The intracellular protozoal parasite Toxoplasma gondii has been associated with worsened cognitive function in animal models and in humans. Despite these associations, the mechanisms by which Toxoplasma gondii might affect cognitive function remain unknown, although Toxoplasma gondii does produce physiologically active intraneuronal cysts and appears to affect dopamine synthesis. Using data from the UK Biobank, we sought to determine whether Toxoplasma gondii is associated with decreased prefrontal, hippocampal, and thalamic gray-matter volumes and with decreased total gray-matter and total white-matter volumes in an adult community-based sample. The results from adjusted multivariable regression modelling showed no associations between Toxoplasma gondii and prefrontal, hippocampal, and thalamic brain gray-matter volumes. In contrast, natural-log transformed antibody levels against the Toxoplasma gondii p22 (b = -3960, 95-percent confidence interval, -6536 to -1383, p < .01) and sag1 (b = -4863, 95-percent confidence interval, –8301 to -1425, p < .01) antigens were associated with smaller total gray-matter volume, as was the mean of natural-log transformed p22 and sag1 titers (b = -6141, 95-percent confidence interval, -9886 to -2397, p < .01). There were no associations between any of the measures of Toxoplasma gondii and total white-matter volume. These findings suggest that Toxoplasma gondii might be associated with decreased total gray-matter in middle-aged and older middle-aged adults in a community-based sample from the United Kingdom.

Background We know little about the pathogenesis and diagnosis of retinal detachment. Purpose To assess spontaneous changes in the cerebral cortex of patients with retinal detachment using voxel-based morphometry and to explore the relationship between retinal detachment and clinical behavioral performance. Material and Methods Patients (14 men, 6 women; average age = 49.15 ± 10.32 years) with rhegmatogenous retinal detachment (duration of 24.05 ± 19.61 days) and 20 matched healthy controls were recruited. All participants underwent repeated functional magnetic resonance imaging scans. The original three-dimensional T1 brain images were analyzed using voxel-based morphometry and whole brain white matter volume and whole brain gray matter volume were compared with those of the control group. A receiver operating characteristic (ROC) curve was used to classify the mean gray matter volume values of the patients with retinal detachment compared with the controls. Results Compared with the controls, whole brain gray matter volume was significantly reduced in patients with retinal detachment, as evidenced by changes in the right inferior frontal gyrus, right superior temporal gyrus, right anterior cingulate gyrus, and right cuneus. In addition, the posterior lobe of the cerebellum, left hippocampus, left cingulate gyrus, and left middle temporal gyrus were also obviously atrophied. Furthermore, whole brain white matter volume of the patients with retinal detachment showed a slight reduction. The ROC curve analysis of each brain region showed that the accuracy of the area under the curve was high. Conclusion We proved that patients with retinal detachment had unusual changes in the gray matter volume and white matter volume in vision-related brain regions, which could reveal potential pathological mechanisms of retinal detachment.

ObjectiveTo assess the association of early morning serum cortisol with cognitive performance and brain structural integrity in community-dwelling young and middle-aged adults without dementia.MethodsWe evaluated dementia-free Framingham Heart Study (generation 3) participants (mean age 48.5 years, 46.8% men) who underwent cognitive testing for memory, abstract reasoning, visual perception, attention, and executive function (n = 2,231) and brain MRI (n = 2018) to assess total white matter, lobar gray matter, and white matter hyperintensity volumes and fractional anisotropy (FA) measures. We used linear and logistic regression to assess the relations of cortisol (categorized in tertiles, with the middle tertile as referent) to measures of cognition, MRI volumes, presence of covert brain infarcts and cerebral microbleeds, and voxel-based microstructural white matter integrity and gray matter density, adjusting for age, sex, APOE, and vascular risk factors.ResultsHigher cortisol (highest tertile vs middle tertile) was associated with worse memory and visual perception, as well as lower total cerebral brain and occipital and frontal lobar gray matter volumes. Higher cortisol was associated with multiple areas of microstructural changes (decreased regional FA), especially in the splenium of corpus callosum and the posterior corona radiata. The association of cortisol with total cerebral brain volume varied by sex (p for interaction = 0.048); higher cortisol was inversely associated with cerebral brain volume in women (p = 0.001) but not in men (p = 0.717). There was no effect modification by the APOE4 genotype of the relations of cortisol and cognition or imaging traits.ConclusionHigher serum cortisol was associated with lower brain volumes and impaired memory in asymptomatic younger to middle-aged adults, with the association being evident particularly in women.
