In vitro screening of Falcataria moluccana (Miq.) was conducted by tissue culture method. Seeds fromtwo different site of community forest, 400 m (S1) and 800 m (S2) above sea level, were used as material.Double concentration of MS (Murashige & Skoog, 1962) with 40 mg/l gall rust (Uromycladium tepperianum(Sacc.) fi ltrate were used for media selection. The results of this research showed that 66 % axenic plantlets invitro from S1 and 27 % from S2 were still survived after 3 months incubation without subculture. The meanof fresh weight (2. 21 ± 0. 26 g) and dry weight (1. 97 ± 0. 12 g) from S1 plantlets lower than the mean of freshweight (2. 87 ± 0,18 g) and dry weight (2. 16 ± 0. 14 g) from S2 plantlets. Qualitative of terpenes, saponins andquantitative of total phenolics were analyzed from those gall rust extract, as source of fi ltrate media, attackedand un-attacked of F. moluccana. They all qualitatively have capability to produce terpenoid and saponin. Itis notice that U. tepperianum, un-cultured pathogen, contain of those compound that may play a role as codeterminantsof pathogenecity. While the highest total phenolic compound were contained in gall rust extract(2. 35 %), followed by attacked F. moluccana branches (1. 18 %) and un-attacked F. moluccana branches (0. 44%). This indicated that phenolic compound in gall rust has higher activity as a response of F. moluccana to U.tepperianum pathogen pressures and result of this study suggest the great value of gall rust fi ltrate for use asmedia selection in vitro.

Objective: To estimate the in vitro total phenolics, flavonoids contents, antioxidant and antimicrobial activities of various solvent extracts from the medicinal plant Physalis minima Linn.Methods: The crude bioactive were extracted from the dried powder of Physalis minima using methanol, ethyl acetate, chloroform and hexane solvents. Total phenolic content (TPC) and total flavonoid content (TFC) were estimated using Folin-Ciocalteu and aluminium chloride colorimetric methods respectively. 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and ferric reducing antioxidant power (FRAP) assays were used to determine the in vitro antioxidant capacity. The antimicrobial assay was done through agar well diffusion; minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using broth microdilution methods against the Gram-negative bacteria (Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, Proteus vulgaris) and Gram-positive bacteria (Staphylococcus aureus).Results: TPC expressed as gallic acid equivalents (GAE) ranged from 60.27±1.73-151.25±2.50 mg GAE/g dry weight, and TFC expressed as quercetin equivalents (QE) ranged from 56.66±0.80-158.84±2.30 mg QE/g dry weight. Methanol extract showed the highest antioxidant activity followed by ethyl acetate, chloroform, hexane extract and the IC50 values of methanol extract for scavenging DPPH and ABTS free radicals were 280.23±5.75-173.40±0.38µg/ml, respectively. All the extracts have shown potent antimicrobial activity for the zone of inhibition ranged from 9-35 mm; MICs and MBCs values ranged from 0.125-4.0 and 0.25-8.0 mg/ml, respectively towards tested pathogenic species.Conclusion: The comprehensive analysis of the present results demonstrated that Physalis minima possess high potential antioxidant properties which could be used as a viable source of natural antioxidants in treating infections caused by above-mentioned pathogens.

In this study, the changes in nutritional composition, phenolic compounds and antioxidant activity in free and bound fractions of shortcrust cookies were investigated. By incorporating ingredients such as kamut, matcha tea, dried mango and jasmine flowers, the contents of crude and neutral-detergent fibre reached up to 2.0 and 5.0%, respectively. Similar increments were observed in phenolic compound contents and 2,2-diphenyl-1-picrylhydrazyl scavenging activity values. Concerning cookies supplemented with matcha tea, the total phenolic compound content raised from 1.0 to 4.8 mg gallic acid equivalent/g and the antioxidant activity value increased from 0.5 to 5.7 mg trolox equivalent/g on a dry weight basis. For determining the antioxidant activity values in water-soluble and insoluble phenolic fractions of the cookies, a photochemiluminiscence assay was separately applied, and they were found up to 0.8 mg ascorbic acid equivalent/g and 3.2 mg trolox equivalent/g, respectively. The main phenolic compounds in all supplemented cookies were neochlorogenic, gallic and vanillic acids.

In this study, the antagonistic effect of five bacteria genera namely Bacillus, Pseudomonas, Azotobacter, Azospirillum, and Streptomyces isolated from rhizosphere of wheat were evaluated against Fusarium graminearum as potential biocontrol agents in vitro. F. graminearum was molecularly diagnosed using the Polymerase chain reaction (PCR) technique. Each bacteria were tested for the production of catalase enzyme, oxidase enzyme, analysis of starch, analyze of gelatin, and the motility, where Azotobacter, Azospirillum, and Bacillus subtilis were positive for all tested. Fungal inhibition tests were performed by using the dual culture method and agar well diffusion technique. Among them, Streptomyces and Azospirillum exhibited potent inhibition to the growth of F. graminearum (72.14% and 66.42%) respectively, followed by B.pumillus, P.fluorescens, B. subtilis and Azotobacter ( 58.28%, 43.23%, 39.71% and 35.71%) respectively as compared with the control treatment (0.0%).The dry weight of the fungus biomass was decreased with bacteria P. fluorescens, Streptomyces sp, Azotobacter sp, Azospirillum sp, B. subtilis, and B. pumillus which reached (0.114, 0.103, 0.147, 0.101, 0.143, and 0.107 g) respectively compared to the control treatment that was 0. 665 g.

