Biochemical Composition of Lathyrus L. Seeds: Antioxidant Activities, Phenolic Profiles, β-ODAP and Protein Contents

Lathyrus taxa are used in different areas including nutritive, agricultural areas, and they are seen as the source of both protein and phenolics. Lathyrus taxa seeds contain a neurotoxic substance called β-N-oxalyl-l-β,β-diaminopropionic acid (β-ODAP) in different amounts. Lathyrus species grown in Turkey is reported to have low β-ODAP and high protein content. In this study, antioxidant activities, phenolic profiles, β-ODAP and protein contents of six Lathyrus taxa were investigated. Total phenolic content (TPC) in Lathyrus taxa used was ranging from 0.17±0.05 to 5.10±0.02. The antioxidant activities were observed in a wide interval that IC50 values were between 7.05±0.11 to 1.15±0.08 mg/mL. The highest TPC and antioxidant activity were recorded for L. clymenum. In HPLC analysis, gallic, p-hydroxybenzoic, caffeic, chlorogenic acid and epicatechin in all of the extracts were determined. Also, β-ODAP and protein contents in seeds of the Lathyrus taxa were found between 0.20–1.18 mg/g and 22.66–29.74%, respectively. β-ODAP contents of investigated Layhyrus taxa were within the safe consumption range in terms of health (plessthan2.00 mg/g). The investigated Lathyrus taxa were found to be excellent protein sources with low β-ODAP content and contain natural antioxidants. As a result, this study has provided important data for wild Lathyrus taxa to be used as a cheap protein source and functional food for promoting health.

The antioxidant activities of methanol and ethanol (10%, 30%, 50% and 96%) extracts of the aerial parts of Melittis melissophyllum L. were determined by DPP H, ABTS and FRAP assays. The content of flavonoids and phenols was also investigated. The total phenolic content in the extracts was determined using Folin-Ciocalteu assay; their amounts ranged between 63.5 and 111.7 mg GAeqv/g, while the concentrations of flavonoids were from 7.33 to 56.00 mg Queqv/g. IC50 values of the DPP H scavenging effect were from 0.109 to 0.664 mg/mL. The DPP H scavenging effect of the extracts was determined and the obtained IC50 values were from 0.109 to 0.664 mg/mL of solution. The values of ABTS radical activity were from 0.45 to 0.89 mg ascorbic acid/g. The FRAP value was within the range 0.160-0.382 mmol Fe/mg. The obtained values were analyzed by means of multivariate analysis, employing a hierarchical cluster analysis and between-groups linkage. The presented results confirmed that M. melissophyllum possesses good antioxidant properties and may serve as a promising source of natural antioxidants.

Since α-mangostin in mangosteen fruits was reported to be the main compound able to provide natural antioxidants, the microwave-assisted extraction process to obtain high-quality α-mangostin from mangosteen pericarp (Garcinia mangostana L.) was optimized using a central composite design and response surface methodology. The parameters examined included extraction time, microwave power, and solvent percentage. The antioxidant and antimicrobial activity of optimized and non-optimized extracts was evaluated. Ethyl acetate as a green solvent exhibited the highest concentration of α-mangostin, followed by dichloromethane, ethanol, and water. The highest α-mangostin concentration in mangosteen pericarp of 121.01 mg/g dry matter (DM) was predicted at 3.16 min, 189.20 W, and 72.40% (v/v). The verification of experimental results under these optimized conditions showed that the α-mangostin value for the mangosteen pericarp was 120.68 mg/g DM. The predicted models were successfully developed to extract α-mangostin from the mangosteen pericarp. No significant differences were observed between the predicted and the experimental α-mangostin values, indicating that the developed models are accurate. The analysis of the extracts for secondary metabolites showed that the total phenolic content (TPC) and total flavonoid content (TFC) increased significantly in the optimized extracts (OE) compared to the non-optimized extracts (NOE). Additionally, trans-ferulic acid and catechin were abundant among the compounds identified. In addition, the optimized extract of mangosteen pericarp with its higher α-mangostin and secondary metabolite concentrations exhibited higher antioxidant activities with half maximal inhibitory concentration (IC50) values of 20.64 µg/mL compared to those of the NOE (28.50 µg/mL). The OE exhibited the highest antibacterial activity, particularly against Gram-positive bacteria. In this study, the microwave-assisted extraction process of α-mangostin from mangosteen pericarp was successfully optimized, indicating the accuracy of the models developed, which will be usable in a larger-scale extraction process.

