidized oil significant decreased the production performance of laying hens; and adding 1.5% of oxidized lard into feeds could destroy the integrity of yolk spheres of cooked yolk. mRNA expression of liver antioxidant-related genes increased when dietary oxidized oils were added into feeds. By comparing different qualities oil effect on antioxidant-related genes, the expression of Glutathione S-Transferase Theta 1 (GSTT1), Glutathione S-Transferase Alpha 3 (GSTA3), Glutathione S-Transferase Omega 2 (GSTO2), and Superoxide Dismutase 2 (SOD2) were increased when dietary oils were oxidized, in which change of the GSTO2 expression was the most with 1.5% of oxidized soybean oil. In conclusion, the ideal type of oil for Hy-line brown layer hens is soybean comparing with lard in a corn-soybean diet, avoiding using of oxidized oil.

Application of different medicinal mushrooms intended to enhance production performance and health status has created an importance demand in poultry production. One goal of using medicinal mushrooms is to get rid of antibiotics in poultry feed without affecting the optimum performance. Increasing concerns about this issue have led to more attention on antibiotic substitutes and a significant demand for them for organic egg production. Thus, supplementation with medicinal mushrooms is a new concept for research in layer production, however, there is still a great deal of confusion about inclusion levels and the mode of action of medicinal mushrooms on production performance and health status in laying hens. Taking this into account, this review outlines the experimental uses of medicinal fungi on the growth performance, laying performance, egg quality, and health status of layer birds based on previous findings to date. Finally, we highlight that supplementation with medicinal fungi can play a role on the immunity, health, and production performance in laying hens.

The effects of different dietary lipids on the fatty acid profiles of eggs produced by 20 and 54 wk old Dekalb laying hens were investigated. Laying hens were subjected to three defined treatments according to the source of lipid added to their diets: soybean oil, beef tallow, and a control diet (without the addition of oil). The experimental design was in a 3x2 factorial arrangement (three treatments and two different ages). The fatty acid composition of the yolks in the eggs produced by the laying hens was analyzed. The eggs produced by laying hens on the soybean oil diet had a large amount of polyunsaturated fatty acids (PUFA) omega-6 (n-6) and omega-3 (n-3) in their yolks (23.55, 2.30% respectively), whereas egg yolks from hens who were given beef tallow had higher percentages of monounsaturated fatty acids (47.53%) compared to soybean oil (47.53%) and the control diet (38.72%). The percentages of trans fats present in the egg yolks in all treatments were considered very low (0.91; 0,11; 0.05%). Young layers are more efficient at depositing n-3 fatty acids (1.40%), specially C22:6 (0.76%) with the best ratio n6:n-3 (13.97) compared to old layers (1.35; 0.72; 14.81% respectively). Based on these results, it was concluded that the amount of fatty acids present in the egg yolks can be modified by the sources of lipids included in the diet and that independent of the sources of lipid in the diet and the age of the chicken, egg yolks have insignificant amounts of trans fatty acids.

To evaluate dietary soybean oil supplementation on production performance, egg quality, and keel bone health in laying hens. Two hundred and four laying hens at 20 weeks of age (WOA) were distributed into 12 cages containing 17 birds each. Birds were either fed a commercial diet (control group, CON) or a diet supplemented with 3% of soybean oil (SO group). Experiments lasted 17 weeks. Body weight, daily feed intake, production performance and egg quality were measured at 25, 29, 33, and 37 WOA. Birds were subsequently assessed for keel bone status by palpation, and keel was excised to measure bone length, microstructure, bone mineral density (BMD), elements contents, and the expression of osteoprotegerin (OPG), receptor activator of nuclear factor kappa-B ligand (RANKL), collagen type II alpha 1 (COL2α1), periostin (POSTN), and sclerostin (SOST). The results showed that dietary SO supplementation did not affect production performance and egg quality (P &gt; 0.05), but improved body weight of hens at 29 and 37 WOA (P &lt; 0.05), and decreased daily feed intake at 33 and 37 WOA (P &lt; 0.05). Incidence of keel bone damage (especially fracture) was higher in hens of SO group. Keel bone length in birds of SO group was significantly decreased compared to CON (P &lt; 0.05). Keel bone of supplemented hens showed increased trabecular separation at 29 WOA and higher levels of V, Mn, Fe, Se, and Ba at 33 WOA (P &lt; 0.05). Moreover, decreased BMD, trabecular number and thickness were observed in keel bone of laying hens receiving supplementation at 29 and 37 WOA (P &lt; 0.05); decreased levels of Li, Ca, Hg, and TI at 33 WOA and trabecular thickness at 37 WOA (P &lt; 0.05) were also identified. mRNA levels of SOST and RANKL and the ratio of RANKL/OPG mRNA levels were increased in birds fed a SO-supplemented diet (P &lt; 0.05); COL2α1, OPG, and POSTN were downregulated at all sampling points (P &lt; 0.05). Taken together, results indicate that feeding laying hens a diet supplemented with soybean oil can decrease daily feed intake and impair keel bone health but not influence production performance and egg quality.

