Curcumin diglutaric acid-loaded polyethylene glycol-chitosan oligosaccharide-coated superparamagnetic iron oxide nanoparticles (CG-PEG-CSO-SPIONs) were fabricated by co-precipitation and optimized using a Box–Behnken statistical design in order to achieve the minimum size, optimal zeta potential (≥ ±20 mV), and maximum loading efficiency and capacity. The results demonstrated that CG-PEG-CSO-SPIONs prepared under the optimal condition were almost spherical in shape with a smooth surface, a diameter of 130 nm, zeta potential of 30.6 mV, loading efficiency of 83.3%, and loading capacity of 8.3%. The vibrating sample magnetometer results of the optimized CG-PEG-CSO-SPIONs showed a superparamagnetic behavior. Fourier transform infrared spectroscopy and X-ray diffraction analyses indicated that the CG physically interacted with PEG-CSO-SPIONs. In addition, the CG-PEG-CSO-SPIONs could be stored dry for up to 12 weeks or in aqueous solution for up to 4 days at either 4 °C or 25 °C with no loss of stability. The CG-PEG-CSO-SPIONs exhibited a sustained release profile up to 72 h under simulated physiological (pH 7.4) and tumor extracellular (pH 5.5) environments. Furthermore, the CG-PEG-CSO-SPIONs showed little non-specific protein binding in the simulated physiological environment. The CG-PEG-CSO-SPIONs enhanced the cellular uptake and cytotoxicity of CG against human colorectal adenocarcinoma HT-29 cells compared to free CG, and more CG was delivered to the cells after applying an external magnetic field. The overall results suggest that PEG-CSO-SPIONs have potential to be used as a novel drug delivery system for CG.

The present study focuses on the effects of the physicochemical properties of superparamagnetic PEG-modified, positively charged, and negatively charged iron oxide nanoparticles (SPIONs) on their reactivity with hydrogen peroxide. Our hypothesis was that the reactivity of SPIONs in this reaction would depend on their surface properties. The comparative study of the nanoparticles with DLS and TEM revealed the average sizes of PEG-modified, positively charged, and negatively charged SPIONs. We observed that the reactivity of negatively charged SPIONs with hydrogen peroxide was less than that of positively charged SPIONs and that of these second nanoparticles was less than that of PEG-modified SPIONs. This difference in the reactivity of these SPIONs with hydrogen peroxide was attributed to the presence of carboxyl or amine groups on their surface. However, the values of the rate constants of the reactions of PEG-modified, positively charged, and negatively charged SPIONs with hydrogen peroxide showed that the reaction of negatively charged SPIONs with hydrogen peroxide was more rapid than that of PEG-modified SPIONs and the reaction of these second SPIONs with hydrogen peroxide was more rapid than that of positively charged SPIONs. The surface study of the SPIONs using XPS showed that the high-resolution spectra of these nanoparticles changed after reaction with hydrogen peroxide, which indicates their surface modifications. These investigations can help develop more appropriate nanoparticles with controlled physicochemical properties.

Abstract Superparamagnetic iron oxide nanoparticles were obtained in the polyethylene glycol environment. An effect of precipitation and drying temperatures on the size of the prepared nanoparticles was observed. Superparamagnetic iron oxide Fe3O4, around of 15 nm, was obtained at a precipitation temperature of 80°C and a drying temperature of 60°C. The presence of functional groups characteristic for a polyethylene glycol surfactant on the surface of nanoparticles was confirmed by FTIR and XPS measurements. Silver nanoparticles were introduced by the impregnation. Fe3O4-Ag nanostructure with bactericidal properties against Escherichia coli species was produced. Interesting magnetic properties of these materials may be helpful to separate the bactericidal agent from the solution.
