Experimental assessment of RSF, UF, RSF-O3 and RSF-H2O2/UV for unrestricted agricultural wastewater reuse in Italy

In this paper, tertiary treatment processes aimed at achieving a wastewater quality suitable for reuse in agriculture in Italy have been investigated, with experimental results generated by means of pilot and bench scale tests. Studies were conducted to assess the removal of mineral oil, total surfactants, total coliforms, Escherichia coli and Salmonella using the following treatment options: rapid sand filtration (RSF), hollow fiber ultrafiltration (UF), RSF followed by ozonation (RSF-O3) and RSF followed by hydrogen peroxide combined with UV radiation (RSF-H2O2/UV). Mineral oil concentration, evaluated by means of the hydrocarbon oil index measurement, indicated an effluent concentration consistently below 0.05 mg/L for all processes studied. While total surfactants in the secondary effluent never exceeded the applicable limit of 0.5 mg/L during the studies, the degree of removal measured in studied treatments ranged from moderate to low, with the greatest removal observed using RSF-O3 (24%) and RSF-H2O2/UV (30%) under applied conditions. Overall, the optimal treatment performances were achieved by the RSF-H2O2/UV combined process using ≥1.5 mg/L H2O2 and UV dose ≥45 mJ/cm2, which provided adequate mineral oil and total surfactants removal, complete removal of measurable total suspended solids (TSS) and Salmonella, and greater than 4-log reduction in total coliforms and Escherichia coli.

Wastewater reuse in arid regions is important for the production of a water resource to be utilised for non-potable purposes and to prevent the environmental transmission of disease-causing agents. This study was conducted to evaluate the effect of water quality on the comparative disinfection efficiency of viruses, bacteria and spores by UV irradiation. Furthermore, the microbial quality of effluent produced by coagulation, high rate filtration (HRF) and either UV irradiation or chlorination was determined. Using low pressure collimated beam, a UV dose of 80 mWs/cm2 was needed to achieve a 3-log10 inactivation of either rotavirus SA-11 or coliphage MS2, whereas over 5-log10 inactivation of E. coli was reached with a dose of only 20 mWs/cm2. B. subtilis inactivation was found to be linear up to a dose of 40 mWs/cm2 and then a tailing up to a UV dose of 120 mWs/cm2 was observed. It is worth noting that effluent turbidity of <5 NTU did not influence the inactivation efficiency of UV irradiation. Operation of a pilot plant to treat secondary effluent by coagulation, HRF and UV disinfection at a UV dose of 80 mWs/cm2 resulted in the production of high quality effluent in compliance with the Israel standards for unrestricted irrigation (<10 CFU/100 mL faecal coliform and turbidity of <5 NTU). Sulphite reducing clostridia (SRC) were found to be more resistant than coliphages and F coliform for UV irradiation. The results of this study indicated that UV disinfection is suitable for the production of effluents for unrestricted irrigation of food crops.

This study examined the effects and interactions of UV light dose (1,800 to 20,331 μJ/cm2) and apple cider pH (2.99 to 4.41) on the inactivation of Escherichia coli ATCC 25922, a surrogate for E. coli O157:H7. A predictive model was developed to relate the log reduction factor of E. coli ATCC 25922 to the UV dose. Bacterial populations for treated and untreated samples were enumerated with the use of nonselective media. The results revealed that UV dose was highly significant in the inactivation of E. coli, whereas pH showed no significant effect at higher UV doses. Doses of 6,500 μJ/cm2 or more were sufficient to achieve a greater than 5-log reduction of E. coli. Experimental inactivation data were fitted adequately by a logistic regression model. UV irradiation is an attractive alternative to conventional methods for reducing bacteria in unpasteurized apple cider.

