The objective of this study was to determine the effect of supplementing Kluyveromyces marxianus CPY1, K. marxianus RSY5 and Pichia kudriavzevii YSY2 isolated from ruminal fluid of dairy cows on transfer of aflatoxin B1 (AFB1) from feed into aflatoxin M1 (AFM1) in milk, DMI, milk production and nutrient digestibility. Four multiparous Holsteins in mid-lactation were used in a 4 × 4 Latin square design trial consisting of 14 days in each experimental period for sample collection. Between each period, 14 clearance days prior to the next treatment were allowed to minimize carryover effects. In each treatment, subsequent supplementation of isolated yeast was compared, i.e., (1) control (without yeast supplementation), (2) K. marxianus CPY1 (K1Y), (3) K. marxianus RSY5 (K2Y) and (4) P. kudriavzevii YSY2 (PY). All diets contained 22.28 µg of AFB1/kg. Treatments were individually fed at the rate of 2 g/day (1 × 109 CFU/g) of yeast biomass or corn meal in the control group. Concentrations of AFM1 in milk was reduced with yeast and averaged 1.54, 0.36, 0.43 and 0.51 µg/L for control, K1Y, K2Y and PY, respectively (p < 0.01). The transfer of AFB1 from feed into AFM1 in milk was higher in control compared with K1Y, K2Y and PY (7.26% vs. 1.18%, 1.44% and 1.69% respectively, p < 0.01). Supplementation of yeast also improved DMI and milk compositions, but no differences were observed in nutrient digestibility or milk yield among treatments. Concentration and yield of milk protein, fat, lactose, solid-not-fat (SNF) and total solids were greater in cows fed yeast compared with the control (p < 0.01). These results indicate that K. marxianus CPY1, RSY5 and P. kudriavzevii YSY2 shows promise as a dietary supplementation to detoxify AFB1 and improve DMI and yield of milk components.

The objective of the study was to determine the effect of soybean-derived phytoestrogens and their metabolites on the activity of sex hormones during the oestrous cycle in multiparous lactating dairy cows. The experiment was carried out on 4 multiparous lactating Holstein cows in the form of replicated Latin square in double reversal design. The experiment in the total length of 168 days was divided into 4 periods of 42 days, each consisting of a 21-day preliminary period and a 21-day collecting period. Cows were divided into 2 groups of 2 cows. The control group (C) was fed a diet based on extruded rapeseed cake while the experimental group (S) was fed a diet containing extruded full-fat soya. The intake of total isoflavones was 3297 mg/d in S and 58.0 mg/d in C (P < 0.001). The concentrations of individual isoflavones, it is daidzein, genistein and equol in plasma were significantly higher in the experimental group S (49.3, 78.7 and 218.8 ng/ml, respectively) than in the control group C (13.5, 42.9 and 18.3 ng/ml, respectively, P < 0.001). Plasma concentration of progesterone throughout the oestrous cycle was not influenced by the diet used (P > 0.05). Plasma concentration of prostaglandine PGFM throughout the oestrous cycle in the experimental group (S) tended to be higher (P = 0.095) than in the control group (C). No differences in the length of the oestrous cycle between the cows fed different diets were observed.

The objective of this study was to determine the effect of supplemental lysine (Lys), methionine (Met) or both added to diet of dairy cows in the form of rumen-protected (RP) tablets on changes in milk fatty acids (FA) profile. The trial was carried out on four lactating Holstein cows in the form of Latin square design and was divided into 4 periods of 14 d (10-d preliminary period and a 4-d experimental period). The four treatments were as follows: C – control without amino acids (AA) supplementation, L – supplement of RP Lys, M – supplement of RP Met and ML – supplement of RP Met and Lys. Cows were fed on a diet based on maize silage, lucerne hay and supplemental mixture. Milk yield in ML (34.18 kg/d) was higher than in L or M (32.46 kg and 32.13 kg, respectively, P < 0.05) and tended to be higher than in C (33.33 kg/d, P > 0.05). Protein yield in ML (1054 g / d) was higher than that found in C, L or M (990, 998 or 968 g / d, respectively, P < 0.05). Milk fat content and yield in C and ML was higher in comparison to L and M (P < 0.05). Content of short-chain FA (C 4:0–C 12:0) was not affected by the treatment except of L that was lower than in C (P < 0.05). Content of medium-chain FA in M was lower compared to C, L or ML (P < 0.05). The content of long-chain FA in M was significantly higher than in other groups (P < 0.05). The total content of SFA in M was lower than in C or ML (P < 0.05) and tended to be lower than in L. Contents of UFA, MUFA and PUFA in M were higher than in C and ML (P < 0.05).

