Type 2 diabetes (T2D) is one of the fast growing diet-related chronic diseases throughout the world. Tea contains several bioactive compounds, some of which render health benefits. Black tea polysaccharides (BTPS), extracted from low grade quality tea leaves, after processing, were examined for their in-vitro antidiabetic activities (α-glucosidase inhibitory and glucose uptake activities), chemical compositions (yield, monosaccharides, amino acids, and minerals), and antioxidant activities as well as toxicity (cytotoxicity and genotoxicity). In addition, 50% lethal dose (LD50) for BTPS was determined using an acute toxicity test to assess the safe use of it as a dietary ingredient. BTPS had strong α-glucosidase inhibitory activity with IC50 value of 3.4 µg/mL. This was much lower than that of the positive control, pharmaceutical glucosidase inhibitor acarbose with IC50 of 1.06 mM. BTPS also increased glucose uptake into the adipocyte differentiated 3T3-L1 MBX cells. Neither cytotoxic nor mutagenic effects were found for BTPS. The LD50 of BTPS for acute toxicity demonstrated that it was safe to use. The present work suggests that BTPS can be used as an antidiabetic dietary ingredient without posing any potential health risk.

Procuring valuable bioactive compounds from low quality green tea is of great interest. Green tea polysaccharides (GTPS), obtained from low quality tea leaves, were examined for their yield, monosaccharide composition, total phenolic content, antioxidant potential, and structures as well as molecular weight distribution and in vitro anti-diabetic activities. GTPS were also evaluated for their toxicity by performing cytotoxicity and genotoxicity. Additionally, 50% lethal dose (LD50) for GTPS was determined using an acute toxicity test to assess the safe use of it as a dietary supplement. Monosaccharides of GTPS were mainly composed of xylose, glucose, ribose, galactose, arabinose, and glucuronic acid. GTPS exhibited 91.86% α-glucosidase inhibition at 0.5 mg/mL concentration. The α-glucosidase inhibitory activity of GTPS was higher than that of pharmacological glucosidase inhibitor, acarbose, which exhibited only 47.86% inhibition at 2.5 mg/mL concentration. Neither cytotoxic nor no mutagenic effects were found for GTPS. The acute toxicity also showed that LD50 of GTPS was greater than 5,000 mg/kg. The present work suggests that GTPS can be considered as an anti-diabetic dietary supplement without posing any potential health risk.

In the present study, we aimed to develop a novel fermented tea (NFT) product and to evaluate their in vitro antioxidant potential and chemical composition. We found that NFT contained a high level of total phenolic compounds (102.98 mg gallic acid equivalents/g extract) and exhibited diverse antioxidant activities, such as scavenging of 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and hydroxyl radicals, as well as reducing power. The total catechins in NFT were comparable to those of Lipton black tea (LBT), but lower than those of Boseong green tea (BGT) or Tieguanyin oolong tea (TOT). Among all catechins tested, epigallocatechin (EGC) and epigallocatechin-3-O-gallate (EGCG) were the predominant compounds in NFT. In particular, the contents of total theaflavins (TFs), theaflavin (TF), theaflavin-3-gallate (TF3G), and theaflavin-3’-gallate (TF3’G) in NFT were significantly higher than that of BGT, TOT, or LBT. NFT had the highest level of total essential amino acid and γ-aminobutyric acid (GABA) compared with BGT, TOT and LBT. Furthermore, the sensory evaluation results showed that NFT had satisfactory color, aroma, taste, and overall acceptability scores. Our results highlight the potential usefulness of this novel fermented tea as a nutraceutical food/ingredient with special functional activities.

Nature has generously offered a wide range of herbs (e.g., thyme, oregano, rosemary, sage, mint, basil) rich in many polyphenols and other phenolic compounds with strong antioxidant and biochemical properties. This paper focuses on several natural occurring phenolic acids (caffeic, carnosic, ferulic, gallic, p-coumaric, rosmarinic, vanillic) and first gives an overview of their most common natural plant sources. A summary of the recently reported antioxidant activities of the phenolic acids in o/w emulsions is also provided as an in vitro lipid-based model system. Exploring the interfacial activity of phenolic acids could help to further elucidate their potential health properties against oxidative stress conditions of biological membranes (such as lipoproteins). Finally, this review reports on the latest literature evidence concerning specific biochemical properties of the examined phenolic acids.

