Glycine max NAC81 (GmNAC81) is a downstream effector of the DCD/NRP-mediated cell death signaling, which interacts with GmNAC30 to fully induce the caspase 1-like vacuolar processing enzyme (VPE) expression, the executioner of the cell death program. GmNAC81 has been previously shown to positively modulate leaf senescence via the NRP/GmNAC81/VPE signaling module. Here, we examined the transcriptome induced by GmNAC81 overexpression and leaf senescence and showed that GmNAC81 further modulates leaf senescence by regulating an extensive repertoire of functionally characterized senescence-associated genes (SAGs). Because the NRP/GmNAC81/VPE signaling circuit also relays stress-induced cell death signals, we examined the effect of GmNAC81 overexpression in drought responses. Enhanced GmNAC81 expression in the transgenic lines increased sensitivity to water deprivation. Under progressive drought, the GmNAC81-overexpressing lines displayed severe leaf wilting, a larger and faster decline in leaf Ψw, relative water content (RWC), photosynthesis rate, stomatal conductance, and transpiration rate, in addition to higher Ci/Ca and lower Fm/Fv ratios compared to the BR16 control line. Collectively, these results indicate that the photosynthetic activity and apparatus were more affected by drought in the transgenic lines. Consistent with hypersensitivity to drought, chlorophyll loss, and lipid peroxidation were higher in the GmNAC81-overexpressing lines than in BR16 under dehydration. In addition to inducing VPE expression, GmNAC81 overexpression uncovered the regulation of typical drought-responsive genes. In particular, key regulators and effectors of ABA signaling were suppressed by GmNAC81 overexpression. These results suggest that GmNAC81 may negatively control drought tolerance not only via VPE activation but also via suppression of ABA signaling.

AbstractDrought stress limits the growth and development of grapevines, thereby reducing productivity, but the mechanisms by which grapevines respond to drought stress remain largely uncharacterized. Here, we characterized a group A bZIP gene from ‘Kyoho’ grapevine, VlbZIP30, which was shown to be induced by abscisic acid (ABA) and dehydration stress. Overexpression of VlbZIP30 in transgenic Arabidopsis enhanced dehydration tolerance during seed germination, and in the seedling and adult stages. Transcriptome analysis revealed that a major proportion of ABA- and/or drought-responsive genes are transcriptionally regulated by VlbZIP30 during ABA or mannitol treatment at the cotyledon greening stage. We identified an A. thaliana G-box motif (CACGTG) and a potential grapevine G-box motif (MCACGTGK) in the promoters of the 39 selected A. thaliana genes up-regulated in the transgenic plants and in the 35 grapevine homologs, respectively. Subsequently, using two grapevine-related databases, we found that 74% and 84% (a total of 27 genes) of the detected grapevine genes were significantly up-regulated by ABA and drought stress, respectively, suggesting that these 27 genes involve in ABA or dehydration stress and may be regulated by VlbZIP30 in grapevine. We propose that VlbZIP30 functions as a positive regulator of drought-responsive signaling in the ABA core signaling pathway.HighlightVlbZIP30 positively regulate plant drought tolerance through regulated the expression of 27 grapevine candidate genes via G-box cis-element (MCACGTGK) in ABA signaling pathway.

Food security is the crucial global issue, especially in developing countries like Pakistan. Since edible oil is an essential food item, its persistent paucity in the country and huge import for meeting domestic requirements, has attained it second largest import item after petroleum products. The aim of present study is qualitative and quantitative evaluation of newly developed short duration and drought tolerant canola quality Brassica juncea lines ZBJ-06012 and ZBJ-08051 to overcome the unfavorable edible oil situation in the country. Thirteen lines were evaluated in randomized complete block design (RCBD) for seed yield, oil quality, maturity period and drought tolerance under different agro-climatic zones both in irrigated and arid areas across the Punjab province at eight locations in Micro Yield Trials during Rabi season 2012-13 and 2013-14. Presently, grown non-canola mustard varieties Khanpur Raya and Anmol Raya were used as check varieties. Brassica napus cultivars Punjab canola and Faisal canola were also included in the trials for comparison study of mustard and rapeseed genotypes. Data for all traits under observation was analyzed through Principle Component Analysis (PCA) to evaluate the best performing lines in irrigated as well as in rain fed areas. Principal Component Analysis showed first 2 PCs having Eigen value >1 explaining 76.4% and 72% of the total variation at irrigated areas and rain fed areas respectively. The mean seed yield was also compared by Least Significant Difference (LSD) test to study the significance at 5% probability level. Canola quality B. juncea lines ZBJ-06012 and ZBJ-08051 have shown good adaptability, early maturity, non-shattering, disease and drought tolerance traits with high yield potential in comparison with presently grown Brassica napus cultivars ?Punjab Canola? and ?Faisal Canola?. Due to these prominent features, these lines have a great scope for motivating farmers to grow canola quality B. juncea when compared with B. napus and non-canola B. juncea. Future challenges demand further development of high yielding, short duration and aphid tolerant mustard cultivars having high oil content and canola quality. There is a great potential of exploiting genetic variability in the existing B. juncea material to achieve the aforesaid goals by using conventional plant breeding techniques.
