Analgesic and adjunct actions of nalbuphine hydrochloride in xylazine or xylazine and acepromazine premedicated horses

The study was conducted in eighteen clinical cases of horses for diagnostic and surgical procedures requiring general anaesthesia were randomly divided into three groups, group I, group II and group III, each consisting of six cases. All the horses were premedicated with glycopyrrolate at the dose rate of 0.02 mg/kg body weight, intravenously. Horses in Group I and Group II were administered xylazine hydrochloride at the dose rate of 1.10 mg/kg body weight intravenously, whereas in Group III at the dose rate of 0.50 mg/kg body weight intravenously. In Group III, acepromazine was injected after xylazine administration, at the dose rate of 0.02mg/kg body weight, intravenously. Before induction of anaesthesia, nalbuphine hydrochloride was administered for Group II and Group III at the dose rate of 0.75 mg/kg body weight intravenously. Ketamine hydrochloride was administered intravenously to induce anaesthesia at the dose rate of 2.20 mg/kg body weight and maintained with 0.50 mg/kg body weight in required cases to maintain for duration of 15 ± 1.04 minutes. The mean time for induction in group I, group II and group III were 1.78 ± 0.27, 1.73 ± 0.10 and 1.85 ± 0.28 minutes respectively. The mean total number of additional doses of ketamine for standard duration of 15 ± 1.04 minutes surgery required in group I, group II and group III were 5.00 ± 0.36, 1.66 ± 0.33 and 2.00 ± 0.36 respectively. The quality of induction was 100 per cent smooth in group III, 83.33 per cent smooth and 16.67 per cent rough in group II and 66.66 per cent smooth and 33.34 per cent rough in group I. The quality of analgesia in group I, group II and group III were 2.83 ± 0.47, 1.83 ± 0.30 and 1.33 ± 0.21 respectively. The quality of muscle relaxation in group I, group II and group III were 3.16 ± 0.30, 1.50 ± 0.22 and 1.33 ± 0.21 respectively. The mean time for recovery in group I, group II and group III were 23.00 ± 1.52, 33.00 ± 0.93 and 41.98 ± 1.32 minutes respectively. The mean number of attempts for unassisted standing in group I, group II and group III were 6.66 ± 0.71, 5.00 ± 0.57 and 5.00 ± 0.36 respectively. The quality of recovery was 83.33 per cent smooth and 16.67 per cent rough in group III, 66.66 per cent smooth and 33.34 per cent rough in group II and 50.00 per cent smooth and 50.00 per cent rough in group I. None of the animals in any groups showed any intra and post operative complication.

The monthly wool growth of three groups of rams was studied at Beltsville, Maryland. Group I received natural daylight (at 38° 53' N.) and was shorn monthly. Group II had a 7:17 hours of daylight to hours of darkness rhythm and was shorn every 6 months, once in winter and once in summer. Group III received natural daylight and was likewise shorn every 6 months. The rams of all groups produced more wool in summer than in winter. This difference was significant (P<0.001). The mean body weight and food intake were both greater in the winter months, which indicated that the seasonal rhythm of wool growth was not a consequence of poorer feeding in winter. The rams which were shorn monthly (group I) grew considerably more wool than the other two groups, but the difference was not statistically significant. The short-day treatment of group II did not increase the annual wool production nor decrease the seasonal rhythm of wool growth. The balance of evidence from this and other experiments indicates that temperature rather than light controls the seasonal rhythm of wool growth.

