Forty non-lactating, cyclic adult Pelibuey ewes were randomly divided into six groups. Estrus was synchronized within each group using intravaginal sponges and prostaglandin F2&alpha; injection at the time of the sponge removal. The sponges were inserted and removed on different dates in each group, but all the groups except the control one were first exposed to rams on the same date (July 17<sup>th</sup>), so that at the time of the first exposure the ewes were either on day 0 (group D0; n = 7), 3 (group D3; n = 7), 8 (group D8; n = 7), 12 (group D12; n = 7) or 14 (group D14; n = 7) of their synchronized estrous cycle. Thereafter the ewes of these groups remained continuously exposed to the males until all the females showed estrus. The ewes in the control group (CG; n = 5) remained isolated from all the males, except for 5-minute periods at the time of estrus detection, which was carried out three times a day. Progesterone concentrations were determined in plasma samples taken daily from two days before the initial exposure to the males until the onset of the next estrus. There were no differences in estrous cycle length between the groups exposed to rams and the control group (P &gt; 0.05). The interval from the assumed onset of the estrous cycle (48 h after sponge removal) until the occurrence of luteolysis was not different between the control group and any of the groups exposed to the males. The interval from luteolysis to estrus was not modified by exposure to the males (P &gt; 0.05). Estrus duration was shorter (P &lt; 0.06) in the control group than in group D3. It is concluded that the exposure of cyclic Pelibuey ewes to males does not advance the time of luteolysis and does not affect the length of the estrous cycle. Therefore, the male effect does not synchronize the next estrus of cyclic Pelibuey ewes. &nbsp;

Terbinafine is a fungicide given orally with the dose of 100 mg/kg body weight per day for each ewe for 60 days. The vaginal smears of ewe, body and ovarian weight were daily administered at slaughter time; ewes were slaughtered at 60th day. Estrous cycle was affected by showing a significant reduce in the estrous cycle length of each phases of estrous cycle with associated significant increase in the diestrus phase in terbinafine treatment as compared with control group (olive oil treatment as adjuvant) of ewes. There was a significant reduce in the number of follicles and a significant raise in the number of atretic follicles in treated group as compared with control group as well as upsurge the progesterone/estrogen ratio. The body and ovarian weight were significant diminished in terbinafine treatment. These observed effects of terbinafine on the ovarian activity may be due to a direct effect as antiproliferative agent or the hypothalamus - hypophysial - ovarian axis causing hormonal inequality.

To evaluate the response of the “male effect” and “temporary weaning” on the synchronization of post-partum ovarian activity in Pelibuey ewes, an experiment was carried out using 78 Pelibuey ewes with suckling lambs. The ewes were separated from their offspring for 48 hours and randomly assigned to one of four treatments derived from the arrangement of two factors, “male effect” and “temporary weaning”, each at two levels. Treatments were: T1 (n = 20) control ewes, without “male effect” and without “temporary weaning”); T2 (n = 19) ewes without “male effect” and with “temporary weaning”; T3 (n = 20) ewes with “male effect” and without “temporary weaning”; and T4 (n = 19), ewes with “male effect” and with “temporary weaning”. The response to oestrus, return to oestrus, gestation rate and lambing rate were analysed using logistic regression. The onset of oestrus was analysed using survival curves. No significant differences were found for lambing rate and prolificacy among treatments. “Temporary weaning” (T2) and “male effect” (T3) did not influence the response to oestrus, rate and duration of return to oestrus, or gestation rate and fertility, and was similar to the control group (T1). The interaction of “male effect” with “temporary weaning” (T4) increased the response to oestrus, reduced the rate and duration of return to oestrus, and the gestation rate, but increased fertility. Synchronizing post-partum ovarian activity with “male effect” and “temporary weaning” reduces the onset of oestrus and the rate of return to oestrus, but increases the response to oestrus and fecundity in Pelibuey ewes.Keywords: Biostimulation, progestogens, prolificacy, prostaglandins, suckling

