This study tested whether the oral administration of glycerol at the time of progestin removal and in the first 6 days of the estrous cycle increased the ovulation rate, prolificacy, and pregnancy rate in goats. Intravaginal sponges impregnated with fluorogestone acetate were inserted into 129 goats for 12 days; upon sponge removal, goats were randomly assigned to one of the two following treatment groups: the glycerol group (n = 65), which received an oral drench of 100 mL of glycerol upon sponge removal and was repeated on days 0, 2, 4, and 6 following estrus (estrus = day 0), and the control group (n = 64), which did not receive glycerol. The goats in estrus were mated, and their ovulation rate was determined by ultrasonography between days 8 and 12 of the estrous cycle. Pregnancy was diagnosed by ultrasonography on day 40, and the prolificacy was determined at birth. In 6 goats treated with glycerol and 5 controls, their insulin concentrations at 0, 2, 4, 8, and 12 h after the glycerol drench were determined by radioimmunoassay. The proportion of goats with multiple ovulations (glycerol = 71 vs control = 64) and the proportion of goats with multiple births (glycerol = 52 vs control = 56) were similar (P > 0.05) between treatments. Likewise, the pregnancy rate was similar (P > 0.05) between treatments (glycerol = 88 vs control = 85%). The insulin concentrations tended to be higher in goats treated with glycerol (P = 0.08). In conclusion, an oral drench of 100 mL of glycerol at the time of progestin withdrawal and in the first 6 days of the estrous cycle did not increase the ovulation rate, prolificacy, or pregnancy rate in goats.

Improving AI pregnancy rates in beef cattle will enhance the profitability of beef herds during these current times of increasing cost inputs by enhancing the quality of the calf crop. This experiment was aimed at determining the effect of calf removal (from CIDR removal to timed AI) during estrous cycle synchronization on AI pregnancy rates. This experiment was conduced at the Dixon Springs Research Station (Simpson, IL) during the fall 2007 breeding season. The beef cows used for this research project were crossbred Angus cows between 2 and 8 years of age that had calved in August to October of 2007. Cows were at least 45 days postpartum at the timed AI (November 2007). All cows were randomly divided into 1 of 2 treatment groups: cows in treatment group 1 (control group, n = 284) were allowed to keep their calves for the entire estrous cycle synchronization and AI program (except during brief times of treatment administration), whereas cows allotted to treatment group 2 (n = 258) had their calves removed at the time of CIDR implant removal/PGF2α treatment and returned following AI. The calves were maintained on creep feed out of sight and sound of their dams. All cows were synchronized by using the CO-Synch + CIDR protocol [CIDR (1.38 g) + GnRH (100 μg) on Day 0 and CIDR removal on Day 7 + PGF2α (25 mg)] and were inseminated at a predetermined time of 60 to 66 h post PGF2α with semen from proven bulls. At AI, cows also received an injection of GnRH (100 μg). All cows were pregnancy checked 50 to 65 days after timed AI via ultrasonography. Statistical comparisons were performed by using the chi-square test of SAS. The pregnancy rate for cows in the control treatment group was 47% (132 of 284) and was 57% (147 of 258) for the calf-removal treatment group. There was a significant increase in pregnancy rate in the treatment group (P < 0.02) that resulted in 10 more calves per 100 synchronized AI breedings. Although excellent facilities are needed to maintain calves away from their dams during this 60 to 66 h and there are additional costs for the feedings required for the calves, there is a savings in time (one calf separation from the cow is eliminated). In conclusion, removing the calves at the onset of estrous cycle synchronization can improve pregnancy rates to synchronized timed AI and can increase the ease of cattle handling during this process by reducing the number of calf removals during routine cow workings. This study examined the effect of administering an additional dose of prostaglandin F2 alpha (PGF2?), in a fixed-time artificial insemination protocol (FTAI), on the fertility of female Nellore cattle. Two experiments were carried out: the first (Experiment I) took place in the state of Tocantins and the second (Experiment II) in the state of Pará, Brazil. In Experiment I (E1), 80 cows were used in three treatments (T1, T2 and T3) in which all received the same FTAI protocol. In T1 (n = 29), the cows received 12.5 mg of Dinoprost on day 9; in T2 (n = 28), they received the additional dose on day 10; and in T3 (n = 23; control group), the animals did not receive the additional PGF2? dose. Experiment II consisted of 147 bovine females distributed into two treatment groups, namely, T1 - 72 animals