The study compared the influence of chitosan sources on rumen fermentation, methane emission and milk production in lactating dairy cows fed a glycerin-based diet. Six, lactating Holstein-Frisian crossbreeds (410 &plusmn; 5.0 kg BW, 120 &plusmn; 21 day-in-milk), were arranged in a 3 x 3 replicated Latin square design. In addition to control, a 2% chitosan extract supplement and a 2% commercial chitosan supplement of dry matter intake were the treatments. The results denoted that no significant differences on daily dry matter, nutrients or estimated energy intake were noted when cows received different sources of chitosan. Nutrient digestibility was not influenced differently by extraction based or commercial chitosan supplements. The pH, temperature, ammonia nitrogen, blood urea and microbial count were similar among treatments. The different sources of chitosan supplements did not change the totals of volatile fatty acids, acetate and butyrate; in contrast, different chitosan sources influenced (P&lt;0.05) propionate content. The ruminal acetate to propionate ratio was markedly (P&lt;0.05) reduced with chitosan supplement, but no change appeared between sources of chitosan. At 4 hours after feeding, the methane estimation significantly decreased with the addition of chitosan supplementation (P&lt;0.05) compared to the control group. The purine derivatives and microbial protein synthesis were not altered by the treatments. No significant differences existed on milk yield, milk composition or milk urea nitrogen when cows received different sources of chitosan (P&gt;0.05). In sum, supplementing extracted chitosan showed more potential than did commercial chitosan for enhancing economic efficiency and recycling shrimp residues, therefore, reducing environmental waste.

ABSTRACTA protocol of infrequent, but strategically timed milk-sampling was established for predicting the time of ovulation, and thus the optimum time for insemination, in lactating dairy cows. In the experimental group (no. = 49), the time of ovulation was predicted on the basis of a fall in milk progesterone concentrations, which was identified by the use of ‘on-farm’ progesterone assay kits. Reproductive performance in these animals was compared with that in control cows (no. = 45) that were inseminated solely on the basis of oestrous detection by stockmen. The accuracy of ovulation prediction, and of oestrous detection, was assessed from progesterone profiles based on milk samples collected three times weekly from all 94 cows. These milk samples were analysed by laboratory enzyme-immunoassay after the end of the trial.Over the period during which milk samples were monitored with ‘on-farm’ progesterone kits, a total of 88 ovulations occurred in the experimental group. The progesterone-testing protocol accurately predicted 87 (99%) of these. Over the same period, there was a total of 81 ovulations in the control group and 63 (78%) of these were associated with correct oestrous detection. Conception rates to correctly timed insemination did not differ significantly between groups, but by the end of the three-cycle experimental period, significantly more of the cows in the experimental group (21·8%) than in the control group (4·8%; P < 0·05). In contrast, the use of ‘on-farm’ progesterone results to confirm oestrus avoided any mistimed inseminations (13% of inseminations in the control group). By using the milk-sampling and ‘on-farm’ progesterone-testing protocol, only 1% of ovulations were not accompanied by a correctly timed insemination. This compared with 22% of ovulations in the control group not associated with an insemination because oestrus was not detected. By ensuring that all ovulations are associated with a correctly timed insemination, herd reproductive performance can be significantly improved.

