Cilj ovog rada je prikazati stanje spektroskopije u bliskom infracrvenom području (NIR spektroskopije) kao tehnologije za procjenu ad libitum konzumacije suhe tvari (ST) i organske tvari (OT) te in vivo probavljivosti ST, OT i OT u ST (Dvrijednosti) voluminozne krme. U usporedbi s provedbom skupih, radno zahtjevnih i dugotrajnih bioloških istraživanja radi utvrđivanja ad libitum konzumacije i in vivo probavljivosti voluminozne krme, NIR spektroskopija je brza, fizikalna, nedestruktivna, multianalitička i eklološki prihvatljiva metoda koja može dati precizne i točne rezultate uz minimalnu ili nikakvu pripremu uzoraka. Biološki se parametri teže procjenjuju NIR spektroskopijom u odnosu na kemijske parametre voluminozne krme radi varijacija između životinja, a posljedično pogrešaka u procjeni reakcija životinja na određenu hranidbu i hranu te činjenice da se niti jedan biološki parametar ne može utvrditi u kemijskom laboratoriju s visokom razinom točnosti. Prikazani kalibracijski modeli su prihvatljive razine pouzdanosti procjene ad libitum konzumacije i in vivo probavljivosti voluminozne krme prema utvrđenim koeficijentima determinacije (R2) (0,7-0,9 i 0,47-0,98 slijedom), standardnim greškama kalibracije (SEC) (4,28-6,89 i 0,01-1,69 slijedom), standardnim greškama unakrsne validacije (SECV) (4,53-7,92 i 0,02-2,15 slijedom) te standardnim greškama procjene (SEP) (5,42-9,59 i 0,026-5,7 slijedom). Ovo istraživanje potvrđuje potencijal NIR spektroskopije za procjenu ad libitum konzumacije ST i OT voluminozne krme i in vivo probavljivosti ST, OT i D-vrijednosti voluminozne krme.

Fasting activates orexigenic neuropeptide Y neurons in the hypothalamic arcuate nucleus (ARC) of mice, which is reversed by 2 h refeeding with standard chow. Here, we investigated the contribution of diet-derived macronutrients and anorectic hormones to the reversal of the fasting-induced ARC activation during 2 h refeeding. Refeeding of 12-h-fasted mice with a cellulose-based, noncaloric mash induced only a small reduction in c-Fos expression. Refeeding with diets, containing carbohydrates, protein, or fat alone reversed it similar to chow; however, this effect depended on the amount of intake. The fasting-induced ARC activation was unchanged by subcutaneously injected amylin, CCK (both 20 μg/kg), insulin (0.2 U/kg and 0.05 U/kg) or leptin (2.6 mg/kg). Insulin and leptin had no effect on c-Fos expression in neuropeptide Y or proopiomelanocortin-containing ARC neurons. Interestingly, CCK but not amylin reduced the ghrelin-induced c-Fos expression in the ARC in ad libitum-fed mice, suggesting that CCK may inhibit orexigenic ARC neurons when acting together with other feeding-related signals. We conclude that all three macronutrients and also non-nutritive, ingestion-dependent signals contribute to an inhibition of orexigenic ARC neurons after refeeding. Similar to the previously demonstrated inhibitory in vivo action of peptide YY, CCK may be a postprandial mediator of ARC inhibition.

We evaluated the effects of 8 mo of calorie restriction [CR: 60% of ad libitum (AL) food intake] on glucose uptake by 14 tissues in unanesthetized, adult (12 mo) F344×BN rats. Glucose metabolism was assessed by the 2-[3H]deoxyglucose tracer technique at 1500 or 2100. Despite an ∼60% decline in insulinemia with CR, plasma 2-[3H]deoxyglucose clearance for CR was greater than for AL at both times. A small, CR-related decrease in glucose metabolic index ([Formula: see text]) occurred only at 1500 in the spleen and heart, and this decrease was reversed at 2100. In some tissues (cerebellum, lung, kidney, soleus, and diaphragm),[Formula: see text] was unaffected by diet, regardless of time. In the other tissues (brown fat, 3 white fat pads, epitrochlearis, plantaris, and gastrocnemius),[Formula: see text] was higher or tended to be higher for CR vs. AL at one or both times. These findings indicate that 8 mo of CR did not cause a continuous reduction in in vivo glucose uptake by any tissue studied, and, in several insulin-sensitive tissues, glucose uptake was at times greater for CR vs. AL rats.

