Triglycerides are the main lipid components that are stored in adipose tissue for energy sources. Increased triglyceride levels can lead to cardiovascular disease. One way to lower triglyceride levels is diet. The intermittent fasting diet is one of the diets that can be done, where this diet is a diet with time and calorie restrictions. The intermittent fasting diet is divided into 3 methods, namely Time restricted feeding, Alternate-day fasting and Modified fasting. This study was conducted to see the effect of an intermittent fasting diet on triglyceride levels. This research is an experimental study using white rats of Sprague Dawley strain as experimental. White rats will be divided into 3 groups of intermittent fasting diet and 1 control group and given a diet according to the group for 1 month. Weights and blood draws were performed at the beginning and end of the study, then the results of the intermittent fasting diet intervention were compared with the control group. There was a significant reduction in body weight with a p value of 0.000 and a significant decrease in triglyceride levels with a p value of 0.035 (p <0.05). Keywords : Triglycerides, Intermittent Fasting Diet, Weight loss.

Background: The individual effects of nicotine and caffeine have been reported in previous studies but their combined effect on tooth movement needs to be elucidated. The objective of this study was to evaluate the combined effect of nicotine and caffeine on the magnitude of orthodontic tooth movement (OTM) in rats. Material and Methods: This experimental study was conducted on Sprague-Dawley rats (Animal House and Pathology Laboratory; Post Graduate Medical Institute, Lahore) in the department of Orthodontics, de’Montmorency College of Dentistry, Lahore from 8th July 2014 to 8th January 2015. Forty male Sprague-Dawley rats were divided into four equal groups: Control group (CR), nicotine group (NT), caffeine group (CF) and combined nicotine and caffeine group (CNC). Closed coil nickel titanium (NiTi) spring was placed between incisor and maxillary molar. Nicotine group (NT) was treated by intraperitoneal injections of nicotine. Caffeine was given to caffeine group and Combined nicotine and caffeine group (CNC) was treated in the same way as individual nicotine and caffeine groups daily for 14 days. All the rats were sacrificed on 15th day. Magnitude of the orthodontic tooth movement was measured using digital Vernier caliper. Means and standard deviation were calculated for orthodontic tooth movement. One-way ANOVA was used to determine the mean difference in OTM. Post hoc Tukey test was used for multiple comparisons among the groups. Results: The mean orthodontic tooth movement (OTM) was 0.32 mm ± 0.05 in control group, 0.56 mm ± 0.04 in nicotine group, 0.52 mm ± 0.034 in caffeine group and 0.8 mm ± 0.06 in combined NC group, respectively. The difference between mean OTM among the groups was statistically significant (P-value <0.001). The mean OTM in CNC group was significantly higher as compared to other groups (CR, NT, CF, NT) (P-value <0.001). Conclusions: In rats, the combined use of nicotine and caffeine results in greater orthodontic tooth movement as compared to their individual use.

Objective: To compare and evaluate the effects of broiler chicken fed with commercially offered feed and chicken fed with organic diet on BMI and weight gain in Sprague Dawley rats. Study Design: A randomized controlled trial. Place and Duration of Study: Multi-Disciplinary Laboratory of Department of Biochemistry and Molecular Biology, Army Medical College, Rawalpindi, and collaborated with Quaid-e-Azam University, Islamabad from November 2017 to April 2019. Methodology: Ninety male early weaned Sprague Dawley rats were arbitrarily assorted into three groups (n=30). Group I control rats were nourished on standard pelleted diet. Group II rats were nourished with organic chicken meat along with their standard pelleted diet. Group III rats were nourished with broiler chicken meat along with their standard pelleted diet. BMI and weight gain were estimated. All variables were calculated as Mean ± SD values. One-way ANOVA was applied to determine the significance among groups followed by Tuckey’s HSD post hoc test. p value ≤ 0.05 was considered significant. Results: The current study demonstrated significant increase in BMI (p ≤ 0.001) and weight gain (p ≤ 0.001) in both experimental groups as compared to control group. Conclusion: Based on the findings of our study we propose that broiler chicken meat consumption could be the probable cause of weight imbalances and out of proportion gain of weight and growth in experimental rats.