The effects of two doses of cloprostenol and two doses of flurogestone acetate sponge on the onset of oestrus, and embryo recovery and quality were evaluated. Thirty-two indigenous ewes (Wera breed) were allocated into four groups of eight. All ewes were synchronised with 100 µg (Group I) or 175 µg (Group II) cloprostenol injection, 9 days apart, or insertion of intravaginal sponges containing 30 mg (Group III) or 45 mg (Group IV) flurogestone acetate (FGA) for 12 days. The ewes were superovulated with 600 IU PMSG intramuscularly 10 days after the second cloprostenol injection or immediately after sponge removal on Day 12. After the detection of oestrus, the ewes were mated naturally at 6 and 12 h, and some ewes were inseminated laparoscopically. Embryos were recovered surgically 5 or 6 days after service. All ewes exhibited oestrus. The onset of oestrus occurred significantly (P < 0.05) earlier in FGA-treated (50.0 ± 1.5 and 48.0 ± 0.00 h) than in cloprostenol-treated groups. There were no significant differences (P > 0.05) in the mean time of onset of oestrus (50.0 ± 1.5 and 48.0 ± 0.00 h) between the two doses of cloprostenol. The mean number of corpora lutea (8.1 ± 1.26) and embryos recovered (6.1 ± 1.00) was significantly (P < 0.05) higher in ewes treated with 45 mg FGA than in ewes treated with cloprostenol. Embryo recovery rate was significantly (P < 0.05) higher in ewes treated with 45 mg FGA (75.4%) than in other groups (41.7% and 51.6% in 100 μg and 175 μg cloprostenol- and 52.7% in 30 mg FGA-treated groups, respectively). Fertilisation rate was 93.3% in ewes given 100 µg cloprostenol, whereas other groups showed 100% fertilisation rate. The highest percentage (100%) of Grade 1 embryos was in FGA groups. In conclusion, despite FGA protocol presenting superior results, cloprostenol protocol was equally efficient in synchronising oestrus. The embryo recovery rate was better after 45 mg FGA than 30 mg FGA or after either dose of cloprostenol.

In this study, genotoxic effects of repeated sevoflurane anaesthesia were investigated in rabbits with or without antioxidant supplementation. Twenty-one New Zealand male rabbits were included in the study and randomized into three groups as: placebo treated (Group I), vitamin E supplemented (Group II) and selenium supplemented (Group III). Vitamin E and selenium were given intraperitoneally for 15 days before anaesthesia treatment. Anaesthesia was administered using 3% sevoflurane in 4 L/min oxygen for a 3-hour period and continued for 3 days. Blood samples were collected before anaesthesia (Sample 1), after the first, second and third days of sevoflurane administration (Sample 2, Sample 3 and Sample 4 respectively) and the last samples were taken 5 days after the last sevoflurane administration (Sample 5). Genotoxic damage was examined using the comet assay. The degree of damage is assessed by grading the cells into three categories of no migration (NM), low migration (LM) and high migration (HM) depending on the fraction of DNA pulled out into the tail under the influence of the electric field. The number of comets in each sample was calculated (1 × number of comets in category NM + 2 × number of comets in category LM + 3 ×number of comets in category HM) and expressed as the total comet score (TCS), which summarizes the damage frequencies. In Group I, a significant increase in the mean TCSs was observed for Samples 3 and 4 as compared with Sample 1. However, there were no significant differences between Samples 1, 2 and 5. The mean TCS of Sample 4 was significantly higher than Sample 1, 2 and 3 in Group II. Group III demonstrated no significant mean TCSs for any experimental conditions. Statistical differences were also observed between the groups with significant P values. This experimental study points out the presence of DNA damage with repeated sevoflurane anaesthesia and the genoprotective role of antioxidant supplementation on DNA damage in mononuclear leukocytes of rabbits by highly sensitive comet assay.

Objective: the clinical value of spinal quantitative CT (sQCT) and the structural patterns of the vertebral bone were studied Material and Methods: sQCT was performed on 246 patients with a mean age of 57 years for whom conventional lateral radiographies of the thoracic and lumbar spine were available. All patients were suffering from back pain of unknown etiology. the bone mineral density (BMD) of the midvertebral section of 3 lumbar vertebral bodies was determined by means of single-energy-(SE)-weighted QCT (85 kV). Spongiosa architecture and density profile analyses were made in the axial images. This was contrasted to BMD values ascertained in SE QCT. the mean BMD was compared to the number of fractures and the patients were divided into three groups: group I — no fracture; group II — one fracture; and group III 1 fracture Results: the mean BMD was: 134.3 (74.1–187.5) mg hydroxyapatite (HA)/ml in group I; 79.6 (58.6–114.3) mg HA/ml in group II; and 52.4 (13.1–79.1)mg HA/ml in group III. A significant deterioration in spongiosa structure was found with increasing demineralization: strongly rarefied patterns predominated in the fracture groups II and III Conclusion: sQCT provides a good risk assessment of the occurrence of vertebral body insufficiency fractures

