The aim of the present study was to evaluate the effects of adding dietary rumen-protected microencapsulated supplements into the ruminal fluid on the milk fat compositions of dairy cows. These supplements comprised linseed oil, vitamin E, rosemary extract, and hydrogenated palm oil (MO; Microtinic® Omega, Vetagro S.p.A, Reggio Emilia, Italy). For in vitro ruminal fermentation, Holstein–Friesian dairy cows each equipped with a rumen cannula were used to collect ruminal fluid. Different amounts (0%, 1%, 2%, 3%, 4%, and 5%) of MO were added to the diets to collect ruminal fluids. For the in vivo study, 36 Holstein–Friesian dairy cows grouped by milk yield (32.1 ± 6.05 kg/d/head), days in milk (124 ± 84 d), and parity (2 ± 1.35) were randomly and evenly assigned to 0.7% linseed oil (LO; as dry matter (DM) basis) and 2% MO (as DM basis) groups. These two groups were fed only a basal diet (total mixed ration (TMR), silage, and concentrate for 4 weeks) (period 1). They were then fed with the basal diet supplemented with oil (0.7 LO and 2% MO of DM) for 4 weeks (period 2). In the in vitro experiment, the total gas production was found to be numerically decreased in the group supplemented with 3% MO at 48 h post in vitro fermentation. A reduction of total gas production (at 48 h) and increase in ammonia concentration (24 h) were also observed in the group supplemented with 4% to 5% MO (p < 0.05). There were no differences in the in vitro fermentation results, including pH, volatile fatty acids, or CH4 among groups supplemented with 0%, 1%, and 2% MO. The results of the in vitro study suggest that 2% MO is an optimal dosage of MO supplementation in cows’ diets. In the in vivo experiment, the MO supplement more significantly (p < 0.01) increased the yield of total w3 fatty acids than LO (9.24 vs. 17.77 mg/100 g milk). As a result, the ratio of total omega-6 to omega-3 fatty acids was decreased (p < 0.001) in the MO group compared to that in the LO group (6.99 vs. 3.48). However, the milk yield and other milk compositions, except for milk urea nitrogen, were similar between the two groups (p > 0.05). Collectively, these results suggest that the dietary supplementation of 2% MO is beneficial for increasing omega-3 fatty acids without any negative effects on the milk yield of dairy cows.

This study quantifies the effects of extruded linseed and soybean (ELS) dietary supplementation on milk yield, composition, and fatty acid profiles, as well as first-service conception rate in Holstein dairy cows. Seventy-eight open Holstein dairy cows were divided into two groups: (1) a control, which received a basal diet; and (2) a test group, which received a basal diet supplemented with the ELS (650 g/kg of extruded linseed and 150 g/kg of extruded soybean) at a rate of 100 g/kg. In the ELS group, milk yield per day and solid not fat (SNF) yield increased by 3.26% and 0.88%, respectively, in relation to the control. Percentage milk fat decreased significantly by 1.4% in the ELS group when compared with the control. The ELS supplement resulted in a decrease in saturated fatty acids (SFAs) and an increase in monounsaturated (MUFAs) and polyunsaturated fatty acids (PUFAs) in milk. In conclusion, the supplementation of dairy cow feed with 100 g/kg of ELS increases milk yield and milk unsaturated fatty acids (especially MUFAs and PUFAs). ELS supplementation also causes a decrease in percentage fat and SFA levels but does not affect the first-service conception rate or the incidence rate of mastitis.

