Colostrum and milk composition during postpartum period in Hill cow, sahiwal and crossbreds cow

The study was undertaken to determine the composition of colostrum and milk of Hill cattle during postpartum period and compared them to Sahiwal and crossbred cow. Fifteen colostrum samples per day from each group of cow (as per availability) were collected during first five postpartum days while as milk samples were collected from eight cow each of Sahiwal and crossbreds and six form Hill cow, at fortnightly interval up to 32 weeks postpartum. These samples were subjected to various physico-chemical analyses. There was a significant (P?0.05) difference between overall mean protein per cent, pH and electrical conductivity in the colostrum of Hill cow, Sahiwal and crossbred cow but lactose per cent did not diffe significantly. In milk there was a significant (P?0.05) difference between overall mean of fat, protein, solid not fat, total solids per cent, pH, electric conductivity, somatic cell count and milk yield of Hill cow, Sahiwal and crossbreds while non-significant (P?0.05) difference was noticed for lactose per cent and specific gravity. The colostrum of Hill cow and Sahiwal were richer in protein, fat, lactose, TS and ash content as compared to crossbred cow. Milk of Hill cow had higher content of protein, fat, SNF, TS and lower pH, EC and SCC compared to crossbreds and Sahiwal.

Values of milk indicators (MIs) can be influenced by sampling errors and milk manipulation. This paper estimated the freezing point depression (FPD) and other MIs drifts which can cause fat movement. That is important for: – preparation of reference milk samples (MSs) for proficiency testing and instrument calibrations; – estimation of the impact of milk treatment as centrifugation in dairy plants on FPD. Five MSs (A = original milk; milk with modified fat (F) content; B = less F, C = low F, D = more F, E = high F) were created (gravitation F separation at 4 °C for 12 hours) with the same milk matrix 12× per year. F averages increased by 4.80% (122.1%) from 1.68 to 6.48% due to manipulation. It increased variability of MIs especially for SNF (solids non fat), L (lactose) and CP (crude protein). SCC (somatic cell count) averages increased by 803 (196.8%) from 9 to 812 thousand.ml−1. Correlation (r) F × SCC was 0.85 (P < 0.001). SNF, L and CP averages decreased by 0.47% (5.3%), 0.31% (6.3%) and 0.17% (5.0%). Correlations were −0.78, −0.75 and −0.64 (P < 0.001). Urea decreased along with F increase by 1.05 mg.100ml−1 (2.9%) but with r −0.13 (P > 0.05). Acetone increased by 1.37 mg.l−1 (47.6%) with r 0.21 (P > 0.05). Electrical conductivity decreased by 0.23 mS.cm−1 (6.0%) with r −0.15 (P > 0.05). Alcohol stability was reduced by 0.14 ml (23.3%) with r −0.15 (P > 0.05). FPD, titration and actual acidity were not influenced.

The aim of the present investigation was to evaluate how the order in which the ewes in a milking group enter the milking parlour affects their milkability and milk composition. Therefore, the order of entry into the milking parlour was evaluated in ewes of one flock (n = 353) during six evening milkings. In all, the sheep were milked in 15 milking groups. The sheep entering the milking parlour in the first milking group achieved 15 points and the sheep of each next milking group one point less, i.e. the sheep of the last group achieved 1 point. In the analysis, only the ewes with the highest and the lowest average number of points were included and assigned to the first (FG, n = 19) and the last group (LG, n = 29), respectively. After the last milking, the individual milk samples were collected from the jar to analyze the composition and somatic cell count. Machine milk yield in 30 s (0.15 ± 0.09 and 0.11 ± 0.05 l) and 60 s (0.26 ± 0.16 and 0.19 ± 0.10 l), peak flow rate (1.04 ± 0.39 and 0.77 ± 0.29 l/min), and latency time (14 ± 3 and 20 ± 13 s) significantly differed (P &lt; 0.05) between FG and LG, respectively. Total milk yield (0.41 ± 0.17 and 0.35 ± 0.14 l) and machine milk yield (0.27 ± 0.15 and 0.22 ± 0.10 l) tended to be higher (P = 0.05 and P = 0.09) in FG than in LG, respectively. No significant differences were observed in milk composition between FG and LG. It seems that ewes which enter the milking parlour in early milking groups have better parameters of milkability than those milked in later groups.

Abstract. The aim of this study was to determine the effect of somatic cell count (SCC) on milk yield and composition using a threshold value of 250 ∙103 cells/ml in dairy ewes of line 05. A total of 1 512 milk samples from udder halves were collected throughout milking periods at monthly intervals. In the present study milk composition (from halves of the udder) and daily milk yield (from udders) was examined in terms of SCC in ewe milk in three groups: from both halves of the udder below 250 ∙103 cells/ml, from one halves of the udder below 250 ∙103 cells/ml and from second half udder above 250 ∙103 cells/ml and from both halves of the udder above 250 ∙103 cells/ml. Milk yield from udders, somatic cell count in milk samples from udder halves and milk composition depended on the level of SCC recorded for halves of the udder. Somatic cell counts in milk from one or both halves of udders exceeding 250 ∙103 cells/ ml resulted in a statistically significant (P<0.01) decrease in daily milk production of ewes, by approx. 15.89 and 30.22 %, respectively. The analysis of variance showed also a significant effect of parity and stage of lactation of ewes on somatic cell count (log SCC) and milk composition from udder halves below 250 ∙103 cells/ml.

