Dianthus giganteiformis subsp. kladovanus is an endemic, endangered, horticulturally appealing perennial plant that can be used for the revegetation of sand dunes of the Danube region. The appropriate method for its effective production is micropropagation. For this reason, the experiments were conducted in order to establish an efficient protocol for the micropropagation of this subspecies. The sterile culture was initiated from seeds collected in situ and the germination percentage was high (88%). According to the results obtained in this study, the multiplication phase should be performed on an MS medium enriched with 0.1 mg/L BAP and 0.1 mg/L NAA. The concentration of MS salts significantly influenced rooting, and higher rooting percentages were obtained on reduced MS media (91.7 - 95%) than on MS media (73.4 - 76.7%). The addition of NAA slightly increased rooting percentage (up to 95%). Obtained microplants were successfully acclimatized (83.3%) in a substrate composed of peat and sand (1: 1; v/v). Using the protocol presented in this paper, the efficient propagation of D. giganteiformis spp. kladovanus can be achieved for rapid plant production aimed at revegetation, biodiversity protection and floricultural production of this species.

Curcuma roscoeana Wall. is a tuberous perennial plant with tuberous rhizomes. It is an endangered species. In nature, it has a very low rate of multiplication. Propagation of C. roscoeana in vitro was done by culturing 0.5 × 1.0-mm shoot tips from young buds onto modified Murashige and Skoog (MS)+ 0.25 mg/L kinetin. Stem explants 10.0 mm in size, measured from the base of the plantlets longitudinally cut in half, were used in the experiments. The first experiment was done by varying the concentration of both kinetin and NAA, in MS liquid medium, at 0–8.0 mg/L and 0–0.05 mg/L, respectively. There were no significant differences of kinetin and NAA concentrations on the number of plantlets obtained. The 0.5-mg/L kinetin treatment gave the highest yield in number of new plantlets (3.1 plantlets/cultured explant). In the second experiment, various concentrations of BAP from 0 to 8.0 mg/l were tested. 2.8–3.7 plantlets were formed in the media with 0.05–2.0 mg/L of BAP. The most-suitable concentration of BAP was at 1.0 mg/L, providing 3.7 plantlets/cultured explants. Kinetin or BAP alone could be used in MS medium for rapid clonal propagation of C. roscoeana. The rooted plantlets could be successfully transferred into growing pots. Digitalis trojana Ivanina is a member of the Plantaginaceae family and known by its common name, Helen of Troy foxglove. It is perennial endemic to Çanakkale and Balıkesir, northwestern Turkey. In order to develop an efficient shoot regeneration protocol, the leaf explants of D. trojana were cultured on Murashige and Skoog (MS) medium containing 6-benzyl adenine (0.1, 0.5, 1.0, 3.0, 5.0 mg/L) and α-naphthalene acetic acid (0.1, 0.5, 1.0 mg/L), 3% (w/v) sucrose and 0.8% (w/v) agar. The highest number of regenerated shoots was obtained from leaf explants that were cultured on MS medium with 3.0 mg/L BA+0.1 mg/L NAA. Regenerated shoots were rooted on MS medium without plant growth regulators. Rooted plants (2–3 cm) were separately transferred to pots containing a mixture of peat and perlite (2:1 v/v) and acclimatized successfully in a growth chamber.

One of the problem of Eurycoma longifolia Jack propagation was low germination percentage due to recalcitrant seeds and slow growth of seedling from cutting propagation. To overcome this problem is required propagation of Eurycoma longifolia via in vitro culture. The objective of this research was to know the effect of Auxin (2,4-D and NAA) and Cytokines (BAP and Kinetin)  on Eurycoma longifolia callus induction via leaf and petiole explants. In this study, we used plant growth regulator of 2,4 D, NAA, BAP and Kinetin in several levels.  The observed variables were appearing callus time, callus color and callus texture. The results of this study showed that MS medium supplemented with 1 ppm NAA+ 1 ppm BAP was able to induce callus formation in leaf explant for 6 months after culture. While MS medium supplemented with 1 ppm 2,4-D, 1 ppm BAP, combination of 2,4-D and Kinetin and combination of 2,4-D and BAP can induce callus formation from petiole. All the callus formation has yellow color and yellow brown color. The petiole explant that is grown in MS medium supplemented with 1 ppm BAP induced of callus in short time (18 days after culture).

