The objective of this retrospective study was to investigate the prevalence of pregnancy loss (PL; between 28–110 pregnancy days) and its relationship with factors: farm, year (2015–2018), season, artificial insemination (AI)-rank, parity, AI-type (fixed-time vs. oestrus-AI), previous PL, days in milk (DIM), fixed-time-AI protocol, AI-technician, bull, and semen-type (sexed vs. conventional). Data of 19,437 Holstein cattle AIs from eight Spanish farms were studied. Overall conception rate was 34.3% (6696/19,437) and PL 12.3% (822/6696). The PL was more likely to occur in primiparous (10.8%, odds ratio (OR) = 1.35; p = 0.04) and multiparous (15.3%; OR = 2.02, p < 0.01) than in heifers (PL = 6.9%, reference). Pregnancies achieved with AI after observed oestrus and natural breedings were associated with less PL than pregnancies after fixed-time-AI (12.7 vs. 11.9%; OR = 0.12, p = 0.01). First AIs related to higher PL than ≥2nd AIs (PL = 13.8% vs. 11.2; OR = 0.73, p < 0.01). The factors season, fixed-time-AI protocol, DIM, bull, AI-technician, or type of semen were not significantly associated with PL. Therefore, farmers and consultants should adapt their preventive strategies relating to PL, particularly, to the parity of the cattle.

 Background:Progesterone (P4) is essential for embryonic development and maintenance of pregnancy when deficiency causes early embryonic loss. In this study, we investigated the ability of hormonal supplementation to improve the fertility of cows subjected to fixed-time artificial insemination (IATF) protocols. Here, we evaluated the effect of long-acting injectable progesterone (iP4) supplementation in the D4 after IATF on pregnancy rate and pregnancy loss in Nellore cows (Bos taurus indicus) from different reproductive categories in Western Amazonia.Materials, Methods & Results:Eight hundred thirteen animals from 5 farms were selected and distributed into 2 groups: control [GC; administration of 0.5 mL of 0.9% saline solution, intramuscularly – IM] (n = 407) and a group that received injectable progesterone (iP4) that was long-acting [GiP4; administration of 0.5 mL of iP4, 300 mg, via IM four days after IATF] (n = 406). Each group contained 3 subgroups: heifers, primiparous cows, and multiparous cows. Of the 407 animals in the CG, 103 were heifers, 107 primiparous, and 197 multiparous. Of the 406 animals in the GiP4 group, there were 101 heifers, 107 primiparous, and 198 multiparous. On a random day of the estrous cycle (D0), an intravaginal device containing 1 g of P4 and 2 mg of estradiol benzoate (BE) was inserted by intramuscular injection. On D8, the P4 device was removed and 150 μg of D-cloprostenol (PGF2α), 300 IU eCG, and 1 mg BE were administered IM. Cows were inseminated at D10, 48-52 h after procedure on D8. Pregnancy diagnosis was made between 35 and 40 days after insemination through ultrasound examination. Between 80 and 90 days after insemination, a new ultrasound examination was performed to assess early pregnancy loss. The data were processed using the SAS 9.2. The conception rate, pregnancy loss, and final conception rate were analyzed using PROC GLIMMIX according to groups (CG and GiP4), categories (heifers, primiparous and multiparous), and their interactions as variables. The differences in the means of least squares were adjusted using the Tukey–Kramer method. Statistical significance was defined as P < 0.05. The general conception rate was 46% (375/816). Regardless of the animal class, GiP4 animals (51.97%) had higher conception rates (P < 0.05) than CG (40.29%). In the subgroups (heifers, primiparous and multiparous cows), there was a difference (P < 0.05) between animals treated with iP4 (52.48%, 57.94%, and 48.48%, respectively) and those who were not (39.81%, 41.12%, and 40.10%, respectively). Gestational losses, regardless of the animal class, were higher in females in the CG (7.93%) [P < 0.05] compared to those in the GiP4 group (2.84%). Regardless of treatment with iP4, the percentage of gestational loss in heifers was significantly higher (10.64%) than that in primiparous and multiparous cows (3.77% and 2.86%, respectively). The final conception rates were higher in animals that received long-acting iP4, which increased the final pregnancy in all parity categories. Discussion: In the present study, the use of iP4 increased the pregnancy rate in Nellore females, regardless of the category. Although there has been no consensus on the use of iP4, there is an agreement that increases in the pregnancy rate are related to the moment of exogenous P4 application. In addition to influencing the pregnancy rate, reduction in pregnancy losses is also attributed to iP4 treatment, a fact demonstrated in the present study, where animals treated with iP4 had a lower pregnancy loss rate than normally occurs in beef cattle. This study aimed to evaluate the effects of rectal temperature (RT) on conception rate (CR), as well as the effects of seasonality (spring-summer vs. autumn-winter) and timing of artificial insemination (AI) (morning vs. afternoon) on RT and CR in crossbred dairy cows (Holstein x Gyr). The experiment was conducted on a dairy farm in Centralina, MG, where 1,219 conventional and fixed-time inseminations were analyzed. The RT was measured immediately before AI using a digital thermometer. Pregnancy diagnosis was performed using ultrasonography between 28 and 60 days after AI. T The effects of seasonality and timing of AI on RT were analyzed with a Mann-Whitney U test and the effects of RT (above or below the average), seasonality and timing of AI on CR were analyzed with a Chi-squared test, both using the SAS program. The RT average was 39.4°C. Cows with RT ? 39.4°C had lower CR than cows with RT < 39.4°C (25.78% vs. 32.54%; P = 0.0096). During spring-summer, cows had higher RT (39.44°C ± 0.025 vs. 39.27°C ± 0.022; P < 0.0001) and lower CR (25.49% vs. 31.75%; P = 0.0146) compared with autumn-winter. Cows inseminated in the morning had lower RT (38.96°C ± 0.022 vs. 39.60°C ± 0.018; P < 0.0001) and higher CR (32.86% vs. 26.06%; P = 0.0102) than cows inseminated in the afternoon. In conclusion, crossbred dairy cows with rectal temperature equal to or greater than 39.4°C had lower conception rate. Moreover, rectal temperature and conception rate were affected by seasonality and insemination time.

