The phytic acid (PA), inorganic phosphorus (Pi), yellow pigment (YP), water soluble phenolics (WSPH) and free protein sulfhydryl groups (PSH) of 15 Triticum aestivum and 15 Triticum durum wheats grown at six different environments were evaluated for variability, intra- and inter- relations with agronomic traits. The most variable properties in bread wheat were YP and WSPH with coefficients of variation (CV) amounting to 14.59% and 14.10%, respectively, while in durum wheat those were PSH and YP with CV of 17.59% and 15.02%, respectively. PA in durum wheat showed the least variability (CV 3.61%). Among others, positive associations were obtained between WSPH and YP in bread and durum wheat, YP and PSH, WSPH and PSH, yield (YLD) and WSPH, YLD and PSH in durum wheat, while negative associations were recorded for PA and YP, PA and WSPH, YLD and antioxidants (YP and WSPH) in bread wheat, PA and antioxidants, PA and YLD in durum wheat.

Fifteen genotypes of bread wheat (Triticum aestivum L.) and fifteen genotypes of durum wheat (Triticum durum Desf.) were evaluated in the multi-environment trial during 2010-11. and 2011-12 vegetation seasons to investigate components of variance, heritability in a broad sense (h2), expected genetic advance (GA), and stability of phytic acid (PA), inorganic phosphorus (Pi), phytic phosphorus (Pp)/Pi relation, yellow pigment (YP), water soluble phenolics (WSPH) and free protein sulfhydryl groups (PSH) content. The field trials were carried out at three locations in Serbia, as randomized complete block design with four replications. The genetic component of variance (?2g) predominated the genotype ? environment interaction (?2ge) component for: Pi in bread wheat (3.0 times higher), Pp/Pi in bread wheat (2.1 times higher) and in durum wheat (1.2 times higher), YP content in bread wheat (2.2 times higher) and in durum wheat (1.7 times higher), and WSPH content in bread wheat (1.4 times higher). The relation ?2g/?2ge for Pi content in durum wheat was equal to one. The ?2ge prevailed ?2g for: PA in bread wheat (1.7 times higher) and in durum wheat (5.7 times higher), PSH in durum wheat (3.7 times higher), and WSPH in durum wheat (5.2 times higher). High h2 coupled with high expected genetic advance as percent of mean (GAM) were observed for: Pi (93.7% and 26.1%, respectively) in bread wheat, Pp/Pi relation in bread wheat (92.4% and 20.7%, respectively) and in durum wheat (87.2% and 20.8%, respectively), YP content in bread wheat (92.6% and 28.0%, respectively) and in durum wheat (90.7% and 28.1%, respectively), and WSPH content (88.9% and 25.8%, respectively) in bread wheat. PA content in bread and durum wheat had medium to medium high h2 (50.5% and 77.9%, respectively), and low expected GAM (9.9% and 3.7%, respectively). GGE biplots with average-environment coordination (AEC) indicated less stability of durum wheat for PA, WSPH and PSH content.

The present study was carried out to evaluate agronomic traits and assessment of genetic variability of some wheat genotypes at Qassim region, Saudi Arabia', during 2010/11 and2011/12 seasons. Fourteen wheat genotypes including five bread wheat and nine durum wheat genotypes were evaluated in randomized complete block design with three replications. The genotypes were evaluated for ten different yield contributing characters viz., days to heading, days to maturity, grain filling period, grain filling rate, plant height, number of spikes m-2, kernels spike-1, 1000-kernel weight, grain yield and straw yield. The combined analysis of variance indicated the presence of significant differences between years for most characters. The genotypes exhibited significant variation for all the characters studied indicating considerable amount of variation among genotypes for each character. Maximum coefficient of variation was observed for number of spikes m-2 (17%), while minimum value was found for days to maturity. Four genotypes produced maximum grain yield and statistically similar, out of them two bread wheat genotypes (AC-3 and SD12) and the other two were durum wheat (AC-5 and BS-1). The genotypes AC-3, AC-5 and BS-1 had higher grain yield and stable in performance across seasons. The estimation of phenotypic coefficient of variation in all the traits studied was greater than those of the genotypic coefficient of variation. High heritability estimates (> 0.5) were observed for days to heading, days to maturity, and plant height, while the other characters recorded low to moderate heritability. The high GA % for plant height and days to heading (day) was accompanied by high heritability estimates, which indicated that heritability is mainly due to genetic variance. Comparatively high expected genetic advances were observed for grain yield components such as number of kernels spike-1 and 1000-kernel weight. Grain yield had the low heritability estimate with a relatively intermediate value for expected genetic advance. The results of principle component analysis (PCA) indicated that the superior durum wheat genotypes for grain yield in the two seasons (AC-5 and BS-1) are clustered in group II (Fig. 2). Also, the superior two bread wheat genotypes (AC-3 and SD12) were in group I. Therefore, it could be future breeding program to develop new high yielding genotypes in bread and durum wheat.

