The toxicity of ammonia/ammonium to the vermifiltration wastewater treatment process

This study was undertaken to assess the toxicity of ammonia/ammonium to key species within the vermifiltration process. The key species, the earthworm Eisenia fetida, was subjected to a series of tests in solid phase mesocosms and full-scale units. The solid phase tests showed a relatively low toxicity to ammonium with ammonium chloride having an LC50 for ammonium of 1.49 g/kg. Ammonium sulfate did not show an effect on mortality at 2 g/kg ammonium. The full-scale units showed that ammonia hydroxide can change the pH and concentration of ammonia in wastewater and while it caused some mortality to the worms its overall affect on system functioning was minimal with no significant difference in terms of worm survival found between treatments. The affect on nitrifying bacteria was also minimal with no linear trend shown with ammonia concentration.

ABSTRACTNitrification is one of the most widely used methods to reduce ammonia concentration in wastewater.The purpose of this study was to determine the effectiveness of high-concentrated ammonia nitrification by the consortium between commercial probiotics and bacteria from the sediment of the catfish pond. The study was conducted on batch system bioreactors with a working volume of 1 liter containing 100 mg/L ammonia solution and 50 grams sediment of catfish pond. This study used a treatment variation of the concentration of commercial probiotics and the bacterial isolate from the catfish pond sediment using glucose as the carbon source. The variations of commercial probiotics added to the bioreactor were 5 ml/L, 10 ml/L and 15 ml/L. The variations of glucose concentration were 0 g/L and 3,9 g/L. Analysis of ammonia concentration was carried out by spectrophotometry using the phenate method. The highest removal efficiency of ammonia was 92.35% in the bioreactor with a mixture of 15 ml/L commercial probiotics and 3.9 g/L glucose with the fastest ammonia rate was on the third day of the experimental period. The addition of glucose in the bioreactors could increase ammonia removal by 57.39%. The result of statistical analysis indicated that variations in the concentration of commercial probiotic indicated no statistically significant difference in ammonia removal (P 0.05), while variations in glucose concentration showed a statistically significant difference in ammonia removal (P 0.05). Three isolates were successfully isolated on specific media for nitrifying bacteria. The result of bacterial identification showed that three isolated bacteria were Bacillus sp., Aeromonas salmonicida, and Burkholderia cepacia.Keywords: ammonia, nitrification, sediment of catfish pond, commercial probioticABSTRAKNitrifikasi merupakan salah satu metode yang paling banyak digunakan untuk mengurangi konsentrasi amonia pada limbah cair. Penelitian ini bertujuan untuk mengetahui efektivitas penambahan sedimen kolam lele dan probiotik komersial dalam nitrifikasi amonia konsentrasi tinggi. Penelitian menggunakan bioreaktor sistem batch dengan volume kerja 1 liter yang berisi larutan amonia 100 mg/L dan 50 gram sedimen kolam lele. Perlakuan pada penelitian ini terdiri dari pemberian variasi konsentrasi probiotik komersial dan sedimen kolam lele dengan sumber karbon glukosa. Variasi probiotik komersial yang ditambahkan pada bioreaktor adalah 5 ml/L, 10 ml/L dan 15 ml/L sedangkan variasi konsentrasi glukosa adalah 0 g/L dan 3,9 g/L. Analisis konsentrasi amonia dilakukan secara spektrofotometri menggunakan Metode Fenat. Hasil penurunan rata-rata amonia tertinggi adalah sebesar 92,35% pada bioreaktor dengan campuran probiotik komersial 15 ml/L dan glukosa 3,9 g/L dengan laju penurunan ammonia tercepat pada hari ke-3 periode eksperimen. Penambahan glukosa pada bioreaktor mampu meningkatkan penurunan amonia sebesar 57,39%. Analisis statistik menunjukkan bahwa variasi konsentrasi probiotik komersial tidak menunjukkan perbedaan signifikan pada penurunan amonia (p0,05) sedangkan variasi konsentrasi glukosa menunjukkan perbedaan yang signifikan terhadap penurunan amonia (p0,05). Tiga isolat berhasil diisolasi menggunakan media spesifik bakteri nitrifikasi. Hasil identifikasi dari tiga bakteri menunjukkan bahwa isolat tersebut adalah Bacillus sp., Aeromonas salmonicida dan Burkholderia cepacia.Kata kunci: amonia, nitrifikasi, sedimen kolam lele, probiotik komersial