De-hydrated forages are often fed to equids in the UK in place of more traditional grass hay, particularly where individual animals are known to have a sensitivity to dusty, mouldy hay which may play a part inducing respiratory problems such as chronic obstructive pulmonary disease (COPD). One such alternative forage is short-chop de-hydrated grass. However, there is very little information available on voluntary feed intake (VFI), apparent digestibility and nutrient intake parameters when de-hydrated grass is offered to equids compared with traditional grass hay. This study examines the VFI and apparent digestibility in vivo of a short-chop de-hydrated grass compared with a traditional grass hay and determines their ability to meet the predicted energy and protein needs of mature ponies.Six mature Welsh-cross pony geldings with a mean liveweight (LW) of 281 kg (s.e.d. 0.89) were individually housed and offered ad libitum access to either short-chop de-hydrated grass (DHG) or traditional grass hay (HAY) plus 60 g/h/d minerals. The DHG and HAY were made from the same 2nd cut perennial ryegrass sward cut on the same day.

Increases in the concentration of malonyl-CoA in skeletal muscle have been observed in the KKAy mouse, an obese rodent with high plasma insulin and glucose levels [Saha et al. Am. J. Physiol. 267 (Endocrinol. Metab. 30): E95-E101, 1994]. To assess whether insulin and glucose directly regulate malonyl-CoA in muscle, soleus muscles from young rats were incubated with insulin and glucose at various concentrations, and their content of malonyl-CoA was determined. In addition, the effect on malonyl-CoA of denervation and electrically induced muscle contractions was assessed. The concentration of malonyl-CoA in the soleus, taken directly from a rat fed ad libitum, was 2.0 +/- 0.2 nmol/g. In muscles incubated for 20 min in a medium devoid of added insulin and glucose, the concentration was decreased to 0.8 +/- 0.2 nmol/g. When the medium contained 0.5, 7.5, or 30 mM glucose, malonyl-CoA levels were 1.3 +/- 0.1, 1.8 +/- 0.1, or 2.4 +/- 0.2 nmol/g, respectively, in the absence of insulin and 1.7 +/- 0.1, 4.6 +/- 0.3, or 5.5 +/- 0.6 nmol/g in its presence (10 mU/ml). Compared with its level in a control muscle, the concentration of malonyl-CoA was increased threefold in the soleus 6-8 h after denervation and remained twofold higher for > or = 48 h. In contrast, muscle contractions induced by sciatic nerve stimulation, in vivo, acutely decreased the concentration of malonyl-CoA by 30-35%. The results indicate that insulin and glucose, and probably contractile activity, regulate the concentration of malonyl-CoA in muscle.(ABSTRACT TRUNCATED AT 250 WORDS)

Utilizaram-se seis ovinos, sem raça definida, para avaliar o consumo e as digestibilidades aparentes totais da matéria seca (MS), matéria orgânica (MO), proteína bruta (PB), extrato etéreo (EE), carboidratos totais (CHO), fibra em detergente neutro (FDN), carboidratos não fibrosos (CNF), fibra em detergente ácido (FDA), celulose (CEL), hemicelulose (HCEL) e energia do feno de Arachis pintoi. Também foi determinado o balanço de nitrogênio. Os animais foram colocados em gaiolas metabólicas e receberam apenas o feno de A. pintoi mais sal mineral como componentes da dieta. O Arachis pintoi foi colhido com aproximadamente 100 dias. O fornecimento do feno foi ad libitum, sendo a quantidade calculada para permitir sobras de 20%. O experimento teve 20 dias de duração, sendo 15 dias de adaptação e cinco dias para coletas de amostras do feno, sobras, fezes e urina. Foi utilizado o óxido crômico, em duas doses diárias de 1 g cada, como indicador externo para estimar a produção fecal. Os consumos de MS e MO do A. pintoi foram 90,17 e 85,67 g/kg0,75, respectivamente. Os teores de PB, nutrientes digestíveis totais (NDT) e energia metabolizável (EM) foram, respectivamente, 14,3%, 66,4% e 2,0 Mcal/kg MS. O balanço de nitrogênio (N) foi de 12,1 g/dia e representou 40,2% de todo N consumido. As digestibilidades aparentes totais da MS, MO, PB, EE, CHO, FDN, CNF, FDA, CEL, HCEL e energia foram 64,4, 68,4, 70,0, 63,4, 68,2, 53,6, 93,3, 47,2, 62,8, 66,8 e 63,7%, respectivamente. O feno de Arachis pintoi apresentou consumo e digestibilidades dos nutrientes elevados para uma forrageira, permitindo assim fornecer nutrientes em quantidades suficientes para ganhos de peso satisfatórios, o que dá maior suporte para o uso dessa leguminosa na alimentação de ruminantes.