The recent availability of a high-throughput milk analyzer performing a partial differential somatic cell count (DSCC) opened new opportunities in investigations on bovine udder health. This analyzer has a potential limitation on the accuracy of measurements when the somatic cell count (SCC) is below 50,000 cells/mL, values characterizing a good proportion of lactating cows in many herds. We obtained data for cows below this threshold, assessed the repeatability of these measurements and investigated the relationship between DSCC and udder health, milk composition and yield. Overall, 3022 cow milk test records performed on a Fossomatic™ 7/DC (Foss A/S, Hillerød, Denmark) were considered; 901 of them had an SCC ≤ 50,000 cells/mL. These latter samples were analyzed by qPCR to identify the presence of bacteria. Overall, 20.75% of the samples (187) were positive. However, the health status did not have any significant association with DSCC. The analysis of the association of DSCC on milk fat, protein and casein showed a significant decrease in their proportions as the DSCC increased, whereas it was not observed for milk yield and lactose. Therefore, DSCC in very low SCC cows may be suggested as a marker to identify early changes in milk composition.

ABSTRACT: This study vised to detect possible changes in chemical and cellular composition of cow’s milk at different milking sessions and to test if proportional or composite milk sampling can be used instead of separate milk analysis. Two experiments were conducted: one in cows milked thrice daily and the other one in cows milked twice daily. In both experiments, three milk samples were collected from each cow in each milking session in order to determine milk composition at each milking; to have a composite sample, i.e., a single sample from the same aliquots of milk from each milking session; and to have a proportional sample, i.e., a single sample from each milking session in a volume proportional to milk volume yielded. Fat content and fat to protein ratio were higher in the afternoon on both experiments. Protein content was larger in the afternoon on cows milked twice daily, but not on cows milked thrice daily. Lactose level was not different across the milking sessions in any of the experiments. The urea level was higher in the afternoon and evening milking on cows milked thrice daily, and in the morning milking on cows milked twice daily. Somatic cell count did not differ between milking in any experiment. No difference in milk composition was noted between proportional and composite samples on cows milked thrice or twice daily. The collection of proportional and composite samples is recommended in twice or thrice daily milking sessions so that the analysis of milk components becomes more easy and cheap.

Activity of lysosomal N-acetyl-β-d-glucosaminidase (NAGase) in milk has been used as an indicator of bovine mastitis. We studied NAGase activity of 808 milk samples from healthy quarters and quarters of cows with spontaneous subclinical and clinical mastitis. Associations between milk NAGase activity and milk somatic cell count (SCC), mastitis causing pathogen, quarter, parity, days in milk (DIM) and season were studied. In addition, the performance of NAGase activity in detecting clinical and subclinical mastitis and distinguishing infections caused by minor and major bacteria was investigated. Our results indicate that NAGase activity can be used to detect both subclinical and clinical mastitis with a high level of accuracy (0·85 and 0·99). Incomplete correlation between NAGase activity and SCC suggests that a substantial proportion of NAGase activity comes from damaged epithelial cells of the udder in addition to somatic cells. We therefore recommend determination of NAGase activity from quarter foremilk after at least six hours from the last milking using the method described. Samples should be frozen before analysis. NAGase activity should be interpreted according to DIM, at least during the first month of lactation. Based on the results of the present study, a reference value for normal milk NAGase activity of 0·1–1·04 pmoles 4-MU/min/μl for cows with ≥30 DIM (196 samples) could be proposed. We consider milk NAGase activity to be an accurate indicator of subclinical and clinical mastitis.

The objective was to evaluate the physico-chemical composition, somatic cell count and electrical conductivity of the milk from lactating Zebu cows. For this, monthly milk samples were collected on the official milk control day intended for analysis of electrical conductivity of milk (ECM), somatic cell count (SCC) and milk composition. We used monthly data from 680 controls, from 10 Gyr and 17 Guzerat cows from a herd located in the city of São Gonçalo do Amarante/RN, from August to November 2013. Analyses of variance, mean test and Pearson correlation analysis were conducted. The levels of fat (% F), protein (% P), lactose (% L), total solids (% TS) and non-fat dry extract (% NFDE) were dependent variables for the ECM and SCC. The milk of Gyr cows for ECM, SCC, GOR, PROT, LACT, TS and NFDE were respectively, 3.88±1.18 mS/cm, 1629±2195 thousands cells/mL, 4.71±1.77%, 3.29±0.31%, 4.5±0.33%, 13.5±1.78%, 8.79±0.37%, and for the Guzerat breed 3.59±0.88 mS/cm 1356±1793 thousands cells/mL, 4.47±0.98%, 3.43±0.32%, 4.52±0.24%, 13.5±1.19%, 9.02±0.48%. No significant correlations between SCC and ECM characteristics were observed. However, when the correlation of each of these parameters and milk composition in different classes for each breed was analyzed, a significant correlation of the SCC was observed with the protein and lactose in Gyr, of the ECM with the lactose and SCC with total solids and fat for the Guzerat cows.

