The study consists of a bioassay of a medium experimentally contaminated with heavy metals using Rhizobium meliloti microorganisms as a test object. In vitro, the viability reaction of clover and alfalfa nodule bacteria to lead, cadmium, copper and zinc was established. The effect of the studied metals on the number of colonies depends on the concentration of the chemical element, physiological action of the microelement and biological characteristics of the strains used. The studies performed have determined a direct relationship of reducing the growth in the rhizobia colonies number of clover and alfalfa with an increase in the concentration of salts of heavy metals. Inhibition of the colonies number had a high regularity when the environment was contaminated with lead, copper and cadmium at a concentration of 0,3%; with a subsequent increase, complete death of microorganisms was observed. With the introduction of zinc into the nutrient medium in small concentrations, a positive resistance tendency of the clover and alfalfa bacteria was observed: thus, at 0,01-0,1% of the zinc salt content, the number of grown colonies was above the control level and amounted to 714-987 pcs. at the control value of 578 pcs. In an environment with a concentration of zinc 0,5%, a significant decrease in the growth of clover rhizobia was noted up to 65 pieces of colonies. At the same time, in the alfalfa rhizobia colonies in these concentrations a lower degree of survival was observed and the environment of zinc 0,3% became critical.

Background: Functional and pathological improvements following rapid rewarming in 42°C water was compared with alterations following slow thawing at room temperature (22°C) after frostbite (–9°C, 15 minutes) in vivo of the rabbit central ear artery. Methods: Following two to ten weeks of in vivo regeneration, vascular segments were tested in vitro. Maximal and dose-dependent isometric contractions were induced by exogenous noradrenaline. Sympathetic nerves in the vascular wall were stained with glyoxylic acid. Vascular ring segments were stained with haematoxylin and eosin. Results: Following slow thawing, the total uptake, the K+ evoked and the spontaneous release of [3H]noradrenaline in the sympathetic nervous system were strongly reduced two weeks after freezing, with a subsequent increase to control level within 3–4 weeks. After rapid rewarming the total uptake, the spontaneous release and the K+ evoked release of [3H]noradrenaline commenced earlier such that after ten weeks the level was twice as high as following slow rewarming. The glyoxylic acid induced catecholamine fluorescence in sympathetic nerves, revealed an earlier regeneration after rapid rewarming. Haematoxylin and eosin-stained segments revealed less intimal hyperplasia three to 20 weeks after rapid rewarming than after slow thawing. Conclusion: Rapid rewarming of in vivo frozen arteries in warm water (42°C) did not prevent immediate vasoparalysis and degeneration of sympathetic nerves. However, nerve regeneration occurred earlier and with higher tissue nerve densities as compared to tissue that had been slowly rewarmed. Myointimal hyperplasia was less pronounced after rapid rewarming. Abnormal sympathetic nerve function and myointimal hyperplasia, as observed in this study, may contribute to a greater understanding of sequelae in the human body following frostbite.

The uptake mechanism of Zn2+ through the gill epithelium of freshwater rainbow trout was investigated both in intact animals and in isolated basolateral membranes. Involvement of the apical Ca2+ uptake sites in Zn2+ uptake was examined in vivo by pharmacological manipulation of the apical Ca2+ permeability. The apical entries of Ca2+ and Zn2+, but not Na2+ and Cl-, were inhibited by addition of La to the water. Addition of 1.0 microM La reduced the influxes of Ca2+ and Zn2+ to 22 +/- 3 and 53 +/- 7% (mean +/- SE) of the control value, respectively. Injection of CaCl2 also reduced the branchial influxes of Ca2+ and Zn2+. This treatment decreased the influx of Ca2- to 45 +/- 4% of the control level and the Zn2+ influx to 68 +/- 5%. These results strongly imply that Zn2+ passes across the apical membrane of the chloride cells of the gills via the same pathway as Ca2+. The presence of an active basolateral transporter for Zn2+ was investigated in vitro on isolated basolateral membranes. There was no ATP-dependent or Na2+(-)gradient driven transport of Zn2+ at physiological Zn2+ activities. The same system was used to study potential effects of Zn2+ on the basolateral Ca2+(-)adenosinetri-phosphatase. Zn2+ was found to be a potent blocker of this transporter, causing a mixed inhibitory effect on the ATP driven Ca2+ transport at a free Zn2+ activity of 100 pM.