receiving the same protocol as T1 of E1; and T2 - 75 animals receiving the same protocol as T3 of E1. Statistical analysis was performed using SAS software, applying the PROC NPARWAY procedure for E1, and means were compared by the Wilcoxon test at the 5% significance level. In Experiment II, the data were subjected to analysis of variance by PROC GLIMMIX and means were compared by the T test at the 5% significance level. The following pregnancy rates were obtained in Experiment I: T1 - 62.06% (18/29); T2 - 57.14% (16/28); and T3 - 52.17% (12/23), with no significant difference observed between treatments. In Experiment II, pregnancy rate in T1 was 66.67% (48/72), whereas in T2 it was 41.33% (31/75), with a significant difference detected (P < 0.05). An additional dose of PGF2? provides an increase in pregnancy rate in Nellore females.

Previously we demonstrated that sex-sorted sperm could produce IVF embryos from juvenile and adult cattle at rates similar to those for unsorted sperm (Fry et al., 2003 Theriogenology 52, 198). In this study we investigated the pregnancy rates of recipient cattle following the transfer of frozen/thawed IVF embryos generated from young heifers using sex-sorted and unsorted sperm. COCs collected from FSH-stimulated Senepol or Beefex heifers by TVR were matured, fertilized with either sex-sorted or unsorted Senepol sperm and cultured for 6 days under our standard laboratory conditions (Fry et al., 2003 Theriogenology 59, 446, Earl et al., 1997 Theriogenology 47, 255). Embryos reaching the blastocyst or expanded blastocyst stage of development were frozen by the CL-V method of vitrification. Briefly, embryos were equilibrated for 5–10min in HEPES-199 media containing 20% FCS (HM), placed in HM containing 10% EG, 10% DMSO for approximately 2 minutes and then in HM containing 20% EG, 20% DMSO for between 20–60sec (Vatja et al., 1997 Cryoletters 18, 191). Vitrification was achieved by collecting between 5–10 IVF embryos in a 3-μL droplet and securing this droplet to a coded CL-V holder. The droplet was vitrified using the CL-V kit (Lindemans et al., 2004 Theriogenology in press) and then sealed in a precooled ‘‘straw’’ for storage in liquid nitrogen. To thaw, the ‘‘straw’’ with specimen was removed from storage;; the specimen droplet was withdrawn from the ‘‘straw’’ and placed directly into HM containg 0.2M sucrose (SM). After approximately 5–10min each embryo was assessed, loaded into a tomcat catheter in SM and transferred surgically into a recipeint cow within 10–15min of thaw. Of 129 Brahman and Brahman cross cows receiving 2 injections of 125μg cloprostenol 11 days apart, 60 exhibited oestrus 2–4 days after the second injection and 53 were deemed suitable for embryo transfer. Pregnancy was determined by ultrasound on Day 40. No difference in pregnancy rate was found between treatment groups (P&gt;0.05; Table 1). The low submission rate (60/129) and pregnancy rate for the in vivo control group indicate that the fertility of the recipient cows may have been compromised by the drought conditions predominating in Central Queensland. Notwithstanding, the CL-V method for the vitrification of IVF embryos produced by either sex-sorted or unsorted sperm gave similar and very promising pregnancy results of around 40%. This provides new opportunities for the rapid banking of large numbers of sexed IVF embryos generated from elite cattle by TVR for user friendly embryo transfer programs. Abstract The aim of the study was to determine the therapeutic efficacy o f simultaneous administration of GnRH and PGF2α in dairy cows with ovarian cysts. Ovarian cyst-affected dairy cows were divided into two experimental groups: 54 cows treated with GnRH and PGF2α, and 42 cows treated with GnRH alone, whereas 22 untreated cows served as the control group. Clinical response and reproductive performance were evaluated. The cumulative disappearance was better in treated cows than in the control group; however, there were no differences between the treatment groups (92.6; 95.2% vs. 72.3%). The mean interval from calving to conception was not significantly shorter (being so by 29 d) in the GnRH/PGF2α group than in the cows treated with GnRH alone (P > 0.05). The intervals from treatment to conception were also similar in these groups. The pregnancy rate in both treated groups was similar (62%) and higher than in the control cows (53%). In the cows with luteal cysts, the total pregnancy rate was higher in all experimental groups; however, only in GnRH-treated cows was this difference statistically significant (77.8% vs. 50.0%, P < 0.05). With time after parturition, the pregnancy rate decreased in all groups. In general, the cows treated with GnRH and PGF2α simultaneously displayed a good clinical response and slight improvement in reproductive performance compared to the single-therapy GnRH group; however, this was not fully convincing.