Trapadil (Mochida Pharmaceuticals, Japan), an antiplatelet drug, suppresses the growth of several cell types and is thought to antagonize platelet-derived growth factor. The effects of trapidil on mesangial-cell proliferation in glomerulonephritis induced by anti-thymocyte serum in Wistar rats were investigated. Control rats were treated with phosphate-buffered saline (group I); group II rats were injected with a single dose of anti-thymocyte serum (8 ml/kg body weight), and group III rats were treated with both a single dose of anti-thymocyte serum (8 ml/kg body weight) and with trapidil (5 mg/kg body weight/day). Three rats in each group were killed on day 3, and the other three on day 10. Control rats showed no significant histological changes on day 3 or day 10. In group II, on day 3, there was a marked decrease in glomerular cell numbers, with mesangiolysis. Histologically severe mesangial-cell proliferation with expansion of mesangial areas was noted on day 10. None of the rats in group III showed mesangial alterations, histologically, indicating that mesangial-cell proliferation was suppressed by trapidil. This suppression may result from antagonism of the binding of platelet derived growth factor to the specific surface receptors in the mesangial cells. Trapidil may have clinical value in the treatment of mesangial-cell proliferative glomerular diseases.

Patients suffering from advanced upper abdominal malignancies have pain as predominant symptom affects their quality of life and survival. USG guided coeliac plexus neurolysis become benevolence in these patients on part of their pain management and quality of life improvement. To compare the efficacy of USG guided coeliac plexus neurolysis for pain relief in upper abdominal malignancies by using different concentration of alcohol (50% vs 75%).This Prospective, comparative, randomised double blinded study was conducted during Sep 2019 – Aug 2020 at our tertiary care centre. Total 60 cases were taken as per following inclusion and exclusion criteria and randomly divided into 2 groups i.e. 30 each group, we compare Visual Analogue Scale (VAS) score, quality of life (QOL) and need of rescue analgesia profile between the groups to know the efficacy of USG guided coeliac plexus block. In our study, we observed that the baseline mean VAS score in group I was 8.26±0.78 while in group II was 8.03±0.76. No significant difference was found in mean VAS score at this time between the groups (p=0.24). The baseline mean QOL score in group-I was 77.46±3.40 while for the cases of group II the mean QOL score was 77.36±3.33. No significant difference was found in mean QOL score at baseline between the groups (p=0.90). The baseline mean morphine consumption in group-I was 113.33±39.24 mg while for the cases of group-II the mean morphine consumption was 120.33±38.37mg. No significant difference was found in mean morphine consumption at this time between the groups (p=0.48).Both groups having 50% alcohol and 75% alcohol decreases the VAS score from baseline in patients having upper abdominal malignancies along with QOL and dosages of rescue analgesia whereas no significant difference in VAS score in patients of both groups.

