The European bison (Bison bonasus) is an endangered species whose restitution requires systematic health monitoring. The most practical way to test clinical material from large wildlife is by serological methods. Recently, the most serious health hazard faced by the European bison population was tuberculosis. However, climate change and globalization have resulted in the arrival of more exotic diseases, one of which is epizootic hemorrhagic disease (EHD), caused by the epizootic hemorrhagic disease virus (EHDV). EHD is an arthropodtransmitted viral disease noted mainly in ungulates, especially ruminants. Clinical signs vary depending on the virus serotype and host. The aim of this study was to determine the prevalence of EHD in European bison by serological testing for antibodies to EHDV. Serum samples were collected from 90 European bison living in different regions of Poland and tested with PrioCHECK™ EHDV Ab Serum Kit (Thermofisher, US). One of the bison (from the Knyszyńska Forest) had a positive result. It cannot be excluded, however, that the result was a false positive. There is a need for further EHDV serology monitoring in European bison and other wildlife, particularly in the Knyszyńska Forest.

ABSTRACT A recent outbreak of hemorrhagic fever in wild ruminants in the northwest United States was characterized by rapid onset of fever, followed shortly thereafter by hemorrhage and death. As a result, a confirmed 1,000 white-tailed deer and pronghorn antelope died over the course of 3 months. Lesions were multisystemic and included severe edema, congestion, acute vascular necrosis, and hemorrhage. Animals that died with clinical signs and/or lesions consistent with hemorrhagic fever had antibody to epizootic hemorrhagic disease virus serotype 2 (EHDV-2) by radioimmune precipitation but the antibody was limited exclusively to class immunoglobulin M. These findings, indicative of acute infection, were corroborated by the observation that numerous deer were found dead; however, clinically affected deer were rarely seen during the outbreak. Furthermore, only in animals with hemorrhagic lesions was EHDV-2 isolated and/or erythrocyte-associated EHDV-2 RNA detected by serotype-specific reverse transcription (RT)-PCR. By using a novel RT in situ PCR assay, viral nucleic acid was localized to the cytoplasm of large numbers of tissue leukocytes and vascular endothelium in tissues with hemorrhage and to vessels, demonstrating acute intimal and medial necrosis. Because PCR amplification prior to in situ hybridization was essential for detecting EHDV, the virus copy number within individual cells was low, <20 virus copies. These findings suggest that massive covert infection characterized by rapid dissemination of virus facilitates the severe and lethal nature of this disease.

Abstract A serological study of twenty three European bison (Bison bonasus) derived from Northern-East Poland for the seroprevalence of Mycobacterium avium subsp. paratuberculosis, Mycoplasma bovis, Mycoplasma mycoides subsp. mycoides SC, Mycoplasma agalactiae and Mycoplasma capricolum subsp. capripneumoniae was conducted. Only specific antibodies to M. bovis were detected in two animals (8.7%) which were connected with the clinical signs and macroscopic anatomopathological lesions.

The European bison (Bison bonasus) is an endangered species which faces a range of health threats. As little is known of exposure of European bison to paratuberculosis caused by Mycobacterium avium spp. paratuberculosis, known to cause losses in cattle, the aim of the present study was to conduct serological survey in Polish bison herds. Between September 2018 and February 2021, blood samples were collected from 165 European bison from different regions of Poland. Samples were taken whenever the animals were immobilized (e.g., putting on telemetry collars) as well as from any dead animal. The serum samples were tested with ELISA. Three individuals, originating from different captive herds, were found to be seropositive. In conclusion, it was found that Mycobacterium avium subsp. paratuberculosis infections are not currently a problem in European bison, especially in free-range herds.

An enzyme-linked immunosorbent assay has been developed to detect antibodies to epizootic hemorrhagic disease of deer virus (EHDV). The assay incorporates a monoclonal antibody to EHDV serotype 2 (EHDV-2) that demonstrates specificity for the viral structural protein, VP7. The assay was evaluated with sequential sera collected from cattle experimentally infected with EHDV serotype 1 (EHDV-1) and EHDV-2, as well as the four serotypes of bluetongue virus (BTV), BTV-10, BTV-11, BTV-13, and BTV-17, that currently circulate in the US. A competitive and a blocking format as well as the use of antigen produced from both EHDV-1-and EHDV-2-infected cells were evaluated. The assay was able to detect specific antibody as early as 7 days after infection and could differentiate animals experimentally infected with EHDV from those experimentally infected with BTV. The diagnostic potential of this assay was demonstrated with field-collected serum samples from cattle, deer, and buffalo.