SummaryA new solid phase enzyme-linked immunosorbent assay (ELISA) was developed for the quantitation of human Factor VII antigen (F VII Ag), using a monospecific rabbit anti-F VII antiserum. Anti-F VII F(ab′)2 fragments were adsorbed to polystyrene plates. The binding of serial dilutions of control or test plasma, containing F VII, was detected by incubation with peroxidase-labeled anti- FV II IgG followed by the addition of hydrogen peroxyde and O-phenylenediamine. This ELISA is specific, sensitive (detection limit: 0.05%) and accurate (coefficient of variation: 1.5-4% for within- and 1.6-9% for between-assays). F VII coagulant activity (F VII C) and F VII Ag were determined in large populations of controls and patients. In normal plasma (n = 38), F VII Ag ranged from 83 to 117% and the correlation coefficient between F VII Ag and F VII C was 0.94. In patients with severe (F VII C inf. 1%) congenital F VII deficiency (n = 5), F VII Ag was undetectable in two cases (inf. 0.05%) and markedly reduced (0.35 to 5.6%) in the three other cases. In patients with liver cirrhosis (n = 15), F VII Ag ranged from 21 to 59% and was in good correlation with F VII C (r = 0.84). In dicoumarol treated patients (n = 15), the levels of F VII Ag ranged from 51% to 79% and a poor correlation (r = 0.52) with F VIIC was observed. In “compensated” DIC (n = 5), levels of F VII Ag varied from 60 to 186%, with significantly higher F VII C levels (from 143 to 189%). In contrast, in “decompensated” DIC (n = 7), low F VII Ag and F VII C levels were observed (from 7 to 27%). In patients with deep-vein thrombosis (n = 25), high levels of F VII Ag (from 102 to 136%) and F VII C (from 110 to 150%) were demonstrated. In surgical patients, no significant difference was observed before and one day after intervention.

AbstractIrradiated roast treatment and the ammonia leaching processing were conducted to deal with the low-grade oxide zinc ores. The ZnCO3phase was hard to be attended, which was the reason for the low leaching rate of the complicated zinc ores. The mineral phase transformation of the ZnCO3phase was generated after the ores irradiated in the microwave at the temperature of 673 K. The irradiated ores generated more small particles, it was the reason that the leaching rate of the complicated zinc ores was increased. When the leaching processing of the irradiated roasted ores was conducted under the conditions of the total ammonia concentration of 6 mol/L, the liquid to solid phase ratio of 11:1, the leaching temperature of 298 K, the leaching time of 150 min and the stirring speed of 400 rpm, 88.3 % of zinc could be achieved, which was the maximum leaching rate of the irradiated ores.

AbstractObjectivesData on the clinical importance of the detection of anti-dsDNA antibodies in patients with negative indirect immunofluorescence on the HEp-2 cell (IIF) are sparse and are especially not available for all common commercially available assays. This study aimed to assess the clinical significance of anti-dsDNA antibodies determined by the Elia™ dsDNA assay in patients with negative IIF.MethodsWe retrospectively examined the medical records of 234 consecutive subjects with detectable anti-dsDNA antibodies determined by the Elia™ dsDNA assay.ResultsA total of 124 subjects with detectable anti-dsDNA autoantibodies were IIF-negative, but yielded positive or borderline results in the Elia™ CTD screen assay for antinuclear antibodies (ANA). Within this group, 6/49 IIF-negative patients (12%) with ANA-associated systemic autoimmune rheumatic disorders (AASARD) and 118/185 subjects (64%) with various other diseases (Non-AASARD) were identified. There was no statistically significant difference with regard to the concentrations of anti-dsDNA antibodies (p=0.53) between the AASARD and the Non-AASARD group. Within the AASARD group, four patients diagnosed with systemic lupus erythematosus (SLE, treated), discoid lupus erythematosus (untreated), indetermined connective tissue disease (untreated) and polymyositis (treated) had positive anti-dsDNA autoantibodies, whereas two patients with treated SLE, thereby one in remission, had borderline concentrations of anti-dsDNA antibodies.ConclusionsOur findings suggest that the detection of anti-dsDNA antibodies in IIF-negative patients can be of clinical relevance in some cases. Our results further support the combined use of IIF and solid-phase assays in screening algorithms for ANA, in order to avoid overlooking potentially important autoantibody entities.