In vitro ovule culture could be used to generate homozygous lines through the production of haploid plants. The present study reports on in vitro regeneration and production of haploid plants through ovule cultures and identification of the regenerated haploids using flow cytometry. The ovules were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of 6-benzyladenine (BA), kinetin (Kin), 2,4-dichlorophenoxyacetic acid (2,4-D), and naphthalene acetic acid (NAA) at 0, 0.5, 1, and 2 mg·L−1 for their gynogenesis. Among different plant growth regulators (PGRs) tested, 2,4-D at 2 mg·L−1 produced direct gynogenesis. The highest callogenesis percentage (100%) was obtained on MS medium containing 1 mg·L−1 2,4-D and 2 mg·L−1 NAA. Flow cytometry analysis was used to identify the regenerated haploids. It also confirmed gynogenic occurrence at 1 and 2 mg·L−1 2,4-D with percentages of 21.7% and 41%, respectively. Therefore, 2,4-D proved effective for the induction of haploids in black cumin. The regenerated haploids were developed on MS medium without PGRs. The obtained results of in vitro gynogenesis and haploid plant production can tremendously facilitate breeding programs of black cumin.

The aim of the work was to determine the effect of fungicides: Topsin M 70 WG, Bravo 500 SC, and Sportak 45 EC in two concentrations on the growth of fungi: Aspergillus niger, Epicoccum purpurascens, Cladosporium sphaerospermum, Mortierella isabellina and Penicillium verrucosum var. verrucosum on PDA and Murashige-Skoog medium (MS). The smallest index of linear growth rate of the fungi had Topsin M 70 WG for fungi: E. purpurascens and P. verrucosum var. verrucosum on PDA and MS medium and for M. isabellina and C. sphaerospermum on MS in concentrations of 71.42 mg/l and 107.13 mg/l. The highest index of linear growth had Bravo 500 SC in concentrations of 0.5 ml/l and 0.75 ml/l on PDA for E. purpurascens and A. niger. The lowest index of linear growth on MS for E. purpurascens, C. sphaerospermum and P. verrucosum var. verrucosum was noted down. The highest effectiveness in control of linear growth all tested species of fungi was found for fungicide Sportak 45 EC in concentration of 0.55 ml/l and 0.825 ml/l on MS and PDA. Among all tested fungicides used in vitro propagation fungicides from imidazoles group are recommended.

A rapid and efficient protocol is outlined for in vitro propagation of muskmelon(Cucumis melo L.) Shoot tips, nodal segments and cotyledonary nodes from invitro grown seedlings were used as explants. The explants were inoculated on MS medium fortified with different combinations and concentrations of growthregulators viz., BAP, NAA, GA3 and IBA for multiple shoot regeneration.Effective result was found on MS medium supplemented with 2.0 mg/l BAP, inwhich 90% and 70% cultures induced multiple shoots from nodal segments andshoot tip explants, respectively. Whereas, 70% cultures of cotyledonary nodeswere found to induced shoots on MS medium with 1.5 mg/l BAP + 0.1 mg/l GA3. In vitro regenerated shoots were subcultured on half strength MS mediumsupplemented with different concentrations of IBA and NAA for successful rootinduction and the effective result (up to 70%) was found in medium with 1 mg/lIBA. Well rooted in vitro grown plantlets were acclimatized in sandy soil, whereas 70% plantlets survived

