Polyphenol production in Kim Tuyen green tea extract was surveyed in this study by optimizing appropriate conditions for Viscozyme L. The optimum amount of water added was 1:20 (w material: w water), the ratio 0.06 v enzyme: w extract, Viscozyme L showed the highest activity at 40 - 450C in 90 min at 60 rpm. Total polyphenol in green tea extract collected from the conditions above reached its peak at 23.49% of dry content (w/w). The total polyphenol and antioxidant activities by carried out by DPPH (1,1-diphenyl-2-picrylhydrazyl) assay of green tea extract (collected by the conditions above) treated with Viscozyme L (23.98%) showed IC50 value at 276.98μL, which was significantly higher antioxidant activities of those treated with non-enzyme extraction (15.75% w/w), with IC50 value at 328.98μL.

Green tea (Camellia sinensis) has many biological activities and may promote diabetic wound healing by regulation of circulating hypoxia responsive microRNAs (HRMs) which triggers the wound repairing process in diabetic and nondiabetic wounds. Thus, in this study, the potential effects of green tea extract (GTE) on the expression of miRNAs; miR-424, miR-199a, miR-210, miR-21, and fibrogenitic markers; hydroxyproline (HPX), fibronectin (FN), and nitric oxide (NO) were evaluated in wounds of diabetic and nondiabetic rats. The animals were topically treated with vaseline, 0.6% GTE, and 5%w/w povidone iodine (standard control). HPX, FN, and NO levels and microRNAs, miR-424, miR-210, miR-199a, and miR-21, were estimated in wound tissues using colorimetric, immunoassay, and molecular PCR analysis. In vitro analysis was performed to estimate active constituents and their antioxidant activities in methanolic green teat extract (GTE). Wounds treated with green tea, a dose of 0.6, healed significantly earlier than those treated with standard vehicle and vaseline treated diabetic wounds. Higher expressions of HRMs, miR-199a, and miR-21, and lower expression of HRMs, miR-424 and miR-210, were significantly reported in tissues following treatment with green tea extract compared to standard control vehicle. The tissues also contained more collagen expressed as measures of HPX, FN, and NO and more angiogenesis, compared to wounds treated with standard control vehicle. Diabetic and nondiabetic wounds treated with green tea (0.6%) for three weeks had lesser scar width and greater re-epithelialization in shorter periods when compared to standard control vehicle. Expression of HRMs, miR-199a, miR-21, and HRMs and miR-424 and miR-210 correlated positively with HPX, fibronectin, NO, better scar formation, and tensile strength and negatively with diabetes. In addition to antidiabetic and antioxidant activities of green tea components, GTE showed angiogenesis promoting activity in diabetic wound healing. In conclusion, Camellia sinensis extracts in a dose of 0.6% significantly promote more collagen and fibronectin deposition with higher expression of NO, promoting angiogenesis process via molecular controlling of circulating hypoxia responsive microRNAs: miR-424, miR-210, miR-199a, and miR-21 in diabetic and nondiabetic wounds. Our results support a functional role of circulating hypoxia responsive microRNAs: miR-424, miR-210, miR-199a, and miR-21 as potential therapeutic targets in angiogenesis and vascular remodeling in diabetic wound healing.

Okra (Abelmoschus esculentus L.) seeds and green tea (Camellia sinensis) have been known have many beneficial functions for human’s health, because of the high antioxidant contents and anti-cholesterol activities that can prevent cardiovascular disease. This research is aimed to make the formulation of healthy drink from okra seed juice with the addition of green tea extract. Green tea used was selected from green tea that has the best antioxidant activity from ten samples of green tea in the market. The best antioxidant activity data was found in Jawa tea extract with IC50 85.28±0.21 µg/mL which was formulated with the extract of okra seeds, made in the ratio of 1: 4. Selected healthy drinks were tested for physical characteristics, antioxidant activities, and sensory tests, and have antioxidant activity IC50 222.16±1.38 µg/mL. Further research was to analyze the potential of drinks in reducing cholesterol in vitro and in vivo ways. In vitro, 500 ppm drinks were able to inhibit cholesterol up to 47.55±0.76% and in vivo, there was a significant decrease in cholesterol levels after drink for 21 days with cholesterol reduction to 35.50±2.37%, triglycerides decrease 19.39±3.10%, LDL decrease 34.76±2.62%, and an increase of HDL to 94.74±16.53%.

Green tea is a kind of plant that have an active polyphenol substance. Polyphenol is a group of compounds that has a role as a natural antioxidant. Solvent comparison used affects the extracted polyphenol content. This study aims to compare the total polyphenol content in green tea from maceration method with solvent concentration variation of 30%, 50%, and 70%. In this research, green tea is extracted by using the maceration method with ethanol – water solvent concentration of 30%, 50%, and 70%. The total polyphenol content determination in green tea was conducted by using the Follin-Ciocalteu method by UV-Vis spectrophotometre, and using gallic acid as a standard. According to this research, the yield calculation of ethanol extract in green tea with the ethanol concentration of 30%, 50%, and 70% are 41.220%; 39.979 %; and 47.971 %. The total polyphenol content test of green tea with the ethanol concentration of 30%, 50%, and 70% are 11.294 mg GAE/100 g; 16.377 mg GAE/100g; and 15.640 mg GAE/100 g. The anova statistical analysis result shows that there is a significant difference of total polyphenol content of green tea extract with ethanol concentration 30% and 50%, also there is no significant difference in the ethanol concentration of 70%. The highest result of total polyphenol of green tea extract with the ethanol concentration of 50%. It is suggested to conduct a further study on the type of the polyphenol substance in green tea.

This brief review provided evidence for the role of green tea extract (GTE)/zinc oxide nanoparticles (ZnO NPs) in a variety of biomedical applications against the toxic effect of monosodium glutamate (MSG). MSG is used to enhance the taste, however it causes oxidative stress in the long-term. Many effects of MSG consumption on the brain, obesity, sex organs, and metabolism have been verified. This review covered the effect of GTE/ZnO NPs on many different organs including the liver, kidney, heart, spleen, testis, brain, and pancreas after being exposed to MSG. The review indicated that the toxicity induced by MSG could be restored by GTE/ZnO NPs in different organs. Accordingly, the green nanoparticles could be attended as a futuristic approach to be used against any toxic substance.