Dialium angolense is used in Bagira for its various medicinal properties particularly in the management of infectious diseases. In this study, the methanol and aqueous extracts of leaves and fruits were evaluated for their in vitro antioxidant and antimicrobial properties and their in vivo toxicity on Cavia porcellus. The major phytochemical classes of extracts were screened using standard in-tube reactions. The antimicrobial study was tested on Candida albicans, Escherichia coli, Salmonella typhi, Staphylococcus aureus and Streptococcus pneumoniae using agar well diffusion and dilution methods, while the antioxidant activity was evaluated by a DPPH assay. For the acute toxicity study, animals (6/group) were orally given in a single dose 5000, 1000 or 15000 mg of extract/kg body weight (BW) then observed for 14 days. In sub-acute toxicity assays, 150 or 300 mg/kg BW/day were orally given, and animals observed for 28 days. Total phenolics and total flavonoids contents ranged 1.19 to 1.61 mg GAE.g-1 and 0.45 to 1.01 mg QEg-1, respectively. The extracts presented antioxidant activity with IC50 ranging 4.9 to 6.9 µg/mL. The minimal inhibitory concentration (MIC) on tested strains ranged from 1.9 to 500 µg/mL with the aqueous extract of fruits as a most active extract: MIC=1.9 µg/mL on E. coli and C. albicans. No signs of toxicity were noted during the acute and sub-acute toxicity assessments, suggesting a maximal tolerate doses (MDT) and LD50 > 15000 mg/kg BW. This study highlights the antioxidant and antimicrobial activities of Dialium angolense and suggests that further studies be directed towards the isolation of active compounds.

Abstract Background Nonetheless curcumin has potential health benefits, its low bioavailability limits the application of conventional turmeric extract with curcumin as major curcuminoid. This is a comparative study to assess the stability, bioaccessibility and biological activity of BDMC in standardized C. longa extract (REVERC3) relative to curcumin in regular turmeric extract (RTE). Here we report the preparation of a standardized Curcuma longa extract (REVERC3™) standardized to contain 75 ± 5 w/w % bisdemethoxycurcumin (BDMC), 1.2 ± 0.8 w/w % curcumin and 10 ± 5 w/w % demethoxycurcumin (DMC). The turmeric extracts were subjected to in vitro gastrointestinal digestion and the curcuminoids in undigested and digested samples were analyzed using HPLC to determine the bioaccessibility. Further, the undigested and digested samples were evaluated for lipase inhibition and antioxidant activities. Male Wistar rats were administered with single dose (1000 mg/kg) of standardized C. longa extract and RTE to determine the plasma concentration of BDMC and curcumin respectively at different time points using LCMS/MS. Results The bioaccessibility of BDMC was significantly higher than curcumin (p < 0.05). BDMC was found superior to curcumin having significant lipase inhibitory effect (p < 0.01), ABTS radical scavenging (p < 0.05), and nitric oxide scavenging activities (p < 0.01). Interestingly, the relative bioavailability of BDMC in standardized C. longa extract was 18.76 compared to curcumin. The Cmax of BDMC was 4.4-fold higher than curcumin. Conclusion BDMC is reported to have higher bioaccessibility and bioavailability than curcumin. Our findings rationalize use of BDMC-enriched standardized C. longa extract for improved physiological benefits counteracting the regular turmeric extract with less bioavailable curcumin as major curcuminoid.

The chemical compositions of the essential oils (EOs) obtained by hydrodistillation from different parts of Cnidium silaifolium ssp. orientale (Boiss.) Tutin were analyzed both by GC-FID and GC/MS, simultaneously. One hundred nine compounds representing 90.1% of the total volatiles in the EOs were identified with the main characteristic compounds α-pinene (50.3%) in the root, germacrene D (20.3%) in the fruit, and β-caryophyllene (18.7%) in the aerial parts of C. silaifolium ssp. orientale. The antimicrobial activity against human pathogenic Gram-negative and Gram-positive bacteria was evaluated by the in vitro microdilution method. Antibacterial succeptibility was observed from the root and aerial part EOs against Staphylococcus aureus (0.039 and 0.156 mg/mL, respectively); while the fruit EO was most effective against Bacillus cereus at 0.07 mg/mL. The antioxidant capacities of the EOs were also evaluated by in vitro DPPH• and ABTS•+ scavenging assays, where no significant activity was observed compared to ascorbic acid and Trolox.

For over a century, peptic ulcer has been one of the most common gastrointestinal tract (GIT) disorder. There are number of drugs are now available for treatment. Drugs of herbal origin reduce the offensive factors and have proved to be safe, clinically effective, relatively less expensive, globally competitive, and with better patient tolerance.This study was performed to assess the anti-ulcer activity on different parts of B.aristata. Apart from that, acute toxicity, qualitative chemical analysis, total phenolic content (TPC), total flavonoid content(TFC) and in vitro antioxidant activities were evaluated. The potentially active plant part was selected for screening as gastro protective, in vivo antioxidant and antisecretory activities in ulcerated rats.The 50% ethanolic extract of B. aristata were subjected to preliminary phytochemical screening, estimation of TFC and TPC. The crude extract from the leaves of B. aristata gave best antiulcer activity among flower and stem. In acute toxicity studies, the administration of the crude extract of B. aristata leaves did not reveal any adverse effects or toxicity in rats at fourteen days observations.The results of these studies have shown that ethylexract of B.aristata leaf (EEBAL) produced a significant dose dependent ulcerprotective, antioxidant and antisecretory activity by blocking the activity of proton pump, protecting from antioxidants produced during stress induced ulcer and by enhancing glycoprotein levels.