Every year around 3.4 million people die from water-related diseases, mainly amoebiasis and diarrhoea caused by bacteria. The assessment of the efficiency of a UV light disinfection process in the inactivation of indicators, pathogen bacteria and amphizoic amoebae in a secondary treated effluent was carried out. Wastewater was irradiated with different doses of UV light using a collimated-beam reactor. Dose-response results showed that a UV dose of 15 mW·s/cm2 was enough to inactivate FC to the limit established in Mexican legislation (<1,000 MPN/100 ml) for irrigation reuse, as well as reaching a 2 log inactivation for faecal streptococci. Also, the final concentration reached for Salmonella typhi was <103 MPN, which is the minimal concentration for a disease response in humans. The isolation from the secondary effluent and the pathogenicity determination of Acanthamoeba were carried out. Amoebae were concentrated to higher concentrations than those of the effluent for irradiation tests. Radiation tests showed that amoebae required higher doses than bacteria for their total inactivation (60 mW·s/cm2 for a 2 log reduction). Results showed that UV light is an alternative to inactivate high contents of bacteria and amoebae trophozoites, although higher doses were needed for the high concentrations of amoebae used.

Among the activities appointed by the EC research-project “Integrated water recycling and emission abatement in the textile industry” (Contract: ENV4-CT95-0064), the effectiveness of ozone for improving the biotreatability of recalcitrant effluents as well as for removing from them toxic and/or inhibitory pollutants has been evaluated at lab-scale. Real membrane concentrates (pH=7.9; TOC=190 ppm; CDO=595 ppm; BOD5=0 ppm; Conductivity=5,000 μS/cm; Microtox-EC20=34%) produced at Bulgarograsso (Italy) Wastewater Treatment Plant by nanofiltering biologically treated secondary textile effluents, have been treated with ozonated air (O3conc.=12 ppm) over 120 min. The results have indicated that during ozonation, BOD5 increases from 0 to 75 ppm, whereas COD and TOC both decrease by about 50% and 30 % respectively. As for potentially toxic and/or inhibitory pollutants such as dyes, nonionic surfactants and halogenated organics, all measured as sum parameters, removals higher than 90% were achieved as confirmed by the complete disappearance of acute toxicity in the treated streams. The only ozonation byproducts searched for and found were aldehydes whose total amount continuously increased in the first hour from 1.2 up to 11.8 ppm. Among them, formaldehyde, acetaldehyde, glyoxal, propionaldehyde, and butyraldehyde were identified by HPLC.

Bacteriophage biocontrol has potential as a means of mitigating the prevalence of Escherichia coli O157:H7 in ruminants. The efficacy of oral administration of bacteriophages for reducing fecal shedding of E. coli O157:H7 by sheep was evaluated using 20 Canadian Arcott rams (50.0 ± 3.0) housed in four rooms (n = 5) in a contained facility. The rams had ad libitum access to drinking water and a pelleted barley-based total mixed ration, delivered once daily. Experimental treatments consisted of administration of E. coli O157:H7 (O157), E. coli O157:H7+bacteriophages (O157+phage), bacteriophages (phage), and control (CON). Oral inoculation of the rams with 109 CFU of a mixture of four nalidixic acid-resistant strains of E. coli O157:H7 was performed on day 0. A mixture of 1010 PFU of bacteriophages P5, P8 and P11 was administered on days -2, -1, 0, 6 and 7. Fecal samples collected on 14 occasions over 21 d were analyzed for E. coli O157:H7, total E. coli, total coliforms and bacteriophages. Sheep in treatment O157+phage shed fewer (P < 0.05) E. coli O157:H7 than did sheep in treatment O157. Populations of total coliforms and total E. coli were similar (P < 0.05) among treatments, implying that bacteriophage lysis of non-target E. coli and coliform bacteria in the gastrointestinal tract did not occur. Bacteriophage numbers declined rapidly over 21 d, which likely reduced the chance of collision between bacteria and bacteriophage. Oral administration of bacteriophages reduced shedding of E. coli O157:H7 by sheep, but a delivery system that would protect bacteriophages during passage through the intestine may increase the effectiveness of this strategy as well as allow phage to be administered in the feed.Key words: Escherichia coli O157:H7, bacteriophage, sheep, environment, coliforms