This study evaluated the effects of precision processing (PP; processing based on kernel size) barley grain on ruminal fermentation and productivity of lactating dairy cows. Twenty multiparous Holstein cows, including eight ruminally cannulated cows, were used in a replicated 4×4 Latin square design with 21-d periods. Diets contained light barley grain processed precisely using a narrow roller setting (LB), heavy barley processed precisely using a wide roller setting (HB), processed HB and LB mixed at equal proportions (PP), or equal parts of light and heavy barley grain processed at a single narrow roller setting (CON). All diets consisted of 40% barley grain, 40% barley silage, and 20% of a supplement premix. Comparisons were made between LB and HB to evaluate the effect of barley quality, and between PP and CON to evaluate the effect of precision processing. Dry matter intake, sorting index, ruminal fermentation characteristics, and nutrient digestibility were not affected by diet. In addition, milk yield and concentrations of milk fat, protein, and lactose were not different, although milk urea nitrogen concentration was greater for PP vs. CON and for LB vs. HB. These results suggest that precision processing barley grain based on kernel size may not drastically affect ruminal fermentation and milk production in lactating dairy cows.

Our objective was to evaluate the effects of providing increasing levels of chitosan on nutrient digestibility, ruminal fermentation, blood parameters, nitrogen utilisation, microbial protein synthesis, and milk yield and composition of lactating dairy cows. Eight rumen-fistulated Holstein cows [average days in lactation = 215 ± 60.9; and average bodyweight (BW) = 641 ± 41.1 kg] were assigned into a replicated 4 × 4 Latin square design, with 21-day evaluation periods. Cows were assigned to be provided with four levels of chitosan, placed into the rumen through the fistula, as follows: (1) Control: with no provision of chitosan; (2) 75 mg/kg BW; (3) 150 mg/kg BW; and (4) 225 mg/kg BW. Chitosan had no effect on dry matter intake (P > 0.73); however, chitosan increased (P = 0.05) crude protein digestibility. Propionate concentration was increased (P = 0.02), and butyrate, isobutyrate, isovalerate and acetate : propionate ratio were decreased (P ≤ 0.04) by chitosan. Chitosan had no effect (P > 0.25) on acetate, pH and NH3 ruminal concentration. Glucose, urea, and hepatic enzyme concentrations in the blood were similar (P > 0.30) among treatments. Nitrogen balance was not affected, but chitosan increased milk nitrogen (P = 0.02). Microbial protein synthesis was not affected by chitosan (P > 0.44). Chitosan increased (P = 0.02) milk yield, fat-corrected milk, protein and lactose production. Chitosan changes ruminal fermentation and improves milk yield of lactating dairy cows; therefore, we conclude that chitosan can be used as a rumen modulator instead of ionophores in diets for dairy cows.

This study evaluates the effects of fresh cassava root (CR) and a solid feed-block containing sulfur (S-FB) on fermentation in the rumen, feed utilization, milk yield, and milk composition in lactating dairy cows. Four Holstein-Friesian cows with 470 ± 50.0 kg body weight (BW), 10 ± 2 kg day−1 average milk yield, and 112 ± 15 days-in-milk were studied. A 2 × 2 factorial combination was arranged in a 4 × 4 Latin square design to evaluate the treatment-related effects. The treatments were obtained from a combination of two factors: (1) levels of CR at 10 g kg−1 BW (CR-1) and 15 g kg−1 (CR-1.5) and (2) levels of sulfur supplementation in solid feed-block at 20 g kg−1 (S-FB-2) and 40 g kg−1 (S-FB-4). The results showed that CR and S-FB had no interaction effect on feed intake, digestibility, fermentation, blood metabolites, milk yield, or its composition. Feeding CR up to 15 g kg−1 of the BW significantly increased (p < 0.05) the milk fat concentration while it decreased (p < 0.05) the somatic cell count. The S-FB-4 of the sulfur significantly (p < 0.05) increased the acid detergent fiber when compared with the S-FB-2 of the sulfur. CR could be fed up to 15 g kg−1 of BW with S-FB containing high sulfur (40 g kg−1) in dairy cows without a negative impact.

