Bioactive substances may constitute an alternative to antibiotics. Probiotics are mixtures of microorganisms that enhance the effectiveness and yield of nutrient absorption by competing for the substrate against pathogens that may cause intestinal infections. Prebiotics are organic substances which include complex carbohydrates and have an influence on the growth and activity of desirable intestinal microflora. Synbiotics are a combination of both of them. The aim of this study was to analyze the impact of synbiotics added to commercial feed on Pectoralis major muscle microstructure in broiler chickens. Research material consisted of 240 one-day-old Ross 308 female chicks. Birds were divided into 2 groups, 12 broilers each (replicated 10 times). The control group (C) was fed a commercial diet, and the SYN group was fed the same diet with added synbiotic: 0.8% of prebiotic RFO (raffinose family oligosaccharides) extracted from lupin seeds and 1% of probiotic Lavipan<sup>®</sup>. Birds were slaughtered at day 42. Immediately after slaughter, samples of the Pectoralis major muscle for histological analysis were taken and frozen in liquid nitrogen. The staining procedures performed were: hematoxylin and eosin staining to evaluate fibre diameter and histopathological changes, oil red staining to determine intramuscular fat content, NADH-TR (tetrazolium reductase) activity to distinguish muscle fibre types and alkaline phosphatase reaction for counting the number of capillaries. The tendency to reduced muscle fibre diameter in chickens supplemented with synbiotics indicates an increase in muscle fibre density, which gives meat a more delicate structure. When assessing the occurrence of histopathological changes, significantly more fibre splitting was observed in the control group. Moreover, the positive effect of feed supplementation with synbiotics on muscle microvascularization could have an important practical meaning, because low physical activity of chickens during rearing may lead to ischemic muscles, increased changes in the structure of muscle fibres, and reduction of meat quality.

The purpose of the study was to analyse the effect of nettle (Urtica dioica L.) leaves and fenugreek (Trigonella foenum-graecum L.) seeds as additives to fodder in order to improve post-slaughter traits and quality parameters of Termond White rabbit meat (n = 60; 30 , 30 ). Three experimental groups were created. The control group (n = 20; 10 and 10 ) was fed ad libitum feed containing corn, bran, wheat, dried alfalfa, soybean meal, sunflower meal, dicalcium phosphate, calcium carbonate and vitamin-mineral premix. The animals from the first experimental group (n = 20; 10 and 10 ) were fed a complete mixture added with 1% of nettle (Urtica dioica L.) leaves. Rabbits from the second group (n = 20; 10 and 10 ) were fed with a complete mixture added with 1% of fenugreek (Trigonella foenum-graecum L.) seeds. Rabbits fed with pellets with the addition of nettle were characterized by a higher slaughter weight, higher weight of hot and cold carcasses, lungs, kidneys and head as well as a higher weight of the fore, middle and hind part of the carcass compared to the other two groups. The carcasses of animals fed with fenugreek and nettle had a higher percentage of the fore and hind parts compared to the carcasses of the animals from the control group. The female carcasses were characterized by a significantly higher percentage of the middle part compared to the male carcasses. For most colour measurement traits, the differences depending on the feeding regime were significant. The effect of gender on meat colour was non significant. The effect of feeding regime and of gender on texture traits such as shear force, hardness, springiness, cohesiveness and chewiness were non significant. Feeding had no effect on muscle fibre diameter, but it affected the muscle fibre type I percentage. Thus, the group fed with pellets containing nettle leaves had higher percentage of type I muscle fibres than the control group. The effect of gender on muscle fibre traits was non significant.

The aim of the study was to analyse the effect of probiotics, prebiotics and synbiotics injected in ovo on day 12 of embryonic development on the microstructure of the superficial pectoral muscle (musculus pectoralis superficialis) from 42-day-old chickens of different genotypes: broilers (Ross 308) and general-purpose type (green-legged partridge (GP) chickens Zk-11, native chickens). Incubated eggs were divided into four groups (each genotype separately) depending on the substance injected in ovo: normal saline (C, control); Lactococcus lactis subsp. cremoris (PRO); galactooligosaccharides, GOS (PRE) or GOS + L. lactis (SYN). After hatching, chicks were placed in eight replicated pens (four pens/genotype group). There were eight birds per pen. In total, 64 birds were used in the experiment. Birds were slaughtered at the age of 42 days, and samples of superficial pectoral muscles were taken for analysis. The microstructure of the pectoral muscles was evaluated using the cryosectioning (frozen tissue sectioning) technique and staining with haematoxylin and eosin. Statistical analysis revealed that the in ovo injection of probiotics, prebiotics and synbiotics had no significant effect on the diameter of muscle fibres from chickens of the two genotypes. The number of fibres in the muscles from green-legged partridge chickens was about three-fold higher than the fibre density in the muscles from broiler chickens, with the fibre diameter being two-fold smaller. This fact may indicate a greater tenderness of meat from GP chickens compared to the meat from Ross 308 broilers. In the case of broilers, a prebiotic (GOS) was the most effective bioactive substance in reducing the number of histopathological changes. Considering muscles from GP chickens, the number of normal fibres was highest in birds treated with the probiotic. These findings indicate that the microstructural features of pectoral muscles depend not only on the type of the injected bioactive substance but also on the genotype of chickens.