Abstract In this work three heavy metals: cadmium (as CdSO4), cobalt (as CoCl2) and zinc (as ZnSO4), were used to determine and compare their toxicity towards two subspecies of barley (Hordeum vulgare subsp. vulgare L. and Hordeum vulgare subsp. distichum L.), focusing on seeds germination, seedlings growth, and cytological parameters. The results indicate that the effect of these heavy metals depends on the metal kind, the metal concentrations and the plant subspecies. Generally, in the case of H. vulgare, the heavy metal salts understudy did not influence significantly seed germination and seedling growth parameters. However, these metal salts influence significantly these parameters for H. distichum. The cytological test showed significant decrease (p < 0.05) in the mitotic index among the increase of the heavy metal concentrations when evaluated with the control for H. vulgare and H. distichum. Consequently, H. vulgare seemed to be more tolerant of the increase of the three heavy metals concentrations than H. distichum.

During four hours of severe respiratory acidosis produced by inhalation of 30% and 40% CO2 in dogs, plasma potassium and inorganic phosphate rose, ultrafiltrable calcium first increased then fell below control level, and total calcium increased slightly. Within 5 minutes after returning the animal to air breathing, potassium showed a further sharp increase, ultrafiltrable calcium a further sharp decrease and total calcium and inorganic phosphate showed no change. With the rise in potassium and fall in ultrafiltrable calcium the UF Ca/K ratio fell to less than one third control value. This appears to be an important factor in the mechanism producing cardiac arrhythmias and ventricular fibrillation in the immediate post-hypercapnic period. In vitro experiments on dog blood indicate that ultrafiltrable calcium varies directly with H+ concentration as expected, and that an increase in inorganic phosphate produces a decrease in ultrafiltrable calcium. Operation of these two factors helps to explain the change in plasma ultrafiltrable calcium observed in the in vivo experiments. It is suggested that the decrease in ultrafiltrable calcium in the presence of the very high phosphate concentrations observed, may be due to the formation of some nonfiltrable calcium phosphate compound. It is possible that the increased phosphate concentration increases the binding of calcium in some other form.

SummaryThe present in vitro study investigated dose-, time- and temperature-dependent effects of two-chain urokinase plasminogen activato(u-PA, urokinase) on normal citrated plasma. When 10 μg/ml u-PA wereadded to pooled normal plasma and incubated for 30 min at an ambient temperature (25° C), α2-antiplas-min decreased to 8% of the control value. Incubation on ice yielded a decrease to 45% of control,whereas α2-antiplasmin was fully consumed at 37° C. Fibrinogen and plasminogen fell to 46% and 39%, respectively, after a 30 min incubation at 25° C. Thrombin time prolonged to 190% of control.Various inhibitors were studied with respect to their suitability and efficacy to prevent these in vitro effects. Aprotinin exhibited a good protective effect on fibrinogen at concentrations exceeding 500 KlU/ml plasma. Its use, however, was limited due to interferences with some haemostatic assays. We could demonstrate that L-Glutamyl-L-Glycyl-L-Arginyl chloromethyl ketone (GGACK) and a specific polyclonal anti-u-PA-antibody (anti-u-PA-IgG) effectively inhibited urokinase-induced plasmin generation without interfering with haemostatic assays. The anti-u-PA-antibody afforded full protection ofα2-antiplasmin at therapeutic levels of u-PA.It is concluded that u-PA in plasma samples from patients during thrombolytic therapy may induce in vitro effects which should be prevented by the use of a suitable inhibitor such as GGACK or specific anti-u-PA-antibody.

SummaryIn vitro, concentration-dependent effects of rt-PA on a range of coagulation and fibrinolytic assays in thawed plasma samples were investigated. In absence of a fibrinolytic inhibitor, 2 μg rt-PA/ml blood (3.4 μg/ml plasma) caused prolongation of clotting time assays and decreases of plasminogen (to 44% of the control value), fibrinogen (to 27%), α2-antiplasmin (to 5%), FV (to 67%), FVIII (to 41%) and FXIII (to 16%).Of three inhibitors tested, a specific polyclonal anti-rt-PA antibody prevented interferences in all fibrinolytic and most clotting assays. D-Phe-Pro-Arg-CH2Cl (PPACK) enabled correct assays of fibrinogen and fibrinolytic parameters but interfered with coagulometric assays dependent on endogenous thrombin generation. Aprotinin was suitable only for a restricted range of both assay types.Most in vitro effects were observed only with rt-PA plasma concentrations in excess of therapeutic values. Nevertheless it is concluded that for clinical application, collection of blood samples on either specific antibody or PPACK is essential for a correct assessment of in vivo effects of rt-PA on the haemostatic system in patients undergoing fibrinolytic therapy.