Objective: To observe the difference of testosterone levels in adult male Sprague Dawley rats treated with combination of cisplatin and vitamin E compared to those treated with cisplatin only. Material & Methods: We used 24 adult male Sprague Dawley rats weight 200–300 grams and randomly assigned into 4 groups (n=6). Rats in negative control group (NC) were given intraperitoneal normal saline injection, while the positive control (PC) group were injected with cisplatin 5 mg/kgBW at the end of the 3rd week. Two other groups, P1 and P2, were injected with cisplatin 5 mg/kgBW and given vitamin E orally 50 mg/kgBW and 200 mg/kgBW, respectively. Cardiac blood was aspirated at the end of the 7th week and processed for analysis of testosterone levels. Results: We recorded a significantly lower testosterone levels in rats treated only with cisplatin 5 mg/kgBW (CP) compared to those in CN group (p=0.006), and those receiving combination of cisplatin and vitamin E 50 mg/kgBW (p=0.003) and 200 mg/kgBW (p=0.001). Though not significant, testosterone levels were higher in P2 group than in P1 group (p=0.702). Conclusion: Exposure to cisplatin can lower testosterone levels in white rats, and the administration of vitamin E gives protection against such effect.

The study was designed to investigate the effect of berberine toxicity on liver enzymes, renal function, serum uric acid and blood cell counts of Sprague-Dawley rats. For this purpose, twenty Sprague-Dawley rats were divided in two groups. Group A (n=10) was given distilled water and Group B (n=10) wassupplemented berberine hydrochloride with dose of 75 mg/kg body weight/day. 24 weeks post-treatment, blood samples were collected by cardiac puncture from both groups and tested for Hb (%), WBC, RBC and platelet counts, SGPT, alkaline phosphatse, serum uric acid, blood urea and serum creatinine. All thenumerical data were expressed as mean± SD. The values in two groups were analyzed with help of unpaired t-test. A p-value ˂ 0.05 was considered significant. After 24 weeks of treatment with berberine hydrochloride, no significant differences were observed in Hb%, platelet count, liver enzymes and RFTs between twogroups. There was a significant increase in RBC count and reduction in serum uric acid and WBC count in experimental group as compared to control group. There were no significant untoward effects of berberine on platelet count, LFTs and RFTs of Spargue-Dawley rats except neutropenia.

