The aim of this research was to evaluate the effect of chayote (Sechium edule) leaves infusion on hemoglobin and hematocrit level of anemic rat (Rattus norvegicus). Twelve anemic hemorrhagic male rats strain Wistar with body weight ranging from 175-200 g were used as sample in this research. Rats were divided randomly into 3 treatment groups, each group consist of 4 rats. The control group (P0) was not given chayote leaves infusion. Group P1 and P2 were given chayote leaves infusion with the concentration of 25% and 50% respectively, with dose of 1 ml per 200 gram body weight. Rats were fed with standard commercial food (Turbo feed) and aquadest ad libitum. The rats were treated anemic by taking 20% blood from total blood volume prior to administration of chayote laeves infusion. Chayote leaves infusion was given for 10 days consecutively. Blood sampling was carried out on day 0, 5, and 10 via caudal vein. Data were analyzed using anlysis of variance (ANOVA) Split-Plot pattern. The result showed that chayote leaves infusion increase level of hemoglobin and hematocrit in experimental rats. In conclusion 25% concentration of chayote leaves infusion is more effective than 50% concentration.Key words: anemia, chayote leaves, hemoglobin, hematocrit

The study was conducted to investigate the effects of caffeine treatment during organogenesis period to the fetal birth weight, using rat (Rattus norvegicus) as the animal model. Thirty-six primipararat obtained from Unit Pengembangan Hewan Percobaan, Gadjah Mada University (UPHP-GMU), 3 month old, 165-200 g body weight, were divided into 6 groups, consisted of 6 rats each. Six of the ratshave been selected based on the estrous cycles, and only rat with regular estrous were use for theexperiment. The rat then were mated, and during day 6-14 of the pregnancies were treated orally withcaffeine diluted in aquadest in dosage: placebo (1 cc aquadest) for group I (control), and 5.4, 10.8, 16.2,21.6, and 27 g/200 g body weight/day for treatment groups II-VI respectively. The pregnant rat bodyweights were determined at day 6 of pregnancies for calculating the caffeine treatment dosages. At day 20thof the pregnancies all of the pregnant rats were caesarotomized, and all of the fetuses were removed and weighed. The results showed that all of the treatment groups have significantly lower birth weightcompare to the groups control group. More over, fetal obtained from the treatment groups also showedserious subcutaneous hemorrhagic.Keywords: organogenesis, Rattus norvegicus, birth weight

Synergy1, a prebiotic composed of Inulin and Oligofructose (1 : 1). Soybean meal is a natural source of isoflavones. The objective was to investigate the effects of feeding Synergy1 and SM on the incidence of azoxymethane- (AOM-) induced aberrant crypt foci (ACF) in Fisher 344 male rats. Rats (54) were randomly assigned to 9 groups (n=6). Control group (C) was fed AIN-93G and treatment groups Syn1 and SM at 5% and 10% singly and in combinations. Rats were injected with two s/c injections of AOM at 7 and 8 weeks of age at 16 mg/kg body weight and killed at 17 weeks by CO2asphyxiation. Colonic ACF enumeration and hepatic enzyme activities were measured. Reductions (%) in total ACF among treatment groups fed combinations were higher (67–77) compared to groups fed singly (52–64). Synergistic mechanisms among phytochemicals may be responsible suggesting protective role in colon carcinogenesis with implications in food product development.

The objective of this study was to determine the effect of centella leaf extract administration on decreased of the molecule cAMP responsive element modulator (CREM) expression in the testicular seminiferous tubules of male rats (Rattus norvegicus). Eight rats, aged 3.5 months with 150-250 grams of body weight (BW) were used in this study. All rats were divided randomly into four groups as if K0 as a control group whereas K1, K2, and K3 were given the centella leaf extract with doses 125, 250, and 500 mg / kg body weight respectivelly that given once daily for 30 days. At the end of the treatment, rats were sacrificed and their testes were collected and subsequently fixed in buffered neutral formalin (BNF) 10% as fixative solution for histological preparation. The CREMs expressions were detected using immunohistochemical methods. The results showed that the number of CREM expression in the seminiferous tubules significantly differ (P <0.05) between K0 and the treatment group (K1, K2, and K3). Conclusion, the administration of centella leaf extract with of the dose 125, 250, and 500 mg/kg BW can decreased CREM expression spermatids of testicular seminiferous tubules in male rat.