The objectives of this study were to determine if washing of whole apples with solutions of three different sanitizers (peroxyacetic acid, chlorine dioxide, or a chlorine-phosphate buffer solution) could reduce a contaminating nonpathogenic Escherichia coli O157:H7 population by 5 logs and at what sanitizer concentration and wash time such a reduction could be achieved. Sanitizers were tested at 1, 2, 4, 8, and 16 times the manufacturer's recommended concentration at wash times of 5, 10, and 15 min. Whole, sound Braeburn apples were inoculated with approximately 1 × 108 or 7 × 106 CFU per apple, stored for 24 h, then washed with sterile water (control) or with sanitizers for the prescribed time. Recovered bacteria were enumerated on trypticase soy agar. Washing with water alone reduced the recoverable population by almost 2 logs from the starting population; this can be attributed to physical removal of organisms from the apple surface. No sanitizer, when used at the recommended concentration, reduced the recovered E. coli population by 5 logs under the test conditions. The most effective sanitizer, peroxyacetic acid, achieved a 5-log reduction when used at 2.1 to 14 times its recommended concentration, depending on the length of the wash time. The chlorine-phosphate buffer solution reduced the population by 5 logs when used at 3 to 15 times its recommended concentration, depending on wash time. At no concentration or wash time tested did chlorine dioxide achieve the 5-log reduction.

Iceberg lettuce is a major component in vegetable salad and has been associated with many outbreaks of foodborne illnesses. In this study, several combinations of lactic acid and hydrogen peroxide were tested to obtain effective antibacterial activity without adverse effects on sensory characteristics. A five-strain mixture of Escherichia coli O157:H7, Salmonella enterica serotype Enteritidis, and Listeria monocytogenes was inoculated separately onto fresh-cut lettuce leaves, which were later treated with 1.5% lactic acid plus 1.5% hydrogen peroxide (H2O2) at 40°C for 15 min, 1.5% lactic acid plus 2% H2O2 at 22°C for 5 min, and 2% H2O2 at 50°C for 60 or 90 s. Control lettuce leaves were treated with deionized water under the same conditions. A 4-log reduction was obtained for lettuce treated with the combinations of lactic acid and H2O2 for E. coli O157:H7 and Salmonella Enteritidis, and a 3-log reduction was obtained for L. monocytogenes. However, the sensory characteristics of lettuce were compromised by these treatments. The treatment of lettuce leaves with 2% H2O2 at 50°C was effective not only in reducing pathogenic bacteria but also in maintaining good sensory quality for up to 15 days. A ≤4-log reduction of E. coli O157:H7 and Salmonella Enteritidis was achieved with the 2% H2O2 treatment, whereas a 3-log reduction of L. monocytogenes was obtained. There was no significant difference (P &gt; 0.05) between pathogen population reductions obtained with 2% H2O2 with 60- and 90-s exposure times. Hydrogen peroxide residue was undetectable (the minimum level of sensitivity was 2 ppm) on lettuce surfaces after the treated lettuce was rinsed with cold water and centrifuged with a salad spinner. Hence, the treatment of lettuce with 2% H2O2 at 50°C for 60 s is effective in initially reducing substantial populations of foodborne pathogens and maintaining high product quality.

ABSTRACT Poor-quality medicines undermine the treatment of infectious diseases, such as tuberculosis, which require months of treatment with rifampin and other drugs. Rifampin resistance is a critical concern for tuberculosis treatment. While subtherapeutic doses of medicine are known to select for antibiotic resistance, the effect of drug degradation products on the evolution of resistance is unknown. Here, we demonstrate that substandard drugs that contain degraded active pharmaceutical ingredients select for gene alterations that confer resistance to standard drugs. We generated drug-resistant Escherichia coli and Mycobacterium smegmatis strains by serially culturing bacteria in the presence of the rifampin degradation product rifampin quinone. We conducted Sanger sequencing to identify mutations in rifampin-resistant populations. Strains resistant to rifampin quinone developed cross-resistance to the standard drug rifampin, with some populations showing no growth inhibition at maximum concentrations of rifampin. Sequencing of the rifampin quinone-treated strains indicated that they acquired mutations in the DNA-dependent RNA polymerase B subunit. These mutations were localized in the rifampin resistance-determining region (RRDR), consistent with other reports of rifampin-resistant E. coli and mycobacteria. Rifampin quinone-treated mycobacteria also had cross-resistance to other rifamycin class drugs, including rifabutin and rifapentine. Our results strongly suggest that substandard drugs not only hinder individual patient outcomes but also restrict future treatment options by actively contributing to the development of resistance to standard medicines.