Abstract: Gentamycin, a frequently used aminoglycoside antibiotics, has a nephrotoxic effect to human beings and animals. The purpose of this research was to find out the microscopic changes of wistar rat kidneys after gentamycin induction. This was an experimental study, using five adult wistar rats, divided into three groups. Group I was the control group; group II consisted of two rats, injected with gentamycin 0,3 ml/day (dose of 60 mg/kg body weight/day) intraperitoneally for seven days; and group III consisted of two rats, injected with gentamycin 0,3 ml/day intraperitoneally for 10 days. Group I and II were terminated at day-8, and group III at day-11. Their kidneys were processed for microscopic slides, stained with hematoxylin eosin and Periodic Acid Schiff. In microscopic evaluation, group II and III showed oedema, necrosis, apoptosis, and basal membrane destruction of tubular epithelial cells. Group III also showed fat vacuoles in these epithelial cells (macrovesicular fatty changes). Conclusion: wistar rats injected with gentamycin 60 mg/kg body weight/day for 7 and 10 days showed oedema, necrosis, apoptosis, and basal membrane destruction of tubular epithelial cells; and macrovesicular fatty changes after 10 days of gentamycin.Key words: gentamycin, necrosis tubular epithelial cells, fatty changesAbstrak: Gentamisin termasuk antibiotik golongan aminoglikosida berspektrum luas yang bersifat nefrotoksik terhadap manusia dan hewan. Tujuan penelitian ini untuk melihat perubahan mikroskopik struktur ginjal tikus Wistar setelah diberikan gentamisin. Metode penelitian eksperimental dengan menggunakan lima ekor tikus Wistar dewasa yang dibagi atas tiga kelompok. Kelompok I tanpa perlakuan; kelompok II terdiri dari dua ekor tikus perlakuan yang diinjeksi dengan gentamisin 0,3 ml/hari (dosis 60 mg/kgBB/hari) secara intraperitonial selama tujuh hari; dan kelompok III terdiri dari dua ekor tikus perlakuan yang diinjeksi dengan gentamisin 0,3 ml/hari secara intraperitonial selama 10 hari. Tikus Wistar kelompok I dan II diteminasi hari ke-8, sedangkan kelompok III diterminasi hari ke-11. Ginjal tikus kelompok I -III kemudian dibuat preparat histopatologik dengan pengecatan rutin hematoksilin eosin dan Periodic Acid Schiff (PAS). Hasil penelitian menunjukkan tikus Wistar perlakuan yang diberikan gentamisin 0,3 ml/hari selama 7 sampai 10 hari secara mikroskopik memperlihatkan pembengkakan, nekrosis, apoptosis, dan destruksi membrana basalis sel epitel tubulus; dan pada hari ke-10 terlihat vakuol-vakuol lemak pada sel epitel sehingga inti terdesak ke tepi (perlemakan makrovesikuler). Simpulan: pemberian gentamisin pada tikus Wistar dengan dosis 60 mg/kg BB/hari selama 7-10 hari menunjukkan pembengkakan, nekrosis, apoptosis sel epitel tubulus, dan membrana basalis tubulus rusak; dan setelah hari ke-10 juga terlihat perlemakan makrovesikuler.Kata kunci: gentamisin, nekrosis sel epitel tubulus, perlemakan makrovesikuler

Interest in medicinal plants for the management of myriad of conditions including reproductive disorders refractory to orthodox medicinal care is on the increase. Ficus syncomorus and Datura metel are two of such plants with folkloric evidence of aiding fertility in human. This study investigated the effect of aqueous stem-bark extracts (200 mg/kg) of F. syncomorus and D. metel respectively on the sperm characteristics of Yankasa rams. Twelve (12) matured (15 – 16 months) old rams were used in this study and randomly assigned into three (I, II, III) groups of four (4) animals each. Group I served as the control while II and III served as the treatment groups and received daily oral doses (200 mg/kg) of F. syncomorus and D. metel extracts respectively for 7 consecutive days. Semen was collected from all the groups at the end of the treatments using Electro-ejaculation method and evaluated by light microscopy. The mean semen volume (68.70+4.2 to 65.62+2.00) and percentage progressive motile cells significantly (p<0.05) reduced 7 days post treatment in group III (84.05+1.3) compared to the control (85.20+1.32) and the group II (86.56+0.40) animals. The mean sperm count, the percentage liveability and the haematological parameters and erythrocytic indices (10.81±0.24 for group 111 to 12.54±0.30 for group1) significantly (p<0.05) decreased in group III compared to the values in the control and group II rams. Abnormal sperm morphology (bent mid-piece, curved tail, headless tail, tailless head) significantly (p<0.05) increased in D. metel group (7.26+0.12) compared to F. syncomorus (5.02+0.04) and control groups (5.62+0.01) respectively. D. metel aquesous extract adversely affected sperm characteristics with significant effect on semen volume, sperm morphology and counts as well as haematological parameters. Exposure of animals to D. metel at the dose used may impair sperm fertilizing ability, thus leading to reduced ram fertility. While F. syncomorus extract appears a potential drug candidate for improving fertility.