Overtraining syndrome is a decrease in physical capacity, emotions and immunity due to training that is too often without adequate periods of rest. Overtraining is often experienced by athletes who daily undergo heavy training with short break periods. This research aims to look at the effect of overtraining aerobic physical exercise on memory in mice. The research method was experimental in vivo with the subject of adult male rat (Rattus Norvegicus) Winstar strain aged 8-10 weeks, body weight 200-250 gr. Divided into three groups, namely the control group, aerobic group and overtraining group. The results of memory tests with water E Maze showed an increase in the duration of travel time and the number of animal errors made by the overtraining group (p = 0.003). This study concludes that overtraining aerobic physical exercise can reduce memory in rat hippocampus.

Phytochemical composition and antioxidant activity of flowers decoction at post-flowering stage (F3D) of Opuntia dejecta were determined. The obtained findings demonstrate that F3D has a marked antioxidant activity in all tested assays. Furthermore, the present study was designed to test the protective activity of F3D against induced Diabetes type 2 (DT2) in male rats. Those metabolic syndromes were induced by a high-fructose diet (HFD) (10% fructose solution) for a period of 20 weeks. F3D was administered orally (100 and 300 mg/kg body weight) daily for the last 4 weeks. Metformin (150 mg/kg body weight) was used as a standard drug and administrated orally for the last 4 weeks. The results showed a significant increase in blood glucose, triglycerides and hepatic markers (ALAT, ASAT and ALK-P) in HFD group. A significant increase in hepatic TBARS and a significant decrease in SOD, CAT and GPX were observed in fructose fed rats compared to control group. Administration of F3D showed a protective effect in biochemical and oxidative stress parameters measured in this study. Also, oral administration of F3D restored the histological architecture of rat liver in comparison with rats fed HFD. In conclusion, F3D attenuated hepatic oxidative stress in fructose-fed rats.

This study examined the effects of increasing levels of dietary fat fed isocalorically on body weight, body composition, and adipose distribution. Adult male rats were weight matched into four groups. One group that was fed a low-fat diet (12%) served as reference controls. The other three groups were fed diets of 24, 36, or 48% fat in amounts to equal the energy intake of the control group. After 6 wk, body weights of the four groups were not significantly different. Intrascapular brown fat did not differ between groups. Total body fat and adipose depot weights, however, increased in proportion to the level of fat in the diet. Total body fat and retroperitoneal and mesenteric depot weights of the 48% fat group were greater than controls (P < 0.05). Mesenteric fat in this group was also significantly increased over all other groups (P < 0.05). These results show that high-fat diets fed to adult animals cause increased body fat in the absence of significant changes in body weight and that mesenteric fat is increased disproportionately.

We tested the hypothesis that endotoxin increases the heterogeneity of gut capillary transit times and impairs oxygen extraction. The gut critical oxygen extraction ratio was determined by measuring multiple oxygen delivery-consumption points during progressive phlebotomy in eight control and eight endotoxin-infused anesthetized pigs. In multiple 1- to 2-g samples of small bowel, we measured blood volume (radiolabeled red blood cells) and flow (radiolabeled 15-microns microspheres) before and after critical oxygen extraction. Red blood cell transit time (= volume/flow) multiplied by morphologically determined capillary/total blood volume gave capillary transit time. During hemorrhage, capillary/total blood volume did not change in the endotoxin group (0.5 +/- 4.5%) but increased in the control group (17.6 +/- 2.5%; P < 0.05) due to a decrease in total gut blood volume. Flow decreased significantly in the endotoxin group (36 +/- 10%; P < 0.05) but not in the control group (12 +/- 10%). Capillary transit-time heterogeneity increased in the endotoxin group (12.3 +/- 4.9%) compared with the control group (-5.8 +/- 7.4%; P < 0.05), predicting a critical oxygen extraction ratio 0.14 lower in the endotoxin group than in the control group (K. R. Walley. J. Appl. Physiol. 81: 885–894, 1996). This matches the measured difference (endotoxin group, 0.60 +/- 0.04; control group, 0.74 +/- 0.03; P < 0.05). Increased heterogeneity of capillary transit times may be an important cause of impaired oxygen extraction.

