Auxotrophic mutants were obtained by exposing Candida fennica FTPT/CCT-8903 to ultraviolet light and treating the cells with nystatin in an enrichment procedure. The mutants were used for protoplast fusion. The highest protoplast regeneration frequency was obtained when cells were treated with 10 mg NovoZym 234/mL in the presence of 0.8 M KCl for 60 min. Fusion frequency was in the range 5.84 × 10−5 to 7.2 × 10−5. Five prototrophic hybrid colonies, designated S9, S15, S17, S18, and S25, were isolated, and their DNA contents were measured. These measurements indicated that the S17 hybrid is haploid, S9 and S18 are polyploid, and S15 and S25 are both aneuploid. ABSTRACT A study of calcium and phosphorus metabolism has been carried out on 13 acromegalic patients, in various stages of the disease. This study was repeated in nine patients following implantation of the pituitary gland with 90Y and in another two patients after deep X-ray therapy and suction removal of a pituitary adenoma respectively. Increased urinary calcium and phosphorus excretion was found in all the patients in whom the disease was in an active phase of evolution. The calcium tolerance test revealed a marked decrease of calcium retention in certain subjects, while in others, calcium retention was found to be increased. Such changes were not found in patients in whom the disease was in a quiescent phase. The blood calcium, phosphorus and alkaline phosphatase were found to be either normal or slightly increased. The implantation of the pituitary gland with 90Y and deep X-ray therapy induced a marked decrease of hypercalciuria, both spontaneous and induced, and of hyperphosphaturia, together with a definite improvement, of the clinical picture and glucose metabolism. It is concluded that the changes in calcium and phosphorus metabolism described above depend either directly or indirectly on a pituitary factor. They may therefore prove a reliable index for assessing pituitary growth hormone activity.

ABSTRACT The response of plasma cortisol, corticosterone and non-protein-bound cortisol in the extracorporeal circulation was investigated in 14 patients. The pre-perfusion levels of plasma cortisol, corticosterone and non-protein-bound cortisol were significantly elevated. During and immediately after perfusion, the levels of cortisol and corticosterone were found to decrease significantly from the pre-perfusion levels, while the percentage of non-protein-bound cortisol was shown to increase significantly. This indicates a marked decrease in cortisol binding capacity of plasma during extracorporeal circulation. Moreover in 200 plasma samples, it was demonstrated that the cortisol level increased markedly and the cortisol binding capacity decreased slightly during and shortly after major surgery without perfusion. It is concluded that stressful situations in major surgery with or without perfusion are associated with markedly increased levels of biologically active non-protein-bound cortisol. The elevated level of non-protein-bound cortisol in surgery seems to be dependent on the increase in the level of plasma cortisol as well as on the decrease in the cortisol binding capacity of plasma. Although the increased plasma cortisol plays the most important role in surgery with no perfusion, the decreased cortisol binding capacity may be the more effective factor involved during perfusion.

ABSTRACT Adult baboons (5 males and 5 females) were exposed to immobilization stress by being strapped to a table in a horizontal position for 2 h. In females the experiment was performed during both the follicular and luteal phase. Peripheral blood was withdrawn at frequent intervals, the first sample just before immobilization, and the last one 3 days later. A number of steroids were measured in blood plasma samples by radioimmunoassay (17-hydroxypregnenolone, 17-hydroxyprogesterone, pregnenolone, testosterone, dihydrotestosterone, progesterone, 20α-dihydroprogesterone, oestrone, oestradiol) or competitive protein binding (cortisol) techniques. The cortisol levels exhibited a marked increase in both sexes. This increase was observed already during the immobilization and lasted for approximately 24 h. A similar, even more pronounced increase was seen in 17-hydroxypregnenolone, 17-hydroxyprogesterone and pregnenolone levels. A marked, long-lasting (72 h) decrease of testosterone and dihydrotestosterone levels was a consistent finding in male baboons. This was not observed in the females which, on the other hand, exhibited a marked decrease (duration 48 h) of progesterone and 20α-dihydroprogesterone levels during the luteal phase, and a significant decrease (duration > 24 h) of oestradiol and oestrone concentrations during the follicular phase. It is concluded that stress has a marked inhibitory action on gonadal function both in male and female baboons. In females the inhibition of steroidogenetic function is exerted both on the ovarian follicles and on the corpus luteum.

