Hypoglycemic Effect of Vitex agnus castus Extract in Diabetic Rats Induced by Streptozotocin

The aim of this study is to evaluate the hypoglycemic activity of the methanolic extract of the aerial parts (leaf + stem) of Vitex on an animal model. Rats of theWistar strain of both sexes, N = 20 females for the short-term test and N = 28 males for the medium-term test, weighing between 125 g and 240 g are rendered diabetic by intraperitoneal injection of streptozotocin (55 mg/kg body weight) and divided into four groups. Triglyceride, total cholesterol, high-density lipoprotein (HDL), and low-density lipoprotein (LDL) cholesterol were determined by the enzymatic method using spectrophotometry, then for the blood glucose is measured by the Accu Chek sensor meter. The oral administration of methanolic extract of Vitex agnus castus at a dose of 300 mg/kg decreased blood glucose after 2 h for the short-term test; with regard to the medium-term test, there was a decrease in the glycemia and an improvement in the lipid profile for 21 days. These results confirm the antidiabetic effects of this plant.

  Objective: The present study was aimed to evaluate antihyperglycemic and antihyperlipidemic activities of methanolic extract of Aristolochia bracteolata (MEAB) against dexamethasone-induced diabetic rat model.Methods: Methanolic extract was prepared by soxhlet extraction and was evaluated for antihyperglycemic and antihyperlipidemic activity using dexamethasone-induced model. The MEAB was administered orally at a dose of 200 and 400 mg/kg body weight glibenclamide was used as standard drug. On 0th and 11th day, blood was collected by retro-orbit plexus.Results: In this model blood glucose levels were determined on 0th and 11th days and MEAB significantly reduced the blood glucose levels in diabetic rats. The effect of MEAB on serum lipid profile such as total cholesterol (TC), triglycerides (TGs), low-density lipoprotein (LDL), very LDL (VLDL), and high-density lipoprotein (HDL) was also measured on the 11th day in the diabetic rats. Significant reduction in TC, TGs, LDL, and VLDL levels and improvement in HDL level were observed in diabetic rats.Conclusion: From the results, it was found that the MEAB possess antihyperglycemic and antihyperlipidemic activities.

Traditionally, in many countries, various parts of the Adansonia digitata (A. digitata) tree have been used in the treatment of many clinical ailments including diarrhea and dysentery. The phytochemical screening has indicated that the leaf extract of A. digitata contains flavonoids, saponins, mucilage, steroids, and alkaloids. Thus, this paper aims to evaluate the hyperglycaemic and hypolipidaemic effects of methanolic extract of A. digitata leaves (200 mg/kg and 400 mg/kg) in diabetic rats. The extract was administered orally for six weeks in the streptozotocin (STZ)-induced diabetic rats. The treatment with the extract caused a significant reduction in the blood glucose, glycosylated hemoglobin, cholesterol, triglycerides, low-density lipoprotein (LDL), interleukin 6 (IL-6), tumor necrosis factor-alpha (TNF-α), and malondialdehyde (MDA) levels by 46.7%, 46.15%, 48.91%, 43%, 60%, 66%, 45.45%, and 30.4%, respectively, as compared to the diabetic group after the sixth week of treatment. The leaf extract also mitigated the decline of high-density lipoprotein (HDL) level, RBCs count, hemoglobin level, packed cell volume (PCV %), and erythropoietin concentration in diabetic rats by 31%, 33.25%, 24.72%, 51.42%, and 220.68% with respect to the diabetic group. Also, the extract maintained the level of antioxidant enzymes, catalase (CAT) and superoxide dismutase (SOD), and reduced glutathione (GSH) in the diabetic rats. It also reduced the elevation in the white blood corpuscles (WBC) count in the STZ-induced diabetic rats. Our study, therefore, indicates that methanolic extract of A. digitata leaf exerts strong antidiabetic and hypolipidaemic properties in a dose-dependent manner by improving the hematological properties and redox parameters in the experimental diabetic rats.

The current study was carried out to determine the hypoglycemic and hypolipidemic activities of methanolic extract P. silhetensis leaves in alloxan-induced diabetic rats. In this study diabetes was induced in rats by intraperitoneal administration of 150 mg/kg of alloxan. Animals were treated for one week with the plant extract at doses of 250 and 500 mg/kg body weight. Metformin (100 mg/kg b.w.) was used as standard for comparison. The anti-diabetes activity was examined by measuring blood glucose level at 0, 3, 5 and 7 days in diabetic induced animal models. Blood samples were collected at day eight of the treatment and analyzed for triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) after sacrifice. Alloxan-induced diabetic rats exhibited a significant decrease in plasma glucose level when treated with P. silhetensis leaf extract as compared to normal, diabetic control and standard rats. All tested lipid parameters were improved after the treatment with methanolic extract of P. silhetensis and metformin as compared to diabetic rats. The results of the present study showed that P. silhetensis possesses significant anti-diabetic properties and has beneficial effects on diabetic hyperlipidemia. This study was done to observe the short tens effect of fresh garlic on different lipid profiles, blood glucose, blood pressure (BP), body weight and haemoglobin (11b) concentration in healthy but overweight (average weight 88.89 kg) adult ( both male and female). Fresh garlic, one clove (average wt. 4 to 6 gm), was taken everyday with night meal by the volunteers for six weeks. The volunteers were mostly students and a few of them were office staff Total number of volunteers were 25. Administration of garlic significantly lowered cholesterol (p<61.001) and low density lipoprotein (p<0.05), but there was no effect on BP, body weight and IIb concentration. Garlic administration reduced the level of blood glucose and triglyceride (TG) to some extent but they were not statistically significant.

