Subterranean and berseem clovers, harvested at early and late stages of maturity, were dried and offered to sheep ad libitum, or at about 90 % of ad libitum intake. Feed consumption and the times spent in ruminating and eating were measured when the forages were offered ad libitum and several aspects of digestion were studied at the restricted level of feeding. The effects of maturity on (i) forage chemical composition, (ii) nutrient digestibility, (iii) parameters relating to the movement of digesta through the stomach, (iv) ruminal volatile fatty acid production, (v) the molar proportions of individual amino acids in abomasal digesta and peripheral blood plasma, (vi) nitrogen balance, and (vii) the time and energy expenditure in ruminating and eating, were generally the same with these clovers as has been observed previously with grasses. Substantial amounts of nutrients were derived from forages harvested in the immature state, the consumption of which provided about 1200 g organic matter per day. It was calculated that under conditions of ad libitum feeding about 500 g of volatile fatty acids was produced in the rumen and 160-230 g of crude protein was digested in the intestines per day. High mean values for nitrogen balance were obtained with immature subterranean clover but the variation between sheep was large. The levels of amino acids in blood plasma were also exceptionally high with this diet. The data obtained on the digestion of immature clover forages were compared with those obtained previously with immature grass forages. The patterns of digestion were generally very similar for both types of forages and any differences observed could usually be ascribed to the higher feed intake with the clovers. The forages had similar capacities to furnish volatile fatty acids in the rumen and digested crude protein in the intestines. The calculated energy expenditure in eating and ruminating, however, was slightly lower for the clovers. The data were interpreted to indicate that if immature clovers are superior to immature grasses in promoting animal production, the superiority cannot be attributed to an inherent capacity of clovers to provide the animal with a more favourable balance of nutrients.

Chain elongation of volatile fatty acids for medium chain fatty acids production (e.g. caproate) is an attractive approach to treat wastewater anaerobically and recover resource simultaneously. Undefined microbial consortia can be tailored to achieve chain elongation process with selective enrichment from anaerobic digestion sludge, which has advantages over pure culture approach for cost-efficient application. Whilst the metabolic pathway of the dominant caproate producer, Clostridium kluyveri, has been annotated, the role of other coexisting abundant microbiomes remained unclear. To this end, an ethanol-acetate fermentation inoculated with fresh digestion sludge at optimal conditions was conducted. Also, physiological study, thermodynamics and 16 S rRNA gene sequencing to elucidate the biological process by linking the system performance and dominant microbiomes were integrated. Results revealed a possible synergistic network in which C. kluyveri and three co-dominant species, Desulfovibrio vulgaris, Fusobacterium varium and Acetoanaerobium sticklandii coexisted. D. vulgaris and A. sticklandii (F. varium) were likely to boost the carboxylates chain elongation by stimulating ethanol oxidation and butyrate production through a syntrophic partnership with hydrogen (H2) serving as an electron messenger. This study unveils a synergistic microbial network to boost caproate production in mixed culture carboxylates chain elongation.

ООО «БИОТРОФ», Санкт-Петербург Аннотация: Приведены данные исследования методом газожидкостной хроматомасс-спектрометрии метаболитов, синтезируемых пробиотическими бактериями, входящими в состав кормовой добавки «Профорт®». Проведен опыт в условиях интенсивного промышленного птицеводства по применению кормовой добавки «Профорт®» (50 тыс. голов бройлеров в группе). Среди метаболитов пробиотических штаммов Enterococcus sp. и Bacillus sp. обнаружены такие полезные вещества, как молочная кислота, уксусная, пропионовая и другие короткоцепочечные (летучие) жирные кислоты, активные пептиды. Результаты зоотехнического опыта показали, что при скармливании бройлерам пробиотика «Профорт®» (500 г/т) живая масса при убое в 40 дней была выше контроля на 6,9%, конверсия корма улучшилась на 3,0%, а европейский индекс продуктивности бройлеров - на 5,69%. Nutritional strategies can be employed to mitigate greenhouse emissions from ruminants. This article investigates the effects of polyphenols extracted from the involucres of Castanea mollissima Blume (PICB) on in vitro rumen fermentation. Three healthy Angus bulls (350 ± 50 kg), with permanent rumen fistula, were used as the donors of rumen fluids. A basic diet was supplemented with five doses of PICB (0%–0.5% dry matter (DM)), replicated thrice for each dose. Volatile fatty acids (VFAs), ammonia nitrogen concentration (NH3-N), and methane (CH4) yield were measured after 24 h of in vitro fermentation, and gas production was monitored for 96 h. The trial was carried out over three runs. The results showed that the addition of PICB significantly reduced NH3-N (p < 0.05) compared to control. The 0.1%–0.4% PICB significantly decreased acetic acid content (p < 0.05). Addition of 0.2% and 0.3% PICB significantly increased the propionic acid content (p < 0.05) and reduced the acetic acid/propionic acid ratio, CH4 content, and yield (p < 0.05). A highly significant quadratic response was shown, with increasing PICB levels for all the parameters abovementioned (p < 0.01). The increases in PICB concentration resulted in a highly significant linear and quadratic response by 96-h dynamic fermentation parameters (p < 0.01). Our results indicate that 0.2% PICB had the best effect on in-vitro rumen fermentation efficiency and reduced greenhouse gas production.

The rumen contains abundant microorganisms that aid in the digestion of lignocellulosic feed and are associated with host phenotype traits. Cows with extremely high milk protein and fat percentages (HPF; n = 3) and low milk protein and fat percentages (LPF; n = 3) were selected from 4000 lactating Holstein cows under the same nutritional and management conditions. We found that the total concentration of volatile fatty acids, acetate, butyrate, and propionate in the rumen fluid was significantly higher in the HPF group than in the LPF group. Moreover, we identified 38 most abundant species displaying differential richness between the two groups, in which Prevotella accounted for 68.8% of the species, with the highest abundance in the HPF group. Functional annotation based on the Kyoto Encyclopedia of Gene and Genome (KEGG), evolutionary genealogy of genes: Non-supervised Orthologous Groups (eggNOG), and Carbohydrate-Active enzymes (CAZy) databases showed that the significantly more abundant species in the HPF group are enriched in carbohydrate, amino acid, pyruvate, insulin, and lipid metabolism and transportation. Furthermore, Spearman’s rank correlation analysis revealed that specific microbial taxa (mainly the Prevotella species and Neocallimastix californiae) are positively correlated with total volatile fatty acids (VFA). Collectively, we found that the HPF group was enriched with several Prevotella species related to the total VFA, acetate, and amino acid synthesis. Thereby, these fulfilled the host’s needs for energy, fat, and rumen microbial protein, which can be used for increased biosynthesis of milk fat and milk protein. Our findings provide novel information for elucidation of the regulatory mechanism of the rumen in the formation of milk composition.