Although we have previously shown that ovarian superstimulation during the first follicular wave resulted in a successful response (Carballo Guerrero D et al. 2009 Reprod. Fertil. 21, 242), the current protocol needs to be optimized in order to be used in the field. Therefore, an experiment was designed to simplify this treatment and to compare it with the traditional superstimulation protocol using progesterone and estradiol. Simmental cows (n = 14) were subjected to 3 superstimulation treatments (2 first wave groups and 1 control group) in a crossover design (i.e. all cows received the 3 treatments and all treatments were represented on each collection day). Cows in Group 1 received a progesterone-releasing device (Cue-Mate®, Bioniche Animal Health, Belleville, Ontario, Canada) along with 0.150 mg of D + cloprostenol (PGF; Bioprost-D®, Biotay, Buenos Aires, Argentina) at random stages of the estrous cycle. A second PGF was injected 5 days after Cue-Mate® insertion, followed by GnRH (0.050 mg of lecirelin; Biosin-OV®, Biotay) 36 h later (i.e. 7 days after Cue-Mate® insertion). Based on previous studies, ovulation was expected to occur 30 to 36 h later. Therefore, superstimulation treatments were initiated 36 h after GnRH (Day 0), with a total dose of 400 mg NIH-FSH-P1 of Folltropin®-V (Bioniche Animal Health) in twice-daily decreasing doses over 4 days. Prostaglandin was administered with the last 2 Folltropin®-V injections and Cue-Mate® devices were removed with the last Folltropin®-V injection. Cows received 12.5 mg of porcine LH (Lutropin®-V, Bioniche Animal Health) 24 h after Cue-Mate® removal and were AI 12 and 24 h later. Ova/embryos were collected 7 days after porcine LH and evaluated following IETS recommendations. Cows in Group 2 were treated similarly to those in the Group 1, except they did not receive the second PGF injection 5 days after Cue-Mate® insertion (thus eliminating the need to handle animals on that day). Finally, cows in Group 3 [estradiol benzoate (EB)+P4 control group] received a Cue-Mate® plus 2.5 mg of EB (Bioestradiol®, Biotay) and 50 mg of progesterone (P4; Lab., Rio de Janeiro, Argentina) at random stages of their estrous cycle. Superstimulation treatments were initiated 4 days later (Day 0) following the same protocol used in Group 1. Data were transformed to square root and analyzed by ANOVA. Mean (± SEM) numbers of ova/embryos collected, fertilized ova, and transferable embryos did not differ among groups (12.9 ± 2.0, 9.8 ± 1.7, and 6.6 ± 1.2; 11.5 ± 1.7, 9.3 ± 1.5, and 7.7 ± 1.6; and 14.5 ± 2.8, 9.4 ± 2.3, and 6.8 ± 1.7 for Groups 1, 2, and 3, respectively). In conclusion, data demonstrated that superstimulation during the first follicular wave can be successfully used in groups of randomly cycling donors without the need for estrus detection or estradiol to synchronize follicular wave emergence. The protocol is easy to follow and embryo production is comparable to that of the estradiol and progesterone protocol.

No experimento foram utilizados 62 animais da raça Charolesa e Caracú, inseminados artificialmente (IA). No 7o dia após a IA, as vacas foram divididas aleatoriamente em 2 grupos: tratado (n=32), que recebeu 2.500 UI de hCG (IM) e controle (n=30), que recebeu 1 ml de solução fisiológica estéril, (IM). O diagnóstico de prenhez foi realizado por toque retal no 45o dia após a IA. Resultados: A duração do ciclo estral diferiu (P0,05) entre o grupo tratado e controle (40,62% e 23,33%, respectivamente). Conclusões: A duração do ciclo estral dos animais tratados foi prolongada em relação aos controles (P0,05) as taxas de prenhez em bovinos de corte. Effect of human chorionic gonadotropin (hCG) on estrous cycle length and pregnancy rates in cattle Abstract In the present experiment a total of 62 cows of Charoles and Caracú breeds, artificially inseminated (AI). At day 7th after AI the cows were aleatory distributed in two groups: treated group (n=32) that receive 2.500 UI of human chorionic gonadotropin (hCG) by the intramuscular route and control group (n=30) to wich 1.0 ml of sterile physiological solution was administered also by the intramuscular route. It has been found that the estrous cycle lenght was different (P0,05) between treated and the control groups (40,6 and 23,3%, respectively). In this way, the estrous cycle lenght of the treated cows was extended in regard to the control cows. The hCG administration did not increase pregnant rates.

This study was designed to examine the effect of the intrauterine infusion of platelet-rich plasma (PRP) or equine lyophilized growth factor (L-GFequina) on the follicular growth, endometrial thickness, estrus cycle length, and pregnancy rate in purebred Arabian mares. A total of 73 purebred Arabian mares who experienced repeat breeding for three successive cycles were randomly divided into the following three groups: control group, without treatment; second Group (PRP group), in which mares were intrauterine infused with 20 mL of fresh PRP on the second day after the end of physic estrus phase; and the third group (L-GFequina Group), consisting of mares that were intrauterine infused with 20 mL of reconstituted lyophilized horse platelets growth factors (L-GFequina) on the second day after the estrus phase. The results showed no significant difference between control and treated groups in the diameter of the preovulatory follicles during the post treatment cycle. The endometrium thickness increased significantly in the L-GFequina and PRP groups ahead of the non-treated group. Intrauterine L-GFequina or PRP administration shortened the estrus cycle length. A higher pregnancy rate was recorded in the L-GFequina and PRP treated mares. In conclusion, the intrauterine infusion of L-GFequina or PRP increased the endometrial thickness and pregnancy rate and could be used to improve fertility in Arabian purebred mares who experienced from repeat breeding.