Xylaria species associated with termite nests or soil have been considered rare species in nature and the few which have been reported upon have been found to act as a rich source of bioactive metabolites. This study evaluated 10 ethyl acetate extracts of five new Xylaria species associated with termite nests or soil for their antioxidant activity, and cytotoxicity against different cancer and normal cell lines. DPPH and ABTS radical scavenging activities of the extracts demonstrated strong capacity with low IC50 values. The highest observed activities belonged to X. vinacea SWUF18-2.3 having IC50 values of 0.194 ± 0.031 mg/mL for DPPH assay and 0.020 ± 0.004 mg/mL for ABTS assay. Total phenolic content ranged from 0.826 ± 0.123 to 3.629 ± 0.381 g GAE/g crude extract which correlated with antioxidant activities. The high total phenolic content could contribute to the high antioxidant activities. Cytotoxicity was recorded against A549, HepG2, HeLa and PNT2 and resulted in broad spectrum to specific activity depending on the cell lines. The highest activities were observed with X. subintraflava SWUF16-11.1 which resulted in 11.15 ± 0.32 to 13.17 ± 2.37% cell viability at a concentration of 100 µg/mL. Moreover, LC-MS fingerprints indicated over 61 peaks from all isolates. There were 18 identified and 43 unidentified compounds compared to mass databases. The identified compounds were from various groups of diterpenoids, diterpenes, cytochalasin, flavones, flavonoids, polyphenols, steroids and derivatives, triterpenoids and tropones. These results indicate that Xylaria spp. has abundant secondary metabolites that could be further explored for their therapeutic properties.

Onion is among the most widely cultivated and consumed economic crops. Onions are an excellent dietary source of polyphenols and nutrients. However, onions phytonutrient compositions vary with cultivars and growing locations. Therefore, the present study involved the evaluation of polyphenol, nutritional composition (proteins, nitrogen, and minerals), sugars, pyruvate, antioxidant, and α-amylase inhibition activities of red onion cultivars, sweet Italian, and honeysuckle grown in California and Texas, respectively. The total flavonoid for honeysuckle and sweet Italian was 449 and 345 μg/g FW, respectively. The total anthocyanin for honeysuckle onion was 103 μg/g FW, while for sweet Italian onion was 86 μg/g FW. Cyanidin-3-(6”-malonoylglucoside) and cyanidin-3-(6”-malonoyl-laminaribioside) were the major components in both the cultivars. The pungency of red onions in honeysuckle ranged between 4.9 and 7.9 μmoL/mL, whereas in sweet Italian onion ranged from 8.3 to 10 μmoL/mL. The principal component analysis was applied to determine the most important variables that separate the cultivars of red onion. Overall results indicated that total flavonoids, total phenolic content, total anthocyanins, protein, and calories for honeysuckle onions were higher than the sweet Italian onions. These results could provide information about high quality and adding value to functional food due to the phytochemicals and nutritional composition of red onions.

In the last years, the interest in Italian monovarietal oils has increased due to their specific organoleptic qualities. Extra virgin olive oils (EVOOs) are rich in phenolic compounds, secondary metabolites well known and studied for their nutraceutical properties. However, among EVOOs, there is great variability in phenolic composition due to the origin, the production technique, and mainly, the genotype. The aim of this work was to evaluate the different phenolic profiles and the antioxidant activities of monovarietal oils. The results confirm this variability. In fact, the overall content of oleuropein varies up to four times between the different genotypes (from 33.80 to 152.32 mg/kg oil), while the oleocanthal content is significant only in two oils. The antioxidant activity, determined with 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) assays, is correlated with the content of total phenolic substances, with half maximal inhibitory concentration (IC50) values for the DPPH test ranging from 160 to 91 mg of oil, while the ORAC test shows values between 5.45 and 8.03 μmol Trolox equivalent (TE)/g oil.

A freshwater macroalga, Spirogyra spp., were analyzed for its phytochemical composition, antioxidant activity and total phenolic content (Folin−Ciocalteu method). Phytochemical analysis of Spirogyra neglecta revealed presence of phenolics, tannins, glycosides and saponins. The crude extraction of Spirogyra spp. was carried out using two solvents via, methanol (methanolic extract) and water (aqueous extract). The total phenolic contents of crude extracts were shown at 346.58±1.61 and 589.77±1.65 mg gallic acid equivalents/g extract in aqueous and methanolic extracts, respectively. The antioxidant capacity of aqueous and methanolic extract was estimated by two different methods; ABTS assay, and DPPH assay. The antioxidant activity of two extracts is affected by the extracting solvent and different assay. In the DPPH scavenging assay and ABTS assay, both extracts showed high antioxidant activity. In addition, the high correlation between results of all antioxidant activities and total phenolic content was found. The rapid TLC assay in methanolic extract is considered as the rapid test to evaluate the antioxidant activity of natural compounds. The compounds showing four bands at Rf = 0.25, 0.35, 0.42, 0.64. This study showed that Spirogyra spp. might constitute an important source of natural antioxidants.