There has recently been increased interest in the potential health effects of omega-3 polyunsaturated fatty acids on the immune system. Paracoccidioidomycosis is the most important endemic mycosis in Latin America. Macrophages have a fundamental role and act as first line of organism defense. The purpose of this study was to analyze the effect of n-3 fatty acids on the production of PGE2and NO by mice infected with Pb18 and fed a diet enriched with LNA for 8 weeks. To study the effect of omega-3 fatty acids on macrophage activity during experimental paracoccidioidomycosis, mice were infected with Pb18 and fed a diet supplemented with LNA. PGE2in the serum of animals was analyzed and NO in the supernatants of macrophages cultured and challengedin vitrowith Pb18 was measured. Omega-3 fatty acids seemed to decrease the production of PGE2in vivoin the infected group fed an LNA-supplemented diet during the 4th and 8th weeks of the experiment. At the same time, we observed an increase in synthesis of NO by peritoneal macrophages in this group. Omega-3 fatty acids thus appear to have an immunomodulatory effect in paracoccidioidomycosis.

Since omega–3 polyunsaturated fatty acids (n-3 PUFAs) can alter ventricular myocyte calcium handling, these fatty acids could adversely affect cardiac contractile function, particularly following myocardial infarction. Therefore, 4 wk after myocardial infarction, dogs were randomly assigned to either placebo (corn oil, 1 g/day, n = 16) or n-3 PUFAs supplement [docosahexaenoic acid (DHA) + eicosapentaenoic acid (EPA) ethyl esters; 1, 2, or 4 g/day; n = 7, 8, and 12, respectively] groups. In vivo, ventricular function was evaluated by echocardiography before and after 3 mo of treatment. At the end of the 3-mo period, hearts were removed and in vitro function was evaluated using right ventricular trabeculae and isolated left ventricular myocytes. The treatment elicited significant ( P < 0.0001) dose-dependent increases (16.4-fold increase with 4 g/day) in left ventricular tissue and red blood cell n-3 PUFA levels (EPA + DHA, placebo, 0.42 ± 0.04; 1 g/day, 3.02 ± 0.23; 2 g/day, 3.63 ± 0.17; and 4 g/day, 6.97 ± 0.33%). Regardless of the dose, n-3 PUFA treatment did not alter ventricular function in the intact animal (e.g., 4 g/day, fractional shortening: pre, 42.9 ± 1.6 vs. post, 40.1 ± 1.7%; placebo: pre, 39.2 ± 1.3 vs. post, 38.4 ± 1.6%). The developed force per cross-sectional area, changes in length- and frequency-dependent behavior in contractile force, and the inotropic response to β-adrenoceptor activation were also similar for trabeculae obtained from placebo- or n-3 PUFA-treated dogs. Finally, calcium currents and calcium transients were the same in myocytes from n-3 PUFA- and placebo-treated dogs. Thus dietary n-3 PUFAs did not adversely alter either in vitro or in vivo ventricular contractile function in dogs with healed infarctions.

Aim of the present study was to evaluate the effect of feeding dairy cows with extruded flaxseed (EF) on milk quality. After a 7-d adaptation period, 40 Holstein dairy cows in midlactation were divided in two experimental groups, based on parity, milk yield and composition, and days in milk. Animals were fed for 28 d a standard total mixed ration containing or not (control) 1.8 kg/cow/d of a supplement based on extruded flaxseed (EF). Milk yield was recorded daily and individual milk samples were collected weekly for analysis. Milk yield was not affected by treatment and averaged 26.2 kg/d throughout the study. Feeding EF reduced milk fat (3.95 vs. 4.24%; P=0.053) and tended to reduce milk protein (3.32 vs. 3.50%; P=0.104). The milk from cows fed EF contained more (P<0.01) stearic (12.0 vs. 7.8 mg/100 mg of fat), oleic (21.2 vs. 18.0 mg/100 mg of fat), ?-linolenic (0.85 vs. 0.41 mg/100 mg of fat) and vaccenic (1.43 vs. 0.62 mg/100 mg of fat) acids than control and also more c9,t11 CLA (0.91 vs. 0.59 mg/100 mg of fat). Conversely, compared with control, feeding EF reduced milk concentrations of palmitic acid (26.5 vs. 33.4 mg/100 mg of fat; P<0.01). The present results show that feeding EF to dairy cows is an efficient strategy to enrich milk in beneficial fatty acids such as vaccenic acid, CLA and omega-3 fatty acids. Nevertheless, the milk fat reducing effect of flaxseed as well as other sources of polyunsaturated fatty acids should be taken into account.