Twenty-nine DLS (1/2 Dorset, 1/4 Leicester, 1/4 Suffolk) ewes selected for extended breeding season, were involved in this study which lasted for 18 mo. According to their lambing date relative to January first, 15 ewes had a high lambing index whereas the others had a low index, the difference being 33 days (P < 0.01). Half the ewes were assigned at random to remain nonpregnant throughout the study, whereas the other half was mated in July and August. From 1 May to 15 July, 20 ewes (69%) were cycling of which 81% were from the high and 54% from the low index group (0.05 > P < 0.10). Two ewes (7%) continued cycling throughout the summer, whereas the remaining 18 ewes experienced a period of summer anestrus which occurred on 27 May (± 3.5 days) and lasted for an average of 63 ± 2.6 days. The first ovulation (unaccompanied by estrus) was observed on 29 July (± 2.6 days) the second ovulation, 20 days later was accompanied by estrus. The breeding season in nonpregnant ewes lasted 194 ± 3.2 days ending on 1 Mar. (± 7.3 days). The average lambing date of the 15 pregnant ewes was 8 Jan. (± 2.8 days). Seven of these ewes exhibited at least one ovulation postpartum, but in only two ewes was the ovulation accompanied by estrus. Introducing rams daily indicated that marker rams were successful in detecting only 88% of the ewes in estrus. Sixty-six percent of the false heats were observed prior to resumption of the regular estrous cycle. Summary This research aimed to examine the effects of thyme, celery and salinomycin on ovarian sex hormones, reproductive traits and antioxidant status during the estrous cycle. Seventy-five mature Barki ewes aged 2–3 years with an average weight of 40 ± 1.5 kg were assigned randomly into five groups (15 head/group). Group 1 was kept as the control; groups 2 and 3 received 20 g/head/day thyme (T) and celery (C) as dried herbs, respectively. Group 4 (T×C) received 10 g thyme + 10 g celery/head/day, and group 5 was treated with salinomycin 1 g/head/day. Blood samples were collected during follicular and luteal phases of the estrous cycle. Thyme and celery and the mixture of T×C increased (P < 0.01) estradiol-17β (E2) during the follicular phase of the estrous cycle, while only the celery group showed a marked (P < 0.001) increase in progesterone (P4) during the luteal phase compared with the control. Salinomycin supplementation decreased (P < 0.05) E2 concentrations during the follicular and luteal phases of the estrous cycle. Supplementation with thyme and celery enhanced (P < 0.001) antioxidant capacity in the luteal phase compared with the follicular stage. The salinomycin group showed increased (P < 0.01) levels of reduced glutathione (GSH) and decreased malondialdehyde (MDA) levels compared with the control group throughout luteal phase. For the interaction between estrous phases and treatments, thyme, celery, and T×C supplementation revealed an increase (P < 0.05) in superoxide dismutase (SOD), GSH, and glutathione disulfide (GSSG) levels compared with the control group during the follicular and luteal phases. Thyme and celery supplementation improved the number of services per conception and fertilization from 1st and 2nd inseminations, respectively. In conclusion, the applied treatment had significant effects on reproductive performance and antioxidant status in ewes throughout the estrous cycle.

AbstractUterine fibroids (UF) is the most common (about 70% cases) type of gynecological disease, with the recurrence rate varying from 11 to 40%. Because UF has no distinct symptomatology and is often asymptomatic, the specific and sensitive diagnosis of UF as well as the assessment for the probability of UF recurrence pose considerable challenge. The aim of this study was to characterize alterations in the lipid profile of tissues associated with the first-time diagnosed UF and recurrent uterine fibroids (RUF) and to explore the potential of mass spectrometry (MS) lipidomics analysis of blood plasma samples for the sensitive and specific determination of UF and RUF with low invasiveness of analysis. MS analysis of lipid levels in the myometrium tissues, fibroids tissues and blood plasma samples was carried out on 66 patients, including 35 patients with first-time diagnosed UF and 31 patients with RUF. The control group consisted of 15 patients who underwent surgical treatment for the intrauterine septum. Fibroids and myometrium tissue samples were analyzed using direct MS approach. Blood plasma samples were analyzed using high performance liquid chromatography hyphened with mass spectrometry (HPLC/MS). MS data were processed by discriminant analysis with projection into latent structures (OPLS-DA). Significant differences were found between the first-time UF, RUF and control group in the levels of lipids involved in the metabolism of glycerophospholipids, sphingolipids, lipids with an ether bond, triglycerides and fatty acids. Significant differences between the control group and the groups with UF and RUF were found in the blood plasma levels of cholesterol esters, triacylglycerols, (lyso) phosphatidylcholines and sphingomyelins. Significant differences between the UF and RUF groups were found in the blood plasma levels of cholesterol esters, phosphotidylcholines, sphingomyelins and triacylglycerols. Diagnostic models based on the selected differential lipids using logistic regression showed sensitivity and specificity of 88% and 86% for the diagnosis of first-time UF and 95% and 79% for RUF, accordingly. This study confirms the involvement of lipids in the pathogenesis of uterine fibroids. A diagnostically significant panel of differential lipid species has been identified for the diagnosis of UF and RUF by low-invasive blood plasma analysis. The developed diagnostic models demonstrated high potential for clinical use and further research in this direction.