Cedera sel yang luas pada luka bakar melepaskan banyak mediator inflamasi. Respon tubuh yang berlebihan pada keadaan tersebut akan memperberat kerusakan sel yang sudah terjadi. Nurtrisi (lemak) yang cukup seperti omega 3 dapat membantu proses penyembuhan luka serta mencegah infeksi pada luka supaya tidak menjadi luka kronis. Maka dari itu peneliti memilih judul penelitian tentang efektifitas kombinasi suplementasi omega 3 dengan restriksi kalori terhadap penyembuhan luka bakar pada tikus Sprague Dawley, ditinjau dari VEGF dan Re-epitelisasi. Untuk mengetahui tingkat efektifitas suplementasi Omega 3 dan retriksi kalori pada penyembuhan luka bakar. Peneliti dalam melakukan penelitian menggunakan metode penelitian eksperimental dengan menggunakan 1 kelas kontrol dan 3 kelas perlakuan tikus Sprague Dawley, yang diinduksi luka bakar kemudian diberi perlakuan, dan diamati secara berkala. Hasil penelitian didapatkan perbedaan yang signifikan antara kelompok kontrol dan kelompok perlakuan yang di uji dengan one way ANOVA mempunyai nilai p = 0,000 (p < 0,05), dan uji Mann-Whitney didapatkan perbedaan yang signifikan antara kelompok perlakuan 1, 2, 3, dan kelompok kontrol p = 0,025; 0,004; 0,004; 0,004; 0,004, nilai p < 0,05. Dan uji Games-Howell ditunjukkan pada kolom rerata ± SD jika memuat huruf yang  berbeda berarti ada perbedaan yang bermakna (p-value <0.05) K1 = 23,3587a ± 0,57556; P1 = 17,1015b ± 2,60108; P2 = 124,1503c ± 15,30168; P3 = 237,5053d ± 0,78263. Kesimpulan yang peneliti dapat bahwa kombinasi suplementasi omega 3 dengan retriksi kalori berpengaruh signifikan terhadap penyembuhan luka bakar pada tikus Sprague Dawley. Therefore the researchers chose the title of the study of the effectiveness combination omega-3 supplementation with calorie restriction for burns in Sprague Dawley rats, in terms of VEGF and Re-epithelialization. To determine the effectiveness of Omega 3 supplementation and calorie restriction when healing burns. The researchers using experimental research methods with 1 control and 3 training classes of Sprague Dawley mice, which were induced by burns were then observed periodically. The results showed a significant difference between the control group and the treatment group tested by one way ANOVA having a value of p = 0,000 (p <0.05), and the Mann-Whitney test found a significant difference between the treatment groups 1, 2, 3, and the control group p = 0.025; 0.004; 0.004; 0.004; 0.004, p value <0.05. Faloak (Sterculia quadrifida R.Br) is a medicinal plant used by people in the province of East Nusa Tenggara to treat hepatitis. This study aims to determine the acute toxicity of faloak (Sterculia quadrifida R. Br) bark ethanol extract in Sprague-Dawley (SD) male white rats. The method used is experimental using post-test only control group design. In this study the parameters used are body weight, clinical symptoms, clinical biochemistry (ALT & AST), histopathology of liver organ and mortality in rats. Twenty male Sprague-Dawley rats were randomly assigned to 5 groups, group 1 as control, group 2 was administered orally faloak bark ethanol extract at dose of 40 mg / kg body weight, group 3 at dose of 200 mg / kg body weight, group 4 at dose of 1000 mg / kg body weight and group 5 at dose of 5000 mg / kg body weight. Results of observation for 24 hours there are no dead rats, so the value of LD50 extract ethanol faloak is > 5,000 mg / kg body weight. Observations were conducted for 14 days to observed delayed occurrence of toxic effects. Oral administration of the faloak bark ethanol extract reduced the level of AST and ALT. The histopathology observation of rats liver cells showed liver cell necrosis at treatment doses of 200 - 5,000 mg / kg body weight.

The objectives of this study were to compare the antidepressant effects between endurance and resistance exercise for optimizing interventions and examine the metabolomic changes in different types of skeletal muscles in response to the exercise, using a rat model of chronic unpredictable mild stress (CUMS)-induced depression. There were 32 male Sprague-Dawley rats randomly divided into a control group (C) and 3 experimental groups: CUMS control (D), endurance exercise (E), and resistance exercise (R). Group E underwent 30 min treadmill running, and group R performed 8 rounds of ladder climbing, 5 sessions per week for 4 weeks. Body weight, sucrose preference, and open field tests were performed pre and post the intervention period for changes in depressant symptoms, and the gastrocnemius and soleus muscles were sampled after the intervention for metabolomic analysis using the 1H-NMR technique. The results showed that both types of exercise effectively improved the depression-like symptoms, and the endurance exercise appeared to have a better effect. The levels of 10 metabolites from the gastrocnemius and 13 metabolites from the soleus of group D were found to be significantly different from that of group C, and both types of exercise had a callback effect on these metabolites, indicating that a number of metabolic pathways were involved in the depression and responded to the exercise interventions.