AbstractDairy cows with ketosis display severe oxidative stress as well as high blood concentrations of non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHB). Cytochrome P4502E1 (CYP2E1) plays an important role in the induction of oxidative stress. The aim of this study was to investigate CYP2E1 expression and activity in the liver of clinically ketotic cows (in vivo) and the effects of NEFA and BHB on CYP2E1 expression and activity in hepatocytes (in vitro). Dairy cows with clinical ketosis exhibited a low blood concentration of glucose but high concentrations of NEFA and BHB. Hepatic mRNA, protein expression, and activity of CYP2E1 were significantly higher in cows with clinical ketosis than in control cows. In vitro, both NEFA and BHB treatment markedly up-regulated the mRNA and protein expressions as well as activity of CYP2E1 in cow hepatocytes. Taken together, these results indicate that high levels of NEFA and BHB significantly up-regulate the expression and activity of hepatic CYP2E1, and may be influential in the induction of oxidative stress in cows with clinical ketosis.

High-yielding dairy cows postpartum face the challenge of negative energy balance leading to elevated free fatty acids levels in the serum and follicular fluid thus affecting the ovarian function. Here, we investigated effects of physiological concentrations of palmitic acid (PA), stearic acid (SA) and oleic acid (OA) on the viability, steroid production and gene expression in a bovine granulosa cell (GC) culture model. Treatment with individual and combined fatty acids increased the CD36 gene expression, while no significant apoptotic effects were observed. Both PA and SA significantly upregulated the expression of FSHR, LHCGR, CYP19A1, HSD3B1, CCND2 and increased 17β-estradiol (E2) production, while OA downregulated the expression of these genes and reduced E2. Interestingly, STAR was equally downregulated by all fatty acids and combination treatment. E2 was significantly reduced after combination treatment. To validate the effects of OA, in vivo growing dominant follicles (10–19 mm) were injected with bovine serum albumin (BSA) with/without conjugated OA. The follicular fluid was recovered 48 h post injection. As in our in vitro model, OA significantly reduced intrafollicular E2 concentrations. In addition, expression of CD36 was significantly up- and that of CYP19A1 and STAR significantly downregulated in antral GC recovered from aspirated follicles. The ovulation rates of OA-injected follicles tended to be reduced. Our results indicate that elevated free fatty acid concentrations specifically target functional key genes in GC both in vitro and in vivo. Suggestively, this could be a possible mechanism through which elevated free fatty acids affect folliculogenesis in dairy cows postpartum.

The Study was conducted to evaluate the additive effect of selected feed additives (Glycerol, a mixture of exogenous fibrolytic enzymes (EFE), a mixture of probiotics and rumen protected methionine (RPM) under both in vitro and in vivo conditions. The optimum dose of pre-mixture of feed additives (PFA) was evaluated by in vitro gas production technique in two stages. The marginal increase in Y-max and in vitro neutral detergent fiber digestibility (IVNDFD) for each incremental increase in PFA was compared graphically and it was found to be 1.7 %. In in vivo trial, 12 crossbred HF cows were equally divided into 3 groups based on milk yield and average days in lactation. The optimized dose of PFA was added to the concentrate supplements without (T1) or with buffer (T2), control group (CG) was fed with the basal diet (roughage: concentrate ratio of 60:40). Results indicated that PFA with or without buffer had no significant effect on nutrient intake and digestibility. Fat corrected milk yield (FCM), milk fat, total solids (TS) and milk protein were significantly (P Lass Than 0.01) increased in TGs than that of CG. From the results, it may be concluded that optimized PFA had an additive effect on milk production efficiency but not on nutrient intake and digestibility. Further, different groups of feed additives can be evaluated for additive effect, which may pave the way for commercial product preparation.