Mathematical modelling is considered a time and cost-saving tool for evaluation of new wastewater treatment concepts. Modelling can help to bridge the gap between lab and full-scale application. Bio-augmentation can be used to obtain nitrification in activated sludge systems with a limited aerobic sludge retention time. In the present study the potential for augmenting the endogenous nitrifying population is evaluated. Implementing a nitrification reactor in the sludge return line fed with sludge liquor with a high ammonia concentration leads to augmentation of the native nitrifying population. Since the behaviour of nitrifiers is relatively well known, a choice was made to evaluate this new concept mainly based on mathematical modelling. As an example an existing treatment plant (wwtp Walcheren, The Netherlands) that needed to be upgraded was used. A mathematical model, based on the TUDP model and implemented in AQUASIM was developed and used to evaluate the potential of this bioaugmentation in the return sludge line. A comparison was made between bio-augmentation and extending the existing aeration basins and anoxic tanks. The results of both modified systems were compared to give a quantitative basis for evaluation of benefits gained from such a system. If the plant is upgraded by conventional extension it needs an increase in volume of about 225%; using a bioaugmentation in the return sludge line the total volume of the tanks needs to be expanded by only 75% (including the side stream tanks). Based on the modelling results a decision was made to implement the bioaugmentation concept at full scale without further pilot scale testing, thereby strongly decreasing the scale-up period for this process.

Aim. To elucidate the role of Th1/Th2 polarization of immune response in LADA patients in the realization of the clinical phenotype of the disease. Materials and methods. 70 patients aged 21-61 (mean 41.3?1.0 yr) with DM diagnosed based on WHO criteria (1999). Groups 1 and 2 included 13 DM1and 57 DM2 patients (34.6?7.2 and 43.6?7.6 yr respectively). 27 DM2 patients (41.2?1.6 yr) presumably had LADA (P. Zimmet's criteria).Serum anti-GAD65, ICA, and IAA antibodies along with C-peptide were measured in fasting sera and 120 min after GTT by solid phase immunoenzymeassays following manufacturer's instructions with the use of a photometer for Multiscan EX microplates (ThermoLabSystems, Finland) at405 nm (for GAG and ICA) and 450 nm (for IAA and C-peptide). GAD, IAA, and C-peptides levels were calculated automatically from calibrationcurves. Mononuclear leukocytes were isolated by centrifugation in the ficoll-verographin density gradient. The cells thus obtained were resuspendedin the complete nutritient medium reducing their concentration to 2.0x10^6/ml. Phytohemagglutinin (Difco, Germany) was added (10 mcg/1 ml) tothe samples to stimulate mononuclear leukocytes; cell suspensions were further incubated for 24 hr. Initial and PGA-induced levels of IL-2, 4, 10 insupernatants of cell cultures were measured by solid phase immunoassay at 450 nm. Results. At least one type of autoantibodies (GAD, ICA or IAA) was identified in 24.3% of all DM patients (17/70) and in 18% of the DM2 patients(10/57). The level of anti-GAD and ICA ABs and percentage of AB-positive patients were higher in the LADA group while that of anti-IAA ABs amongDM1 patients without LADA. Two AB types at a time were found in 17% (4/23) of the patients with autoimmune DM in the absence of significantdifference between LADA and DM1. Patients with LADA had a significantly lower basal C-peptide level than DM2 patients. The was a tendencytoward lower level of stimulated C-peptide secretion in LADA patients compared with DM2 ones. It suggests impairment of beta-cell secretory functionaffected by the autoimmune process. We observed enhanced basal production of IFN-y by blood mononuclear leukocytes in all DM patients in theabsence of significant difference between the groups. Mitogen-activated production in all CD patients was lower than normal without inter-groupdifferences. Patients with DM2 had the inverted type of IL-2 secretion unlike those with autoimmune diabetes. In both cases it was significantly differentfrom normal values. There was a tendency toward higher basal production of IL-4 by mononuclear leukocytes in LADA and DM2 comparedwith CD1 which reflects pathogenetic peculiarities of beta-cell function in LADA differing from those in DM1 and responsible for slower impairment ofbeta-cell function in this condition. Basal and PGA- induced production of IL-10 was higher in LADA and DM2 than in DM1. It suggests enhancedsuppressor activity of leukocytes that may protect beta-cells from autoimmune destruction and determines gradual development of clinical symptoms ofinsulin deficiency. In contrast, low production of IL-10 in DM1 gives evidence of polarization of the immune response. Conclusion. The loss of functional parenchyma and manifestation of insulin deficiency in LADA occur at a relatively low rate due to the peculiarcharacter of cytokine-mediated cell interactions. It suggests the necessity of an active and careful diagnostic strategy with the use of immunologicalmethods for examination of elder patients presenting with a variety of pathogenetic variants of DM.