Oestrus induction is a challenging task during the nonbreeding season in goats. Therefore, our objective was to evaluate the efficacy of gonadotropin-releasing hormone (GnRH) plus a 7-day CIDR-based protocol with or without eCG for the induction of oestrus and fertility in acyclic Beetal goats during the nonbreeding season under subtropical conditions. Based on the absence of corpus luteum via ultrasonography, goats (n=56; mean weight=32.8±0.5 kg; body condition scre=2.5±0.1) were inserted with a CIDR (300mg progesterone) along with GnRH (Day 0), and randomly treated either with eCG (400 IU; n=35) or without eCG (control; n=21) on Day 7. The CIDR was removed on Day 7 and all does were administered single dose of prostaglandin F2α (75µg cloprostenol; Fatro, Italy). Following CIDR insertion, ovarian follicular dynamics was monitored daily through a 7.5-MHz transrectal transducer (Honda HS 1500, Japan) until ovulation in a subset of eCG (n=11) and control goats (n=10). Oestrus was detected at every 12h after CIDR removal with aproned bucks and does were bred naturally. Pregnancy was detected 25 days after breeding by transrectal ultrasound. Data were analysed by t-test, ANOVA, and Chi-squared test using SPSS (version 20.0, SPSS Inc., Chicago, IL, USA). Oestrus occurred more often following CIDR removal in eCG v. control goats (94v. 57%; P&lt;0.05). Similarly, onset of oestrus was earlier (36.0±2.1v. 54.2±4.7 h; P&lt;0.05) and the duration of oestrus was longer in eCG v. control goats (i.e. 30.2±1.3v. 22.1±2.3 h; P&lt;0.05). However, diameter of preovulatory follicles and interval to ovulation after CIDR removal were similar between eCG and control goats (P&gt;0.05). In contrast, growth rate of preovulatory follicle after CIDR removal until ovulation in eCG goats was greater, and ovulation rate was higher than control goats (0.7±0.1v. 0.3±0.1 mm/day; 2.2±0.2v. 1.4±0.2, respectively; P&lt;0.05). In eCG goats, the dominant follicle grew rapidly between Days 8 to 9 (P&lt;0.05), whereas within control goats the dominant follicle grew at consistent rate (P&gt;0.05). After CIDR removal, percentage of ovulating goats in eCG was higher as compared to the control group (100v. 44%; P&lt;0.05). Within control goats, size of the dominant follicle in ovulatory goats was greater at the time of CIDR removal than anovulatory goats (5.7±0.3v. 5.0±0.15 mm; P&lt;0.05), whereas wave emergence tended to be earlier after CIDR insertion in anovulatory goats (4.2±1v. 2.6±0.4 days; P=0.09). Pregnancy rate 25 days postbreeding did not differ between eCG and control goats [i.e. 49% (16/33) v. 25% (3/12); odds ratio 2.9/1; P&gt;0.05]. In conclusion, the eCG protocol resulted in higher oestrus response, ovulation rate, and 3 times higher pregnancy rate in anestrous goats during the nonbreeding season.