This study evaluated the replacement of Tifton hay with xiquexique (0, 12, 24 and 36%) on feeding behavior of lactating dairy cows. To this end, eight cows with average milk production of 15 kg milk day-1 and average body weight of 465.2 ± 39.4 kg were distributed in a double 4x4 Latin square. Each experimental period lasted 16 days, ten for adaptation and six for data collection. The record of behavioral variables was made every five min. for 24 hours. The times spent in feeding, ruminating and total chewing, expressed in hours day-1, as well as feeding (FE) and rumination (RE) efficiencies, expressed in g NDF h-1, the number of cuds and cud chews per day showed a linear decrease according to the levels of xiquexique in the diet. There was a linear increase in time in idleness. Xiquexique levels in the diet for dairy cows did not influence FE and RE, expressed in g DM h-1, number and time spent in chewing per cud. The replacement of Tifton hay with xiquexique in the diet alters the feeding behavior of dairy cows due to the reduction in fiber content.

This study examined the effects of partly replacing grass silage (GS) with maize silage (MS), with or without rapeseed oil (RSO) supplementation, on methane (CH4) emissions, production performance, and rumen microbiome in the diets of lactating dairy cows. The effect of individual pre-trial CH4-emitting characteristics on dietary emissions mitigation was also examined. Twenty Nordic Red cows at 71 ± 37.2 (mean ± SD) days in milk were assigned to a replicated 4 × 4 Latin square design with four dietary treatments (GS, GS supplemented with RSO, GS plus MS, GS plus MS supplemented with RSO) applied in a 2 × 2 factorial arrangement. Partial replacement of GS with MS decreased the intake of dry matter (DM) and nutrients, milk production, yield of milk components, and general nutrient digestibility. Supplementation with RSO decreased the intake of DM and nutrients, energy-corrected milk yield, composition and yield of milk fat and protein, and general digestibility of nutrients, except for crude protein. Individual cow pre-trial measurements of CH4-emitting characteristics had a significant influence on gas emissions but did not alter the magnitude of CH4 emissions. Dietary RSO decreased daily CH4, yield, and intensity. It also increased the relative abundance of rumen Methanosphaera and Succinivibrionaceae and decreased that of Bifidobacteriaceae. There were no effects of dietary MS on CH4 emissions in this study, but supplementation with 41 g RSO/kg of DM reduced daily CH4 emissions from lactating dairy cows by 22.5%.

Objective: To investigate the effect of antibiotics and/or probiotics on broiler performance, some serum metabolites, cecum microflora composition, and ileum histomorphology under the Egyptian conditions. Design: Randomized controlled experimental study. Animals: Two hundred forty 1-day-old Ross (308) chicks were reared till 35 days of age. Procedures: The birds were randomly allocated into four main groups: a control diet without additives (CON); probiotic (Lactobacillus acidophilus) supplemented diet (PRO); antibiotic (Avilamycin) supplemented diet (ANT) and a mix group (AP) that received antibiotic in the diet form 1 to 4 days of age and treated during the rest of the experimental period with probiotics. Results: Chickens fed on probiotic or antibiotic diets had linear improvement in live body weight (LBW) and feed conversion ratio (FCR) compared with the control group, while the best LBW and FCR were in the AP group. An improvement in the nutrient digestibility was observed in the probiotic added groups (PRO and AP). Serum cholesterol and low-density lipoprotein cholesterol contents decreased when antimicrobial (probiotic or antibiotic) supplementations were used, while there was an increase in high-density lipoprotein cholesterol contents, serum total protein, and albumin levels. Among all groups, cecum Clostridium perfringens and Escherichia coli counts decreased; however, there was an increase in Lactobacillus count compared to the control group. In probiotic supplemented groups (PRO and AP), a significant (P<0.05) improvement in ilea architecture. Conclusion and clinical relevance: Using probiotic after initial treatment with an antibiotic in broiler diets had a positive effect on broiler growth performance, gut health (improved cecum microbial populations and ileum histomorphology), and nutrient digestibility.