Background: Among venomous elapid snakes, cobras have the highest public awareness, as their venom represents a combination of proteins, peptides, and enzymes that have a range of biochemical and pharmacological roles and are also the main constitutes of biological activity and lethal toxicity. Objectives: The study aimed to evaluate the effect of the venom of Egyptian Spitting Cobra, Naja nubiae, on the vascular permeability based on the extravasation of the azo dye Evans blue (EB) into the tissues of the liver and kidneys of animals envenomed with low (¼ LD50; 0.32 mg/kg) and high (½ LD50; 0.65 mg/ kg) doses at three sampling times (30, 120, 360 min) post-injection of the venom. Methods: Fifty-four adult male Albino rats (8 weeks old and 180±2 0 g body weight) were divided into three main groups (n=6). In the control group, rats were subcutaneously (SC) injected with saline solution. Envenomed groups were SC injected, one group with 0.32 mg/kg and the other group with 0.65 mg/kg body weight of crude venom, respectively. Rats were I.V injected with EB dye 20 minutes before SC injection with saline solution as control animals and with Naja nubiae venom as treatment groups. Results: The results illustrated a high significant rate of EB extravasation to hepatic and renal tissues by the colorimetric determination of EB dye concentration. Conclusion: The venom of Naja nubiae can cause increased hepatic and renal vascular permeability which may explain the inflammatory effect induced by this venom.

ABSTRACT This study aims to determine the effect of infusion of conjoined pumpkin leaves (Sechium edule) on reducing total blood cholesterol of male white rats (Rattus norvegicus). The subjects of this study were 15 male white rats with an average body weight of 200 grams which were divided into 5 groups, each group consisted of 3 mice. The method used is a laboratory experiment with a completely randomized design. The results were obtained from 2 measurements of blood cholesterol levels, namely measurements before and after treatment. The treatment begins with the provision of high-fat foods for 48 days. On the 49th day a blood cholesterol level was measured before treatment. Furthermore, treatment was given to each group, namely aquades in the negative control group, simvastatin in the positive control group, and squash leaves infusion with their respective doses in the dose group I (40%), the dose group II (20%), and the dose group III (10%). Measurement of cholesterol levels after treatment was carried out on day 54. Data were analyzed by Paired t-test and One Way ANOVA. The analysis showed that there were no significant differences between treatment groups. Judging from the change in average and percentage, 40% infusion dose of siamese pumpkin leaves gives the best reduction in cholesterol levels. Keywords: Cholesterol, pumpkin leaves, male white mouse infusion. ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh pemberian infusa daun labu siam (Sechium edule ) terhadap penuruan kolesterol darah total tikus putih jantan (Rattus norvegicus). Subjek penelitian ini berupa tikus putih jantan berjumlah 15 ekor dengan berat badan rata-rata 200 gram yang dibagi dalam 5 kelompok, masing-masing kelompok sebanyak 3 ekor. Metode yang digunakan yaitu eksperimen laboratorium dengan rancangan acak lengkap. Hasil penelitian diperoleh dari 2 kali pengukuran kadar koleterol darah yaitu pengukuran sebelum dan sesudah perlakuan. Perlakuan dimulai dengan pemberian makanan tinggi lemak selama 48 hari. Pada hari ke49 dilakukan pengukuran kadar kolesterol darah sebelum perlakuan. Selanjutnya diberikan perlakuan pada tiap kelompok yaitu aquades pada kelompok kontrol negatif, simvastatin pada kelompok kontrol positif, dan infusa daun labu siam dengan dosis masing-masing pada kelompok dosis I (40 %), kelompok dosis II (20 %), dan kelompok dosis III (10%). Pengukuran kadar kolesterol sesudah perlakuan dilakukan pada hari 54. Data diananlisis dengan Paired t-test dan One Way ANOVA. Hasil analisa menunjukan tidak terdapat perbedaan yang bermakna antar kelompok perlakuan. Dilihat dari perubahan rerataan dan presentase, dosis infusa  40% daun labu siam memberikan penurunan kadar koleterol terbaik. Kata kunci : Infusa daun Labu siam, kolesterol, tikus putih jantan.
