A new reactive oxygen species (ROS)-responsive drug delivery systems (DDSs) needs development and has excellent application prospect. In this study, FA-PEG-TK-PLGA copolymer containing a ROS-responsive TK linker was designed and generated. The polymer self-assembled into stable small-sized DDSs into with hydrophilic PEG shells and hydrophobic PLGA cores, which exhibited high stability in aqueous solutions, besides an adequate drug loading capacity, excellent ROS sensitivity. high stability and good compatibility. Folic acid (FA) in the DDS presented here was used to target cancer cells. The polymer presents great potential as a novel DDSs for Oral squamous cell carcinoma (OSCC) therapy.