Assay on ability of endophytic bacteria isolated from peanut to inhibit Sclerotium sp. growth in peanut seedlings.   A study on assay of ability of endophytic bacteria to inhibit Sclerotium sp. in peanut seedling has been done. The bacteria were isolated from peanut healthy plants, while Sclerotium sp. was isolated from infected peanaut plant. Antagonistic assay was conducted by dual culture method.  In vivo assay of inhibiting Sclerotium sp. was conducted by dipping peanut seed in bacterial solution, and planting the seed in soil:compost (3:1) growing media. Six endophytic bacterial isolates showed to inhibit the growth of Sclerotium sp. in vitro. LN1 seemed to inhibit more of Sclerotium sp., while LN5 showed to inhibit less. Two potential isolates LN1 of gram-negative and LN2 of gram-positive using for further study showed to decrease more of dumping off. It also seemed that the isolates increased the seedling height, number of leaves, and dry weight.

Wild edible mushrooms are becoming endangered all over the world. Very few wild edible mushrooms are found in natural habitat. Volvariella bombycina is an edible and medicinal mushroom. The mushroom was collected in natural habitat growing on Populus tree. Mycelium of the mushroom was developed in PDA slant tubes by tissue culture method, incubated at 25°C for 1-2 weeks. Spawn was developed in wheat grains after incubation at 25°C for 2-3 weeks. Substrates were formulated for the development of fruiting bodies by combination of paddy straw, saw dust and rice husk. Fruiting bodies of V. bombycina was cultivated in these substrates after incubation at 28 ± 2°C for 2-4 weeks. Abstract 1. An in vitro method to observe radiosensitivity of stem cells was developed in the present study. In vivo and in vitro effect of 60Co irradiation on the erythropoietin-induced stem cell differentiation into erythroblasts was observed, using a tissue culture method of polycythemic mouse spleen. Response to erythropoietin was demonstrated by an appearance of heme synthesis and erythroblasts in spleen fragments. 2. A significant correlation between the rate of appearance of erythroblasts and heme synthesis of the spleen fragments was observed. 3. After irradiation, marked impairment of both heme synthesis and production of erythroblasts was observed, yielding D37 values in the vicinity of 70 r in vivo and 120 r in vitro irradiation, respectively. 4. Marked recovery of erythropoietin-induced heme synthesis in the polycythemic mouse spleen was observed 9 days after 300 r irradiation, with an "overshooting" phenomenon on the 12th day.

Aim. The present study aimed to isolate flavonoid fraction from the aerial parts ofCissus quadrangularisand to evaluate its antioxidant and anticancer potential usingin vitroassay system.Methods. Total phenolic and flavonoid contents were calculated for the drug. Flavonoid fraction was isolated using column chromatography and analysed using HPLC.In vitro, antioxidant activity of the ethanol extract and isolated flavonoid fraction was investigated by nitric oxide, DPPH and hydroxyl radical scavenging assays. Breast cancer (MCF 7) cell line was used as thein vitrocancer model for MTT assay.Result. The amount of total phenolic content and total flavonoid content in the ethanol extract showed 28.6 mg/g dry weight expressed as gallic acid equivalents, and 15.8 mg/g was expressed as quercetin equivalents, respectively. The tested extract showed good dose-dependent free radical scavenging property in all the models with the IC50values of 98 μg/mL, 125 μg/mL, and 96 μg/mL for ethanol extract and 10 μg/mL, 12 μg/mL, and 10 μg/mL for flavonoid fraction, respectively. The flavonoid fraction possess potent anticancer property against breast cancer cells (MCF7) with IC50value of 40 μg/mL.Conclusions. It can be concluded that the aerial part ofCissus quadrangularishas potential antioxidant and anticancer activities.

In this study, the effects of different fruit parts and extraction conditions on the antioxidant properties of jujube (Ziziphus jujuba Mill.) fruit were investigated. Five in vitro antioxidant models and statistical analyses were performed. The results revealed that jujube peel with pulp (peel pulp) exhibited better antioxidant capacity than did seeds. Overall, jujube peel pulp extracted using 50% ethanol at 60 °C exhibited the best antioxidant capacity in terms of 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (0.3 ± 0 mg/mL), 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity (0.5 ± 0 mg/mL), total phenolic content (38.3 ± 0.4 mg gallic acid equivalent per gram dry weight), total flavonoid content (43.8 ± 0.2 mg quercetin equivalent per gram dry weight), and reducing power (41.9 ± 2.2 mg ascorbic acid equivalent per gram dry weight). The results indicated that jujube peel pulp is a more potential natural antioxidant than seeds.

The total phenolics, flavonoids and in vitro antioxidant activity of petroleum ether, benzene, ethyl acetate, methanol and ethanol extracts of stem of Drypetes sepiaria were determined using various antioxidant model systems viz, DPPH, hydroxyl, superoxide, ABTS and reducing power. Total phenolic content was estimated by Folin-Ciocalteau method. Flavonoids were determined by Aluminium chloride method. The total phenolics and flavonoids contents were found to be 0.81 g 100 g-1 and 1.12g 100 g-1 respectively in the methanol extract. Among the solvent tested, methanol and ethanol extracts of D. sepiaria stem showed potent in vitro antioxidant activities. This investigation explored that D. sepiaria stem is a potential source of natural antioxidant.