Purpose Mushrooms have always been treasured and appreciated, thanks to their nutritional as well as medicinal properties. The purpose of this paper is to reveal these properties such as antioxidant activities and enzyme inhibition potentials of three mushroom species from Ordu, Turkey. Furthermore, phenolic profiles which are believed to be source of these properties are investigated. Design/methodology/approach The assays followed to determine total phenolic, flavonoid and antioxidant contents, antioxidative and enzyme inhibition activities are spectrophotometric methods. Phenolic profiles of mushroom samples were investigated by using the HPLC system. Findings The highest phenolic content was found for Lactarius deliciosus. Pyrogallol was the major phenolics in all three samples. Total phenolic content values supported that finding. But, total flavonoid content was the maximum for Cantharellus cibarius. The magnitude of the antioxidant activities revealed by the different tests varied between species. Furthermore, inhibition potential degrees on acetylcholinesterase, butrylcholinesterase, tyrosinase and urease activities were worth mentioning. Lactarius pyrogalus had anti-tyrosinase activity with a 26.229 percent inhibition rate. IC50 values for urease inhibition effect of mushroom samples were in the range of 0.37–0.90 mg/mL. Originality/value Enzyme inhibitory potentials for L. pyrogalus was revealed first time. On the other hand, studies on anti-tyrosinase activity of mushrooms are very rare, while almost no studies have been conducted on anti-urease activity. So, the results of present study are very valuable. Thus, the obtained data not only increase the ecological importance of the region but also provide significant contributions to the literature in terms of new sources rich in antioxidants.

The purpose of this study was to evaluate the antioxidant effect, total phenolic content of the ethanol, methanol and aqueous extract Dracocephalum polychaetum in flowering stage. The total phenolic content of the plant was measured colorimetrically. Antioxidant capacity of the plant extract was evaluated by DPPH method. In the present study, the highest antioxidant effect was to the extract of methanol, ethanol and aqueous, respectively. IC50 values in extracts of ethanol, methanol and water were respectively 11.89, 8.07 and 24.04. This value was obtained 0.86 μg /ml for BHT. The maximum level of phenol was to methanol, ethanol and aqueous extract respectively. Positive correlation was seen between the value of phenol and antioxidant capacity. The results show the various extracts of D. polychaetum have an antioxidant effect and could be considered as a potential source of natural antioxidants for the treatment of some diseases.DOI: http://dx.doi.org/10.3126/ijls.v9i5.12688

Abstract There is considerable interest in identifying new antioxidants from plant materials. Several studies have emphasized the antioxidant activity of species belonging to the Myrtaceae family. However, there are few reports on these species from the Cerrado (Brazilian savanna). In this study, the antioxidant activity and phenolic content of 12 native Myrtaceae species from the Cerrado were evaluated (Blepharocalyx salicifolius, Eugenia bimarginata, Eugenia dysenterica, Eugenia klotzschiana, Hexachlamys edulis, Myrcia bella, Myrcia lingua, Myrcia splendens, Myrcia tomentosa, Psidium australe, Psidium cinereum, and Psidium laruotteanum). Antioxidant potential was assessed using the antioxidant activity index (AAI) by the DPPH method and total phenolic content (TPC) by the Folin–Ciocalteu assay. There was a high correlation between TPC and AAI values. Psidium laruotteanum showed the highest TPC (576.56 mg GAE/g extract) and was the most potent antioxidant (AAI = 7.97, IC50 = 3.86 µg·mL−1), with activity close to that of pure quercetin (IC50 = 2.99 µg·mL−1). The extracts of nine species showed IC50 of 6.24–8.75 µg·mL−1. Most species showed TPC and AAI values similar to or higher than those for Camellia sinensis, a commonly consumed tea with strong antioxidant properties. The results reveal that the analyzed Myrtaceae species from the Cerrado possess high phenolic contents and antioxidant activities. Thus, they are a potential source of new natural antioxidants.

The entire phenolic profiles and antioxidant activities of different organs of the edible tree peony flowers (Fengdan Bai (FDB)) were analyzed. HPLC-quadrupole time-of-flight mass spectrometer (Q-TOF-MS/MS) analyses of individual phenolic compounds revealed that the petal and stamen contained higher levels of flavonoid glycosides than other organs (p < 0.05). Kaempferol-3,7-di-O-glucoside was the dominant flavonoid in these two organs, however, the calyx and ovary contained higher contents of gallic acid derivatives than other organs (p < 0.05). Hexa-O-galloyl-glucose was the dominant species in the calyx and ovary. At the same concentration of total phenolic extract (TPE), the stamen had the highest protection effect on Caco-2 cell oxidative damage induced by H2O2. The antioxidant effect was attributed to potent antioxidant capability; restored redox state due to the increased expression of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD); and improved barrier function of Caco-2 cell owing to increased zonula occludens-1 (ZO-1), CLDN3 (Claudin 3), and occludin mRNA expression. As a new resource food, the edible tree peony flower is a potential functional food material and natural antioxidants resource.