Background: This study was conducted to evaluate the efficacy of letrozole as an ovulation inducing agent and to compare it with clomiphene citrate (CC) in infertile women.Methods: This study includes 100 women referred to gynecology OPD of Pt. B.D Sharma, PGIMS, Rohtak with   infertility due to dysovulation. The patients were divided in two groups each comprised of 50 patients, Alternate women were enrolled in study group (Letrozole). Ultrasonic follicular monitoring was done on day 10, 12, 14, 16 of menstrual cycle to measure the number, size of mature follicles. Endometrial thickness and trilaminar pattern of endometrium was compared in between the groups. Inj. Gonadotrophin (hCG) was given as a trigger intramuscularly when follicle size was between 18 to 21mm. After 36 hours of hCG administration ovulation was confirmed on ultrasound.Results: Mean age, parity, and the duration of infertility were similar in both groups. Ovulation rate was 81.6% in letrozole group and was higher than control group(p<.01). The average number of follicles in the control group was 1.90±0.77and 1.17±0.47 in the study group(p<.001). Endometrial thickness in the study group was 7.55±1.12mm and in the control group it was 6.06±0.87(p<.01). Pregnancy rate in study group was 48 % and control group 16%(p<.05).Conclusions: Aromatase inhibitors (Letrozole) is a new group of drugs to join the arsenal of infertility treatments. The result of this preliminary study suggests that letrozole is associated with higher ovulation rate, higher endometrial thickness and trilaminar pattern thus resulting in higher pregnancy rate. Clomiphene citrate may be replaced by letrozole as primary treatment for ovulation induction in infertile patients.

Hormonal treatment of cows at the coastal region of Barisal district of Bangladesh was performed to assess the improvement of pregnancy rate. A total of 100 cows and heifers with irregular history of cyclicity were selected randomly and divided into five treatment groups. The groups were A (treated with anthelmintic), B (treated with anthelmintic, vitamin ADE and multivitamin powder), C (treated with PGF2α), D (treated with GnRH) and E (treated with GnRH and PGF2α). Each group comprised of 20 animals. The age, breed and parity of experimental cows were considered during treatment. In the study, the cows treated with both GnRH and PGF2α (group E) showed significantly (p<0.05) higher estrus (80%) and pregnancy rate (60%) than that of group A, B, C and D. The overall estrus rates of local and crossbred cows were 64% and 70%, respectively and the pregnancy rates were 40 and 52%, respectively. The crossbred cows responded significantly (p<0.05) to hormonal treatment than that of local cows. Parity-2 cows showed higher estrus sign than that of other parities. However, the pregnancy rates were higher significantly (p<0.05) in parity-2 and parity ≥4 cows than that of parity-0, parity-1 and parity-3 cows. The pregnancy rate was also found higher in case of 4 to <5 years old cows than that of 2 to <3, 3 to <4, 5 to <6, and ≥6 years old. It may conclude that the hormonal regimen increases the pregnancy rate as well as decreases the undesired waiting of estrus and conception. Further study with more sample size will reveal the more effective treatment for cows at the coastal areas of Bangladesh.

In order to increase the pregnancy rate in the assisted reproductive technology, the selection of one embryo with the highest implantation potential it is very important. Time-lapse microscopy (TLM) is a tool for selecting quality embryos for transfer. This study aimed to assess the benefits of single-embryo transfer of autologous oocytes performed on day 5 of embryo incubation in a TLM-equipped system in IVF and ICSI programs. Single-embryo transfer following incubation in a TLM-equipped incubator was performed in 282 patients, who formed the main group; the control group consisted of 461 patients undergoing single-embryo transfer following a traditional culture and embryo selection procedure. We assessed the quality of transferred embryos, the rates of clinical pregnancy and delivery. The groups did not differ in the ratio of IVF and ICSI cycles, average age, and infertility factor. The proportion of excellent quality embryos for transfer was 77.0% in the main group and 65.1% in the control group (p = 0.001). In the subgroup with receiving eight and less oocytes we noted the tendency of receiving more quality embryos in the main group (р = 0.052). In the subgroup of nine and more oocytes the quality of the transferred embryos did not differ between two groups. The clinical pregnancy rate was 60.2% in the main group and 52.9% in the control group (p = 0.057). The delivery rate was 45.0% in the main group and 39.9% in the control group (p > 0.050).