Abstract Development of efficient DNA markers plays an important role in modern breeding projects of many crops, including cultivated hexaploid bread wheat (BW) and tetraploid durum wheat (DW). Findings of genome-wide association studies on major polyploid crops, such as BW, may also help in molecular breeding studies in relative cultivated species with a similar genetic background, including DW. Therefore, the validation of identified quantitative trait loci or marker-trait associations is an important preliminary step in marker-assisted selection (MAS) oriented projects. In this study, thirty-two SNP (single nucleotide polymorphism) markers of six agronomic traits identified in BW, harvested in Kazakhstan, were converted to KASP (Kompetitive Allele-Specific PCR) as-says. Generated 32 KASP assays were used in the analysis of 29 DW accessions from Kazakhstan. Firstly, the group of DW accessions was tested using replicated and randomised one-metre blocks in field conditions of southeast Kazakhstan and evaluated for main agronomic traits. The analysis showed that 14 KASP assays were polymorphic in the scoring of 29 DW accessions. The t-test suggested that the segregation in eight KASP assays was significantly associated with five agronomic traits. The study confirms robustness of KASP assays in MAS of DW breeding projects for the improvement of yield potential.

Herbicide application in seed maize requires special attention due to their larger sensitivity than hybrid maize. The aim of study was to examine and define the sensitivity/tolerance of the five maize inbred lines with different susceptibility to herbicides (belonging to different heterotic groups), based on alterations of secondary metabolites (phenolics, protein sulfhydryl groups, phytic and inorganic phosphorus). Two groups of herbicides: triketons (mesotrione and topramezone) and sulfonylureas (rimsulfuron and foramsulfuron) were tested. Lines from independent heterotic group, which were sensitive to herbicides expressed visible damages together with significant reduce in grain yield, mainly induced by sulfonylurea herbicides. Parallel with that, significant increase in phenolics, phytic and inorganic phosphorus, as well as drop in protein sulfhydryl groups were observed in their leaves. Tolerant lines (belonging to Lancaster group) had mainly insignificant grain yield reduce, also with lesser variations in sulfhydryl groups, content of phytic and inorganic phosphorus, as well as increase in phenolics content. Among examined secondary metabolites, phytate is the main factor, contributing to herbicide tolerance in maize lines. Owing to lesser yield decrease and variation in content of examined secondary metabolites, expressed in treatments with triketone herbicides, they usage could be safe in maize lines.

Saharan wheats have been studied particularly from a botanical viewpoint. Genotypic identification, classification and genetic diversity studies to date were essentially based on the morphology of the spike and grain. For this, the allelic variation at the glutenin loci was studied in a set of Saharan bread and durum wheats from Algerian oases where this crop has been traditionally cultivated. The high molecular weight and low molecular weight glutenin subunit composition of 40 Saharan bread and 30 durum wheats was determined by SDS-PAGE. In Saharan bread wheats 32 alleles at the six glutenin loci were detected, which in combination resulted in 36 different patterns including 17 for HMW and 23 for LMW glutenin subunits. For the Saharan durum wheats, 29 different alleles were identified for the five glutenin loci studied. Altogether, 29 glutenin patterns were detected, including 13 for HMW-GS and 20 for LMW-GS. Three new alleles were found in Saharan wheats, two in durum wheat at the Glu-B1 and Glu-B3 loci, and one in bread wheat at the Glu-B1 locus. The mean indices of genetic variation at the six loci in bread wheat and at the five loci in durum wheat were 0.59 and 0.63, respectively, showing that Saharan wheats were more diverse. This information could be useful to select Saharan varieties with improved quality and also as a source of genes to develop new lines when breeding for quality.