This study investigated the effects of plastic-net housing system (NRS) and floor-litter housing system (LRS) on the production performance, serum parameters and intestinal morphology of Shendan laying hens. A total of 1200 30-week-old hens were randomly allocated to the NRS and LRS groups, each of which included five replicates with 120 chickens in each replicate. The experiment was conducted from 32 to 40 weeks of age. Indoor airborne parameters were measured every 2 weeks, and indoor ground contamination was measured monthly. The laying rate and mortality of hens were recorded daily, and egg quality traits and serum parameters were measured every 2 weeks. At 40 weeks of age, four birds per replicate from each experimental group were selected for intestinal morphological observation. The results showed that the airborne bacteria number in the LRS was significantly higher than that in the NRS (p < 0.05) for most of the experimental period (except at 32 and 38 weeks of age), and the bacterial numbers on the surfaces of the floor and floor eggs in the LRS were approximately 10 times higher than those in the NRS (p < 0.05). Compared with the LRS, the NRS improved the laying rate (p < 0.05), reduced serum malondialdehyde (MDA) (p < 0.05) and corticosterone (CORT) concentrations and increased serum glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities, indicating favourable effects on antioxidative status. The NRS was significantly associated with an increased villus height (VH), villus height to crypt depth ratio (VCR) in the small intestine (p < 0.05) and increased VCR in the caecum (p < 0.05). Overall, the lower rate of bacterial contamination in the NRS than in the LRS indicated better environmental hygiene. The NRS enhanced the laying performance and antioxidant capacity of hens and was superior to the LRS in improving intestinal health. The current findings support the advantages of the NRS for the health and welfare of Shendan chickens during the peak laying period.

Abstract Background: The cuticle formed in the uterus is the outermost layer as the first defense line of eggshell against microbial invasions in most avian species, and analyzing its genetic regulation and influencing factors are of great importance to egg biosecurity in poultry production. The current study compared the uterine transcriptome and proteome of laying hens producing eggs with good and poor cuticle deposition (GC and PC, the top and tail of the cuticle deposition distribution), and identified several genes involved with eggshell cuticle deposition.Results: Overall, transcriptomic analysis identified 53 differentially expressed genes (DEGs) between PC vs GC group hens, among which 25 were up-regulated and 28 were down-regulated. No differences were found in the uterine proteome. Several up-regulated DEGs of PC vs GC hen uterus, including PTGDS, PLCG2, ADM and PRLR related to uterine functions and reproductive hormones, were validated by qPCR analysis. Phenotypic measurements for verification between GC and PC hens showed GC hens had longer laying interval between two consecutive ovipositions (25.64 ± 1.23 vs 24.94 ± 1.12 h), thicker eggshell thickness (352.01 ± 23.04 vs 316.20 ± 30.58 μm) and more laying days (20.67 ± 2.58 vs 18.48 ± 2.46) within 27 laying days (P < 0.05). Apart from eggshell traits, other egg quality traits didn’t differ. The results above suggested that the expression patterns of PTGDS, PLCG2, ADM and PRLR genes in the uterus regulated the muscular activity rhythm and secretion function, which ensured the eggshell and cuticle formation duration, and lead to the good eggshell and cuticle quality of the GC hens.Conclusions: The present study demonstrated eggshell and cuticle secretion duration in the uterus is one of the major factors affecting eggshell cuticle deposition in laying hens. PTGDS, PLCG2, ADM and PRLR genes were discovered and might play crucial roles in cuticle deposition by regulating the uterine muscular activities and secretion function. Moreover, the cuticle is associated with the eggshell quality and laying performance, implying the importance of selection to the cuticle trait in laying hens. The findings in present study provide new insights into the genetic regulation of cuticle deposition in laying hens and establish a foundation for further investigations.

One of the most important factors that determine feed utilization by chickens is the feed form. Although it is generally believed that pellet diets have a positive effect on chicken growth, there are some studies that have indicated no difference between pellet and mash on chickens performance. This study was conducted to assess the effects of feed form on production performance, egg quality, nutrient metabolism and intestinal morphology in two breed laying hens. Two hundred and sixteen 25-week-old Hy-Line brown (n = 108) and Hy-Line grey (n = 108) hens were selected. Each breed was randomly allocated into two treatments with 6 replications (9 birds in each replication), which were fed mash and pellet diets, respectively. Production performances were recorded daily and egg quality traits were measured every two weeks. At 42 weeks of age, one bird per replication from each experimental group was selected for metabolism determination and intestine morphology observation. Compared with mash diets, pellet diets improved laying rate (p < 0.05), ADFI (average daily feed intake, p < 0.05), egg weight, shell strength, yolk proportion and Haugh unit (p < 0.05) in both breeds and reduced the FCR (feed conversion ratio, p < 0.05) in Hy-Line grey. The apparent digestibility of DM% (dry matter) and CP% (crude protein) were significantly higher (p < 0.05) in both breed laying hens fed pellet than those fed mash. The apparent digestibility of P% (phosphorus) and Ca% (calcium) was higher in Hy-Line grey fed pellet and was higher in Hy-Line brown fed mash. Compared to mash diets, pellet diets increased the VH (villus height), CD (crypt depth) and VCR (ratio of villus height to crypt depth) of the small intestine of Hy-Line grey, and increased the VH and CD of duodenum and ileum of Hy-Line brown. Overall, pellet diets improved production performance and nutrition metabolism through positive changes in the laying rate, feed intake, egg albumen quality and apparent digestibility of laying hens. The current findings provided support for the advantages of feeding pellets during the peak egg laying period for the two popular laying hen strains, Hy-Line brown and Hy-Line grey.
