We determined the contents of total glutathione (tGSH), reduced glutathione (GSH), and oxidized glutathione (GSSG) and values of the glutathione redox index (GSH RI) in hepatopancreas, gills, and muscle of three freshwater crayfish species: noble crayfish (Astacus astacus) from the Southern Morava River, stone crayfish (Austropotamobius torrentium) from the Krajkovacka River, and spinycheek crayfish (Orconectes limosus) from the Danube River. The obtained data show strong tissue and species specifity of investigated parameters: tGSH, GSH, GSSG, and GSH RI in the hepatopancreas, gills, and muscle of the indicated crayfish species. Our work represents the first study of its kind and showed that the investigated parameters can be considered suitable biomarkers of the cellular glutathione redox status in of freshwater crayfish species.

AbstractUsing column and thin-layer chromatography, the authors investigated the presence of carotenoids in the carapaces and muscles of freshwater crayfish species. Fifteen carotenoids were found, among which canthaxanthin, adonixanthin, and astaxanthin were predominant. The highest carotenoid content was revealed in the carapaces of Pacifastacus leniusculus and Astacus astacus and in the muscles of Astacus leptodactylus and Orconectes limosus. En utilisant la chromatographie sur colonnes et en couches minces, les auteurs ont recherche la presence de carotenoodes dans les carapaces et les muscles de plusieurs especes d'ecrevisses. Quinze carotenoodes ont ete trouves, parmi lesquels la canthaxanthine, l'adonixanthine et l'asthaxantine etaient dominantes. Le contenu en carotenoodes le pluseleve a ete trouve dans les carapaces de Pacifastacus leniusculus et Astacus astacus et dans les muscles de Astacus leptodactylus et Orconectes limosus.

Introduction of invasive North American crayfish species and their pathogen Aphanomyces astaci has significantly contributed to the decline of European freshwater crayfish populations. In this study, noble crayfish, a susceptible native European species, and marbled crayfish, an invasive disease-resistant species, were challenged with haplogroup A (low virulence) and haplogroup B (high virulence) strain of A. astaci. Hepatopancreatic tissue was isolated 3 and 21 days post-challenge. Our results revealed strong up-regulation in expression levels of the prophenoloxidase cascade immune-related genes in the haplogroup B challenged noble crayfish 3 days post-challenge. In the marbled crayfish, we observed an up-regulation of immune system relevant genes (DSCAM, AP, ALFs, CTLs and hemocyanin) 3 days post-challenge. This response highlights the marbled crayfish capability of building the immune tolerance. Furthermore, we successfully characterised several novel immune related gene groups in both crayfish species, contributing to our current understanding of crayfish immune related genes landscape.

Abstract Several methods of sampling are commonly used to detect freshwater crayfish (Decapoda: Malacostraca). Many of them are laborious, time-consuming, and require dedicated equipment. The aims of this contribution are i) to compare visual encounter surveys and baited trap success in the detection of the noble crayfish, Astacus astacus (L.), which is endangered in Poland, and ii) to assess the time needed to detect the invasive spiny-cheek crayfish, Orconectes limosus (Raf.). The study is based on data collected between 2016–2018 in various habitats of Astacus astacus and Orconectes limosus in Poland. Visual encounter surveys are at least as effective in assessing the presence of A. astacus as the trapping method. The modal value for the detection time of O. limosus at all sites and all surveys was two minutes. Sample rarefaction showed that one survey covered 11.33 (SD = 0.43) of a maximum of 12 detections per survey. This suggests that, despite some limitations, visual detection might be an efficient method for determining crayfish presence/absence for a wide range of applications.

Noble crayfish is the most widespread native freshwater crayfish species in Europe. It is threatened in its entire distribution range and listed on the International Union for Concervation Nature- and national red lists. Reliable monitoring data is a prerequisite for implementing conservation measures, and population trends are traditionally obtained from catch per unit effort (CPUE) data. Recently developed environmental DNA (eDNA) tools can potentially improve the effort. In the past decade, eDNA monitoring has emerged as a promising tool for species surveillance, and some studies have established that eDNA methods yield adequate presence-absence data for crayfish. There are also high expectations that eDNA concentrations in the water can predict biomass or relative density. However, eDNA studies for crayfish have not yet been able to establish a convincing relationship between eDNA concentrations and crayfish density. This study compared eDNA and CPUE data obtained the same day and with high sampling effort, and evaluated whether eDNA concentrations can predict relative density of crayfish. We also compared two analytical methods [Quantitative real-time PCR (qPCR) and digital droplet PCR (ddPCR)], and estimated the detection probability for eDNA monitoring compared to trapping using occupancy modeling. In all lakes investigated, we detected eDNA from noble crayfish, even in lakes with very low densities. The eDNA method is reliable for presence-absence monitoring of noble crayfish, and the probability of detecting noble crayfish from eDNA samples increased with increasing relative crayfish densities. However, the crayfish eDNA concentrations were consistently low and mostly below the limit of quantification, even in lakes with very high crayfish densities. The hypothesis that eDNA concentrations can predict relative crayfish density was consequently not supported. Our study underlines the importance of intensified sampling effort for successful detection of very low-density populations, and for substantiating presumed absence, inferred from negative results. Surprisingly, we found a higher likelihood of eDNA detection using qPCR compared to ddPCR. We conclude that eDNA monitoring cannot substitute CPUE data, but is a reliable supplement for rapid presence-absence overviews. Combined with eDNA analyses of alien crayfish species and diseases such as crayfish plague, this is a cost-efficient supplement offering a more holistic monitoring approach for aquatic environments and native crayfish conservation.