The objective of this work was to obtain prevalence estimates of cloudy vaginal mucus in artificially inseminated Holstein cows raised under intense heat, in order to assess the effect of meteorological conditions on its occurrence during estrus and to determine its effect on conception rate. In a first study, an association was established between the occurrence of cloudy vaginal mucus during estrus and the conception rate of inseminated cows (18,620 services), raised under intense heat (mean annual temperature of 22°C), at highly technified farms, in the arid region of northern Mexico. In a second study, data from these large dairy operations were used to assess the effect of meteorological conditions throughout the year on the occurrence of cloudy vaginal mucus during artificial insemination (76,899 estruses). The overall rate of estruses with cloudy vaginal mucus was 21.4% (16,470/76,899; 95% confidence interval = 21.1-21.7%). The conception rate of cows with clean vaginal mucus was higher than that of cows with abnormal mucus (30.6 vs. 22%). Prevalence of estruses with cloudy vaginal mucus was strongly dependent on high ambient temperature and markedly higher in May and June. Acceptable conception rates in high milk-yielding Holstein cows can only be obtained with cows showing clear and translucid mucus at artificial insemination.

The studies aimed to evaluate the pregnancy rate (PR) for timed artificial insemination (TAI) after G7G-Ovsynch, modified G7G-Ovsynch (MG7G-Ovsynch) and Ovsynch protocols and to assess the accuracy of using pregnancy-associated glycoproteins (PAGs) and plasma progesterone (P4) in pregnancy diagnosis compared with ultrasonography (US). In study 1, Holstein cows (n = 37) were bred by TAI following the G7G-Ovsynch protocol (n = 19) or MG7G-Ovsynch (n = 18). Pregnancy was evaluated by US at days 31, 59, and 87 after breeding. The PR was not different for the G7G-Ovsynch and MG7G-Ovsynch. Blood and milk samples were collected on day 3 after insemination and then weekly through day 59 post TAI in cows diagnosed as not pregnant on day 31 and through day 87 in pregnant cows. PAGs were measured using ELISA and P4 by radioimmunoassay (RIA). In the second study, Holstein cows (n = 212) were bred by TAI following G7G-Ovsynch protocol (n = 110) or standard Ovsynch (n = 102). Cows were subjected to pregnancy diagnosis on days 30, 60, and 90. A subset (n = 15 in each group) was subjected to blood and milk samples on days 30, 45, 60, 75, and 90 to measure PAGs and P4. In study 2, PR was not significantly different between synchronization protocols on days 30, 60, and 90. Pregnancy loss averaged 15% between day 30 and day 90. The use of PAGs and P4 proved equally effective in diagnosis of pregnancy. Thus, G7G-Ovsynch was deemed the protocol of choice in postpartum cows, and PAGs assayed in milk or plasma could be used to diagnose pregnancy.