Methanol poisoning is a hazardous intoxication characterized by visual impairment and formic acidemia. The therapy for methanol poisoning is alcohol dehydrogenase (ADH) inhibitors to prevent formate accumulation. Ranitidine has been considered to be an inhibitor of both gastric alcohol and hepatic aldehyde dehydrogenase enzymes. This study aimed at testing ranitidine as an antidote for methanol acute toxicity and comparing it with ethanol and 4-methyl pyrazole (4-MP). This study was conducted on 48 Sprague-Dawley rats, divided into 6 groups, with 8 rats in each group (one negative control group [C1], two positive control groups [C2, C3] and three test groups [1, 2 and 3]). C2, C3 and all test groups were exposed to nitrous oxide by inhalation, then, C3 group was given methanol (3 g/kg orally). The three test groups 1, 2 and 3 were given ethanol (0.5 g/kg orally), 4-MP (15 mg/kg intraperitoneally) and ranitidine (30 mg/kg intraperitoneally), respectively, 4 hours after giving methanol. Rats were sacrificed and heparinized, cardiac blood samples were collected for blood pH and bicarbonate. Non-heparinized blood samples were collected for formate levels by high performance liquid chromatography. Eye balls were enucleated for histological examination of the retina. Ranitidine corrected metabolic acidosis (p = .025), decreased formate levels (p = .014) and improved the histological findings in the retina induced by acute methanol toxicity.

P165 Aldosterone through its interaction with the mineralocorticoid receptor (MR) plays a critical role in the development of hypertension and cardiovascular injury (CVI). Normally, MR is protected by 11β-hydroxysteroid dehydrogenase (11β-HSD) which inactivates glucocorticoids preventing their binding to MR. We hypothesis that if activation of MR by either aldosterone or glucocorticoids induces hypertension and CVI, then the inhibition of 11β-HSD with glycyrrhizin (GA), a natural inhibitor of 11β-HSD, should induce damage similar to that observed with aldosterone. Sprague-Dawley rats were uninephrectomized, and treated for 4 weeks with 1% NaCl (in drinking water) for the control group, 1% NaCl + aldosterone infusion (0.75 μg/h), or 1% NaCl + GA (3.5 g/l in drinking water). After 4 weeks, aldosterone and GA caused significant increases in blood pressure compared to control rats ([mean ± SEM] 211± 9, 205 ± 12, 120 ± 9 mmHg, respectively, p<0.001). Both aldosterone- and GA-treated rats had a significant increase in proteinuria (152.2 ± 8.7 and 107.7 ± 19.5 mg/d, respectively) versus controls (51.2 ± 9.5 mg/d). There was a significant increase (p<0.001) in heart to body weight ratio in the rats treated with aldosterone or GA compared with control (3.92 ± 0.10, 3.98 ± 0.88, and 3.24 ± 0.92 mg/g, respectively). Hearts of GA and aldosterone treated rats showed similar histological changes consisting of biventricular myocardial necrosis and fibrinoid necrosis of small coronary arteries and arterioles. These data suggests that in rodents activation of MR by either aldosterone or corticosterone leads to severe hypertension, vascular injury, proteinuria and myocardial infarction. Thus, 11β-HSD plays an important role in protecting the organism from injury.

PURPOSE: To investigate synergistic suppression of donor liver pre-perfusion with recipient serum (RS) and cobra venom factor (CVF) treatment on hyperacute rejection (HAR) following liver xenotransplantation. METHODS: Guinea-pigs (GP, n=24) and Sprague-Dawley rats (SD, n=24) were recruited. Before transplantation, serum was collected from SD rats and used for preparation of inactivated complements. GP and SD rats were randomly assigned into four groups (n=6), respectively: RS group, CVF group, RS+CVF group and control group. Orthotopic liver xenotransplantation was performed with modified two-cuff technique. The survival time and liver function of recipients, morphological and pathological changes in rat livers were investigated. RESULTS: There was no piebald like change in the recipient livers in all experiment groups. The survival time of recipients in all experiment groups was longer than that in control group (p<0.05). Moreover, the survival time in the RS+CVF group was markedly longer than that in the RS group (p<0.01) and CVF group (p<0.05). The serum ALT level in all experiment groups were lower than that in the control group (p<0.05). Furthermore, the ALT level in the RS+CVF group was significantly lower than that in the CVF group (p<0.05) and RS group (p<0.01). The histological damages were significantly improved when compared with the control group, and the histological damages in the RS+CVF group were milder than those in the remaining groups (p<0.05) CONCLUSION: Pre-perfusion of donor liver with recipient serum and cobra venom factor treatment can exert synergistic suppressive effects on the hyperacute rejection following liver xenotransplantation.