Abstract The aim of the study was to evaluate the effect of one-week quantitative feed restriction (in days 8-14 of age) on histological characteristics of musculus pectoralis major in broiler chickens during the fattening period. Cockerels of Ross 308 (1215 chicken) were divided into 3 groups: ad libitum fed (AL; 3 × 135), restricted 80% of AL (R80; 3 × 135), and with limited feed intake 65% of AL (R65; 3 × 135). Eight cockerels per group were slaughtered in weekly intervals from day 14 of age till the end of experiment at 35 days to determine changes in the muscle fibre characteristics. The number of muscle fibres in pectoralis major decreased (P ≤ 0.001) with increasing age. Fibre cross sectional area was s ignificantly (P ≤ 0.027) affected by the interaction of group and age. At the end of feed restriction at 14 days all groups had similar fibre area (519-539 μm2), the differences between groups were observed at 35 days of age with the largest fibre area in R65 chickens (2296 μm2), while R80 did not differ from AL (1728 μm2 and 1667 μm2). There was no effect of feeding regime on giant muscle fibre incidence.

Cefotaxime (CTX)-resistant Enterobacteriaceae are still an ongoing challenge in human and veterinary health. High prevalence of these resistant bacteria is detected in broiler chickens and the prevention of their dissemination along the production pyramid is of major concern. The impact of certain on-farm interventions on the external bacterial contamination of broiler chickens, as well as their influence on single processing steps and (cross-) contamination, have not yet been evaluated. Therefore, we investigated breast skin swab samples of broiler chickens before and during slaughter at an experimental slaughter facility. Broiler chickens were previously challenged with CTX-resistant Escherichia coli strains in a seeder-bird model and subjected to none (control group (CG)) or four different on-farm interventions: drinking water supplementation based on organic acids (DW), slow growing breed Rowan × Ranger (RR), reduced stocking density (25 kg/sqm) and competitive exclusion with Enterobacteriales strain IHIT36098(CE). Chickens of RR, 25 kg/sqm, and CE showed significant reductions of the external contamination compared to CG. The evaluation of a visual scoring system indicated that wet and dirty broiler chickens are more likely a vehicle for the dissemination of CTX-resistant and total Enterobacteriaceae into the slaughterhouses and contribute to higher rates of (cross-) contamination during processing.

AbstractThe primary aim of this study was to investigate the impact of three dietary levels of lactose (LAC) originating from conventional dried whey (DW) and the duration of these treatments (from 8 to 21 or to 42 days of age) on growth performance, basic post-slaughter traits and excreta quality of broiler chickens kept in cages. A secondary purpose was to investigate the effect of LAC level on some parameters of the caecal micro-environment and gross morphology in these birds. A total of 560 Ross 308 chickens (sex ratio 1:1) were assigned to 7 dietary combinations with 10 replicate cages of 8 birds per cage. The control group was fed basal diets consisting of maize, wheat and soybean meal. The other 6 groups received the same basal diets with DW added in amounts equivalent to a LAC dietary levels of 1, 2 or 3%. Only continuous feeding (day 8 to 42) with 1% and 2% levels of LAC was found to yield the overall body weight gain (BWG) during the whole 42-day rearing period, which was significantly higher than that on the control diet, with a larger share of breast meat in carcass at a 2% LAC. However, these effects were associated with greater faecal score values indicating more watery excreta compared with the control. Increasing levels of LAC augmented the relative caecal weight and length. A reduction in the caecal pH was confirmed at day 21 for birds fed 1% and 2% of dietary LAC. The lower pH values were correlated to an increased sum of total volatile fatty acids (VFA), causing large increases in the concentration of undissociated forms of individual VFA. The decline in plate counts of coliform bacteria was observed with 2% and 3% LAC, whereas the counts of lactic acid-producing bacteria (LAB) were higher at these two LAC levels. The present findings lead to the conclusion that the dietary level of 2% LAC originated from DW is the most effective in enhancing the productivity of broilers, with moderate occurrence of undesirable side effects.