<p>Rationale: The profile of acquisition for MDMA self-administration differs from that of amphetamine and cocaine self-administration in that fewer rats meet an acquisition criterion and the latency to acquisition is longer. These characteristics of MDMA self-administration may be because it preferentially stimulates serotonin (5HT) release whereas self-administration has generally been attributed to enhanced dopamine (DA) neurotransmission. Because 5HTergic agonists are not self-administered and increased synaptic 5HT decreased self-administration of other drugs, MDMA self-administration may be initially inhibited by the pronounced 5HT response. Accordingly, the acquisition of MDMA self-administration might proceed as a result of deficits in 5HT neurotransmission and a corresponding disinhibition of DA neurotransmission.  Objective: The primary objective was to determine the role of 5HT in the acquisition and maintenance of MDMA self-administration.  Methods: MDMA-induced increases of extracellular 5HT and DA and their primary metabolites were measured in the DA terminal regions of the nucleus accumbens (NAc) using in vivo microdialysis, prior to the commencement of MDMA self-administration. The relationship between MDMA-induced increases of neurotransmitter levels and the acquisition of MDMA self-administration was assessed. A subsequent study depleted brain 5HT by administering the neurotoxin, 5,7 – DHT, or vehicle into the lateral ventricle of the left hemisphere, prior to the commencement of MDMA self-administration. The proportion of subjects that acquired MDMA self-administration and the latency to acquire MDMA self-administration was compared for the two groups. In order to determine effects of MDMA self-administration on 5HT and DA responses, behaviours that reflect 5HT and/or DA neurotransmission were measured 5 or 14 days after self-administration of 165 mg/kg MDMA, or 14 days after vehicle self-administration. These time periods were chosen because they reflect a period of 5HT deficits (5 days) and recovery (14 days). Finally, the effect of abstinence on MDMA self-administration was measured.  Results: The MDMA-induced increase of extracellular 5HT was significantly lower for the group that subsequently acquired MDMA self-administration but the MDMA-induced increase in DA was not different from the group that failed to acquire self-administration. 5, 7-DHT administration significantly decreased tissue levels of 5HT, but not DA. MDMA self-administration was facilitated by the lesion; 100% of the lesion group acquired MDMA self-administration, whereas only 50% of the control group acquired self-administration. Five days following the last MDMA self-administration session, DAergic behaviours were enhanced and 5HTergic behaviours were reduced relative to the control group. These differences in 5HTergic mediated behaviours were not apparent 14 days after self-administration but the DAergic behaviours remained elevated. The pattern of self-administration did not differ as a function of the length of the abstinence period.  Conclusions: The variability in acquisition of MDMA self-administration was related to the magnitude of the 5HT response evoked by initial exposure to MDMA. These findings suggested that predisposing differences in the 5HT response might explain differences in the variability in acquisition of MDMA self-administration. The negative impact of 5HT on the acquisition of MDMA self-administration was clearly demonstrated following a 5, 7-DHT lesion. Thus, 5HT limits the development of MDMA self-administration. With repeated exposure to self-administered MDMA, behavioural responses indicative of 5HT activation were reduced whereas behavioural indices of DA activation were increased. The maintenance of MDMA self-administration was comparable regardless of whether there was a forced abstinence period or not. These data are consistent with the hypotheses that 5HT is inhibitory to the acquisition, but not the maintenance, of MDMA self-administration. Rather, the maintenance of self-administration might reflect sensitised DA responses that became apparent following repeated exposure.</p>

The oestrus is defined as a complex of physiological signs and changes of behavior occurring immediately before the ovulation. It is the period of sexual receptivity of the cow and this period characterized by, typical cow behaviour which is standing when mounted by a bull or companion cow. Cow indicates this willingness by a slight arching of the back and immobility when approached. The bovine estrus cycle averages 20 days in heifers and 21 days in cows, but the fluctuations in the cycle length is laying between 18 and 25 days. As a result of this progress often only 50 % of the estrus cycles are recognized, and it is extremely hard to find all cows on heat. We state a few methods for estrus detection such as visual observation, locomotion activity and rumination time measurement and other.