Цель исследования - изучение перепрограммирования мононуклеарных лейкоцитов на модели системного ювенильного идиопатического артрита (сЮИА), воспроизводимой у крыс Wistar с использованием полного адъюванта Фрейнда и липополисахарида. Методика. сЮИА воспроизведен у 6-месячных крыс-самцов Wistar. На 40-е сут. эксперимента животные были разделены на 3 группы: 1-я группа - контроль; 2-я - группа доксициклина; 3-я - группа дексаметазона. Взятие проб крови у животных проводили на нулевые, 41-е и 55-е сут. Мононуклеарные клетки периферической крови выделяли гравиметрически, после чего окрашивали их на маркеры и внутриклеточные цитокины. Дифференцировали моноциты (CD3-CD4+) и Т-хелперы (CD3+CD4+). Анализировали динамику внутриклеточной экспрессии интерлейкина IL-4 (рассматривали как маркер про-М2 фенотипа, так как в случае выделения из клетки ИЛ-4 служит стимулятором М2 поляризации макрофагов) и IFN-g (как маркер про-М1 фенотипа) по данным проточной цитофлуориметрии. Применяли непараметрический статистический тест Mann-Whitney-Wilcoxon в программе R для статистической обработки данных. Результаты и заключение. При моделировании сЮИА выявлено закономерное изменение фенотипа моноцитов. Применение же доксициклина и дексаметазона приводило к более ранней поляризации их по про-М2-пути в отношении моноцитов (на 41-е сут.) в сравнении с контролем. Про-М1 эффект (на 55-е сут., в сравнении с контролем) выявлен также в группах доксициклина и дексаметазона. У животных разных групп обнаружены характерные динамические изменения внутриклеточной экспрессии цитокинов. Важно, что различная направленность поляризации фенотипа при сЮИА и применении препаратов наблюдается не только у моноцитов, но и у Т-хелперов. The study objective was to evaluate targeted reprogramming of peripheral blood mononuclear cells in systemic juvenile idiopathic arthritis (sJIA) modeled in 6-month-old male Wistar rats by co-administration of complete Freund’s adjuvant and lipopolysaccharide. Methods. On day 40 of the experiment, rats were divided into three groups: control, doxycycline, and dexamethasone groups. Blood samples were collected on days 0, 41, and 55. Peripheral blood mononuclear cells were isolated gravimetrically and stained for markers and cytokines. Monocytes (CD3-CD4+) and T-helpers (CD3+CD4+) were differentiated as target cells. IL-4 was considered a marker for the pro-M2 phenotype since IL-4 can activate M2 macrophage polarization; IFN-g was considered a marker for the pro-M1 phenotype. Time-related changes in the intracellular expression of IL-4 and IFN-g were studied using flow cytometry. Data were analyzed using nonparametric statistical tests (Mann-Whitney-Wilcoxon) in the R environment for statistical computing. Results and conclusions. Monocytes (like macrophages) underwent reprogramming during the development of modeled sJIA disease. In monocytes of doxycycline and dexamethasone treatment groups, pro-M2 effects were observed earlier (day 41) than in the control group. Pro-M1 effects were observed in monocytes of doxycycline and dexamethasone groups on day 55, as compared with the control group. Characteristic time-related changes of intracellular cytokine expression were described for different groups. Importantly, the differently directed phenotype polarization was observed in sJIA and treatment groups for both monocytes and T-helpers.