Purpose: To evaluate the impact of Vicia ervilia (L.) Willd (Fabaceae) lyophilized extract on various biomarkers of streptozotocin-induced diabetic rats. Methods: Adult male rats (36) were divided into 6 groups, viz, normal control (NC), diabetes control (DC), and treatment groups. Experimental diabetes was induced with streptozotocin. Blood samples were taken from the tail veins daily for 25 days. Fasting and postprandial blood glucose levels of rats were determined. The rats were sacrificed and blood, kidney and liver were taken for biochemical analysis. Results: Blood glucose decreased in DC groups compared to NC group (p ≤ 0.05). In diabetic extract groups, glycated Hb decreased compared to DC group (p ≤ 0.05). Alpha glucosidase activity was reduced in all treatment groups compared to DC group (p ≤ 0.05). Only triglyceride levels of diabetes extract (400 mg/mL) group decreased compared to DC (p ≤ 0.05). Total cholesterol levels fell compared to NC and DC groups (p ≤ 0.05). Low density lipoprotein was lower compared to NC (p ≤ 0.05), while aspartate aminotransferase and alanine aminotransferase levels rose in diabetic extract groups compared to NC, but fell in some diabetic extract groups compared to DC group (p ≤ 0.05). Lactate dehydrogenase decreased in all diabetic extract groups compared to NC and DC (p ≤ 0.05). Serum urea and creatinine levels were higher in diabetic extract groups compared to NC, but decreased in DC group (p ≤ 0.05). Conclusion: These findings demonstrate that Vicia ervilia lyophilized extract haas positive effects on blood glucose and biochemical parameters in diabetic rats compared to DC of V. ervilia seed extract.

Objective: Moringa oleifera is a medicinal plant species that has potential as an antidiabetic. The present study was designed to evaluated molecullar mechanism antidiabetic ethyl acetate fraction of Moringa oleifera leaves (EAFML).Methods: The study was conducted on 30 male wistar rats which were induced with the combination of streptozotocin and nicotinamide by intraperitoneal administration to make a model of type 2 diabetes. After the diabetic induction, all rats were divided into six group and once daily administered orally EAFML in doses standardized quercetine. The treatment was given for ten days, and on the final treatment, all rats were checked their blood glucose level and lipid profile. The skeletal muscles and liver were taken to examine glucose transporter-4 (GLUT4) expression by immunohistochemistry.Results: The result of this study shows that the blood glucose level in diabetic rats with the treatment of EAFML decreased significantly from 355.8±83.7 mg/dL to 177.5±89.3 mg/dL. The cholesterol decreased significantly from 105.2±47.4 mg/dL to 58.6±6.9 mg/dL and low-density lipoprotein (LDL) in diabetic rats with the treatment of EAFML decreased significantly compared to control group from 175.6±41.9 mg/dL to 95.3±8.0 mg/dL. The expression of GLUT4 in skeletal muscles increased from 0.7±1.0 to 3.9±1.1 and in the liver increased significantly from 1.8±1.3 to 2.9±1.9.Conclusion: The EAFML can decrease blood glucose level, cholesterol and low-density lipoprotein in type 2 diabetic rat model. Other than that, this fraction can improve insulin sensitivity by the increase of GLUT4 expressions.

ABSTRACTObjectives: Diabetes mellitus is associated with profound alterations in the levels of plasma lipids and the lipoprotein profile. Herbs and herbal drugpreparations find wide use in alleviating the complications of uncontrolled diabetes owing to their low cost and lesser side effects. This study wasaimed at establishing the hypolipidemic effect of methanolic extract of sea weed Gracilaria corticata on streptozotocin (STZ)-induced diabetic rats.Methods: STZ-induced Wistar male albino rats were administered with methanolic extract of Gracilaria corticata (MGC) at a dosage of 100 mg/kgBW/day per orally for 45 days. Glibenclamide (600 μg/kg BW/day) administered rats served as standard drug control. Animals were sacrificed on46th day, biochemical parameters associated with lipid metabolism were analyzed in blood and tissue samples, statistical significance determined.Results: Elevated levels of total cholesterol, triglycerides, and free fatty acids were observed in the plasma and tissues of diabetic rats as compared toMGC and glibenclamide treated groups. The diabetic rats also had elevated levels of low-density lipoprotein (LDL-C) and very low-density lipoprotein(VLDL-C) and decreased levels of high-density lipoprotein (HDL-C) while the MGC and glibenclamide treated rats showed near normal levels of theseindices.Conclusion: This study establishes the hypolipidemic potential of MGC in STZ-induced diabetic rats.Keywords: Gracilaria corticata, Diabetes mellitus, Hypolipidemic, Streptozotocin.