The method of soil samples preparation for measuring the (wetting) contact angle (CA) of the soil solid phase surface using membrane filters is proposed. The samples of kaolinite, a standard sample of chernozem and samples of agro-chestnut soil were taken for the experiment. The results of the CA measurements using two types of sample preparation for the analysis were compared. The first method of sample preparation was to apply a sample to a double-sided adhesive tape; the second method involved the deposition of suspensions of the studied samples of certain concentrations on membrane filters. The advantages and disadvantages of each sample preparation method are described. The significant difference in the obtained CA values depending on the sample preparation for measurement was revealed. The method of sample preparation with the use of membrane filters developed by the authors made it possible to reduce the CA measurement error by more than 2 times. Reducing the variation of the CA value of a single sample will allow comparing similar soil samples, including soils of the same type, but involved in different land use systems.

Benzo(a)pyrene (BaP) has been recognized as a marker for the detection of carcinogenic polycyclic aromatic hydrocarbons. In this work, a novel monolithic solid-phase extraction (SPE) sorbent based on graphene oxide nanoparticles (GO) in starch-based cryogel composite (GO-Cry) was successfully prepared for BaP analysis. Rice flour and tapioca starch (gel precursors) were gelatinized in limewater (cross-linker) under alkaline conditions before addition of GO (filler) that can increase the ability to extract BaP up to 2.6-fold. BaP analysis had a linear range of 10 to 1000 µgL−1 with good linearity (R2 = 0.9971) and high sensitivity (4.1 ± 0.1 a.u./(µgL−1)). The limit of detection and limit of quantification were 4.21 ± 0.06 and 14.04 ± 0.19 µgL−1, respectively, with excellent precision (0.17 to 2.45%RSD). The accuracy in terms of recovery from spiked samples was in the range of 84 to 110% with no significant difference to a C18 cartridge. GO-Cry can be reproducibly prepared with 2.8%RSD from 4 lots and can be reused at least 10 times, which not only helps reduce the analysis costs (~0.41USD per analysis), but also reduces the resultant waste to the environment.

CONTEXT: The association between Helicobacter pylori infection and colon neoplasia has been the subject of recent investigations which have produced controversial results. OBJECTIVE: To evaluate the prevalence of H. pylori infection in patients with colonic adenomas and also in patients whose colonoscopy exams were normal. METHODS: After colonoscopy, the individuals were distributed into two groups: patients with colon adenomas (cases) and patients whose colons were normal (controls). The groups were similar regarding age and gender. The individuals of both groups were subjected to a dosage of IgG antibody against H. pylori. The dosage was applied according to the solid phase, chemiluminescent immunometric assay. The chi-square test was used to analyze the data. RESULTS: There were 30 men and 64 women in each group (94 cases and 94 controls). The mean age of the cases was 59.79 ± 12.25 years and that of the controls was 58.98 ± 11.55 years. The H. pylori serology was positive for 66 (70.21%) of the cases and for 51 (54.25%) of the controls. There was a significant difference (P = 0.024). The odds ratio was 1.98 (CI 95%, 0.82-3.15). The prevalence of H. pylori in cases and controls according to gender, histological type and location of the colon lesions showed a significant difference only among women (P = 0.03), among patients with tubular adenomas (P = 0.03), and in those with distal adenomas (P = 0.038). CONCLUSION: There is a positive association between H. pylori infection and colonic adenomas. This association is more evident in women, especially for tubular adenomas and distal colonic location.