Recent trials with cows fed grass silage have shown significant increases in intake and in yields of milk and milk solids when the crude protein (CP) concentration in a supplementary concentrate was raised. Giving additional CP in the concentrate was a more effective strategy for improving yields of milk and milk protein than giving extra energy (Aston et al 1992). The objective of this trial was to examine the influence of changes to the amount and pattern of distribution of CP supplied in a fixed concentrate ration given with grass silage ad libitum.Fifty-five Holstein-Friesian cows in their second and subsequent lactations were given a standard diet for two weeks from calving and then were used in a continuous feeding trial from weeks 4 to 21. The cows received 5 kg of fresh concentrate daily containing 156 (LP), 245 (MP) or 338 (HP) g CP per kg dry matter (DM), MP comprised equal amounts of LP and HP. Concentrate carbohydrate sources were cereals and digestible fibre and CP was increased by a mixture of 3:1 soya:fish meals. Grass silage contained 253 g toluene DM/kg, 162 g CP/kg DM, D value in vivo 0.723, pH 3.7, fermentation acids 135 g/kg DM of which 0.85 was lactic acid and NH3-N 91 g/kg total N.

Many mature, non-pregnant, non-lactating equids are often kept in circumstances where they are expected to perform only light physical work or activity eg: a childs pony. Consequently their maintenance energy and protein needs can often be met at very restricted feed intake levels. Conversely, when they are housed during the winter months it is believed desirable to manage such animals on unrestricted ad libitum feeding regimes in order to allow the animals to exhibit their natural feed intake pattern and consume forage on a little and often basis throughout the daily feeding period. However, ad libitum access to the diet may lead to such animals becoming excessively fat. These conflicting needs of low energy and protein requirement coupled with the desirability of unrestricted access to the diet could both be met, at least in part, if a low quality forage is available ad libitum. This study examines the voluntary feed intake and apparent digestibility in vivo of a mature threshed grass hay offered ad libitum and determines its ability to meet the predicted energy and protein needs of mature ponies.

SUMMARY1. Four experiments have been carried out to determine the effect of treatment of spring-sown barley straw (var. Deba Abed) with sodium hydroxide (NaOH) solution and subsequent neutralization with hydrochloric acid (HCl) on its digestibility and intake by sheep.2. In Experiment 1, chopped straw was mixed with 4·5 or 9·0 g NaOH in 200 or 800 ml water/100 g, straw, and after 24 hr neutralized with HCl and left for a further 24 hr before being offered to castrated male sheep (wethers) in a maintenance diet containing 35% concentrates. Organic-matter digestibility of straw significantly increased, by 8 and 11 percentage units respectively, after treatment with 200 ml solution containing 4·5 and 9·0 g NaOH. Volume of solution did not affect digestibility.3. In Experiment 2, straw treated as in Experiment 1 was given ad libitum. The highest intake was for straw treated with 200 ml solution containing 4·5 g NaOH/100 g straw, treatment with 9·0 g NaOH giving a significantly lower intake but higher than that of untreated straw.4. In Experiment 3, the in vitro digestibility of milled straw, treated as in Experiments 1 and 4, increased with increasing volumes of solution up to 120 ml/100 g straw, but the response to successive increments of NaOH declined progressively.5. In Experiment 4 chopped straw was mixed with 4·5, 6·75 or 9·0 g NaOH in 30, 60 or 120 ml water/100 g straw and offered as in Experiment 1. Treatments significantly increased straw digestibility, by 8 to 16 percentage units. Increasing the volume of water from 30 to 60 ml significantly improved digestibility, by 5 percentage units at the two lower levels of NaOH. The response to an increase in the level of NaOH was less, and inconsistent. In vitro and in vivo digestibilities were significantly correlated, but it is concluded that the in vitro technique used overestimates the digestibility of treated straw.

Variability in bodyweight (BW) among pigs complicates the management of feeding strategies and slaughter. Including variability among individuals in modelling approaches can help to design feeding strategies to control performance level, but also its variability. The InraPorc model was used to perform simulations on 10 batches of 84 crossbred pigs each to characterise the effect of feeding strategies differing in amino acid supply or feed allowance on the mean and variation in growth rate. Results suggested that a feed restriction reduces the coefficient of variation of BW at first departure for slaughter (BW1) by 34%. Growth performance obtained from an in silico simulation using ad libitum and restricted feeding plans was compared with results obtained in an in vivo experiment on a batch of 168 pigs. Pigs were offered feed ad libitum or were restricted (increase in feed allowance by 27 g/day up to a maximum of 2.4 and 2.7 kg/day for gilts and barrows, respectively). A two-phase feeding strategy was applied, with 0.9 and 0.7 g of digestible lysine per MJ of net energy (NE) in diets provided before or after 65 kg BW, respectively. Actual growth was similar to that obtained by simulation. Coefficient of variation of BW1 was similar in vivo and in silico for the ad libitum feeding strategy but was underestimated by 1 percentage point in silico for the restriction strategy. This study confirms the relevance of using simulations performed to predict the level and variability in performance of group housed pigs.