AbstractThymus persicus (Ronniger ex Rech. f.) Jalas is a valuable and endangered natural source of antitumor pentacyclic triterpenoids, i.e., betulinic acid, oleanolic acid and ursolic acid, which grows in northwest Iran. As the plant has a low propagation rate in nature, a suitable method for in vitro-propagation is needed. With the aim of identifying a suitable system for regenerating T. persicus via direct organogenesis, Murashige & Skoog (MS) medium supplemented with different plant growth regulators (PGRs) was tested. In vitro-grown shoot tips were exposed to the cytokinins 6-benzylaminopurine (BAP), kinetin (KN), and thidiazuron (TDZ), alone or in combination with the auxins 1-naphthalene-acetic acid (NAA), 2,4-dichlorophenoxyacetic (2,4-D), indole-3-butyric acid (IBA) and indole-3-acetic acid (IAA). The highest shoot formation (7.1 ± 0.9) was obtained with a medium fortified with 8.9 μM BAP plus 2.7 μM NAA. Regenerated shoots were easily rooted on the different tested media, with the most abundant (16.6 ± 1.4) and strongest roots obtained on half-strength MS medium containing 2.5 μM IBA. The rooted plantlets were successfully acclimatized (76.6%) in a greenhouse before transference to natural conditions. Homogeneity and phytochemical productivity of the in vitro regenerated plantlets were confirmed by random amplified polymorphic DNA (RAPD) profiles and high performance liquid chromatography (HPLC), respectively.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

Factors such as slow growth, low rates of sexual and asexual reproduction, and viability of seeds among others limit the massive propagation of Agave americana L. by conventional methods. In this study, callus induction and shoot proliferation was determined in A. americana using Murashige and Skoog (MS) medium supplemented with dicholorophenoxyacetic acid (2,4-D) and 6-benzyl adenine (BA). Meristematic tissue was used as the explants, and were placed on MS medium supplemented with 30.0 g·L−1 sucrose with 0.11, 0.18, or 0.45 μm 2,4-D and 11.0, 22.0, 38.2, 44.0, 58.7, or 73.3 μm BA. Treatments were implemented according to factorial experimental design 3 × 6. After 1 month, the number of explants with callus was determined, whereas the numbers of shoots per explant were monitored after 4, 16, 20, and 36 weeks. The maximum percent of explants with callus was obtained with 0.11 μm 2,4-D and 58.7 and 73.3 μm BA, whereas the maximum numbers of shoots per explant (71) were obtained with 0.11 μm 2,4-D and 73.3 μm BA. The effect of different concentrations of indolebutyric acid (IBA) in the rooting of shoots was evaluated. There were no significant effects of IBA on the number of roots, root length, and axillary roots. Plantlets were acclimatized in the glasshouse and they did not show any phenotypic alteration. This is a highly efficient protocol for the in vitro propagation of A. americana via indirect organogenesis.

Marumi kumquat (Fortunella Japonica) is culture for its valuable nutritional value and medicinal importance in many regions of the world. The current study aimed to evaluate the effect of two types of media enriched with different concentrations of fructose and different plant growth regulators and different fructose concentration on in vitro propagation of Fortunella Japonica. The findings showed that the most effective treatment for explant surface sterilization was by using 0.1% HgCl2 for ten minutes which give best results for production contamination-free explants at the initiation cultures. At multiplication stage, WPM medium gave better results at all tested BA levels as compared with MS medium. No significant differences were showed by using BA alone or in combination with GA3 in the measured parameters. It has been observed that WPM medium supplemented with 0.5mgl-1 BA with the presence of 30mgl-1 fructose was able to give the highest shoot length (1.56cm) with maximum shoots number/explant 9.0 and highest leaves number/explant (21.0). The proliferated shoots were exposed to full strength MS medium salts supplemented with 2mgl-1 NAA which showed the highest ratio of rooting. In vitro rooted plantlets were gradually acclimatized and transferred to open air conditions, which recorded a high survive rate reached to 92%