BACKGROUND: Mastitis is an inflammation of the breast tissue, usually caused by bacteria. Mastitis stimulates pro-inflammatory cytokines. The cytokine interleukin-1β (IL-1β) is a crucial mediator of the inflammatory response. This cytokine has adverse effects of hosting immunity that mediates resistance to pathogens and also exacerbates damage during chronic disease and acute tissue injury. Scaevola taccada (Gaertn.) Roxb. has been used as an ethnomedicine for healing sores in several provinces in Indonesia. AIM: This study aimed to assess the profile of pro-inflammatory cytokine IL-1β through the treatment effect of leaf extracts of S. taccada (Gaertn.) Roxb. as adjuvant for healing mastitis. METHODS: This study was a true control group experiment using the pre-test-post-test control design aimed to measure the effect of hydroalcoholic compounds in leaf extracts of S. taccada on the systemic pro-inflammatory activity of interleukin-1β (IL-1β). The treated animals were 18 mice of Sprague Dawley strain induced by Staphylococcus aureus. These treated mice were divided into three groups in which each group consisted of six mice. The mice in the Group I (negative control) were given 1 ml aquabides/250 g body weight, those in the Group II (positive control) were delivered with 9.6 mg/ml amoxicillin/250 g body weight, and those in the Group III (experimental) were given 9.6 mg amoxicillin/250 g body weight + 400 mg/ml leaf extracts of S. taccada/g body weight for 5 days, respectively. Pathological examinations were carried out from the inflamed tissues to prove the healing process of the treated mice. IL-1Β levels were determined using the enzyme-linked immunosorbent assay method. Data were analyzed using ANOVA and post hoc tests. RESULTS: There were statistically significant differences of IL-1β levels after the administration of leaf extracts of S. taccada among all the treated mice groups at p < 0.05. The Group III had the lowest IL-1β level with the mean value ± 1.45 pg/ml compared to the IL-1β level in the Group II (positive control) with the mean value ± 3.82 pg/ml and the IL-1β level in the Group I (negative control) with the with mean value ± 5.22 pg/ml. The pathological analysis of breast tissues of the treated mice proved that leaf extracts of S. taccada (Gaertn Roxb.) could reduce damaged tissues, cellular infiltration, and subcutaneous edema induced by this pathogenic microorganism. CONCLUSION: Leaf extracts of S. taccada had a significant function as adjuvant for healing mastitis by reducing pro-inflammatory cytokine IL-1β.

Background: The non-availability of the sophisticated anaesthetic machine and the necessary equipment to administer inhalant anaesthetic in the field hospitals make their use practically unfeasible for the field veterinarians. Therefore, the present study was undertaken to evaluate the effect of propofol, ketamine and their combination ‘Ketofol’ as a TIVA on certain haematological, serum biochemical and hormonal profiles in atropine and xylazine premedicated dogs. Methods: The study was conducted in eighteen clinical cases of dogs of either sex. The animals were randomly divided into three groups with six animals in each group. All the three groups were premedicated with Atropine sulphate @ 0.04mg/kg body weight and xylazine HCl @ 0.5mg/kg body weight intramuscularly. In group-I, propofol @ 5mg/kg body weight, in group-II, ketamine @ 5mg/kg body weight and in group-III, ketofol @ 4mg/kg body weight was administered intravenously for induction after 15 minutes of pre-anesthetic administration. Surgical anaesthesia was maintained for 90 minutes in all three groups viz. group-I, group-II and group-III with propofol @ 2.5mg/kg. b.w., ketamine @ 2.5mg/kg b.wt. and ketofol @ 2mg/kg b.wt. respectively by intermittent bolus injection (IBI) technique. Haematological, serum biochemical and hormonal profile were evaluated before administration of the anaesthetic agent (0 minutes) then at 15, 30, 60 and 90 minutes during and after administration of anaesthetic agents. Result: The study revealed that Hb, PCV and TEC were significantly decreased in all the groups at 60 mints and 30 mints respectively. The biochemical evaluation revealed that blood glucose level was significantly increased in all the groups until the end of the experiment. BUN and creatinine value was a significant increase in group-I and group-II than group-III at different time intervals up to the end of the experiment. In all the groups’ alanine aminotransferase (ALT) values significantly increased up to 60th minutes during TIVA whereas AST value was significantly increased in group-II at 30th and 60th minute of the experiment in compare to group-I and group-III. A higher level of cortisol values was recorded in group-I animals for the entire period of observation. There were no changes observed in the case of T3. Transient variables of haemato-biochemical have been reported following propofol, ketamine and their combination (ketofol) as total intravenous anaesthesia (TIVA). Thus, it has been concluded that diligent monitorization and electrolyte support are essential during the period of anaesthesia.
