Two polymorphic esterase loci, EstA and EstB, of the olive-fruit fly Dacus oleae were studied in a natural population. The analysis of about 500 individuals revealed the presence of 15 alleles for EstA and 12 alleles for EstB. A ‘silent’ allele was found segregating at both loci. Segregation data for most of the alleles are presented. The allele frequency distribution follows the same pattern at both loci: one allele of each gene has a frequency of nearly 0·50, a few have frequencies between 0·05 and 0·15 and many are below 0·05. Two main hypotheses, those of overdominance and selective neutrality, were examined in order to explain these polymorphisms. We deduced that under both hypotheses a relatively high mutation rate is necessary to balance the result of random drift. This rate was estimated to be higher than 4 × 10−5 for the EstA locus. Since homozygotes for the ‘silent’ allele at the first or at the second locus were found in the population in expected frequencies, it was concluded that these alleles are not inferior to active ones under natural conditions.

ABSTRACTA metagenomic library was constructed from microorganisms associated with the brown algaAscophyllum nodosum. Functional screening of this library revealed 13 novel putative esterase loci and two glycoside hydrolase loci. Sequence and gene cluster analysis showed the wide diversity of the identified enzymes and gave an idea of the microbial populations present during the sample collection period. Lastly, an endo-β-1,4-glucanase having less than 50% identity to sequences of known cellulases was purified and partially characterized, showing activity at low temperature and after prolonged incubation in concentrated salt solutions.

SUMMARY The genetics of anodal esterase (Est), cathodal esterase (cEst), cathodal acid phosphatase (cAcp) and malate dehydrogenase (Mdh) has been studied in mature seeds and leaves (genetics of cAcp and Mdh has not been studied in leaves) of sunflower (Helianthus annuus L.). A total of ten loci (four loci of anodal esterase, two loci of cathodal esterase, three loci of malate dehydrogenase and one locus of cathodal acid phosphatase) have been identified and described. Five esterase loci (Est1, Est2, Est3, Est4, cEst5), three malate dehydrogenase loci and one locus of cathodal acid phosphatase are expressed in seeds. Three esterase loci (Est2, cEst5 and cEst6) are expressed in leaves. The analysis of linkage between these loci has been made. Two linkage groups have been found. The sequence of the loci in the first linkage group was Mdh2-Est1- Est2-Est3-cEst5. In the second linkage group it was Est4-cAcp1. Linkages have been analyzed between three isoenzymatic loci expressed in leaves and between two loci controlling morphological traits (branched stem and male fertility restoration). The linkage between morphological traits and isoenzymatic loci has not been revealed. It has been revealed in Br-Rf pair.

Chromosome 2 inversion, Est-l and Est-2 haplotype frequencies were determined for 19 wild D. buzzatii collections from the known range of this species in Australia. Three different chromosome 2 sequences (ST, j, j z3) were polymorphic across the collections. They occurred at overall frequencies which approximated those in the ancestral New World and colonized Old World populations, which indicated that no radical genetic change was associated with the colonization of Australia by D. buzzatii. Linkage disequilibria of Est-l and Est-2 alleles with the inversions tended to be strong, and consistent in direction, in almost all collections. The distributions of conditional allele frequencies within the different inversions were consistent with stochastic historical explanations for the linkage disequilibria. Significant linkage disequilibria between Est-l and Est-2 were evident after correcting for the inversions, but these disequilibria largely were restricted to southern (higher latitude) populations, and were inconsistent in direction among the collections. Hence, population bottlenecks may underlie these genic disequilibria.