AbstractFood fraud is becoming a prominent topic in the food industry. Thus, valid methods for detecting potential adulterations are necessary to identify instances of food fraud in cereal products, a significant component of human diet. In this work, primer–probe systems for real-time PCR and droplet digital PCR (ddPCR) for the detection of these cereal species: bread wheat (together with spelt), durum wheat, rye and barley for real-time PCR and ddPCR were established, optimized and validated. In addition, it was projected to validate a molecular system for differentiation of bread wheat and spelt; however, attempts for molecular differentiation between common wheat and spelt based on the gene GAG56D failed because of the genetic variability of the molecular target. Primer–probe systems were further developed and optimized on the basis of alignments of DNA sequences, as well as already developed PCR systems. The specificity of each system was demonstrated on 10 (spelt), 11 (durum wheat and rye) and 12 (bread wheat) reference samples. Specificity of the barley system was already proved in previous work. The calculated limits of detection (LOD95%) were between 2.43 and 4.07 single genome copies in real-time PCR. Based on the “three droplet rule”, the LOD95% in ddPCR was calculated to be 9.07–13.26 single genome copies. The systems were tested in mixtures of flours (rye and common wheat) and of semolina (durum and common wheat). The methods proved to be robust with regard to the tested conditions in the ddPCR. The developed primer–probe systems for ddPCR proved to be effective in quantitatively detecting the investigated cereal species rye and common wheat in mixtures by taking into account the haploid genome weight and the degree of milling of a flour. This method can correctly detect proportions of 50%, 60% and 90% wholemeal rye flour in a mixture of wholemeal common wheat flour. Quantitative results depend on the DNA content, on ploidy of cereal species and are also influenced by comminution. Hence, the proportion of less processed rye is overestimated in higher processed bread wheat and adulteration of durum wheat by common wheat by 1–5% resulted in underestimation of common wheat.

AbstractThe genetic improvement of durum wheat and enhancement of plant performance often depend on the identification of stable quantitative trait loci (QTL) and closely linked molecular markers. This is essential for better understanding the genetic basis of important agronomic traits and identifying an effective method for improving selection efficiency in breeding programmes. Meta-QTL analysis is a useful approach for dissecting the genetic basis of complex traits, providing broader allelic coverage and higher mapping resolution for the identification of putative molecular markers to be used in marker-assisted selection. In the present study, extensive QTL meta-analysis was conducted on 45 traits of durum wheat, including quality and biotic and abiotic stress-related traits. A total of 368 QTL distributed on all 14 chromosomes of genomes A and B were projected: 171 corresponded to quality-related traits, 127 to abiotic stress and 71 to biotic stress, of which 318 were grouped in 85 meta-QTL (MQTL), 24 remained as single QTL and 26 were not assigned to any MQTL. The number of MQTL per chromosome ranged from 4 in chromosomes 1A and 6A to 9 in chromosome 7B; chromosomes 3A and 7A showed the highest number of individual QTL (4), and chromosome 7B the highest number of undefined QTL (4). The recently published genome sequence of durum wheat was used to search for candidate genes within the MQTL peaks. This work will facilitate cloning and pyramiding of QTL to develop new cultivars with specific quantitative traits and speed up breeding programs.

Abstract We report the map-based cloning of the leaf rust resistance gene Lr21, previously mapped to a generich region at the distal end of chromosome arm 1DS of bread wheat (Triticum aestivum L.). Molecular cloning of Lr21 was facilitated by diploid/polyploid shuttle mapping strategy. Cloning of Lr21 was confirmed by genetic transformation and by a stably inherited resistance phenotype in transgenic plants. Lr21 spans 4318 bp and encodes a 1080-amino-acid protein containing a conserved nucleotide-binding site (NBS) domain, 13 imperfect leucine-rich repeats (LRRs), and a unique 151-amino-acid sequence missing from known NBS-LRR proteins at the N terminus. Fine-structure genetic analysis at the Lr21 locus detected a noncrossover (recombination without exchange of flanking markers) within a 1415-bp region resulting from either a gene conversion tract of at least 191 bp or a double crossover. The successful map-based cloning approach as demonstrated here now opens the door for cloning of many crop-specific agronomic traits located in the gene-rich regions of bread wheat.

Abstract The present investigation was carried out to evaluate agronomic performance and oil quality of seven sunflower genotypes at Shandaweel Research Station, Agricultural Research Center, Sohag, Egypt during 2018 and 2019 summer seasons. These genetic materials were sown in a randomized complete block design having three replications. Significant genetic variations among evaluated sunflower genotypes for agronomic traits and oil quality were observed. The superior sunflower genotypes were Line 120 for seed yield per hectare (3102.38 kg), Sakha 53 for seed oil content (44.63 %) and Line 125 for oil quality where it contained the highest proportion of unsaturated fatty acids (89.20 %). The phenotypic coefficients of variation were slightly higher than genotypic coefficients of variation for all studied traits. High heritability (exceeded 60%) and genetic advance as percent of mean (ranged from medium to high, exceeded 10%) was observed for most studied traits. Seed yield per plant positively correlated with plant height, stem diameter, head diameter, and 100-seed weight and most chemical traits at phenotypic and genotypic levels. Maximum phenotypic direct effects on seed yield per plant were observed for 100-seed weight, head diameter and total unsaturated fatty acids. While, the highest genotypic direct effect on seed yield per plant was observed for head diameter. Hence, most studied traits could be employed as selection criteria for improving evaluated sunflower genotypes.