ABSTRACT: Two experiments were conducted to evaluate the impact of two vaccines on the reproductive performance of multiparous beef cows in Mato Grosso do Sul, Midwest Brazil. In Experiment 1,765 calved multiparous Nellore cows were subjected to the same fixed-time artificial insemination (FTAI) protocol and rebred. Ultrasound pregnancy diagnosis was performed 30 and 90 days post-FTAI (DPI). Rates of pregnancy and pregnancy loss were determined for three periods: from 30 to 90 DPI, from 30 DPI to calving, and from 90 DPI to calving. The cows were assigned to three groups with different vaccination protocols - namely, Group VACMULT (n=250): vaccine against bovine herpesvirus 1 (BoHV-1), bovine viral diarrhea virus (BVDV), and leptospirosis; Group VACL (n=245): vaccine against leptospirosis alone; Group NOVAC (n=270): no vaccination. Serum antibody titers for BoHV-1, BVDV, and leptospirosis, measured in 57 cows from each group indicated active infection, suggesting circulation of these pathogens in the herd. No differences in pregnancy rates were observed across groups. Pregnancy loss rates did not differ significantly across groups within any of the periods investigated (30 to 90 DPI, 30 DPI to calving, or 90 DPI to calving). In Experiment 2, two vaccination protocols for each vaccine were investigated. Group VACGEST was vaccinated on day zero of FTAI (D0) and again 30 days post-FTAI (30 DPI). Group VACPREV was vaccinated on D0 and again on the day of insemination (D11). No significant difference was observed between groups, or significant gestational loss in the group that received the second vaccine on the day of insemination. The results revealed that neither vaccine interfered with the reproductive performance of multiparous cows. No differences were observed between vaccination carried out on both D0 and D11 and that performed on both D0 and D30. The performance of the IBR/BVDV/Leptospirosis vaccine on the day of the artificial insemination did not cause adverse effects on the reproductive parameters.

The aim of the present study was to determine the effects of one or two doses of prostaglandin F2α (PGF) on the pregnancy rate (PR) in Simmental cows subjected to the Ovsynch or Ovsynch + Controlled internal drug release (CIDR) protocols. On d 0, 100 µg gonadorelin diacetate tetrahydrate (GnRH) was injected into cows in group 1 (n = 50) and group 2 (n = 40) and CIDR was intravaginally inserted. On d 7, 25 mg dinoprost tromethamine (PGF2α, PGF) was administered, and GnRH was injected 56 h later. Fixed-time artificial insemination (TAI) was performed 16 h after GnRH injection. In group 3 (n = 62) and group 4 (n = 63), PGF was administered 7 d following GnRH treatment, and GnRH was injected 56 h later. TAI was performed 16 h after GnRH injection. In groups 2 and 4, a second dose of PGF was injected 24 h after the first dose. Pregnancy was determined by transrectal ultrasonography on d 30 and 60 post-TAI. The pregnancy rate (PR) on d 30 post-TAI was significantly different among the groups: 46%, 55%, 29%, and 36.5%, in groups 1, 2, 3, and 4, respectively (P = 0.048). In addition, the PR was 36.3% and 43.7% in the single PGF and double PGF injection groups, respectively. There was no statistical difference in the PR on d 60 post-TAI (P > 0.05). Pregnancy loss was between 4.34% and 11.11% (P > 0.05). The highest PR in the primiparous cows was observed in group 3 (57.7%). The PR was not significantly different among multiparous cows (P > 0.05). In conclusion, two injections of PGF administered 24 h apart in Simmental cows may increase the PR in the Ovsynch or Ovsynch + CIDR protocols.