Abstract Background Diabetes mellitus is clinically underlined by hyperglycemia and dyslipidemia. In view of this, the current study assessed the glycemic and lipidemic control potentials of methanol extract of Sruchium sparganophora leaves (SPA) in the alloxan-induced diabetic model using male Wistar rats. Experimental diabetes was induced through a single intraperitoneal injection of 120 mg/kg freshly prepared alloxan. Thirty-six rats were randomly assigned into six groups of normoglycemic control, untreated diabetic group, and diabetic treated with (i) metformin (12 mg), (ii) metformin 12 mg + SPA 300 mg/kg, (iii) SPA 300 mg/kg, and (iv) SPA 600 mg/kg per os twice at 9.00 and 18.00 h daily for 10 days. Fasting blood glucose (FBG) level and markers of dyslipidemia, and oxidative stress markers were determined. Results SPA at selected doses decreased fasting blood glucose which was significantly (p < 0.05) raised by alloxan. Increase in plasma total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and triglyceride (TG) concentrations and decrease in HDL-cholesterol (HDL-C) concentration (p < 0.05) caused by alloxan were significantly moderated by SPA at selected doses. Glutathione-s-transferase (GST), superoxide dismutase (SOD), and catalase (CAT) activities reduced by alloxan (p < 0.05) in both the liver and pancreas were reversed by SPA 300 and 600, and its combination with metformin. Decreased reduced glutathione (GSH) concentration in alloxan diabetic rats was also reversed by the extract, while the level of malondialdehyde (MDA) exacerbated by alloxan (p < 0.05) in the tissues was decreased by the extracts. Conclusion Struchium sparganophora possesses considerable antihyperglycemic, antidyslipidemic, and antioxidant potentials without compromising organ functionality.

The aim of this study was to explore the potency of salacca vinegar made from various Indonesian salacca fruit extracts as therapeutic agent for hyperglycemia and dyslipidemia for STZ-induced diabetic rats. The rats were grouped into untreated rats, STZ-induced diabetic rats without treatment, and STZ-induced diabetic rats treated with Pondoh salacca vinegar, Swaru salacca vinegar, Gula Pasir salacca vinegar, Madu salacca vinegar, or Madura salacca vinegar. Parameter observed included blood glucose, total cholesterol (TC), high density lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TG), malondialdehyde (MDA), superoxide dismutase (SOD), and pancreas histopathology of the samples. The results demonstrated that all salacca vinegars were capable of reducing blood sugar (from 25.1 to 62%) and reducing LDL (from 9.5 to 14.8 mg/dL), TG (from 58.3 to 69.5 mg/dL), MDA (from 1.1 to 2.2 mg/dL), and TC (from 56.3 to 70.5 mg/dL) as well as increasing HDL blood sugar of STZ-induced diabetic Wistar rats (from 52.3 to 60 mg/dL). Various salacca vinegars were also capable of regenerating pancreatic cells. Nevertheless, the ability of Swaru salacca vinegar to manage hyperglycemia and dyslipidemia appeared to be superior to other salacca vinegars. Swaru salacca vinegar is a potential therapeutic agent to manage hyperglycemia and dyslipidemia of STZ-induced diabetic rats.

The present study is an extension of our previous work carried out onCynodon dactylon. This study deals with the critical evaluation of glycemic potential of ethanolic extract of defattedC. dactylon. The doses of 250, 500 and 750 mg kg−1bw of the extract were administered orally to normal as well as Streptozotocin-induced diabetic rats to study its glycemic potential. The effect of repeated oral administration of the same doses of ethanolic extract was also studied on serum lipid profile of severely diabetic (SD) rats. The dose of 500 mg kg−1bw was identified as the most effective dose as it lowered the blood glucose levels of normal by 42.12% and of diabetic by 43.42% during fasting blood glucose (FBG) and glucose tolerance test respectively. The SD rats were also treated daily with this identified dose of 500 mg kg−1bw for 2 weeks and a significant reduction of 56.34% was observed in FBG level. Total cholesterol, low density lipoprotein and triglyceride levels were also decreased by 32.94, 64.06 and 48.46% respectively in SD rats whereas, cardioprotective high density lipoprotein increased by 16.45%. The reduced urine sugar level and increased body weight are additional advantages. These evidences clearly indicate that the ethanolic extract of defattedC. dactylonhas high antidiabetic potential along with good hypolipidemic profile.