A large number of compounds with glycosidic linkages have been tested as substrates for the ,8-glucosidase of Stachybotrys atm. The enzyme appears to be specific for ,8-glucosides and all configurational alterations to the D-glucopyranose ring or substitutions in it lead to non-substrates; phenyl-fl-thioglucoside is a substrate, however. Aryl-,8-glucosides have a higher affinity for the enzyme than alkyl-,8-glucosides and no hydrolysis of cellobiose by the enzyme can be demonstrated. arrha-Substitution in aryl-fl-glucosides leads to a marked decrease in the affinity between enzyme and substrate.

To identify potential mechanisms for hepatic removal of circulating glutathione (GSH) conjugates, uptake and metabolism of S-2,4-dinitrophenylglutathione (DNP-SG) were examined in isolated perfused livers from rat and guinea pig. Guinea pig livers perfused with 5 mumol of DNP-SG in a recirculating system (50 microM initial concn) rapidly cleared the conjugate from the perfusate (half time 3.7 min), whereas clearance was considerably slower in rat liver (half time 35 min). Disappearance of DNP-SG from the perfusate was accompanied by a simultaneous appearance of DNP-SG and its metabolites in bile. Addition of acivicin, an inhibitor of gamma-glutamyltransferase (gamma-GT), to the perfusate resulted in a marked decrease in DNP-SG clearance by guinea pig liver but had no effect in rat liver, suggesting that in the guinea pig this process is largely dependent on sinusoidal gamma-GT activity. However, even in the presence of acivicin, rat and guinea pig livers removed nearly one-half of the administered DNP-SG from the recirculating perfusate over 30 min. High concentrations of DNP-SG were found in bile (up to 3.7 mM), indicating that the liver is capable of transporting the intact conjugate from the circulation. When rat livers were perfused with higher concentrations of DNP-SG (100 and 250 microM), biliary excretion of DNP-SG increased dose dependently, with concentrations in bile reaching 10 mM at the higher dose. An analysis has been made of the organ weights of normal rabbits exposed to a constant illumination having none of the shorter ultraviolet rays and of other rabbits kept in darkness for periods of 2 to 12 weeks. The environment of constant illumination was associated with a well marked decrease in the relative weights of most organs, and in certain instances this occurred when the organ weights of the controls were becoming increasingly large. There was also an associated effect of stabilization of organ weight. The majority of the organs of rabbits caged in constant darkness also showed a tendency toward decreased and stabilized weights, but these effects were less pronounced than in the rabbits caged under conditions of constant illumination. A notable exception to this general result was the weight of the liver which was markedly increased. The results of this experiment support the conception that there is a relationship between light and the physical state of the animal organism which may be expressed in the concrete form implied by the trend or direction of organ weight.

AbstractA counterargument to the importance of climate change for malaria transmission has been that regions where an effect of warmer temperatures is expected, have experienced a marked decrease in seasonal epidemic size since the turn of the new century. This decline has been observed in the densely populated highlands of East Africa at the center of the earlier debate on causes of the pronounced increase in epidemic size from the 1970s to the 1990s. The turnaround of the incidence trend around 2000 is documented here with an extensive temporal record for malaria cases for both Plasmodium falciparum and Plasmodium vivax in an Ethiopian highland. With statistical analyses and a process-based transmission model, we show that this decline was driven by the transient slowdown in global warming and associated changes in climate variability, especially ENSO. Decadal changes in temperature and concurrent climate variability facilitated rather than opposed the effect of interventions.

The COVID-19 pandemic has led to delays in cancer diagnosis, in part due to postponement of cancer screening. We used Google Trends data to assess public attention to cancer screening during the first peak of the COVID-19 pandemic. Search volume for terms related to established cancer screening tests (“colonoscopy,” “mammogram,” “lung cancer screening,” and “pap smear”) showed a marked decrease of up to 76% compared to the pre-pandemic period, a significantly greater drop than for search volume for terms denoting common chronic diseases. Maintaining awareness of cancer screening during future public health crises may decrease delays in cancer diagnosis.
