General control nonderepressible 2 (GCN2) is an environmental sensor controlling transcription and translation in response to nutrient availability. Although GCN2 is a putative therapeutic target for immuno-oncology, its role in shaping the immune response to tumors is poorly understood. Here, we used mass cytometry, transcriptomics, and transcription factor–binding analysis to determine the functional impact of GCN2 on the myeloid phenotype and immune responses in melanoma. We found that myeloid-lineage deletion of GCN2 drives a shift in the phenotype of tumor-associated macrophages and myeloid-derived suppressor cells (MDSCs) that promotes antitumor immunity. Time-of-flight mass cytometry (CyTOF) and single-cell RNA sequencing showed that this was due to changes in the immune microenvironment with increased proinflammatory activation of macrophages and MDSCs and interferon-γ expression in intratumoral CD8+ T cells. Mechanistically, GCN2 altered myeloid function by promoting increased translation of the transcription factor CREB-2/ATF4, which was required for maturation and polarization of macrophages and MDSCs in both mice and humans, whereas targeting Atf4 by small interfering RNA knockdown reduced tumor growth. Last, analysis of patients with cutaneous melanoma showed that GCN2-dependent transcriptional signatures correlated with macrophage polarization, T cell infiltrates, and overall survival. Thus, these data reveal a previously unknown dependence of tumors on myeloid GCN2 signals for protection from immune attack.

Rigid filters made of ceramic or metal are widely used to remove solid particles from hot gases at temperature above 260 °C in the petrochemical and coal industries. Pulse-jet cleaning of fine dust from rigid filter candles plays a critical role in the long-term operation of these filters. In this study, an experimental apparatus was fabricated to investigate the behavior of a 2050 mm filter candle, which included monitoring the variation of pressure dynamic characteristics over time and observing the release of dust layers that allowed an analysis of the cleaning performance of ISO 12103-1 test dusts with different particle size distributions. These results showed the release behavior of these dusts could be divided into five stages: radial expansion, axial crack, flaky release, irregular disruption and secondary deposition. The cleaning performance of smaller sized dust particles was less efficient as compared with larger sized dust particles under the same operating conditions primarily because large, flaky-shaped dust aggregates formed during the first three stages were easily broken into smaller, dispersed fragments during irregular disruption that forced more particles back to the filter surface during secondary deposition. Also, a “low-pressure and long-pulse width” cleaning method improved the cleaning efficiency of the A1 ultrafine test dust from 81.4% to 95.9%.

Electrodialysis (ED) desalination performance of different conventional and laboratory-scale ion exchange membranes (IEMs) has been evaluated by many researchers, but most of these studies used their own sets of experimental parameters such as feed solution compositions and concentrations, superficial velocities of the process streams (diluate, concentrate, and electrode rinse), applied electrical voltages, and types of IEMs. Thus, direct comparison of ED desalination performance of different IEMs is virtually impossible. While the use of different conventional IEMs in ED has been reported, the use of bioinspired ion exchange membrane has not been reported yet. The goal of this study was to evaluate the ED desalination performance differences between novel laboratory‑scale bioinspired IEM and conventional IEMs by determining (i) limiting current density, (ii) current density, (iii) current efficiency, (iv) salinity reduction in diluate stream, (v) normalized specific energy consumption, and (vi) water flux by osmosis as a function of (a) initial concentration of NaCl feed solution (diluate and concentrate streams), (b) superficial velocity of feed solution, and (c) applied stack voltage per cell-pair of membranes. A laboratory‑scale single stage batch-recycle electrodialysis experimental apparatus was assembled with five cell‑pairs of IEMs with an active cross-sectional area of 7.84 cm2. In this study, seven combinations of IEMs (commercial and laboratory-made) were compared: (i) Neosepta AMX/CMX, (ii) PCA PCSA/PCSK, (iii) Fujifilm Type 1 AEM/CEM, (iv) SUEZ AR204SZRA/CR67HMR, (v) Ralex AMH-PES/CMH-PES, (vi) Neosepta AMX/Bare Polycarbonate membrane (Polycarb), and (vii) Neosepta AMX/Sandia novel bioinspired cation exchange membrane (SandiaCEM). ED desalination performance with the Sandia novel bioinspired cation exchange membrane (SandiaCEM) was found to be competitive with commercial Neosepta CMX cation exchange membrane.

The recent stringent regulations on vehicle safety and reducing CO2 emissions have led to a continuous increase in the application of press-hardened steel (PHS) in automobiles. Similar to other high-strength steels, assembling PHS components using the common welding techniques employed in automotive production lines is significantly difficult because of the surface coating layers and the additives within. This difficulty in post-processing, attributed to its high strength, also limits the mechanical fastening of PHS components. Therefore, this study aims to develop a process for forming a structure enabling mechanical fastening by sequentially applying piercing and hole-flanging operations during the hot stamping process. Our experimental apparatus was designed to perform the hole-flanging operation after the piercing operation within a single stroke at a specific temperature during the quenching process of PHS. At high temperatures of 440 °C or higher, the hole-flanging process was conducted in a direction opposite to that of the piercing operation for creating the pilot hole. An extruded collar with a height of 8.0 mm and a diameter of 17.5 mm was achieved, which is hole expansion ratio(HER) of 82.5%.

Intermediate Temperature Solid Oxide Fuel Cell (IT-SOFC) technology offers interesting opportunities in the panorama of a larger penetration of renewable and distributed power generation, namely high electrical efficiency at manageable scales for both remote and industrial applications. In order to optimize the performance and the operating conditions of such a pre-commercial technology, an effective synergy between experimentation and simulation is fundamental. For this purpose, starting from the SIMFC (SIMulation of Fuel Cells) code set-up and successfully validated for Molten Carbonate Fuel Cells, a new version of the code has been developed for IT-SOFCs. The new release of the code allows the calculation of the maps of the main electrical, chemical, and physical parameters on the cell plane of planar IT-SOFCs fed in co-flow. A semi-empirical kinetic formulation has been set-up, identifying the related parameters thanks to a devoted series of experiments, and integrated in SIMFC. Thanks to a multi-sampling innovative experimental apparatus the simultaneous measurement of temperature and gas composition on the cell plane was possible, so that a preliminary validation of the model on local values was carried out. A good agreement between experimental and simulated data was achieved in terms of cell voltages and local temperatures, but also, for the first time, in terms of local concentration on the cell plane, encouraging further developments. This numerical tool is proposed for a better interpretation of the phenomena occurring in IT-SOFCs and a consequential optimization of their performance.
