SummaryThe characteristics are given of a Chaddar cheese defect known as ‘tallowy discoloration’ or ‘white streak’. In the tallowy portions of the cheese the copper content is lower and the iron content higher than in the adjacent normal cheese. The copper probably migrates in cheese following oxidation of the sulphydryl compounds in the tallowy area while the unexpected distribution of iron may be explained by changes that occur during salting of the curd. The proportion of ferrous to ferric iron tends to increase as the cheese matures and tallowy portions of cheese do not contain less ferrous iron than adjacent portions.The addition of iron salts to cheese milk did not increase the incidence of tallowy discoloration, but the addition of sufficient copper salts reduced it. Although the sulphydryl content of cheese appeared to be related to tallowy discoloration, little evidence of the part it plays was obtained. The oxidation of sulphydryl groups and the formation of peroxides occurred more readily at lower temperatures.The significance is discussed of ferrous iron in a fat-oxidizing system where copper does not increase oxidation.

SummaryPlatelet membrane glycoproteins (GP) IIb/IIIa and rap1b, a 21 kDa GTP binding protein, associate with the triton-insoluble, activation-dependent platelet cytoskeleton with similar rates and divalent cation requirement. To examine the possibility that GPIIb/IIIa was required for rap1b association with the cytoskeleton, experiments were performed to determine if the two proteins were linked under various conditions. Chromatography of lysates from resting platelets on Sephacryl S-300 showed that GPIIb/IIIa and rap1b were well separated and distinct proteins. Immunoprecipitation of GPIIb/IIIa from lysates of resting platelets did not produce rap1b or other low molecular weight GTP binding proteins and immunoprecipitation of rap1b from lysates of resting platelets did not produce GPIIb/IIIa. Finally, rap1b was associated with the activation-dependent cytoskeleton of platelets from a patient with Glanzmann’s thrombasthenia who lacks surface expressed glycoproteins IIb and IIIa. Based on these findings, we conclude that no association between GPIIb/IIIa and rap1b is found in resting platelets and that rap1b association with the activation-dependent cytoskeleton is at least partly independent of GPIIb/IIIa.

Improved removal of particles during the treatment of natural aquatic suspensions has been achieved by pre-ozonation and the addition of small quantities of iron salts (βFe ≤ 0.1 mg.L-1; “Fe(III)-assisted filtration”) followed by rapid filtration. As shown by investigations on a large-scale installation at Lake Constance Water Supply, this procedure reliably reduces suspended solids by at least 2-3 powers of ten in long-term use. However, the high efficacy of Fe(III)-assisted filtration cannot be explained on the basis of known coagulation mechanisms (like adsorption-charge neutralization, co-precipitation). Instead, the essential step was found to be the conditioning of the filter medium by coating it with colloids containing Fe(OH)3, and this “Fe coating” process occurs only in the presence of alkaline earths (especially Ca2+). According to further experiments, the enhanced solid-liquid separation was ultimately traced to chemical interactions such as the formation of calcium-organic association structures between the iron hydroxides and other solids. For design of Fe(III)-assisted filtration steps, finally, a βCa/DOC ratio above 40 mg.mg-1 and pre-oxidation with ozone dosages not exceeding 2 mg O3/mg DOC was recommended.