AbstractIn this research, bioremediation of tsunami-affected polluted soil has been conducted by using collective microorganisms and recycled waste glass. The Tohoku earthquake, which was a mega earthquake in Japan triggered a huge tsunami on March 11th, 2011 that caused immeasurable damage to the geo-environmental conditions by polluting the soil with heavy metals and excessive salt content. Traditional methods to clean this polluted soil was not possible due to the excess cost and efforts. Laboratory experiments were conducted to examine the capability of bioremediation of saline soil by using recycled waste glass. Different collective microorganisms which were incubated inside the laboratory were used. The electrical conductivity (EC) was measured at different specified depths. It was noticed that the electrical conductivity decreased with the assist of the microbial metabolisms significantly. Collective microorganisms (CM2) were the highly capable to reduce salinity (up to 75%) while using recycled waste glass as their habitat.

AbstractDue to the special sensitivity of typical ecologically fragile areas, a series of human life, mining, and other activities have a greater impact on the environment. In this study, three coal mines in Ordos City on the Loess Plateau were selected as the study area, and the pollution levels of heavy metals in the area were studied by measuring As, Hg, Cr, Cd, Cu, Ni, and Pb in the soil of 131 sampling points. Combined with the concept of “co-occurrence network” in biology, the level of heavy metals in soil was studied using geostatistics and remote sensing databases. The results showed that the concentrations of Hg, Cr, Ni, Cu, and Pb in more than half of the sampling points were higher than the local environmental background value, but did not exceed the risk control value specified by China, indicating that human factors have a greater influence, while Cd and As elements are mainly affected Soil parent material and human factors influence. Heavy metal elements have nothing to do with clay and silt but have an obvious correlation with gravel. Cd, Pb, As and Ni, Cd, Cr are all positively correlated, and different heavy metals are in space The distribution also reflects the autocorrelation, mainly concentrated in the northeast of the TS mining area and the middle of the PS mining area.

Decidualisation of uterine stromal cells is a prerequisite for implantation of the embryo in mice. Here we have used an in vitro culture system in which stromal cells decidualise as indicated by a number of markers, including an increase in alkaline phosphatase (ALP) activity. The latter was used as a quantitative marker of decidualisation in the presence of low (2%) fetal calf serum. Prostaglandin E(2) (PGE(2)), which is known to induce decidualisation, increased ALP activity, and this effect was blocked in a dose-dependent manner by indomethacin. Leukemia inhibitory factor (LIF) was then examined, but it had no effect on PGE(2) secretion. However, LIF suppressed ALP activity in a dose-dependent manner in the presence of 2% serum, while an inhibitor of LIF that competes for binding to its receptor reversed the effect of LIF and increased ALP activity above the control level. In serum-free cultures, stromal cells differentiated rapidly, and no differences were observed between LIF-treated and untreated cultures. Stromal cells produce LIF during in vitro culture, and this peaked at 48 h. Freshly collected stromal cells from both day-2 and -4 pregnant mice expressed mRNA for the LIF receptor, and the transcript level was higher in cells isolated on day 4. However, no differences were observed in the relative levels of transcripts in cells from day 2 and day 4 after culture, nor were there differences between the LIF-treated cultures and controls. Therefore, in this study, we have shown that LIF suppresses decidualisation of murine uterine stromal cells in the presence of serum, this is not due to the regulation of PGE(2) secretion by stromal cells.

The replacement of affected blood vessels of the polymer material can cause imbalances in the blood haemostatic system. Changes in blood after the implantation of vascular grafts depend not only on the chemical composition but also on the degree of surface wettability. The Dallon® H unsealed hydrophilic knitted vascular prosthesis double velour was assessed at work and compare with hydrophobic vascular prosthesis Dallon®. Spectrophotometric studies were performed in the infrared and differential scanning calorimetry, which confirmed the effectiveness of the process of modifying vascular prostheses. Determination of the parameters of coagulation time of blood after contact in vitro with Dallon® H vascular prosthesis was also carried out. Prolongation of activated thromboplastin time, decreased activity of factor XII, IX and VIII, were observed. The prolonged thrombin and fibrinogen were reduced in the initial period of the experiment. The activity of plasminogen and antithrombin III and protein C were at the level of control value. The observed changes in the values of determined parameters blood coagulation do not exceed the range of referential values for those indexes. The observed changes are the result of considerable blood absorptiveness by the prosthesis of blood vessels and their sealing.