A total of 50 sheep milk samples during lactations were used to investigate the impact of subclinical mastitis induced Somatic Cell Count (SCC) increase on changes in chemical composition in milk. Samples were collected and analyzed for fat, protein, lactose, solids non-fat (SNF) and total solids (TS) and SCC. Within three stages of lactation, all milk samples were analyzed for three times. The highest average of SCC was recorded in ewes aged 4 years (46.67) while lowest average was recorded at the age of 3-3.5 years (34.24%). Protein content was highest at ewes aged 1-1.5, whereas highest lactose percentage recorded at ewes aged 3-3.5. In conclusion, in high SCC milk, lactose content may be more indicative of SCC level than milk fat, protein, SNF and TS during lactation. Age of ewes is an important factor affecting milk composition, thus is a necessary parameter in optimizing sheep milk quality in conditions of subclinical mastitis.

Background: During present investigation burfi was prepared from different combinations of cow milk khoa and pumpkin pulp [viz., 100:00 (T1), 75:25 (T2), 70:30 (T3), 65:35 (T4) and 60:40 (T5)] and the samples were subjected to sensory evaluation, chemical analysis and cost was computed considering the ingredients and processing costs. The pumpkin pulp (as per treatments) and sugar (30% by weight of khoa) were mixed with khoa at different stage of khoa preparation. Then, the mixture was heated till burfi was obtained. Methods: For preparation of burfi fresh cow milk was obtained from Livestock Instructional Farm, Akola. The milk was standardized to 4.0 per cent fat and 9.0 per cent SNF for the preparation of burfi. The khoa blended with pumpkin (Cucurbita pepo) pulp at different ratios, the product without using pumpkin pulp served as control. The burfi was analyzed for chemical composition (viz., fat protein, total sugars, ash and total solids). The organoleptic attributes of burfi were analyzed in terms of its flavor, body and texture and color and appearance, wherein the total score was out of 100; the judging panel comprised on 5 members. Result: The total sensory scores of burfi obtained were 87.81, 90.46, 95.46, 92.99 and 84.30 for the treatment T1, T2, T3, T4 and T5 respectively. The burfi prepared from various combinations involving use of up to 70 per cent cow milk khoa in blend with pumpkin pulp was found acceptable. The fat content of burfi’s was 19.61, 15.40, 14.38, 13.40 and 12.37 per cent, protein content was 14.89, 12.64, 12.11, 11.61 and 11.04 per cent, total sugar content was 32.78, 36.73, 37.59, 38.45 and 39.31 per cent, ash content was 2.69, 2.55, 2.53, 2.49 and 2.45 per cent, total solids content was 69.94, 67.30, 66.60, 65.94 and 65.17 per cent for the treatment T1, T2, T3, T4 and T5 respectively. The fat, protein, ash and total solids of burfi tended to decrease while total sugar and moisture content tended to increase with an increase in the level of pumpkin pulp used as additive. The per kg cost of production of pumpkin based burfi decreased with increasing level of incorporation of pumpkin pulp in burfi formulation i.e. ₹ 240 (100:00 khoa), ₹ 204.51 (75:25, khoa: pulp), ₹ 199.11 (70:30 khoa: pulp), ₹ 194.33 (65:35 khoa: pulp), ₹ 189.25 (60:40, khoa : pulp) for the treatment T1, T2, T3, T4 and T5 respectively. The most acceptable burfi i.e. T3 was computed to be priced at ₹ 199.11 per kg.

This study investigated the effects of subclinical mastitis on milk yield and milk composition parameters in dairy camels. A total of 140 camel milk samples were collected from multiparous she-camels (7-10 years old) and were subjected to bacteriological culture studies; 76 samples displayed subclinical mastitis and 64 samples were healthy. Furthermore, the samples were analyzed via standard procedures to determine the yield and composition parameters of the milk such as pH, electrical conductivity, protein, fat, lactose, and ash content. The results showed that the milk yield was significantly low (p < 0.05) in infected animals compared to healthy animals (3.80 vs 4.32 L·day-1). Moreover, compared to the values observed in healthy milk samples, milk from animals with subclinical mastitis showed significantly decreased (p < 0.05) protein (3.33 vs 3.40%) and fat (3.67 vs 3.74%) contents. However, no significant changes (p > 0.05) were observed in pH, electrical conductivity, lactose, or ash content. The results of the uninfected milk samples revealed that there was significant correlation between the protein and fat content (r = 0.781; p < 0.01) and between the lactose and ash content (r = 0.701; p < 0.01). Conversely, the infected ones showed that there was higher significant correlation between the protein and fat contents (r = 0.807; p < 0.01) and a medium correlation between the lactose and ash contents (r = 0.603; p < 0.01). In conclusion, the results suggest that subclinical mastitis is negatively associated with a reduced milk yield and lower protein and fat content. Thus, these parameters can be used to diagnose mastitis in dairy camels.