The immune-suppressive effects of omega-3 (n-3) polyunsaturated fatty acids (PUFAs) on T cells have been observed via multiple in vitro and in vivo models. However, the precise mechanism that causes these effects is still undefined. In this study, we investigated whether n-3 PUFAs regulated T cell receptor (TCR) and peptide-major histocompatibility complex (pMHC) interactions. The expansion of anti-viral CD8+ T cells that endogenously synthesize n-3 PUFAs (FAT-1) dramatically decreased upon lymphocytic choriomeningitis virus (LCMV) infection in vivo. This decrease was not caused by the considerable reduction of TCR expression or the impaired chemotactic activity of T cells. Interestingly, a highly inclined and laminated optical sheet (HILO) microscopic analysis revealed that the TCR motility was notably reduced on the surface of the FAT-1 CD8+ T cells compared to the wild type (WT) CD8+ T cells. Importantly, the adhesion strength of the FAT-1 CD8+ T cells to the peptide-MHC was significantly lower than that of the WT CD8+T cells. Consistent with this result, treatment with docosahexaenoic acid (DHA), one type of n-3 PUFA, significantly decreased CD8+ T cell adhesion to the pMHC. Collectively, our results reveal a novel mechanism through which n-3 PUFAs decrease TCR-pMHC interactions by modulating TCR mobility on CD8+ T cell surfaces.

Aim: The present study was undertaken to evaluate the effect of variable levels of silkworm pupae oil and roughage: concentrate ratio on in vitro methane production, fermentation characteristics, and rumen protozoa population. Materials and Methods: In vitro gas production study (24 h) was performed with graded levels of silkworm pupae oil, namely, 0.5, 1, 2, 4, and 5% of the basal diet and four variable dietary regimes consisting roughage and concentrate in different proportions (70:30, 60:40, 50:50, and 40:60). At the end of incubation, gas samples were analyzed for methane, while fermented rumen liquor was used for protozoa enumeration. A separate set of incubations was carried out for the determination of in vitro dry matter digestibility. Results: Results from the in vitro studies revealed no adverse impact of the silkworm pupae oil supplementation up to 2% level on total gas production. However, supplementation beyond 2% has shown a reduction in total gas production. Incubation with variable levels (0.5-5%) of silkworm pupae oil with different dietary regimes indicated negligible (3-5%) to a substantial reduction (25-30%) on methane production. A graded decrement in methane production was recorded with increasing levels of silkworm pupae oil. Similarly, the protozoal populations were decreased from 10 to 51.5% with graded levels of silkworm pupae oil in different dietary regimes as studies did not reveal any significant (p>0.05) variation between 2 and 4% of oil supplementation. Conclusion: The silkworm pupae oil supplementation at 2% level decreases methane production by 12-15% without any adverse impact on feed fermentation. Oil supplementation may have a more pronounced effect on methane reduction if added to high roughage diet at in vitro conditions. However, in vivo, studies in ruminants are warranted to confirm the methane reduction with silkworm pupae oil supplementation.

Omega-3 fatty acids, which are abundant in fish oil, improve the prognosis of several chronic inflammatory diseases although the mechanism for such effects remains unclear. These fatty acids, such as eicosapentaenoic acid (EPA), are highly polyunsaturated and readily undergo oxidation. We show that oxidized, but not native unoxidized, EPA significantly inhibited human neutrophil and monocyte adhesion to endothelial cells in vitro by inhibiting endothelial adhesion receptor expression. In transcriptional coactivation assays, oxidized EPA potently activated the peroxisome proliferator-activated receptor α (PPARα), a member of the nuclear receptor family. In vivo, oxidized, but not native, EPA markedly reduced leukocyte rolling and adhesion to venular endothelium of lipopolysaccharide (LPS)–treated mice. This occurred via a PPARα-dependent mechanism because oxidized EPA had no such effect in LPS-treated PPARα-deficient mice. Therefore, the beneficial effects of omega-3 fatty acids may be explained by a PPARα-mediated anti-inflammatory effect of oxidized EPA.