AbstractThe neuromuscular junction (NMJ) is the highly specialised peripheral synapse formed between lower motor neuron terminals and muscle fibres. Post-synaptic acetylcholine receptors (AChRs), which are found in high density in the muscle membrane, bind to acetylcholine released into the synaptic cleft of the NMJ, ultimately facilitating the conversion of motor action potentials to muscle contractions. NMJs have been studied for many years as a general model for synapse formation, development and function, and are known to be early sites of pathological changes in many neuromuscular diseases. However, information is limited on the diversity of NMJs in different muscles, whether muscle fibre type impacts NMJ morphology and growth, and the relevance of these parameters to neuropathology. Here, this crucial gap was addressed using a robust and standardised semi-automated workflow called NMJ-morph to quantify features of pre- and post-synaptic NMJ architecture in an unbiased manner. Five wholemount muscles from wild-type mice were dissected and compared at immature (post-natal day, P7) and early adult (P31-32) timepoints. Post-synaptic AChR morphology was found to be more variable between muscles than that of the motor neuron terminal and there were greater differences in the developing NMJ than at the mature synapse. Post-synaptic architecture, but not neuronal morphology or post-natal synapse growth, correlates with fibre type and is largely independent of muscle fibre diameter. Counter to previous observations, this study indicates that smaller NMJs tend to innervate muscles with higher proportions of fast twitch fibres and that NMJ growth rate is not conserved across all muscles. Furthermore, healthy pre- and post-synaptic NMJ morphological parameters were collected for five anatomically and functionally distinct mouse muscles, generating reference data that will be useful for the future assessment of neuromuscular disease models.Graphical Abstract

The article presents the results of studying the impact of the spore-forming probiotic “Olin” and the prebiotic “Eсofiltrum” on the meat quality of broiler chicken carcasses of the Ross-308 cross. In the course of the research, it has been found that the inclusion of these preparations in the broiler diet has a positive effect on the studied indicators: the pre-slaughter weight increased by 4.3-5.7%, the weight of the gutted carcass by 4.6-6.2%, the muscle weight 4.8-6.5%, the weight of the loin muscles by 5.7-8.2%. The ratio of edible parts to inedible parts is 4.14 in the third experimental group, 4.08-in the second, and 4.04-in the control. Based on the results of the anatomical cutting of broiler carcasses and according to the fatness indicators the poultry carcasses of the third and second experimental groups, 87.5% and 87.2%, respectively, belong to the first grade, and the yield of the first grade carcasses is 86.2% in the control group. Meat products obtained from both the control poultry and the poultry of both experimental groups has no off-aroma odor and taste, the meat is juicy, tender and aromatic. The products obtained from broiler chickens, whose ration includes the probiotic “Olin” and the prebiotic “Ecofiltrum”, do not have any deviations from the established standards and norms. During the research, it has been found that the use of the studied drugs has a positive effect on the meat quality of poultry carcasses. However, broiler chickens receiving the probiotic “Olin” has more pronounced meat qualities.

The study evaluated the effects of nucleotide dietary supplementation on the physical and nutritional characteristics of the Pectoralis major muscle of male broiler chickens (n = 60 000), divided into two homogeneous groups: Control (C) and Nucleotides (N). The animals of the two groups, from the birth (24 h of age) to the slaughtering age (52 days), received the same diet, supplemented (N) or not (C) with 0.1% of a Nucleotide pool. At the slaughtering, on a sample of 130 animals per group, randomly selected, the physical and nutritional characteristics of Pectoralis major muscle were determined. The meat of the N group showed significantly higher redness and Hue values, lower shear force values, higher lipid and ash percentages and iron content. Moreover, nucleotides significantly increased monounsaturated acids and linolenic acid and decreased eicosapentanoic and docosahexanoic acids. The unsaturation degree was higher in the Nucleotides group and Atherogenic index was positively influenced by the nucleotide supplementation. Nucleotide dietary supplementation improved the physical and nutritional characteristics of the Pectoralis major muscle of broiler chickens.

1. An investigation is reported in which the relationship was examined between body size and muscle fibre size of nineteen newborn lambs and including twelve males and seven females. Body size was measured in terms of the dead body weight and that of the dressed carcass, while muscle fibre size was estimated on the basis of the mean crossdiameter of 150 fibres per lamb.2. It was shown that the dressing (or carcass) percentage tends to increase with an increase in body weight, from 42·26% at a body weight of 2000 g. to 49·04% at 7000 g.3. Of the 2850 cross-diameters recorded, the majority (33·8%) of the fibres measured between 8·0 and 9·6 μ, while individual fibres varied in size from 1·6 to 22·4 μ.4. Highly significant, positive correlations were shown to exist between both body (r = 0·996) and carcass (r = 0·946) weight, and mean muscle fibre diameter, indicating that differences in size between the lambs may be accounted for largely by corresponding variations in the size of individual muscle fibres.5. Of the three muscles sampled m. gastrocnemius had the largest mean fibre diameter (10·38μ), followed in decreasing order by m. rectus femoris (9·72 μ) and m. longissimus dorsi (9·09 μ). These inter-muscle differences were significant at the 1% level of probability.6. Ram lambs had significantly thicker muscle fibres (10·32 μ) than ewe lambs (8·72 μ), but also weighed the heavier and produced heavier dressed carcasses. Some evidence was produced, however, in support of the theory that at comparable weights males possess thinner individual, and therefore a greater number of fibres than females.