Ischemic stroke (IS) and type 2 diabetes mellitus are factors that affect the homeostasis of low-molecularweight aminothiols (cysteine, homocysteine, glutathione etc.). It has already been shown that IS in the acute period led to a decrease a level of reduced forms of aminothiols, but it is not clear whether type 2 diabetes mellitus has a noticeable effect there. Objective: to reveal the features of homeostasis of aminothiols in patients with type 2 diabetes mellitus in acute IS. Material and methods. The study involved 76 patients with primary middle cerebral artery IS in the first 10–24 hours after development of neurological symptoms. Group 1 included 15 patients with IS and type 2 diabetes mellitus, group 2 — 61 patients with IS and stress hyperglycemia. Their total plasma levels of cysteine, homocysteine, and glutathione, their reduced forms, and redox status were determined at admission (in the first 24 hours after IS). Results. There was a decrease in the level of total glutathione level (1.27 vs. 1.65 μM, p = 0.021), as well as its reduced form (0.03 vs. 0.04 μM, p = 0.007) in patients with IS and type 2 diabetes mellitus. Patients with type 2 diabetes mellitus who had a low redox status of homocysteine (0.65–1.2%) and glutathione (0.7–2.0%) were also characterized by a decrease in total glutathione level (p = 0.02; p = 0.03). Conclusion. Thus, type 2 diabetes mellitus is associated with a decrease in the level of total glutathione in acute IS. Probably, type 2 diabetes mellitus is characterized by a particular relationship between the metabolism of homocysteine, glutathione and glucose. Therefore, the search for homocysteine-dependent approaches to correct glutathione metabolism in type 2 diabetes mellitus may be of interest as an adjuvant therapy for IS.

Under pathological conditions, the induction of nitric oxide synthase (NOS) in macrophages is responsible for NO production to a cytotoxic concentration. We have investigated changes to, and the role of, intracellular glutathione in NO production by the activated murine macrophage cell line J774. Total glutathione concentrations (reduced, GSH, plus the disulphide, GSSG) were decreased to 45% of the control 48 h after cells were activated with bacterial lipopolysaccharide plus interferon γ. This was accompanied by a decrease in the GSH/GSSG ratio from 12:1 to 2:1. The intracellular decrease was not accounted for by either GSH or GSSG efflux; on the contrary, rapid export of glutathione in control cells was abrogated during activation. The loss of intra- and extracellular glutathione indicates either a decrease in synthesis de novo, or an increase in utilization, rather than competition for available NADPH. All changes in activated cells were prevented by pretreatment with the NOS inhibitor l-N-(1-iminoethyl)ornithine. Basal glutathione levels in J774 cells were manipulated by pretreatment with (1) buthionine sulphoximine (glutathione synthase inhibitor), (2) acivicin (γ-glutamyltranspeptidase inhibitor), (3) bromo-octane (glutathione S-transferase substrate) and (4) diamide/zinc (thiol oxidant and glutathione reductase inhibitor). All treatments significantly decreased the output of NO following activation. The degree of inhibition was dependent on (i) duration of treatment prior to activation, (ii) rate of depletion or subsequent recovery and (iii) thiol end product. The level of GSH did not significantly affect the production of NO, after induction of NOS. Thus, glutathione redox status appears to plays an important role in NOS induction during macrophage activation.

Exposure ofAzollaplants to UV-B radiation for 6 h resulted in a decrease in biomass and relative growth rate (RGR), which coincided with an increase in doubling time (DT) as compared with the control. Also, the protein content decreased. On the other hand, hydrogen peroxyde (H2O2) and malondialdehyde (MDA) accumulated significantly in UV-treatedAzollaplants. Conversely, the addition of selenium (Se) at 1 ppm resulted in a significant increase in biomass and protein content of untreated and UV-treatedAzollaplants, and a significant reduction in both H2O2and MDA. Moreover, the addition of Se to UV-treated and untreatedAzollaplants resulted in a significant increase in total ascorbate and total glutathione (GSH) contents compared with the control and UV-stressedAzollaplants. Also, glutathione redox potential (GSH/TG) increased significantly in UV-treatedAzollaplants in the presence of Se. There also was a significant increase (38%) in ascorbate peroxidase (APX) activity in UV-treated plants compared with the control. APX activity in the presence of Se did not change significantly compared with the control. Glutathione reductase (GR) activity increased significantly in UV-treatedAzolla, while glutathione peroxidase (GSH-PX) activity did not. On the other hand, both GSH-PX and GR activity in untreated and UV-treatedAzollaplants were significantly enhanced by the application of Se to the nutrient media at a concentration of 1 ppm. Therefore, we can conclude that Se protectsAzollaplants from UV-B stress.

The role of oxidative stress (OS) in cancer is a matter of great interest due to the implication of reactive oxygen species (ROS) and their oxidation products in the initiation of tumorigenesis, its progression, and metastatic dissemination. Great efforts have been made to identify the mechanisms of ROS-induced carcinogenesis; however, the validation of OS byproducts as potential tumor markers (TMs) remains to be established. This interventional study included a total of 80 colorectal cancer (CRC) patients and 60 controls. By measuring reduced glutathione (GSH), its oxidized form (GSSG), and the glutathione redox state in terms of the GSSG/GSH ratio in the serum of CRC patients, we identified significant changes as compared to healthy subjects. These findings are compatible with the effectiveness of glutathione as a TM. The thiol redox state showed a significant increase towards oxidation in the CRC group and correlated significantly with both the tumor state and the clinical evolution. The sensitivity and specificity of serum glutathione levels are far above those of the classical TMs CEA and CA19.9. We conclude that the GSSG/GSH ratio is a simple assay which could be validated as a novel clinical TM for the diagnosis and monitoring of CRC.
